Deregulated oxidative metabolism is a hallmark of leukaemia. While tyrosine kinase inhibitors (TKIs) such as imatinib have increased survival of chronic myeloid leukaemia (CML) patients, they fail to eradicate disease-initiating leukemic stem cells (LSCs). Whether TKI-treated CML LSCs remain metabolically deregulated is unknown. Using clinically and physiologically relevant assays, we generate multi-omics datasets that offer unique insight into metabolic adaptation and nutrient fate in patient-derived CML LSCs. We demonstrate that LSCs have increased pyruvate anaplerosis, mediated by increased mitochondrial pyruvate carrier 1/2 (MPC1/2) levels and pyruvate carboxylase (PC) activity, in comparison to normal counterparts. While imatinib reverses BCR::ABL1-mediated LSC metabolic …

Methicillin-resistant Staphylococcus aureus (MRSA) is an important cause of complicated urinary tract infection (UTI) associated with the use of indwelling urinary catheters. Previous reports have revealed host and pathogen effectors critical for MRSA uropathogenesis. Here, we sought to determine the significance of specific metabolic pathways during MRSA UTI. First, we identified four mutants from the Nebraska transposon mutant library in the MRSA JE2 background that grew normally in rich medium but displayed significantly reduced growth in pooled human urine (HU). This prompted us to transduce the uropathogenic MRSA 1369 strain with the transposon mutants in sucD and fumC (tricarboxylic acid …
Variability Of Total Mercury Concentrations In Steller Sea Lion Bone Locations And Bone Elements, Mary P. Keenan
Variability Of Total Mercury Concentrations In Steller Sea Lion Bone Locations And Bone Elements, Mary P. Keenan
Marine Biology
Mercury (Hg) is a contaminant of global concern with impacts on the health and resilience of marine wildlife. Some forms of ingested Hg are bioaccumulated and biomagnified with greatest concentrations observed in long-lived top predators. High concentrations of Hg in mammals impair immune function, neurochemistry, and reproduction. Steller sea lions (Eumetopias jubatus, SSL) are a long-lived top predator, and some populations have total mercury concentrations ([THg]) in fur that are above the threshold values of concern for toxic effects. While this information is available for present-day populations of SSLs, [THg] in historic and ancient populations of SSLs are unknown. Bone …

On The Right Or Left: Benjamin And The Scapegoat, Terrence L. Szink, John W. Welch
Insights: The Newsletter of the Neal A. Maxwell Institute for Religious Scholarship
Over the years, scholars have found in King Benjamin's speech much that is at home in the ancient Israelite autumn festivals surrounding the Day of Atonement. An essential part of that day was the scapegoat ritual.

The Feminist Keyword Project: Literature Reviews As Feminist Praxis, Krystal Cleary
Feminist Pedagogy
The Feminist Keyword Project is a scaffolded series of assignments that culminates in a literature review of the student’s chosen keyword. It is designed for intermediate undergraduate students who acquired a foundational feminist vocabulary in their introductory classes. Building upon this baseline fluency, the project shifts students’ understanding of course concepts from a glossary of terms to politicized feminist gatherings. The project is informed by the tradition of the keyword entry genre to move students beyond merely defining their chosen term to narrating it as a site of feminist discourse in and outside of academia. In doing so, the Feminist …
A Framework For Creating Virtual Reality Models For More Effective Coastal Flood Risk Communication, Tina Korani, Alexandrea Martinez
A Framework For Creating Virtual Reality Models For More Effective Coastal Flood Risk Communication, Tina Korani, Alexandrea Martinez
CSU Journal of Sustainability and Climate Change
Coastal cities are exposed to increasing risks of flooding from sea-level rise. Climate change is expected to double the frequency of coastal flooding within the next decade, and some areas could experience floods of a magnitude 100 times higher than currently (Vitousek et al., 2017). People living in at-risk areas often ignore the impact of climate change on flood intensity and frequency. Immersive visual storytelling techniques proved promising and powerful tools to engage with and raise awareness of flood hazards. Here, we are introducing a framework to use Virtual Reality (VR) to reach better people living in coastal cities and …

