Identification Of Mitotic Phosphatases And Cyclin K As Novel Molecular Targets In Pancreatic Cancer, Yi Xiao May 2024

Identification Of Mitotic Phosphatases And Cyclin K As Novel Molecular Targets In Pancreatic Cancer, Yi Xiao

Theses & Dissertations

Pancreatic cancer is a highly lethal disease worldwide. Given the limited effectiveness of current regimens in restricting tumor progression, it is imperative that potential molecular targets be identified to offer valuable insights into alternative therapeutics. By using a phosphate-binding tag (Phos-tag) technique, previous studies have identified several Hippo pathway-related proteins and kinases required for cancer growth or paclitaxel resistance. This work has screened various phosphatases and cyclins/cyclin-dependent kinases (CDKs) as potential cancer targets, but specifically focuses on characterizing the roles of carboxy-terminal domain small phosphatase like 2 (CTDSPL2), apoptosis-stimulating protein of p53-2 (ASPP2), and Cyclin K in pancreatic cancer.

Therapeutic Effects Of Bet Protein Inhibition In B-Cell Malignancies And Beyond, Audrey L. Smith May 2024

Therapeutic Effects Of Bet Protein Inhibition In B-Cell Malignancies And Beyond, Audrey L. Smith

Theses & Dissertations

Chronic lymphocytic leukemia (CLL) develops through a combination of intrinsic genetic defects, constitutively active survival/proliferation signaling pathways, and complex microenvironment interactions. Redundant tumor microenvironment (TME) immuno-suppressive mechanisms and epigenetic maintenance of terminal T-cell dysfunction also greatly hinder anti-tumor T-cell responses in CLL. Small-molecule agents targeting the B-cell receptor (BCR) pathway and anti-apoptotic proteins alone or in combination with chemo-immunotherapy have revolutionized the management of CLL, yet there is still a significant need for novel therapeutic strategies to yield durable tumor elimination. Bromodomain and extra-terminal domain (BET) proteins are epigenetic readers that bind to acetyl-lysine residues on histones to regulate gene …

Identifying The Molecular Determinants Of Lung Metastatic Adaptation In Prostate Cancer, Grace M. Waldron May 2024

Identifying The Molecular Determinants Of Lung Metastatic Adaptation In Prostate Cancer, Grace M. Waldron

Theses & Dissertations

Prostate cancer (PC) stands as the primary diagnosed cancer in men in the US at approximately 299,010 cases in 2024 and ranks second globally, posing a significant public health challenge. Clinical presentations vary widely, from indolent to aggressive forms, necessitating stage-specific treatment regimens. Understanding its metastatic nature is critical due to the impact of cancer cell dissemination on disease morbidity, with bone and visceral organs serving as key sites of metastasis. Despite bone metastasis being the most common site for metastasis, visceral metastases at sites such as the liver and lungs correlate with poorer survival, emphasizing the role of microenvironmental …

Beating The Heat: The Use Of Coral-Benefitting Bacteria For Heat-Stressed Anemones And Inspiring Future Biologists, Navah Eshraghi May 2024

Beating The Heat: The Use Of Coral-Benefitting Bacteria For Heat-Stressed Anemones And Inspiring Future Biologists, Navah Eshraghi

Undergraduate Honors Theses

The warming of ocean temperatures has caused mass coral-bleaching events characterized by the expulsion of vital algal symbionts from coral tissues. However, the possibility of algal reuptake and existence of bacteria that support coral-algal symbiosis are two avenues of potential restoration efforts. Using the sea anemone Exaiptasia diaphana as a model for corals, I tested whether the coral-benefitting bacterium Endozoicomonas montiporae could enhance algal uptake under ambient (27°C) and heat-stressed (32°C) conditions. I then turned acquired results into an article modeled after National Geographic Kids to introduce the experiment to younger ages. While temperature was a larger determinant of algal-uptake …

This paper examines and reevaluates the lasting impacts of Priscus of Panium’s eyewitness account of his ambassadorial trip to Atilla the Hun in 449 CE, dubbed Priscus at the Court of Attila. Through meticulous analysis, this paper attempts to contextualize the presence and military movements of Huns across Europe based on Priscus’ original work. I clarify that Atilla's encampment was in Wallachia while detailing the location's significance and the significance of Hunnic military movements in Media. Moving forward, I use Priscus’ work as a tool to observe the social norms of Byzantium and Scythia ranging from things like their female …

Optimizing Biofuel Formula For Ideal C/N Conditions And Maximum Net Energy Output, Kylie Kirkpatrick, Zamzam Alabdullah, Saleh Bubshait, Jackson Hopper

Creative Collaborations: Annual Showcase of Undergraduate Research

In spring 2023, using 25mL small-scale digesters, a team of students used a dry matter mixture (92.8% solid matter) consisting of kelp, animal waste and beer waste water to produce biomethane gas. These experiments resulted in biogas output (at standard temperature and pressure) that was three times the biomass input. When scaled up (by a factor of x28K) the energy input to mix the solid matter could substantially reduce the net energy of the system. To reduce the energy input to the system, we need to move to a more liquid mixture. We want to compare the carbon to nitrogen …

Ceremonial Sexual Sacrifice To Commercial Prostitution: The History Of Prostitution And The Social, Economic, And Religious Progress That Revolved Around The Profession, Katelyn E. Crowell

Young Historians Conference

From its believed origin in Ancient Mesopotamia, prostitution has not only survived but is a profession that has continued to play a culturally defining role through the centuries. While its initial emergence was through an act of religious ritual and sacrifice, it transformed into a commercial profession. Prostitution, despite it becoming a representation of sexual deviance, not only persevered but thrived across vast regions, cultures, and time periods. The profession's social ‘taboo’ and the forbiddenness of being associated with the institution has carried forward through time and across varying societal constructs, the attempts to hide or extinguish prostitution has never …

