Heart disease involves irreversible myocardial injury that leads to high morbidity and mortality rates. Numerous cell-based cardiac in vitro models have been proposed as complementary approaches to non-clinical animal research. However, most of these approaches struggle to accurately replicate adult human heart conditions, such as myocardial infarction and ventricular remodeling pathology. The intricate interplay between various cell types within the adult heart, including cardiomyocytes, fibroblasts, and endothelial cells, contributes to the complexity of most heart diseases. Consequently, the mechanisms behind heart disease induction cannot be attributed to a single-cell type. Thus, the use of multi-cellular models becomes essential for creating …

Plant Model Of Α-Synucleinopathy: Expression Of Α-Synuclein A53t Variant In Hairy Root Cultures Leads To Proteostatic Stress And Dysregulation Of Iron Metabolism, Jasmina Kurepa, Kristen A. Bruce, Greg A. Gerhardt, Jan A. Smalle

Neurology Faculty Publications

Synucleinopathies, typified by Parkinson’s disease (PD), entail the accumulation of α- synuclein (αSyn) aggregates in nerve cells. Various αSyn mutants, including the αSyn A53T variant linked to early-onset PD, increase the propensity for αSyn aggregate formation. In addition to disrupting protein homeostasis and inducing proteostatic stress, the aggregation of αSyn in PD is associated with an imbalance in iron metabolism, which increases the generation of reactive oxygen species and causes oxidative stress. This study explored the impact of αSyn A53T expression in transgenic hairy roots of four medicinal plants (Lobelia cardinalis, Artemisia annua, Salvia miltiorrhiza, and Polygonum multiflorum). In all …

Department of Horticulture & Landscape Architecture Faculty Publications

Sweetpotato [Ipomoea batatas (L.) Lam.] is a staple crop that provides nutritional benefits to humans globally, but it is subject to yield loss when competing with weeds, especially during the early stage of establishment. Yield loss can vary widely based on the cultivar, production environment, weed species, and management techniques. To address this challenge, we conducted field research at the Samuel G. Meigs Horticulture Research Farm, Lafayette, IN, and at the Southwest Purdue Agricultural Center, Vincennes, IN, in 2022 to determine the effect of sweetpotato cultivar on the critical weed-free period. The experiment was a split-plot design, with weed-free interval …

Mechanical Property Simulations Of Ploybenzimidazole Membranes., Blake Alexander Clark May 2024

Mechanical Property Simulations Of Ploybenzimidazole Membranes., Blake Alexander Clark

Electronic Theses and Dissertations

This work explores mechanical properties of membranes made of polybenzimidazole utilizing molecular simulations. These simulations researchers to narrow down polybenzimidazole membrane compositions to viable candidates. These simulations are quicker than current purely experimental methods. The increase in throughput would be achieved by eliminating compositions that have unfavorable mechanical properties through preliminary simulation work before moving onto experimental methods. Determining polybenzimidazole membranes’ mechanical properties experimentally would, therefore, allow for researchers to speed up research in areas such as fuel cells and electrolyzers that make use of polybenzimidazole membranes. This study uses Materials Studio and the COMPASS III forcefield to perform these …

China's Nature-Based Solutions In The Global South: Evidence From Asia, Africa, And Latin America, Annah Lake Zhu, Niklas Weins, Juliet Lu, Tyler Harlan, Jin Qian, Fabiana Barbi Seleguim

Urban and Environmental Studies Faculty Works

China increasingly engages in environmental diplomacy through South-South cooperation across the developing world. Since 2019, the rise of the discourse of Nature-based Solutions (NbS) within this cooperation has been exponential. Coined just over ten years ago, NbS refers to the underexplored potential of leveraging the natural world to address socio-environmental challenges. The concept finds particular resonance in China, where it demonstrates strong parallels with the domestically-pioneered concept of Ecological Civilization – the ruling paradigm when it comes to all realms of Chinese environmental governance. Building on the global discourse, NbS has been adapted to the Chinese context, creating what some …

Afraid For The Dark: Regulating Light Pollution Under The Clean Water Act, Katrina Umstead May 2024

Afraid For The Dark: Regulating Light Pollution Under The Clean Water Act, Katrina Umstead

William & Mary Law Review

Currently, light pollution is only regulated at the state and local level. However, not all states implement legislation to mitigate the adverse effects of ALAN [Artificial light at night]. Nineteen states, the District of Columbia, and Puerto Rico have implemented laws to reduce light pollution. In states without such laws, or in federal waters, light-intensive activities remain unchecked. The rapid increase in light pollution in recent years illustrates the inadequacies of existing state and local regulatory schemes and calls for a new understanding of ALAN as a pollutant to marine ecosystems.

