Using Unmanned Aircraft Systems (Uas) To Assess Body Condition Of Common Bottlenose Dolphins (Tursiops Truncatus Truncatus) In The Indian River Lagoon, Florida, Jessica Jane Provenzano
Theses and Dissertations
Common bottlenose dolphins (Tursiops truncatus truncatus) in the Indian River Lagoon (IRL) have experienced four Unusual Mortality Events (UMEs). This stock is considered immunocompromised and is routinely subjected to persistent anthropogenic stressors such as fishing gear entanglement, vessel strikes, contaminants, and harmful algal blooms. Previous body condition assessments of this stock have involved invasive capture-release examinations or subjective methods using lateral body images. To improve precision, we investigated the use of photogrammetry data collected from noninvasive unmanned aircraft systems (UAS) combined with models developed from capture-release data to estimate morphometric parameters and subsequently determine the body condition of …

Assessing The Gut Content Of Styela Plicata: A Benthic Filter Feeder In The Indian River Lagoon, Honor Lily Triumph Edmands
Theses and Dissertations
The Indian River Lagoon (IRL) has experienced declining water quality due to human activities involving nutrient input, leading to Harmful Algal Blooms (HABs). The Living Docks program affixes oyster mats to dock pilings to promote the settlement and recruitment of benthic filter feeders for water quality improvement. However, questions remain about filtering efficiencies of organisms that make up the mat communities, and whether organisms remove harmful species from the IRL. The pleated sea squirt, Styela plicata, is a common oyster mat resident. The goal of this thesis was to investigate what phytoplankton and bacteria S. plicata consumes, and if …
Bracketed Assessments In Behavioral Gerontology: Evaluating Preference With Present And Absent Stimuli, Flavia Eduarda Del Castillo Troche
Bracketed Assessments In Behavioral Gerontology: Evaluating Preference With Present And Absent Stimuli, Flavia Eduarda Del Castillo Troche
Theses and Dissertations
The present study evaluated the use of bracketed assessments to assess the activity preference of three individuals with dementia. The study proposed an alternative method to traditional stimulus preference assessments, to bridge the gap in behavior analytical literature within behavioral gerontology. Thus, bracketed assessments were conducted using two presentation methods, verbal plus visual presentation and verbal-only presentation. Results suggested that while participants showed consistency within low preferred activity selection in the bracketed assessment, variability was observed among participants’ preferences for high and moderate preferred activities. Both presentation methods yield similar selection results for one participant, however, the other two participants …
Cultural Selection Of Ethical Self-Control Toward Sustainability, Michael Jonathan Passage
Cultural Selection Of Ethical Self-Control Toward Sustainability, Michael Jonathan Passage
Theses and Dissertations
This study consisted of three experiments to improve our understanding of the conditions that foster cooperation related to sustainability. In Experiment 1, participants completed a demographic questionnaire and an environmental and social discounting task. Results (a) support the utility of the environmental discounting task; (b) suggest responding to the discounting task is dissimilar; and (c) show that responding to hypothetical consequences on the social discounting task correlates to actual responding. Experiment 2 evaluated whether interlocking behavioral contingencies (by microculture members) associated with sustainable and non-sustainable aggregate products (Condition A & B) could be selected by cultural consequences (i.e., 20¢ toward …

This study used a marsupial Monodelphis domestica, which is born very immature and most of its development is postnatal without placental protection. RNA-sequencing (RNA-Seq) was used to identify the expression of influx and efflux transporters (ATP-binding cassettes [ABCs] and solute carriers [SLCs]) and metabolizing enzymes in brains of newborn to juvenile Monodelphis. Results were compared to published data in the developing eutherian rat. To test the functionality of these transporters at similar ages, the entry of paracetamol (acetaminophen) into the brain and cerebrospinal fluid (CSF) was measured using liquid scintillation counting following a single administration of the drug along with …

PURPOSE: CD25KO mice are a model of Sjögren disease (SjD) driven by autoreactive T cells. Cathepsin S (CTSS) is a protease crucial for major histocompatibility complex class II presentation that primes T cells. We investigated if a diet containing CTSS inhibitor would improve autoimmune signs in CD25KO mice.
METHODS: Four-week female CD25KO mice were randomly chosen to receive chow containing a CTSS inhibitor (R05461111, 262.5 mg/kg chow) or standard chow for 4 weeks. Cornea sensitivity was measured. Inflammatory score was assessed in lacrimal gland (LG) histologic sections. Flow cytometry of LG and ocular draining lymph nodes (dLNs) investigated expression of …

Introduction: Social workers, the largest group of mental health clinicians in the United States, play a pivotal role in mental health promotion. Despite the importance of sleep for mental health, there is no empirical research on sleep education interventions for social workers. Method: We designed an online sleep health education intervention to equip social work students to promote healthy sleep practices among their clients. An interdisciplinary team of experts devised the 90 min intervention using an empirically supported behavioral change theoretical model (COM-B). The intervention discusses multi-level factors that impact sleep and emphasizes considerations for health disparities in populations commonly …