Groundwater Resource Assessment For West Africa Using Earth Observations And Groundwater Modeling, Sergio Armando Barbosa Casas
Groundwater Resource Assessment For West Africa Using Earth Observations And Groundwater Modeling, Sergio Armando Barbosa Casas
Theses and Dissertations
Groundwater is a crucial global resource, particularly in semiarid regions where surface water is scarce. As a result, effective groundwater management is essential. However, the challenge lies in a lack of data and information for proper decision-making. This dissertation present studies related to groundwater storage changes and recharge in West Africa, using data from NASA's Gravity Recovery and Climate Experiment (GRACE) mission. In Niger, this research reveals little change in groundwater storage from 2002 to 2010 but a steep rise from 2010 to 2021, attributed partially to increased precipitation. Groundwater resources in Niger are found to be underutilized, with estimated …
Feasibility Of Rna-Based Pathogen Detection In An Infant Formula Matrix, Gweneth Claire Gustafson
Feasibility Of Rna-Based Pathogen Detection In An Infant Formula Matrix, Gweneth Claire Gustafson
Theses and Dissertations
Background: The dairy processing industry is currently hindered by slow, antiquated testing for high-risk pathogens, including Listeria monocytogenes and Cronobacter sakazakii. Powder infant formula's low water activity and substantial fat content enhance the survival and thermotolerance of such foodborne pathogens. As DNA-based detection methods can produce false positives even after cell death, a more accurate approach is needed. The objective of this study is to evaluate the suitability of an RNA-based platform for the detection of pathogens in a dairy powder matrix. Materials and Methods: A unique phenol-chloroform extraction method was used to obtain total RNA from heat-killed and unheated …

Brain-Derived Neurotrophic Factor Dysregulation As An Essential Pathological Feature In Huntington's Disease: Mechanisms And Potential Therapeutics, Andrew Speidell, Noman Bin Abid, Hiroko Yano
2020-Current year OA Pubs
Brain-derived neurotrophic factor (BDNF) is a major neurotrophin whose loss or interruption is well established to have numerous intersections with the pathogenesis of progressive neurological disorders. There is perhaps no greater example of disease pathogenesis resulting from the dysregulation of BDNF signaling than Huntington's disease (HD)-an inherited neurodegenerative disorder characterized by motor, psychiatric, and cognitive impairments associated with basal ganglia dysfunction and the ultimate death of striatal projection neurons. Investigation of the collection of mechanisms leading to BDNF loss in HD highlights this neurotrophin's importance to neuronal viability and calls attention to opportunities for therapeutic interventions. Using electronic database searches …

Critical Engagements With Award Winning Picturebooks: My Journey In Creating A More Equitable Classroom Library, Carrie Elizabeth Crowe
Critical Engagements With Award Winning Picturebooks: My Journey In Creating A More Equitable Classroom Library, Carrie Elizabeth Crowe
Theses and Dissertations
Research consistently shows that students from historically marginalized populations have difficulty accessing texts that represent the realities of their lives. Concerned that such might be the case for my students, I conducted an inventory of my classroom library and sought out award-winning texts that could make my library more diverse and inclusive. I then analyzed these award-winning texts using Bishop's (1992) categories of multicultural literature in order to better understand the picturebooks' underlying messages and ideologies. My examinations showed that the majority of the texts fit into the same category (culturally specific), though I was able to identify award-winning texts …

Omega-3 (n-3) and omega-6 (n-6) polyunsaturated fatty acids (PUFAs) play critical roles in human health. Prior genome-wide association studies (GWAS) of n-3 and n-6 PUFAs in European Americans from the CHARGE Consortium have documented strong genetic signals in/near the FADS locus on chromosome 11. We performed a GWAS of four n-3 and four n-6 PUFAs in Hispanic American (n = 1454) and African American (n = 2278) participants from three CHARGE cohorts. Applying a genome-wide significance threshold of P < 5 × 10

