Globally, river restoration has become a popular tool for improving the health of a watershed and restoring ecosystem services, but still has significant knowledge gaps. In certain areas and scientific communities, special attention has been given to the response of macroinvertebrates as a measure of restoration success. This systematic literature review aims to highlight and discuss the patterns in studies that have comparable before-and-after restoration data on macroinvertebrates after reconnecting stream channels to their floodplains. Background: Physical activity and metformin pharmacotherapy are associated with improved clinical outcomes in breast and colorectal cancer survivors. Myokines are cytokines secreted from skeletal muscle that may mediate these associations. Methods: This hypothesis-generating analysis used biospecimens collected from a multi-centre 2 × 2 factorial randomized design of 116 patients with stage I–III breast and colorectal cancer who were randomized to 12 weeks of (1) aerobic exercise (moderate intensity titrated to 220 min/week); (2) metformin (850 mg daily for 2 weeks and then titrated to 850 mg twice per day); (3) aerobic exercise and metformin; or (4) control. Inhibitors of Bruton's tyrosine kinase (BTKi) and chimeric antigen receptor T-cell (CAR-T) therapy targeting CD19 are paradigm-shifting advances in treating patients with aggressive mantle cell lymphoma (MCL). However, clinical relapses following BTKi and CD19-directed CAR-T treatments are a fast-growing medical challenge. Development of novel therapies to overcome BTKi resistance (BTKi-R) and BTKi-CAR-T dual resistance (Dual-R) are urgently needed. Our single-cell RNA sequencing data revealed major transcriptomic reprogramming, with great enrichment of MYC-targets evolving as resistance to these therapies developed. The best-known mode of action of calmodulin (CaM) is binding of Ca2+ to its N- and C-domains, followed by binding to target proteins. An underappreciated facet of this process is that CaM is typically bound to proteins at basal levels of free Ca2+, including the small, intrinsically disordered, neuronal IQ-motif proteins called PEP-19 and neurogranin (Ng). PEP-19 and Ng would not be effective competitive inhibitors of high-affinity Ca2+-dependent CaM targets at equilibrium because they bind to CaM with relatively low affinity, but they could influence the time course of CaM signaling by affecting the rate of association of CaM with …

Lung metastases are the primary cause of death for osteosarcoma (OS) patients. We recently validated interleukin-11 receptor α (IL-11Rα) as a molecular target for the inhibition of OS lung metastases. Since there is no clinically approved antibody against this receptor, we sought to identify downstream targets that mediate the effects of IL-11Rα signaling. We used shRNA to deplete IL-11Rα from OS cells; as a complementary approach, we added IL-11 exogenously to OS cells. The resulting changes in gene expression identified EZH2 as a downstream candidate. This was confirmed by knockdown of IL-11Rα in OS cells, which led to increased expression …

Background: Aging significantly elevates the risk of developing neurodegenerative diseases. Neuroinflammation is a universal hallmark of neurodegeneration as well as normal brain aging. Which branches of age-related neuroinflammation, and how they precondition the brain toward pathological progression, remain ill-understood. The presence of elevated type I interferon (IFN-I) has been documented in the aged brain, but its role in promoting degenerative processes, such as the loss of neurons in vulnerable regions, has not been studied in depth.

Resistance to clinical malaria takes years to develop even in hyperendemic regions and sterilizing immunity has rarely been observed. To evaluate the maturation of the host response against controlled repeat exposures to P. falciparum (Pf) NF54 strain-infected mosquitoes, we systematically monitored malaria-naïve participants through an initial exposure to uninfected mosquitoes and 4 subsequent homologous exposures to Pf-infected mosquitoes over 21 months (n = 8 males) (ClinicalTrials.gov# NCT03014258). The case hinged on whether HPD adequately considered environmental impacts under SEQRA. The court held HPD's assessment met legal standards by identifying relevant environmental concerns and taking a reasoned approach, including mitigating open space deficiencies. The decision reinforced that judicial review of agency actions under SEQRA is limited to procedural adherence and rationality, emphasizing agencies' discretion in environmental assessments. The ruling clarified that SEQRA does not require agencies to address every potential environmental impact comprehensively.