A pervasive view is that undifferentiated stem cells are alone responsible for generating all other cells and are the origins of cancer. However, emerging evidence demonstrates fully differentiated cells are plastic, can be coaxed to proliferate, and also play essential roles in tissue maintenance, regeneration, and tumorigenesis. Here, we review the mechanisms governing how differentiated cells become cancer cells. First, we examine the unique characteristics of differentiated cell division, focusing on why differentiated cells are more susceptible than stem cells to accumulating mutations. Next, we investigate why the evolution of multicellularity in animals likely required plastic differentiated cells that maintain …

Polarized vesicular trafficking directs specific receptors and ion channels to cilia, but the underlying mechanisms are poorly understood. Here we describe a role for DLG1, a core component of the Scribble polarity complex, in regulating ciliary protein trafficking in kidney epithelial cells. Conditional knockout of Dlg1 in mouse kidney causes ciliary elongation and cystogenesis, and cell-based proximity labeling proteomics and fluorescence microscopy show alterations in the ciliary proteome upon loss of DLG1. Specifically, the retromer-associated protein SDCCAG3, IFT20, and polycystin-2 (PC2) are reduced in the cilia of DLG1-deficient cells compared to control cells. This phenotype is recapitulated in vivo and …

PURPOSE: Serotonin (5-HT
METHODS: Ondansetron (10 mg/kg) and tariquidar (7.5 mg/kg) were administered intravenously, plasma and tissue samples were collected and analyzed by HPLC. A mathematical model with brain, spinal cord, cerebrospinal fluid and two systemic disposition compartments was developed to describe the data.
RESULTS: The results demonstrate that tariquidar at 7.5 mg/kg resulted in a complete inhibition of Pgp efflux of ondansetron in the brain and spinal cord. The compartmental model successfully captured pharmacokinetics of ondansetron in wild type and Pgp knockout (KO) animals receiving the drug alone or in wild type animals receiving the ondansetron and tariquidar combination. …

Fibrillin microfibrils play a critical role in the formation of elastic fibers, tissue/organ development, and cardiopulmonary function. These microfibrils not only provide structural support and flexibility to tissues, but they also regulate growth factor signaling through a plethora of microfibril-binding proteins in the extracellular space. Mutations in fibrillins are associated with human diseases affecting cardiovascular, pulmonary, skeletal, and ocular systems. Fibrillins consist of up to 47 epidermal growth factor-like repeats, of which more than half are modified by protein O-glucosyltransferase 2 (POGLUT2) and/or POGLUT3. Loss of these modifications reduces secretion of N-terminal fibrillin constructs overexpressed in vitro. Here, we investigated …

Purpose: Existing resources that characterize the essentiality status of genes are based on either proliferation assessment in human cell lines, viability evaluation in mouse knockouts, or constraint metrics derived from human population sequencing studies. Several repositories document phenotypic annotations for rare disorders; however, there is a lack of comprehensive reporting on lethal phenotypes.
Methods: We queried Online Mendelian Inheritance in Man for terms related to lethality and classified all Mendelian genes according to the earliest age of death recorded for the associated disorders, from prenatal death to no reports of premature death. We characterized the genes across these lethality categories, …

General Agreement on Tariffs and Trade (GATT) Article X has been consistently overlooked in literature even though it is a unique provision in the GATT-regime: it acknowledges standing for private actors who can challenge trade-related administrative action by World Trade Organization (WTO) members. This provision is an unusual instantiation of the need to provide transparency for private actors about state policy. It is equally eccentric in seeming to provide a right for private actors to challenge governments, albeit only before domestic fora. How did the GATT end up with this provision? And are these apparent private rights meaningful in practice? …
Safe And Efficient Operation Of Mobile Robots In Indoor Environments: A User-Centric Shared Control System With High-Level Navigation Capabilities, Ahmet Saglam
Electrical & Computer Engineering Theses & Dissertations
Hospitalization and isolation can be a traumatic experience for immunocompromised children, especially because they are separated from their families and friends. Social robots have been proposed as a way to improve the quality of care for children hospitalized in isolation by providing alternative means of social interaction and support. Remote control of such robots in a hospital setting, particularly where safety is a major concern, can be a daunting task for young patients.
This dissertation introduces a multilevel shared control system for mobile robots, specifically companion robots in hospital-like indoor spaces. The system integrates user inputs with algorithmic semi-autonomous control …

Lafora disease (LD) is a syndrome of progressive myoclonic epilepsy and cumulative neurocognitive deterioration caused by recessively inherited genetic lesions of EPM2A (laforin) or NHLRC1 (malin). Neuropsychiatric symptomatology in LD is thought to be directly downstream of neuronal and astrocytic polyglucosan aggregates, termed Lafora bodies (LBs), which faithfully accumulate in an age-dependent manner in all mouse models of LD. In this study, we applied home-cage monitoring to examine the extent of neurobehavioral deterioration in a model of malin-deficient LD as a means to identify robust preclinical endpoints that may guide the selection of novel genetic treatments. At 6 weeks, ∼6-7 …

