Deletion Of Tissue Factor Pathway Inhibitor Isoform Beta Or Gamma, But Not Alpha, Improves Clotting In Hemophilic Mice, Irem Eldem, Lilian Antunes-Heck, Renumathi Subramanian, Nina M Lasky, Katrina Ashworth, Jorge Di Paola, Thomas J Girard Oct 2024

Deletion Of Tissue Factor Pathway Inhibitor Isoform Beta Or Gamma, But Not Alpha, Improves Clotting In Hemophilic Mice, Irem Eldem, Lilian Antunes-Heck, Renumathi Subramanian, Nina M Lasky, Katrina Ashworth, Jorge Di Paola, Thomas J Girard

2020-Current year OA Pubs

BACKGROUND: Tissue factor pathway inhibitor (TFPI) regulates tissue factor-triggered coagulation. Humans and mice express transcripts encoding for multidistributed (endothelial, platelet, and plasma) 3-Kunitz domain TFPIα and endothelial membrane-anchored 2-Kunitz TFPIβ. Mice express a third transcript, γ, that encodes plasma lipoprotein-associated 2-Kunitz TFPI. In humans, proteolysis of α and/or β produces plasma lipoprotein-associated 2-Kunitz TFPI at lower levels. In clinical trials, monoclonal antibodies that target all TFPI isoforms extend coagulation and correct bleeding in hemophilic patients but with some thrombosis risks.

OBJECTIVES: To determine the impact of TFPI isoform-specific deletions on promoting clotting in hemophilic mice.

METHODS: Engineered TFPI isoform-specific, hemophilic …

Amyloid Accelerator Polyphosphate Fits As The Mystery Density In Α-Synuclein Fibrils, Philipp Huettemann, Pavithra Mahadevan, Justine Lempart, Eric Tse

Open Access archive

Aberrant aggregation of α-Synuclein is the pathological hallmark of a set of neurodegenerative diseases termed synucleinopathies. Recent advances in cryo-electron microscopy have led to the structural determination of the first synucleinopathy-derived α-Synuclein fibrils, which contain a non-proteinaceous, “mystery density” at the core of the protofilaments, hypothesized to be highly negatively charged. Guided by previous studies that demonstrated that polyphosphate (polyP), a universally conserved polyanion, significantly accelerates α-Synuclein fibril formation, we conducted blind docking and molecular dynamics simulation experiments to model the polyP binding site in α-Synuclein fibrils. Here, we demonstrate that our models uniformly place polyP into the lysine-rich pocket, …

The Embrace Of Religion And Spirituality In Indonesia, Olivia Cotton Oct 2024

The Embrace Of Religion And Spirituality In Indonesia, Olivia Cotton

Indonesia: Arts, Religion, and Social Change

This thesis offers a contemporary investigation of the extent to which historical contexts contribute to Indonesian people’s reservations towards political conversations and social movements, such as protests and demonstrations. The historical contexts in question are focused on the Soeharto dictatorship from 1966-1999 and its long reign of censorship, citizen violence, and general human rights violations. The research employs a mixed-methods approach focused on cultural and participant observation, social media analysis, and interviews. Social media analyses offer insights into activism, particularly that of students and the youth, that are not generally available in scholarly sources. Interviews with Indonesian citizens and local …

Is The Constitution Of 1787 A White Supremacist Document? Against Essentialism In Constitutional Interpretation, David S. Schwartz Oct 2024

Is The Constitution Of 1787 A White Supremacist Document? Against Essentialism In Constitutional Interpretation, David S. Schwartz

William & Mary Bill of Rights Journal

A curious convergence is emerging in legal academia around the conclusion that the 1787 Constitution is a white supremacist document. Although most originalists would deny that contention, their methodology strongly favors, if it does not compel, an agreement with progressive, “neo-Garrisonian” scholars that the Constitution of 1787 is indeed a white supremacist document. Both the neo-Garrisonian and originalist elements of this implicit convergence stem from their “essentialism” in Constitutional interpretation: the idea that the Constitution or its terms or provisions carry a uniquely and objectively correct meaning, invariant over time, and independent of our evolving normative commitments.

This Article argues …

Life, Liberty, And Freedom From Non-Consensual Pelvic Exams?, Michaela Cotton

William & Mary Bill of Rights Journal

[T]here is a need for the federal government to intervene and finally ban the practice of performing non-consensual pelvic exams, and it can do so under the Due Process Clause of the Fourteenth Amendment, which guarantees the right to refuse involuntary medical treatment. Part I will provide a brief overview of pelvic exams and informed consent; Part II draws on policy considerations for banning this practice and tackles counter-arguments; Part III will discuss the relevant federal law and case precedent; Part IV demonstrates how federal law applies to non-consensual pelvic exams; and, finally, Part V will further explain the need …

Excluded But Equal, Gali Racabi Oct 2024

Excluded But Equal, Gali Racabi

William & Mary Bill of Rights Journal

Work law is excessively exclusionary. Canonical examples include agriculture workers, independent contractors, and domestic workers. But work law excludes far more workers and employers than those familiar categories. Whole sectors, occupations, business sizes, wage arrangements, geographical areas, and more are excluded from the basic tenets of economic citizenship, including federal, state, and local minimum wage; overtime; and safety and labor protection laws.