Global food calorie supply per person is more than 2900 kcal per day on average, but we have failed to ensure equitable access to these calories. Yet how uneven food access is within countries has remained poorly understood, as has the progress made over the past five decades on closing the access gap. Using publicly available data, we developed a theoretically-grounded statistical model to estimate the cross-national relationship between average per capita expenditure and per capita food availability and used this to estimate within-country access to food for income deciles in 135 countries. BACKGROUND: p53, the most frequently mutated gene in cancer, lacks effective targeted drugs.
METHODS: We developed monoclonal antibodies (mAbs) that target a p53 hotspot mutation E285K without cross-reactivity with wild-type p53. They were delivered using lipid nanoparticles (LNPs) that encapsulate DNA plasmids. Western blot, BLI, flow cytometry, single-cell sequencing (scRNA-seq), and other methods were employed to assess the function of mAbs in vitro and in vivo.
RESULTS: These LNP-pE285K-mAbs in the IgG1 format exhibited a robust anti-tumor effect, facilitating the infiltration of immune cells, including CD8+ T, B, and NK cells. scRNA-seq revealed that IgG1 reduces immune inhibitory signaling, increases …

Hyperpolarization and cyclic nucleotide (HCN) activated ion channels are critical for the automaticity of action potentials in pacemaking and rhythmic electrical circuits in the human body. Unlike most voltage-gated ion channels, the HCN and related plant ion channels activate upon membrane hyperpolarization. Although functional studies have identified residues in the interface between the voltage-sensing and pore domain as crucial for inverted electromechanical coupling, the structural mechanisms for this unusual voltage-dependence remain unclear. Dietary protein is a critical regulator of metabolic health and aging. Low protein diets are associated with healthy aging in humans, and dietary protein restriction extends the lifespan and healthspan of mice. In this study, we examined the effect of protein restriction (PR) on metabolic health and the development and progression of Alzheimer's disease (AD) in the 3xTg mouse model of AD. Here, we show that PR promotes leanness and glycemic control in 3xTg mice, specifically rescuing the glucose intolerance of 3xTg females. PR induces sex-specific alterations in circulating and brain metabolites, downregulating sphingolipid subclasses in 3xTg females. PR also …

Teaching cultural literacy skills provides students an opportunity to develop capabilities to competently function in our current ever-changing global society. A digital escape room was created for a fully online Latin American Humanities course offered in the fall 2022 semester, after the peak of the COVID-19 pandemic. This article aims to envision how archives could better reflect and represent neurodivergent records creators—people whose brains function in ways that diverge significantly from dominant societal standards of “normal". It discusses how neurodivergent records creators and their recordkeeping behaviors often do not fit within traditional archival paradigms which center verbal, written, and linear documents in specific organizational systems. The article then brings together sources from disability studies, feminist and gender studies, library studies, literary analysis, and archival scholarship to imagine ways in which the principle of provenance could be expanded to fit the archives of neurodiverse creators (and archives that …

Purpose
This study aims to understand the level of graduate students’ awareness and usage of e-books purchased by the authors’ university library since the COVID-19 pandemic.
Design/methodology/approach
The authors created a Qualtrics survey with 13 questions that was sent to the university’s graduate student email list. The survey was sent in the fall of 2023 and was open for one month. The list had 3,318 subscribers; 113 complete responses were received, for a response rate of 3.4%.
Findings
The results found that doctoral students (80%) are more aware than master’s students (64%) of e-book availability through the library, and usage …

Myxine limosa is a burrowing species of hagfish that occurs in the western North Atlantic in areas with muddy substrate and at depths generally greater than 100 meters. Burrowing of M. limosa has been observed from submersibles, but little is known about the behavior of these animals within the substrate or the biomechanical mechanisms involved. Here, we investigated burrowing in M. limosa by observing individuals as they burrowed through transparent gelatin. A photoelastic setup using crossed polarizers allowed us to visualize stress development in the gelatin as the hagfish moved through it. We found that M. limosa created U-shaped burrows …