Fragments Of A Dream: Armenia And The Shadow Of Genocide, Ada A. Camp

Young Historians Conference

Amidst the shadows of the war in Ukraine, in September of 2023, Azerbaijan’s military advancement into an ethnic Armenian enclave called Nagorno-Karabakh ended a thirty year conflict in just one violent day. The next morning, hundreds of thousands of Armenians fled, fearing ethnic cleansing and retaliatory killings. While the more recent history of this conflict is tied to the fall of the Soviet Union, Armenia and Azerbaijan’s disagreements have lasted for generations. The threat of ethnic violence and forced migration is not new to the Armenian people, and unfortunately still remains relevant. This paper deals not only with questions of …

Solar Energy Industries Association V. Federal Energy Regulatory Commission, Brandy Keesee May 2024

Solar Energy Industries Association V. Federal Energy Regulatory Commission, Brandy Keesee

Public Land & Resources Law Review

In Solar Energy Industries Association v. Federal Energy Regulatory Commission (“Solar Energy”), the court grappled with a complex web of regulatory and environmental considerations. The overall dispute was the promulgation and implementation of Order 872, a directive issued by the Federal Energy Regulatory Commission (“FERC” or “Commission”), and its alignment with the Public Utility Regulatory Policies Act of 1978 (“PURPA”) and the Administrative Procedure Act (“APA”). The dispute in Solar Energy is about FERC’s interpretation and application of PURPA in managing qualifying facilities (“QFs”). The crux of the contention was whether FERC’s 2020 rule revisions set forth in Order 872 …

Effects Of Leaf Species And Conditioning State Of Fresh Leaves On Colonization By Stream And Pond Macroinvertebrates, Austin R. Hoffman, Erika V. Iyengar May 2024

Effects Of Leaf Species And Conditioning State Of Fresh Leaves On Colonization By Stream And Pond Macroinvertebrates, Austin R. Hoffman, Erika V. Iyengar

Pacific Faculty Work

Fresh, green leaves are increasing as resources in aquatic ecosystems due to more frequent severe spring and summer storms, but research on allochthonous resources typically uses senescent leaves. We examined macroinvertebrate colonization of green leaves of three native deciduous trees (red maple, red oak, and tulip poplar) over two weeks within both a stream and fishless pond. Leaf colonization varied depending on the taxa of leaves and colonizers, submersion time, and the ecosystem examined. Within the stream, the densities of isopods (Lirceus sp.) and snails (mostly the invasive Potamopyrgus antipodarum) did not vary significantly across leaf species. In contrast, mayflies …

Toxicological Impacts Of River Chemical Spills In Affected West Virginia Counties, Janice Lorraine Taylor Peters May 2024

Toxicological Impacts Of River Chemical Spills In Affected West Virginia Counties, Janice Lorraine Taylor Peters

Walden Dissertations and Doctoral Studies

Factors such as geographical location/proximity and exposure to external environmental events, as well as poor regulatory processes, are known to increase the risk for cancers among affected populations. There is still a gap in the literature related to the incidence of cancers in specific regions with heavy industrialization and environmental health hazards. In West Virginia, chemical contamination events have occurred in or near water systems. The goal of this quantitative, retrospective study was to discover whether there were any differences in the incidence of disease or adverse health effects and cancers due to chemical contamination in four counties (Kanawha, Mingo, …

Faculty Research 2024

Immunotherapy with chimeric antigen receptor T cells for pediatric solid and brain tumors is constrained by available targetable antigens. Cancer-specific exons present a promising reservoir of targets; however, these have not been explored and validated systematically in a pan-cancer fashion. To identify cancer specific exon targets, here we analyze 1532 RNA-seq datasets from 16 types of pediatric solid and brain tumors for comparison with normal tissues using a newly developed workflow. We find 2933 exons in 157 genes encoding proteins of the surfaceome or matrisome with high cancer specificity either at the gene (n = 148) or the alternatively spliced …

Cultivating Hope: The Influence Of Positive Outlooks In K-12 Climate Change Education, Agnes Brown May 2024

Cultivating Hope: The Influence Of Positive Outlooks In K-12 Climate Change Education, Agnes Brown

Student Theses 2015-Present

Abstract

Students studying the environment know and feel the emotional toll that it can take. This can easily contribute to mental health issues and utter defeat. This paper addresses the exclusion of optimism from environmental education and the impacts this has on childhood development and students as they advance to higher levels of education. Through this analysis, the goal is to find a role for hope in a topic typically framed in an apocalyptic manner. The shift from denying the existence of climate change to denying that it can be addressed, particularly through the usage of “doomsday language,” has created …

Assessing The Effects Of Cold Storage Regimes Of North American Bombus Gynes, Thuy Tien T. Lindsay, Marcia M. Hagen, Joyce N. Knoblett, Joseph P. Rinehart, Karen M. Kapheim, James P. Strange May 2024

Assessing The Effects Of Cold Storage Regimes Of North American Bombus Gynes, Thuy Tien T. Lindsay, Marcia M. Hagen, Joyce N. Knoblett, Joseph P. Rinehart, Karen M. Kapheim, James P. Strange

Biology Faculty Publications

Bumble bees are the primary commercial pollinators for greenhouse-grown crops and managing them requires insight into the Bombus life cycle. However, some aspects of the life cycle are not fully understood. Once mated, adult Bombus gynes can be stored for a short term at a constant low temperature (CLT). However, survival rates for some species are low during CLT, particularly when storage periods exceed a month. To address this, we tested the hypothesis that Bombus gynes stored in a fluctuating thermal regime (FTR) have increased survival compared to gynes stored in CLT. Specifically, the FTR had a baseline temperature of …