Microbiomes have emerged as a key component essential for maintaining the health of an organism. Additionally, the roles of microbiomes are multifaceted, some unique to specific body areas and organs while others, particularly the gut microbiome, having broader effects on the entire organism. Comparative literature is emerging that compares microbiomes across mammals and birds. Domestic poultry have been the most extensively studied relative to their role in production agriculture. These data have provided a great deal of information about the effects of diet and nutritional requirements relative to the gut microbiome, productivity, and resilience to diseases. Conversely, limited research has …

Parents and teachers concerns about the development and socialization of lying aligns with research suggesting that persistent lying is related with delinquency, aggression, and conduct problems. We were interested in examining the difference between two groups: Honest and Praise (HP), and Control Group (CG) for the extent to which exposure to the experimental condition influenced a child’s decision to be honest and confess after a transgression. The study was designed to examine the extent to which observing peers of similar age receiving praise for being honest can promote honesty amongst children. Further, the study determined if gender influences honest reporting. …

Vascular remodeling to match arterial diameter to tissue requirements commonly fails in ischemic disease. Endothelial cells sense fluid shear stress (FSS) from blood flow to maintain FSS within a narrow range in healthy vessels. Thus, high FSS induces vessel outward remodeling, but mechanisms are poorly understood. We previously reported that Smad1/5 is maximally activated at physiological FSS. Smad1/5 limits Akt activation, suggesting that inhibiting Smad1/5 may facilitate outward remodeling. Here we report that high FSS suppresses Smad1/5 by elevating KLF2, which induces the bone morphogenetic protein (BMP) pathway inhibitor, BMP-binding endothelial regulator (BMPER), thereby de-inhibiting Akt. Polymyxins are a last-resort treatment option for multidrug-resistant gram-negative bacterial infections, but they are associated with nephrotoxicity. Gelofusine was previously shown to reduce polymyxin-associated kidney injury in an animal model. However, the mechanism(s) of renal protection has not been fully elucidated. Here, we report the use of a cell culture model to provide insights into the mechanisms of renal protection. Murine epithelial proximal tubular cells were exposed to polymyxin B. Cell viability, lactate dehydrogenase (LDH) release, polymyxin B uptake, mitochondrial superoxide production, nuclear morphology, and apoptosis activation were evaluated with or without concomitant gelofusine. A megalin knockout cell line was …

SummaryPregnancy during or soon after treatment with weight loss medication, particularly glucagon-like peptide-1 receptor agonists (GLP-1 RAs), is contraindicated due to potential teratogenicity. The aim of this scoping review is to investigate what is known about the use of weight loss medication in women of childbearing age in relation to reproductive health outcomes, focusing on the three medications licenced in the United Kingdom at the time of the search. A systematic search of studies that assessed reproductive health outcomes in women taking either orlistat, liraglutide or semaglutide was undertaken in July 2023 and updated in January 2024 across MEDLINE, Embase, …

Autophagy is a conserved catabolic process where double membrane-bound structures form around macromolecules or organelles targeted for degradation. Autophagosomes fuse with lysosomes to facilitate degradation and macromolecule recycling for homeostasis or growth in a cell autonomous manner. In cancer cells, autophagy is often up-regulated and helps cancer cells survive nutrient deprivation and stressful growth conditions. Here, we propose that the increased intracellular pH (pHi) common to cancer cells is sufficient to induce autophagic cell death. We previously developed tools to increase pHi in the Drosophila eye via overexpression of DNhe2, resulting in aberrant patterning and reduced tissue size. We examined …

BACKGROUND: The Global Program to Eliminate Lymphatic Filariasis (GPELF) is the largest public health program based on mass drug administration (MDA). Despite decades of MDA, ongoing transmission in some countries remains a challenge. To optimise interventions, it is critical to differentiate between recrudescence and new infections. Since adult filariae are inaccessible in humans, deriving a method that relies on the offspring microfilariae (mf) is necessary.
METHODS: We developed a genome amplification and kinship analysis-based approach using Brugia malayi samples from gerbils, and applied it to analyse Wuchereria bancrofti mf from humans in Côte d'Ivoire. We examined the pre-treatment genetic diversity …

The emergence of 2D materials has significantly expanded the wide range of nanomaterials with diverse applications. Notably, their high conductivity, catalytic efficiency, and hydrophobicity have fueled heightened research interests for water treatment applications. This research aimed to investigate the synthesis and characterization of MXene and reduced graphene oxide (rGO) nanocomposites with silver nanoparticles (Ag) for enhanced catalytic activity in the decomposition of Direct Blue-24 dye. In this study, we employed well-established methods, previously documented in the literature, to prepare two distinct nanocomposites. Novel nanocomposites, namely reduced graphene oxide–silver nanoparticles (rGO–Ag) and MXene–silver nanoparticles (MXene–Ag), were synthesized using the hydrothermal and …
