Our sense of hearing is mediated by cochlear hair cells, of which there are two types organized in one row of inner hair cells and three rows of outer hair cells. Each cochlea contains 5-15 thousand terminally differentiated hair cells, and their survival is essential for hearing as they do not regenerate after insult. It is often desirable in hearing research to quantify the number of hair cells within cochlear samples, in both pathological conditions, and in response to treatment. Machine learning can be used to automate the quantification process but requires a vast and diverse dataset for effective training. …

Parkinson's disease (PD) is the second most common neurodegenerative disorder. Dopamine (DA) neurons in the substantia nigra pars compacta, which have axonal projections to the dorsal striatum (dSTR), degenerate in PD. In contrast, DA neurons in the ventral tegmental area, with axonal projections to the ventral striatum, including the nucleus accumbens (NAcc) shell, are largely spared. This study aims to uncover the relative contributions of glycolysis and oxidative phosphorylation (OxPhos) to DA release in the striatum. Mangroves serve as important nursery habitats for several species of subtropical fishes and invertebrates. Southeastern Florida mangrove environments have experienced an increasing frequency of strong hot and cold weather front systems, but little information is known about the temperature thresholds for mangrove-associated prey species that lethally disrupts homeostasis. To assess the vulnerability of mangrove-associated prey species to these short-duration temperature fluctuations, critical thermal methodology (CTM) was used to determine thermal tolerances for two fishes and two invertebrates, using loss of equilibrium (LOE) as the endpoint to determine CTmax and CTmin values as a survivable proxy for death. Background Managing genetic diversity is critically important for maintaining species fitness. Excessive homozygosity caused by the loss of genetic diversity can have detrimental effects on the reproduction and production performance of a breed. Analysis of genetic diversity can facilitate the identification of signatures of selection which may contribute to the specific characteristics regarding the health, production and physical appearance of a breed or population. Sudden Acquired Retinal Degeneration Syndrome (SARDS) is a perplexing and debilitating ocular condition that affects canine patients primarily in the form of the sudden onset of total vision loss. Veterinary literature will be reviewed to discuss the basic anatomy of the canine eye and retina, as well as to investigate various diagnostic techniques to evaluate ophthalmic function and hypotheses of the pathogenesis of SARDS. Fires are becoming larger and more frequent in the deserts of North America, driven largely by the expansion of invasive annual grasses. Rodent consumers can curb invasive grass expansion through plant herbivory, but the top-down effects of this pressure on soil resources are understudied. In shrub dominated deserts, islands of fertility under shrub canopies are surrounded by nutrient poor interspace soils that dictate patterns of vegetative growth. Little is known of how top-down controls from rodents and fire may interact with fertile island topography. The basal breast cancer subtype is enriched for triple-negative breast cancer (TNBC) and displays consistent large chromosomal deletions. Here, we characterize evolution and maintenance of chromosome 4p (chr4p) loss in basal breast cancer. Analysis of The Cancer Genome Atlas data shows recurrent deletion of chr4p in basal breast cancer. Phylogenetic analysis of a panel of 23 primary tumor/patient-derived xenograft basal breast cancers reveals early evolution of chr4p deletion. Mechanistically we show that chr4p loss is associated with enhanced proliferation. Gene function studies identify an unknown gene, C4orf19, within chr4p, which suppresses proliferation when overexpressed-a member of the PDCD10-GCKIII kinase module …

Introduction: In healthcare, while the suffering of patients is often evident, the suffering of clinicians receives less focus. Some sources of clinician distress are directly related to constant exposure to patient suffering, but others are caused by the health care system, and thus potentially preventable. Looking at clinician suffering through the lens of compassion fosters a new paradigm of individual, team, and organizational capabilities, and moves the responsibility to alleviate this suffering from the individual onto the organization and team. Yet research into the impact of organizational compassion in healthcare has been extremely limited.

Background: Elevated lipoprotein(a) (Lp[a]) concentrations are associated with increased cardiovascular event risk even in the presence of well-controlled low-density lipoprotein cholesterol levels, but few treatments are documented to reduce this residual risk.
Objectives: The aim of this post hoc analysis of REDUCE-IT (Reduction of Cardiovascular Events with Icosapent Ethyl-Intervention Trial) was to explore the cardiovascular benefit of icosapent ethyl (IPE) across a range of Lp(a) levels.