This Note argues that the existing tools in the …

Unreasonable Traffic Stops, Sam Kamin May 2024

Unreasonable Traffic Stops, Sam Kamin

William & Mary Law Review

In 1996, the Supreme Court announced in Whren v. United States that a traffic stop is constitutional if there is probable cause to believe a traffic infraction has occurred. So long as the officers who stop an individual can point—even after the fact—to any violation of the traffic laws, their actual, subjective motivations for initiating a stop are legally irrelevant. Case-by-case determination of reasonableness is unnecessary in the traffic stop context, the Court concluded, because the balancing of interests has already been done. Unlike warrantless entries into homes, the use of deadly force, or unannounced warranted entries, a traffic stop …

Comparing The Effectiveness Of Different Forms Of Vitamin E Supplementation (Liquid Vs. Powder) On Serum Alpha-Tocopherol Concentrations In Developing Horses, Emilia Fong-Gallagher

Honors Scholar Theses

Vitamin E is an essential antioxidant in equines that has a significant role in the nutritional requirements of horses to keep bodily systems functioning at optimal levels. Vitamin E deficiency is often associated with serious incurable neuromuscular diseases such as equine motor neuron disease and equine protozoal myeloencephalitis. Many horses stabled in urban areas are often deficient due to the lack of adequate grazing time as their natural source of vitamin E is from grass, and therefore should receive supplementation. This study evaluated the effectiveness of different forms of Elevate vitamin E supplement on the serum alpha-tocopherol concentrations in fourteen …

Honors Scholar Theses

This literature review explores the utilization of CRISPR-Cas9 gene editing technology in addressing Sickle Cell Disease (SCD) and Beta-Thalassemia. CRISPR, leveraging the Cas-9 enzyme inspired by bacterial defense mechanisms, serves as a precise molecular tool for targeted genetic modifications. The process involves guide RNA directing Cas-9 to specific DNA sites, allowing for meticulous gene editing. Focusing on the specific applications of CRISPR in SCD and Beta-Thalassemia, the review critically analyzes recent methods, including CRISPR-Cas9 editing targeting the BCL11A enhancer for SCD and the HBG1/HBG2 promoters for both SCD and Beta-Thalassemia. The synthesis of findings highlights the potential and challenges of …

Intuition In Modal Epistemology: Concept-Based Or Imagination-Based?, Jeremy Bachman May 2024

Intuition In Modal Epistemology: Concept-Based Or Imagination-Based?, Jeremy Bachman

Honors Scholar Theses

In both George Bealer’s moderate modal rationalism (MMR) and Peter Kung’s modal evidence from imagination (MEI) intuition plays an indispensable role in modal knowledge. However, these modal epistemologies differ in whether modal knowledge is based on intuitions yielded by concepts or by imaginations. Both MMR and MEI take intuitions to be sui generis propositional attitudes, basic epistemic units, reasons for belief, and sources of evidence. However, MEI holds that sensory and intellectual content can prompt a rational intuitions. On the other hand, MMR is restricted to intuitions yielded by non-sensory content. According to MMR, intellectual intuitions about privileged concepts give …

How Does Hummock Creation In Submerging Salt Marshes Alter Nitrous Oxide Fluxes?, Juliette Doyle May 2024

How Does Hummock Creation In Submerging Salt Marshes Alter Nitrous Oxide Fluxes?, Juliette Doyle

