Effects Of Stocking Rates On Water Use Efficiency Of Established Species In Stipa Breviflora Desert Steppe, M. Zhao, G. M. Yin, S. B. Liu, W. J. Meng

IGC Proceedings (1977-2023)

Plant water use efficiency reflects the survival adaptation strategies adopted by plants to obtain maximum carbon harvest. Understanding the water and resource utilization strategies of Stipa breviflora in desert steppe will help people understand its growth mechanism, and it is of great significance to analyze the ecological countermeasures of Stipa breviflora to cope with disturbance. In this study, the water use efficiency (WUE) of the established populations of Stipa breviflora in desert steppe was measured by stable carbon isotope method and photosynthometer method, and the adaptation mechanism of Stipa breviflora under different stocking rates was discussed based on the basic …

The Hindgut Microbiome Of Grazing Horses, J. R. Weinert, C. A. Williams Feb 2024

The Hindgut Microbiome Of Grazing Horses, J. R. Weinert, C. A. Williams

IGC Proceedings (1977-2023)

The hindgut microbiome plays an essential role in horses consuming forage-based diets high in fiber, such as pasture. The equine hindgut harbors a large microbial community that ferments dietary fiber and produces by-products which provide a substantial portion of daily energy requirements. Despite the importance in nutrition and health, research evaluating the hindgut microbiome of grazing horses is relatively limited. Grazing horse microbiome research has primarily focused on inter-diet comparisons with mixed diets including concentrates as well as with other forms of forage. Recent research has begun to explore responses of the gut microbiome to different pasture forage species and …

Horse And Pasture Responses To Stocking Methods: Rotational Versus Continuous, C. A. Williams, J. R. Weinert-Nelson Feb 2024

Horse And Pasture Responses To Stocking Methods: Rotational Versus Continuous, C. A. Williams, J. R. Weinert-Nelson

IGC Proceedings (1977-2023)

Rotational grazing management offers potential advantages for economic and environmental sustainability, as well as for horse health. Rotational grazing has the potential to increase pasture yields and available forage for grazing, and therefore reduce supplemental feed costs. A rotational grazing management strategy can also prevent or mitigate negative effects of overgrazing, with implications for pasture productivity and the environment. Overgrazing can reduce vegetative cover over time, increasing the potential for erosion and nutrient run-off. Overgrazing may also result in long-term decreased forage stand persistence and weed invasion, necessitating more frequent pasture renovation. All of this can negatively impact the horse’s …

Energetic Value Estimates Of Wild And Domesticated Food Items Consumed By Congolese Bayaka Foragers, Lucie Benoit, Sheina Lew-Levy, Yann R. Ouamba, Alyssa N. Crittenden

Journal of Ecological Anthropology

We present a dataset that systematically estimates the energetic value of wild and domesticated food resources collected and consumed by Congolese BaYaka foragers. This dataset is unique in that it accounts for everyday practices of food processing and consumption. Reported values of inedible portions, moisture content, and estimated energetic values (kcal/g) were extracted from field research and the published food chemistry and nutrition literature. These data are central to understanding how nutritional needs are met in rainforest environments and will allow for current and future tracking of diet composition in this and similar ecologies. The dataset is available at https://doi.org/10.6084/m9.figshare.19446938.v9

Neurons of the basal forebrain nucleus basalis and posterior substantia innominata (NBM/SIp) comprise the major source of cholinergic input to the basolateral amygdala (BLA). Using a genetically-encoded acetylcholine (ACh) sensor in mice, we demonstrate that BLA-projecting cholinergic neurons can 'learn' the association between a naïve tone and a foot shock (training) and release ACh in the BLA in response to the conditioned tone 24h later (recall). In the NBM/SIp cholinergic neurons express the immediate early gene, Fos following both training and memory recall. Cholinergic neurons that express Fos following memory recall display increased intrinsic excitability. Chemogenetic silencing of these learning-activated …

Soluble 'SOSIP'-stabilized HIV-1 envelope glycoprotein (Env) trimers elicit dominant antibody responses targeting their glycan-free base regions, potentially diminishing neutralizing responses. Previously, using a nonhuman primate model, we demonstrated that priming with fusion peptide (FP)-carrier conjugate immunogens followed by boosting with Env trimers reduced the anti-base response. Further, we demonstrated that longer immunization intervals further reduced anti-base responses and increased neutralization breadth. Here, we demonstrate that long trimer-boosting intervals, but not long FP immunization intervals, reduce the anti-base response. Additionally, we identify that FP priming before trimer immunization enhances antibody avidity to the Env trimer. We also establish that adjuvants Matrix …

KRAS mutations drive oncogenic alterations in numerous cancers, particularly in human pancreatic ductal adenocarcinoma (PDAC). About 93% of PDACs have KRAS mutations, with G12D (∼42% of cases) and G12V (∼32% of cases) being the most common. The recent approval of sotorasib (AMG510), a small-molecule, covalent, and selective KRASG12C inhibitor, for treating patients with non-small cell lung cancer represents a breakthrough in KRAS targeted therapy. However, there is a need to develop other much-needed KRAS-mutant inhibitors for PDAC therapy. Notably, Mirati Therapeutics recently developed MRTX1133, a small-molecule, noncovalent, and selective KRASG12D inhibitor through extensive structure-based drug design. MRTX1133 has demonstrated potent …