Equal protection claims govern the authority of legislatures to determine who is in and who is out of work laws. Excluded workers and included employers now routinely use state and federal constitutional equal protection arguments against exclusionary work …

Approaching A U.S. Regulatory Framework For Sporeless Seaweed Biotechnology, Gabriela Martinez, Terra Bowling, Catherine M. Janasie, Charlie Doering-Powell, Amalia Aruda Almada

William & Mary Environmental Law and Policy Review

Seaweed farming is the fastest-growing aquaculture sector in the United States. The development of sterile, selectively bred (sporeless) seaweed aims to support the economic viability of future large offshore seaweed operations and minimize the environmental risks of these operations to wild seaweed beds. Farmers have implemented analogous breeding techniques to control and enhance most terrestrial crops and common aquaculture products (e.g., oysters and carp). This Article draws from existing federal and state aquaculture policies to explore four pillars critical to a future sporeless seaweed regulatory framework: ensuring maintained sterility, tracing the geographic origin of seed, securing offshore permits, and facilitating …

Multi-Generation Queer Families: Foregrounding The Lgbtqia+ Children Of Lgbtqia+ People, Bella Mancini Pori, Edward Stein

Articles

In the last several years, state legislatures have passed a disturbing array of anti-queer laws that target queer (especially trans) children. The basic fear animating these laws is that exposure to LGBTQ people or LGBTQ supportive ideas will somehow turn straight children queer. LGBTQ activists have responded to the concern that exposure to queer people or ideas will make children queer by arguing that sexual orientations are innate or immutable. This immutability argument is problematic, first, because it avoids confronting the normative judgment at the heart of the fear of a queer child by failing to say that there is …

An Introduction To Open-Source Geographic Information Systems (Gis): Qgis, Ashley York Oct 2024

An Introduction To Open-Source Geographic Information Systems (Gis): Qgis, Ashley York

Library Workshops

In general, Geographic Information Systems (GIS) provide a framework for gathering, organizing, and analyzing spatial data, that is visualized through maps. By arranging spatial information into coincident layers, GIS analyses help reveal deeper insights into a study area by identifying not only where features of interest are located, but also why patterns or processes are occurring. However, a number of various GIS software and online mapping platforms exist, each with a different range of analysis capabilities, but also with different accessibility limitations in terms of user cost (e.g., open-source vs. commercial). In this introductory workshop, participants will become familiar with …

What Freedom? Freedom-A Disease Without Cure. Slavoj Žižek. London: Bloomsbury Academic. 2023. Pp. 328., Edward Sankowski, Betty J. Harris

International Dialogue

Slavoj Žižek's outpouring of books continues, including the volume, Freedom-A Disease Without Cure (2023). The title is somber, but the contents do not signal an unrelievedly depressed attitude on the part of the author. Rather, the book manifests expanding curiosity and far-reaching originality. That is, however, supplemented by some self-criticism, e.g., when the author broods about the apathy in himself and others that he claims is a result of the hyperactivity enjoined by and observed in contemporary capitalism. We hope to go beyond critical interpretation and evaluation of Žižek, to express our own new insights beyond those formulated or hinted …

Revolution And Dictatorship: The Violent Origin Of Durable Authoritarianism Steven Levitsky And Lucan Way. Princeton: Princeton University Press, 2022. Pp. 656., Joseph L. Derdzinski

International Dialogue

W h y do s o m e ––a nd not other, of course— regimes survive? Surveying a comprehensive range of historical and more contemporary examples, Levitsky and Way's Revolution and Dictatorship: The Violent Origins of Durable Authoritarianism is an important contribution to scholarship that seeks to answer this fundamental question. While much of comparative political specifically, and political science more broadly, centers on democratic regimes and their fragility (and cases of stability, to be fair), why some authoritarian regimes "succeed" over multiple generations is less well understood. By focusing on the authoritarian regimes that emerge from social revolutions, Levitsky …

Rogue Ai Patents And The Uspto’S Rejection Of Alice Joshua A. Z, Joshua A. Zuchniarz Oct 2024

Rogue Ai Patents And The Uspto’S Rejection Of Alice Joshua A. Z, Joshua A. Zuchniarz

University of Chicago Law Review

AI inventions, from ChatGPT to self-driving taxis, have taken the world by storm. Many of these inventions are protected by patents, and the number of AI patents is rapidly growing. Yet a large number of AI patents are flawed, prone to invalidation in court. This Comment argues that many AI patents were granted on legally flawed grounds, pursuant to United States Patent and Trademark Office (USPTO) regulations that systematically departed from controlling case law. The existence of these flawed patents poses a growing problem. Courts may invalidate the patents, upsetting expectations of an important, nascent industry. On the other hand, …

