Impacts Of Lake Elevation Decline On Tui Chub, A Critical Forage Species For Lahontan Cutthroat Trout In Pyramid Lake, Nevada, Usa, Sarah Barnes May 2024

Impacts Of Lake Elevation Decline On Tui Chub, A Critical Forage Species For Lahontan Cutthroat Trout In Pyramid Lake, Nevada, Usa, Sarah Barnes

All Graduate Theses and Dissertations, Fall 2023 to Present

Lake level decline affects lakes worldwide, changing the availability and character of nearshore habitat used by fish to spawn, and increasing total dissolved solids (TDS), similar to salinity, a factor that negatively impacts fish health. Lake level decline can affect different lakes in different ways, but typically when lake level declines significantly, there is less nearshore habitat overall, and what nearshore habitat remains has less diverse habitat for fish. We investigated whether both impacts of lake level decline may be causing declines of Tui Chub Siphateles bicolor, a large minnow native to Pyramid Lake that spawns in nearshore habitat. …

Acoustic-Perceptual Relations Between Fundamental Frequency And Expressiveness In Speakers With Hypokinetic Dysarthria, Alena Portnova May 2024

Acoustic-Perceptual Relations Between Fundamental Frequency And Expressiveness In Speakers With Hypokinetic Dysarthria, Alena Portnova

All Graduate Theses and Dissertations, Fall 2023 to Present

Hypokinetic dysarthria is a motor speech disorder that occurs in patients with Parkinson’s disease (PD) and affects not only their speech intelligibility but also how their attitudes and emotions are perceived by listeners. People with PD have been judged as less happy, involved, friendly, and interested based only on their speech samples. A lack of speech expressiveness is one of the characteristics that is likely to be related to these negative listener judgments. Specifically, it has been suggested that a lack of fundamental frequency (F0) variation reduces speakers’ ability to express various emotions. To investigate whether speech expressiveness is related …

One Year Follow Up Of A Cross-Sectional Study Of Hookworm Infection And Response To Deworming In Beposo, Ghana, Taylor Clarkson May 2024

One Year Follow Up Of A Cross-Sectional Study Of Hookworm Infection And Response To Deworming In Beposo, Ghana, Taylor Clarkson

Capstone Experience: Master of Public Health

Hookworms are soil-transmitted helminths that infect over 500 million people worldwide, primarily those in low-income countries. Infection with hookworm can cause anemia, malnutrition, and stunted physical and cognitive development. Deworming campaigns have attempted to combat hookworm infections in endemic areas for decades. However, there is growing variability in the effectiveness of deworming drugs, and post-treatment reinfections are abundant. There are significant gaps in our understanding of critical host-parasite factors that mediate hookworm infection, reinfection, and response to deworming treatment in endemic communities. A five-year prospective study was undertaken to better understand drivers of hookworm infection in rural Ghana and thus …

Reynolds Revisited: The Original Meaning Of Reynolds V. United States And Free Exercise After Fulton, Clark B. Lombardi May 2024

Reynolds Revisited: The Original Meaning Of Reynolds V. United States And Free Exercise After Fulton, Clark B. Lombardi

Articles

This Article calls for a profound reevaluation of the stories that are being told today about the Supreme Court’s free exercise jurisprudence starting with the Court’s seminal 1879 decision in Reynolds v. United States and proceeding up to the present day. Scholars and judges today agree that the Supreme Court in Reynolds interpreted the Free Exercise Clause of the First Amendment to protect only religious belief and not religiously motivated action. All casebooks today embrace this interpretation of the case, and the Supreme Court has regularly endorsed it over the past twenty years, most recently in 2022. However, this Article …

Coarctation Duration And Severity Predict Risk Of Hypertension Precursors In A Preclinical Model And Hypertensive Status Among Patients, Arash Ghorbannia, Hilda Jurkiewicz, Lith Nasif, Abdillahi Ahmed, Jennifer Co-Vu, Mehdi Maadooliat, Ronald K. Woods, John F. Ladisa Jr.

Mathematical and Statistical Science Faculty Research and Publications

BACKGROUND:

Coarctation of the aorta (CoA) often leads to hypertension posttreatment. Evidence is lacking for the current >20 mm Hg peak-to-peak blood pressure (BP) gradient (BPGpp) guideline, which can cause aortic thickening, stiffening, and dysfunction. This study sought to find the BPGpp severity and duration that avoid persistent dysfunction in a preclinical model and test if predictors translate to hypertension status in patients with CoA.