Sensory neurons detect mechanical forces from both the environment and internal organs to regulate physiology. PIEZO2 is a mechanosensory ion channel critical for touch, proprioception, and bladder stretch sensation, yet its broad expression in sensory neurons suggests it has undiscovered physiological roles. To fully understand mechanosensory physiology, we must know where and when PIEZO2-expressing neurons detect force. The fluorescent styryl dye FM 1-43 was previously shown to label sensory neurons. Surprisingly, we find that the vast majority of FM 1-43 somatosensory neuron labeling in mice in vivo is dependent on PIEZO2 activity within the peripheral nerve endings. We illustrate the …
The Pathophysiology Of Equine Glandular Gastric Disease: Investigating The Role Of The Mucosal Microbiome And Bile Acid Reflux, Linda Paul
LSU Doctoral Dissertations
Equine glandular gastric disease (EGGD) is a common disease of athletic horses that can affect performance and welfare. Clinical signs are non-specific and diagnosis is made using gastroscopy after a prolonged fasting period. Effective prevention and treatment strategies for EGGD are lacking despite its widespread prevalence and negative effects. The goal of this research was to investigate the pathophysiology of EGGD. Objectives included: 1) compare the gastric glandular mucosal microbiome in horses with and without EGGD, 2) assess the influence of dietary and management factors on the gastric microbiome, and 3) evaluate the pyloric gastric fluid bile acid profile in …

Xenon (Xe) has shown great potential as a stroke treatment due to its exceptional ability to protect brain tissue without inducing side effects. We have previously developed Xe-loaded liposomes for the ultrasound-activated delivery of Xe into the cerebral region and demonstrated their therapeutic efficacy. At present, the sole FDA-approved thrombolytic agent for stroke treatment is recombinant tissue plasminogen activator (rtPA). In this study, we aimed to investigate the potential of combining Xe-liposomes with an intravenous rtPA treatment in a clinically relevant embolic rat stroke model. We evaluated the combinational effect using an in vitro clot lysis model and an in …

The beneficial effects of physical activity on brain ageing are well recognised, with exerkines, factors that are secreted into the circulation in response to exercise, emerging as likely mediators of this response. However, the source and identity of these exerkines remain unclear. Here we provide evidence that an anti-geronic exerkine is secreted by platelets. We show that platelets are activated by exercise and are required for the exercise-induced increase in hippocampal precursor cell proliferation in aged mice. We also demonstrate that increasing the systemic levels of the platelet-derived exerkine CXCL4/platelet factor 4 (PF4) ameliorates age-related regenerative and cognitive impairments in …

BACKGROUND: Serological surveys are used to ascertain influenza infection and immunity, but evidence for the utility of mucosal immunoglobulin A (IgA) as a correlate of infection or protection is limited.
METHODS: We performed influenza-like illness (ILI) surveillance on 220 individuals living or working in a retirement community in Gainesville, Florida from January to May 2018, and took pre- and postseason nasal samples of 11 individuals with polymerase chain reaction (PCR)-confirmed influenza infection and 60 randomly selected controls. Mucosal IgA against 10 strains of influenza was measured from nasal samples.
RESULTS: Overall, 28.2% and 11.3% of individuals experienced a 2-fold and …
Liposome-Based Carriers For Crispr Genome Editing, Shao-Ling Huang
Liposome-Based Carriers For Crispr Genome Editing, Shao-Ling Huang
Faculty, Staff and Student Publications
The CRISPR-based genome editing technology, known as clustered regularly interspaced short palindromic repeats (CRISPR), has sparked renewed interest in gene therapy. This interest is accompanied by the development of single-guide RNAs (sgRNAs), which enable the introduction of desired genetic modifications at the targeted site when used alongside the CRISPR components. However, the efficient delivery of CRISPR/Cas remains a challenge. Successful gene editing relies on the development of a delivery strategy that can effectively deliver the CRISPR cargo to the target site. To overcome this obstacle, researchers have extensively explored non-viral, viral, and physical methods for targeted delivery of CRISPR/Cas9 and …

Objective: RANK-RANKL-OPG signaling is being considered as the pathway periodontitis induces osteoclastogenesis. Alveolar bone resorption and increased osteoclast activity are both triggered by a greater RANKL/OPG ratio in periodontitis. Gambir contains catechins and quercetin, which have antimicrobial, antioxidant, and anti-inflammatory properties. This study aimed to analyze the anti-osteoclastogenesis effect of gambir extract gel through RANKL and OPG serum levels, the number of osteoblasts, and osteoclasts in alveolar bone tissue of Wistar rats. Material and Methods: Twenty-four male Wistar rats were divided into four equal groups, each receiving a different treatment: Group I received a placebo gel, Group II received Tetracycline …