Stable isotope analysis in bioarchaeology of human bone is generally used to reconstruct diet or migratory patterns of certain populations. Although this type of analysis is now standard practice, little research has been done regarding how cultural and mortuary practices may affect chemical composition of bone. This study aims to determine whether different simulated mortuary contexts have an effect on stable isotope levels in bone, using seven pig rib bone samples as proxy for human bone. Dysregulated nuclear-cytoplasmic trafficking has been shown to play a role in oncogenesis in several types of solid tumors and hematological malignancies. Exportin 1 (XPO1) is responsible for the nuclear export of several proteins and RNA species, mainly tumor suppressors. KPT-330, a small molecule inhibitor of XPO1, is approved for treating relapsed multiple myeloma and diffuse large B-cell lymphoma. Cutaneous T-cell lymphoma (CTCL) is an extranodal non-Hodgkin lymphoma with an adverse prognosis and limited treatment options in advanced stages. Aerodynamic forces developed by automobiles have destabilizing effects at high speed. These forces tend to skew toward a vehicle’s rear which can present safety concerns, especially for rear-wheel-drive automobiles like the Mazda Miata. To address oversteer and high-speed instability, a vehicle’s design can be tailored to bring about aerodynamic balance and improve traction. LiDAR was used to bring the physical automobile into the digital space. Then, a splitter and diffuser were added to reduce the magnitude of the destabilizing forces. Border-associated macrophages (BAMs) are tissue-resident macrophages that reside at the border of the central nervous system (CNS). Since BAMs originate from yolk sac progenitors that do not persist after birth, the means by which this population of cells is maintained is not well understood. Using two-photon microscopy and multiple lineage-tracing strategies, we determine that CCR2+ monocytes are significant contributors to BAM populations following disruptions of CNS homeostasis in adult mice. After BAM depletion, while the residual BAMs possess partial self-repopulation capability, the CCR2+ monocytes are a critical source of the repopulated BAMs. In addition, we demonstrate the existence of CCR2+ …

Corticospinal neurons (CSNs) synapse directly on spinal neurons, a diverse assortment of cells with unique structural and functional properties necessary for body movements. CSNs modulating forelimb behavior fractionate into caudal forelimb area (CFA) and rostral forelimb area (RFA) motor cortical populations. Despite their prominence, the full diversity of spinal neurons targeted by CFA and RFA CSNs is uncharted. Here, we use anatomical and RNA sequencing methods to show that CSNs synapse onto a remarkably selective group of spinal cell types, favoring inhibitory populations that regulate motoneuron activity and gate sensory feedback. Background: Lipoprotein(a) (Lp[a]) is a causal genetic risk factor for atherosclerotic cardiovascular disease (ASCVD). There are limited long-term follow-up data from large U.S. population cohorts.
Objectives: This study examined the relationship of Lp(a) with ASCVD outcomes in a large, pooled, multi-ethnic U.S.
Methods: The study included data on Lp(a) and ASCVD outcomes from 5 U.S.

The global incidence of cancer continues to rise, posing a significant public health concern. Although numerous cancer therapies exist, each has limitations and complications. The present study explores alternative cancer treatment approaches, combining hyperthermia and photodynamic therapy (PDT). Magnetic nanoparticles (MNPs) and amine-functionalized carbon quantum dots (A-CQDs) were synthesized separately and then covalently conjugated to form a single nanosystem for combinational therapy (M-CQDs). The successful conjugation was confirmed using zeta potential, Fourier transform infrared spectroscopy (FT-IR), and UV-visible spectroscopy. Morphological examination in transmission electron microscopy (TEM) further verified the conjugation of CQDs with MNPs. Energy dispersive X-ray spectroscopy (EDX) revealed …

Many studies infer the role of neurons by asking what information can be decoded from their activity or by observing the consequences of perturbing their activity. An alternative approach is to consider information flow between neurons. We applied this approach to the parietal reach region (PRR) and the lateral intraparietal area (LIP) in posterior parietal cortex. Two complementary methods imply that across a range of reaching tasks, information flows primarily from PRR to LIP. This indicates that during a coordinated reach task, LIP has minimal influence on PRR and rules out the idea that LIP forms a general purpose spatial …

Our sense of hearing is mediated by cochlear hair cells, of which there are two types organized in one row of inner hair cells and three rows of outer hair cells. Each cochlea contains 5-15 thousand terminally differentiated hair cells, and their survival is essential for hearing as they do not regenerate after insult. It is often desirable in hearing research to quantify the number of hair cells within cochlear samples, in both pathological conditions, and in response to treatment. Machine learning can be used to automate the quantification process but requires a vast and diverse dataset for effective training. …

A growing body of literature on African Pentecostals in sub-Saharan Africa is highlighted in this article, highlighting a more defined contribution of African Pentecostals to the development landscape. Until recently, the African Pentecostal development landscape recorded little visibility at the national level, on the assumption that their experience only highlights cultural and theological relevance. Jakarta Bay is a bay supporting the economy of thousands of nearby fishermen. Therefore, the reclamation in Jakarta Bay raises many environmental, economic, and social problems. During this reclamation, dredging and stockpiling activities increase the turbidity of the waters. This research aims to determine the changes in the distribution of total suspended solids (TSS) after reclamation using Sentinel 2A satellite imagery from 2016 to 2022. The three research questions guiding this project are: What cognitive and language abilities do children in the early childhood stage of development have? What are the most effective strategies for communicating information about mental health disorders to children ages 4 to 7 through storytelling?