BACKGROUND: Despite serious health and social consequences, effective intervention strategies for habitual alcohol binge drinking are lacking. The development of novel therapeutic and preventative approaches is highly desirable. Accumulating evidence in the past several years has established associations between the gut microbiome and microbial metabolites with drinking behavior, but druggable targets and their underlying mechanism of action are understudied.
RESULTS: Here, using a drink-in-the-dark mouse model, we identified a microbiome metabolite-based novel treatment (sodium valerate) that can reduce excessive alcohol drinking. Sodium valerate is a sodium salt of valeric acid short-chain fatty acid with a similar structure as γ-aminobutyric acid …

Ancient Sanskrit literature has rich discussion of measurements by different authorities. Examples from the Vedic Age and the period of Itihāsas can be found in Ṛgveda (c. 1500 BCE), Rāmāyaṇa, and Mahābhārata. A subsequent, and more elaborate, exercise of codification of measurements, relevant to finance and administration, is observed in Arthaśāstra, written by Kauṭilya, also known as Cāṇakya and Viṣṇugupta (c. 350-275 BCE).
The author, a scholar from Takṣaśilā, was the advisor during Cāndragupta Maurya’s reign (c. 321-297 BCE). After millennia of hibernation, Kauṭilya’s Arthaśāstra resurfaced, in the beginning of the twentieth century. Ever since, Arthaśāstra has been a subject …

Nursing students experience moderate to severe anxiety related to the rigor of nursing programs. Anxiety often leads to poor performance on exams and skills evaluations. When anxiety is not limited, it can contribute to nursing students being unable to progress in the nursing program and high attrition rates. The National League of Nursing (2021) estimates the attrition rate for nursing students in the United States at 20%. This work explores the fabrication route of curcumin-loaded carboxymethyl cellulose sodium/sodium alginate/palladium chloride (CMC-Na/SA/PdCl2) composite hydrogel as localized delivery system for cancer treatment. Herein, an approach was established for improving encapsulation capacity and exploiting therapeutic efficiency. CMC-Na/SA/PdCl2 composite hydrogels were prepared by gelation method using ionic crosslinker (PdCl2). Results proved that the optimized formula (CMC-Na:SA (4:1)/PdCl2 0.4%) recorded a gel fraction of 91% with a swelling ratio ~ 3000% after 96 h, while XRD analysis exhibited no remarks of sharp peaks which confirmed an amorphous phase of tested hydrogel. When postmenopausal women are under stress conditions, this exacerbates mood disorders and issues with neuroimmune systems. The porcine placenta is known to relieve menopausal depression in clinical trials, but its underlying mechanisms for depression and anti-inflammatory functions remain poorly defined. The present study was designed to examine the anti-inflammatory effects of enzymatic porcine placenta hydrolysate (EPPH) on LPS-induced levels of nitric oxide (NO), prostaglandin E2 (PGE2), corticosterone (CORT), and pro-inflammatory cytokine interleukin-1 beta (IL-1β) in RAW 264.7 macrophage cells. Agricultural diversification in the Midwestern Corn Belt has the potential to improve socioeconomic and environmental outcomes by buffering farmers from environmental and economic shocks and improving soil, water, and air quality. However, complex barriers related to agricultural markets, individual behavior, social norms, and government policy constrain diversification in this region. This study examines farmer perspectives regarding the challenges and opportunities for both corn and soybean production and agricultural diversification strategies. STING agonists can reprogram the tumor microenvironment to induce immunological clearance within the central nervous system. Using multiplexed sequential immunofluorescence (SeqIF) and the Ivy Glioblastoma Atlas, STING expression was found in myeloid populations and in the perivascular space. The STING agonist 8803 increased median survival in multiple preclinical models of glioblastoma, including QPP8, an immune checkpoint blockade-resistant model, where 100% of mice were cured. Ex vivo flow cytometry profiling during the therapeutic window demonstrated increases in myeloid tumor trafficking and activation, alongside enhancement of CD8+ T cell and NK effector responses.