Honors Scholar Theses

Climate change is altering ecosystems and the services they provide. Salt marsh ecosystems typically protect coastal areas and filter nitrogen out of water, but are rapidly submerging due to rising sea levels and human development that prevents landward migration. Recent restoration efforts to preserve salt marshes attempt to build elevation capital and promote vegetation and animal habitat, but it is unclear how such efforts affect salt marsh biogeochemistry and dynamics of nitrous oxide, a potent greenhouse gas. To better understand how adding sediment to submerging salt marshes may alter nitrous oxide fluxes, I leveraged a salt marsh hummock creation experiment …

Use Of Cre-Lox Mice As A Model For Ependymogenesis, Poorva Bagchee May 2024

Use Of Cre-Lox Mice As A Model For Ependymogenesis, Poorva Bagchee

Honors Scholar Theses

Stem cells in the lateral ventricles of the brain undergo a process of development within the ventricular-subventricular zone. In early development neuroepithelial cells divide to form more neuroepithelial cells or convert to radial glial cells (RGCs). These RGCs, or stem cells, then divide to form neurons or other glial cells such as ependymal cells, which form a single-layered ependyma along the ventricular wall. These stem cells divide in two ways: symmetrically, producing two ependymal cells or asymmetrically, yielding one neural stem cell and ependymal cell. To further study this process the Cre-lox system in mice will be used. By breeding …

Visions Of Our Future: A Manifesto For Environmentalist Cinema In The Climate Era, Daniel Milovic May 2024

Visions Of Our Future: A Manifesto For Environmentalist Cinema In The Climate Era, Daniel Milovic

Honors Scholar Theses

Environmentalism has strengthened as a discipline during the climate era, a period defined by the ongoing effects of climate change, and it is time for a fully environmental activist cinema to emerge. This type of filmmaking must be committed to generating images of our future along a utopian or apocalyptic continuum. This approach allows filmmakers to maintain the gravity of the climate crisis while generating inspiring, alternative futures that are not tied to the current global culture of capitalist exploitation of natural resources and marginalized peoples. Environmentalist filmmakers must then articulate climate change as a cultural issue—one that is produced …

Exploring Essentialization, Stereotyping, And Forms Of Dehumanization In American Sign Language, Madeline Haefele

Honors Scholar Theses

This paper investigates the philosophical implications of evolving and removing certain signs in American Sign Language (ASL), with a focus on those considered problematic due to their potential to perpetuate stereotypes or essentialism. Through an analysis of signs for words such as Jewish, Chinese, Japanese, Korean, and Black, this study explores how language shapes and reflects social attitudes, particularly within the Deaf community. The research situates these signs within philosophical frameworks, identifying their potential contributions to social essentialism, prejudice, or forms of dehumanization.

While addressing the challenges of limited first-hand accounts from Deaf individuals, the …

Archives Of Facade, Arielle Nord May 2024

Archives Of Facade, Arielle Nord

Masters Theses and Doctoral Dissertations

Fantasy is a genre that seldom includes black characters. When black characters are present, they often lack the same narrative significance, reliability and character growth that is given to white characters. This can lead to poorly written black characters that lack purpose or fall victim to stereotypical roles and troupes. Depicting black characters in this way can further isolate readers who wish to read about black characters, that are more than just stereotypes, within the fantasy genre. This thesis will examine the writing techniques of character coding and the subversion of stereotypes and how they present a way for black …

Fate And Treatability Of Engineered Nanoparticles In Urban Stormwater, Laura Wright May 2024

Fate And Treatability Of Engineered Nanoparticles In Urban Stormwater, Laura Wright

Masters Theses and Doctoral Dissertations

Despite the tremendous benefits of engineered nanoparticles (ENPs), their inevitable release into the environment may be detrimental and hence cause for concern of these emerging contaminants. In this thesis research, the fate and transport of nano-silver (n-Ag) and nano-titanium dioxide in source specific stormwater is explored, and the influence of n-Ag on the treatability of co-pollutants is examined. Through the analysis of particle size distribution, it was noticed that metallic ENPs tend to aggregate initially, while extended turbulence in the system leads to disaggregation. Stormwater samples with added ENPs exhibited larger particles than those in stormwater without added ENPs. The …