Stimulator of interferon genes (STING) is an immune adaptor protein that senses cyclic GMP-AMP (cGAMP) in response to self or microbial cytosolic DNA as a danger signal. STING is ubiquitously expressed in diverse cell populations including cancer cells with distinct cellular functions such as activation of type I interferons, autophagy induction, or triggering apoptosis. It is not well understood whether and which subsets of immune cells, stromal cells, or cancer cells are particularly important for STING-mediated antitumor immunity. Here using a polymeric STING-activating nanoparticle (PolySTING) with a “shock-and-lock” dual activation mechanism, we show type 1 conventional dendritic cell (cDC1) is …

The TP53 tumor suppressor gene is mutated early in most of the patients with triple-negative breast cancer (TNBC). The most frequent TP53 alterations are missense mutations that contribute to tumor aggressiveness. Here, we used an autochthonous somatic TNBC mouse model, in which mutant p53 can be toggled on and off genetically while leaving the tumor microenvironment intact and wild-type for p53 to identify physiological dependencies on mutant p53. In TNBCs that develop in this model, deletion of two different hotspot p53R172H and p53R245W mutants triggers ferroptosis in vivo, a cell death mechanism involving iron-dependent lipid peroxidation. Mutant p53 protects cells …

Pathological consequences of circadian misalignment, such as shift work, show considerable individual differences, but the lack of mechanistic understanding hinders precision prevention to prevent and mitigate disease symptoms. Here, we employed an integrative approach involving physiological, transcriptional, and histological phenotypes to examine inter-individual differences in pre-symptomatic pathological progression, preceding irreversible disease onset, in wild-type mice exposed to chronic jet-lag (CJL). We observed that CJL markedly increased the prevalence of hepatic steatosis with pronounced inter-individual differences. Stratification of individual mice based on CJL-induced hepatic transcriptomic signature, validated by histopathological analysis, pinpoints dysregulation of lipid metabolism. Moreover, the period and power of …

Oligopeptides of l-lysine have the potential for applications in various scientific and technical areas. The number of residues in polycationic compounds such as oligolysine is also reported to have an effect on its biological properties. Hence, there is a necessity for developing efficient oligolysine synthesis methods where the oligopeptide dispersity can be tailored, along with optimum yield values. The ability of proteases to reverse their proteolytic activity to synthesize peptides has been reported in the literature. However, protease-catalyzed synthesis of oligopeptides of basic amino acids such as lysine in aqueous buffers is hindered by unfavorable thermodynamics. In this work, a …

Pancreatic ductal adenocarcinoma (PDAC) is a lethal disease with limited effective treatment options, potentiating the importance of uncovering novel drug targets. Here, we target cleavage and polyadenylation specificity factor 3 (CPSF3), the 3′ endonuclease that catalyzes mRNA cleavage during polyadenylation and histone mRNA processing. We find that CPSF3 is highly expressed in PDAC and is associated with poor prognosis. CPSF3 knockdown blocks PDAC cell proliferation and colony formation in vitro and tumor growth in vivo. Chemical inhibition of CPSF3 by the small molecule JTE-607 also attenuates PDAC cell proliferation and colony formation, while it has no effect on cell proliferation …

Background

In this priority-setting exercise, we sought to identify leading research priorities needed for strengthening future pandemic preparedness and response across countries.

Methods

The International Society of Global Health (ISoGH) used the Child Health and Nutrition Research Initiative (CHNRI) method to identify research priorities for future pandemic preparedness. Eighty experts in global health, translational and clinical research identified 163 research ideas, of which 42 experts then scored based on five pre-defined criteria. We calculated intermediate criterion-specific scores and overall research priority scores from the mean of individual scores for each research idea. We used a bootstrap (n = 1000) to …

Despite six decades of the use of exogenous oxytocin for management of labor, little is known about its effects on the developing brain. Motivated by controversial reports suggesting a link between oxytocin use during labor and autism spectrum disorders (ASDs), we employed our recently validated rat model for labor induction with oxytocin to address this important concern. Using a combination of molecular biological, behavioral, and neuroimaging assays, we show that induced birth with oxytocin leads to sex-specific disruption of oxytocinergic signaling in the developing brain, decreased communicative ability of pups, reduced empathy-like behaviors especially in male offspring, and widespread sex-dependent …

Investigating The Phytochemicals In Sargassum (Brown Algae) Against The Therapeutic Targets Of Oral Squamous Cell Carcinoma Metastasized From Breast: An Approach, Mahema S

Annual Research Symposium

ABSTRACT:

Purpose: Oral metastases are relatively rare. In women, the most common oral metastases originate from breast cancer, the most diagnosed malignancy, and the second leading cause of death. Sargassum is a genus of brown algae which exhibits its natural therapeutic potential with anticancer properties. This study aimed to identify a therapeutic target for OSCC metastasized from breast cancer through network pharmacology and to evaluate potent phytochemicals against the identified target.

Methods: DEGs associated with OSCC and breast cancer were obtained from the Gene Expression Omnibus. The upregulated DEGs were then intersected to identify common targets between OSCC and BC. …