METHODS:

Rabbits (n=75; 5–12/group) were exposed to mild, intermediate, or severe CoA (≤12, 13–19, ≥20 mm Hg BPGpp) for ≈1, 3, or 22 weeks using dissolvable and permanent sutures with thickening, stiffening, contraction, and endothelial …

Impact Of Strategic Postweaning Supplementation On March- And May-Born Yearling Range Heifer And Calf Growth And Performance In The Nebraska Sandhills, Josie Nicole Crouch

Department of Animal Science: Dissertations, Theses, and Student Research

Three experiments were conducted at the Gudmundsen Sandhills laboratory near Whitman, Nebraska. Experiment 1, a retrospective regression analysis, evaluated the impact of heifer percentage of mature body weight (BW) at breeding on heifer performance, subsequent pregnancy rates, and calf production. Overall pregnancy rates were greater (P < 0.01) for heifers at 60 to 70% of mature BW than 50 to 55%, with no differences in pregnancy rates (P ≥ 0.15) after 2-years of age. Heifers at 50 to 60% of mature BW had a greater (P = 0.05) proportion give birth in the first 21-d. A higher percentage of mature BW at breeding resulted in greater (P < 0.01) BW at birth and weaning for offspring.

Experiments 2 and 3 were 3-year studies that evaluated the impact …

Artificial Insemination And Fetal Programming In Beef Production Systems, Dempster M. Christenson May 2024

Artificial Insemination And Fetal Programming In Beef Production Systems, Dempster M. Christenson

Department of Animal Science: Dissertations, Theses, and Student Research

Pregnancy to artificial insemination and late gestation fetal programming of the next calf crop are central to genetic development, reproductive longevity, calf crop productivity, and efficiency of cow/calf ranching. The purposes of this research are to find methods to improve pregnancy rate to artificial insemination, the growth and reproductive health of pregnant heifers, and the productivity of their offspring. In the first of four studies we found that extending the period of progesterone administration in an estrus synchronization protocol did not significantly hasten estrus response or increase pregnancy rate to artificial insemination, but timing of estrus within the melengestrol acetate …

Adding Parasite Resistance To A Hair Sheep Breeding Objective, Robert M. Forbes May 2024

Adding Parasite Resistance To A Hair Sheep Breeding Objective, Robert M. Forbes

Department of Animal Science: Dissertations, Theses, and Student Research

The United States Hair Sheep Index was designed to increase total weight of lamb weaned per ewe lambing (TW) using estimated breeding values (EBV) for direct (DWWT) and maternal (MWWT) weaning weight, and number of lambs born (NLB) and weaned (NLW), as selection criteria. Producers are interested in adding post-weaning fecal egg count EBV (PFEC), an indicator of parasite resistance, to the index to achieve genetic change in both TW and PFEC.

First, the effects of adding PFEC alongside TW to the breeding objective of indexes that used either phenotypes or EBV as selection criteria were studied. Since MWWT …

Feeding Dried Distillers' Grains With Solubles To Lactating Dairy Cattle: Whole Animal Energy Utilization And Manure Biogas Production, Grant Michael Fincham May 2024

Feeding Dried Distillers' Grains With Solubles To Lactating Dairy Cattle: Whole Animal Energy Utilization And Manure Biogas Production, Grant Michael Fincham

Department of Animal Science: Dissertations, Theses, and Student Research

When DDGS are fed to lactating dairy cattle, the observed response in energy supply, milk production, and methane production varies among studies. One potential reason for these observed discrepancies is the nature of the diet formulation itself. Furthermore, little research has been conducted to examine how dietary changes can affect dairy cattle manure composition and methane production in an anaerobic manure digester. In order to gain a better understanding of these topics, we conducted two experiments. The first experiment was conducted to test how the manipulation of lactating dairy cattle diets affects energy utilization, milk production, methane production, and manure …

Global citizenship education is one way to better prepare individuals to learn about global systems, understand their own place in those systems, and act with others to create a more just world. While global citizenship education has big aspirational goals, for this to be effective, educators must better understand what pedagogical strategies impact the development of global citizenship capacities in students. This study aims to understand the potential of one emerging pedagogical strategy in the context of higher education –online community-based learning– for fostering global citizenship capacities in an increasingly interconnected and digitized world. This research is guided by a …

Comparing The Regulatory Effects Of Overexpressed Micrornas And Xenobiotic Drugs On Cell Cycle And Apoptotic Regulators In Pc-3 Cells, Tommie Johnson May 2024

Comparing The Regulatory Effects Of Overexpressed Micrornas And Xenobiotic Drugs On Cell Cycle And Apoptotic Regulators In Pc-3 Cells, Tommie Johnson

Dissertations (2016-Present)

MicroRNA was first discovered in C. elegans as small temporal RNA (stRNA) that does not code for protein. Since being discovered they have played a significant role in regulating gene expression at the post-transcriptional level. miRNAs are found in various organisms, and they bind to the 3' untranslated regions to inhibit translation and cause mRNA degradation. Some drugs are involved in cell cycle regulation such as Palbociclib, Ribociclib (LEE011), and Abemaciclib (LY2835219), that causes G1 arrest impeding cell proliferation. Cyclin D1 (CCND1) is supported by the cell cycle making it into a functional product. When undergoing a chemical reaction, the …

Faculty, Staff and Student Publications

Ferroptosis and apoptosis are key cell-death pathways implicated in several human diseases including cancer. Ferroptosis is driven by iron-dependent lipid peroxidation and currently has no characteristic biomarkers or gene signatures. Here a continuous phenotypic gradient between ferroptosis and apoptosis coupled to transcriptomic and metabolomic landscapes is established. The gradual ferroptosis-to-apoptosis transcriptomic landscape is used to generate a unique, unbiased transcriptomic predictor, the Gradient Gene Set (GGS), which classified ferroptosis and apoptosis with high accuracy. Further GGS optimization using multiple ferroptotic and apoptotic datasets revealed highly specific ferroptosis biomarkers, which are robustly validated in vitro and in vivo. A subset of …

The rise of neuroinflammatory disorders highlights the importance of early detection and intervention for more effective treatment options. Neuroinflammation is associated with the pathogenesis of many neurological disorders, including Major Depressive Disorder, Alzheimer's disease, and Multiple Sclerosis. There has been a focus on neurons to advance our understanding of the underlying mechanisms of neuroinflammation and its role in neurodegeneration. However, recent studies have highlighted the pivotal role of glial cells, particularly microglia, in neuroinflammation due to their active participation in the immune response. This study investigates glial-specific indicators of morphology, metabolic changes, and drug efficacy in neuroinflammatory conditions. By analyzing …

Gain-of-function mutations activating JAK/STAT signaling are seen in the majority of patients with myeloproliferative neoplasms (MPN), most commonly JAK2V617F. Although clinically approved JAK inhibitors improve symptoms and outcomes in MPNs, remissions are rare, and mutant allele burden does not substantively change with chronic therapy. We hypothesized this is due to limitations of current JAK inhibitors to potently and specifically abrogate mutant JAK2 signaling. We therefore developed a conditionally inducible mouse model allowing for sequential activation, and then inactivation, of Jak2V617F from its endogenous locus using a combined Dre-rox/Cre-lox dual-recombinase system. Jak2V617F deletion abrogates MPN features, induces depletion of mutant-specific hematopoietic …

Intermittent fasting is a dietary intervention that is increasingly being tested for positive outcomes in patients receiving cancer treatment. In this review, we examine the impact of intermittent fasting on symptoms, toxicities, and quality of life in patients undergoing cancer therapy and highlight unmet investigative areas to prompt future research. While current evidence is preliminary and conclusions mixed, some promising clinical studies suggest that intermittent fasting interventions may improve fatigue and reduce gastrointestinal toxicities in certain patients with cancer. Emerging clinical evidence also demonstrates that intermittent fasting may reduce off-target DNA damage, and induce favorable cellular-level immune remodeling. Furthermore, intermittent …

Repair of large bone defects caused by severe trauma, non-union fractures, or tumor resection remains challenging because of limited regenerative ability. Typically, these defects heal through mixed routines, including intramembranous ossification (IMO) and endochondral ossification (ECO), with ECO considered more efficient. Current strategies to promote large bone healing via ECO are unstable and require high-dose growth factors or complex cell therapy that cause side effects and raise expense while providing only limited benefit. Herein, we report a bio-integrated scaffold capable of initiating an early hypoxia microenvironment with controllable release of low-dose recombinant bone morphogenetic protein-2 (rhBMP-2), aiming to induce ECO-dominated …

Zanidatamab is a bispecific human epidermal growth factor receptor 2 (HER2)-targeted antibody that has demonstrated antitumor activity in a broad range of HER2-amplified/expressing solid tumors. We determined the antitumor activity of zanidatamab in patient-derived xenograft (PDX) models developed from pretreatment or postprogression biopsies on the first-in-human zanidatamab phase I study (NCT02892123). Of 36 tumors implanted, 19 PDX models were established (52.7% take rate) from 17 patients. Established PDXs represented a broad range of HER2-expressing cancers, and in vivo testing demonstrated an association between antitumor activity in PDXs and matched patients in 7 of 8 co-clinical models tested. We …