Aging Skeletal Muscle: Influence Of Obesity And Role Of The Exercise Responsive Nuclear Receptor Nor-1, Hector G. Paez

Alternative Theses and Dissertations (AETDs)

Skeletal muscle mass and function are critical components of overall health and quality of life. Advanced age is associated with skeletal muscle disease characterized by a loss of muscle mass, function, and quality. Indeed, low muscle mass and function is associated with poor disease outcomes and greater mortality, greater incidence of falls, and a high fiscal and societal cost. Aging contributes to a decline in skeletal muscle health through several intrinsic and extrinsic mechanisms including impaired nutrient sensing, mitochondrial dysfunction, inflammation, physical inactivity, and proteostatic imbalance. Obesity, an increasingly prominent public health concern, is becoming more common in the elderly …

Climate Change Adaptation Through Economic Democracy, Shriraksha Mohan Oct 2024

Climate Change Adaptation Through Economic Democracy, Shriraksha Mohan

Student Papers in Local and Global Regional Economies

As global climate change poses a serious threat to humanity's survival, equitable and just climate change adaptation has become the focus of several climate movements, social justice groups, governments, and international organizations. Climate sciences and studies have concluded that climate change not only impacts the planet's natural ecosystems, but also has cascading negative consequences on society and the economy. The existing paradigm of economic development has created vast wealth and resource inequality. The inequality further exacerbates the ill effects of climate change and makes vulnerable communities bear a disproportionate amount of the burden of climate catastrophes. This necessitates replacing the …

Reimagining Science As A Natural Tool For Connection: A Restorative Approach, Zachary C. Schafer, Lawrence C. Scharmann

Department of Teaching, Learning, and Teacher Education: Faculty Publications

This paper explores a restorative-type intervention with a high school student who is suspended from school for repeated threats to other students. Throughout the paper, we explore the use of restorative practices in the context of science teaching. Science in process and content is used in tandem with various restorative practices to provide the student a lens through which to better understand what led to her suspension, learn science content, and find perspective change in the process as it pertains to her developing sense of self. The methods and tools are then transformed into practical use through a modified version …

Examining The Effects Of Externally Mounted Biologgers On At-Sea Behavior And Reproductive Success In Adult Female Northern Elephant Seals, Mirounga Angustirostris, Using Computational Fluid Dynamics, Kali N. Prescott

Capstone Projects and Master's Theses

Over the past five decades, biologgers have been used to study animal behavior, physiology, and ecology. Externally mounted biologgers may have unforeseen effects on behavior, habitat use, foraging success, and reproductive success. Although guidelines exist for the recommended mass of biologgers on terrestrial animals, these guidelines are inappropriate for marine animals, which are more affected by buoyancy and drag. However, drag is difficult to measure directly necessitating the use of proxies based on physical characteristics of biologgers like frontal area. Northern elephant seals are a model species for studying the impacts of biologgers due to an existing long-term dataset collected …

Low Prevalence Of Soil Transmitted Helminth Infection In Ugandan Children Hospitalized With Severe Malaria, Drew Capone, Nuzrath Jahan, Ruth Namazzi, Robert Opoka, Chandy John

Paediatrics and Child Health, East Africa

Co-infection by intestinal helminths and Plasmodium spp. may be common in endemic communities. Several studies have identified a relationship between helminth infection, Plasmodium spp. infection and malaria severity. However, the relationship is not well defined, and results are inconclusive. We analyzed stool samples from a cohort of children with severe malaria enrolled in two hospitals in Uganda from 2014-2017 and asymptomatic community children from the same household or neighborhood and enrolled at the same time, all 6 months to 48 months of age. We investigated if intestinal helminth infection modified risk of severe malaria. We extracted nucleic acids from stool …

Local land-use plans help guide future development, but it is often difficult to compare content across jurisdictions, making regional coordination and plan evaluation challenging. This research reviews federal, state, and local data infrastructure guidance for land-use plans and compares such guidance to compliance with a California use-case. Findings indicate a number of obstacles to fostering data sharing and comparative analysis of plans: there is currently no central repository of land-use plans; plans are not uniform in format and are often out of date; many plans are not machine-readable thereby inhibiting text extraction, and planning language varies so greatly that there …

Understanding the neurobiology of semantic knowledge is a major goal of cognitive neuroscience. Taxonomic and thematic semantic knowledge are represented differently within the brain's conceptual networks, but the specific neural mechanisms remain unclear. Some neurobiological models propose that the anterior temporal lobe is an important hub for taxonomic knowledge, whereas the TPJ is especially involved in the representation of thematic knowledge. However, recent studies have provided divergent evidence. In this context, we investigated the neural correlates of taxonomic and thematic confrontation naming errors in 79 people with aphasia. We used three complementary lesion-symptom mapping (LSM) methods to investigate how structure …

Reports of overuse and antimicrobial resistance have fuelled some clinicians to adopt alternative wound dressings termed to be non-medicated or non-antimicrobials, which still claim antimicrobial or antibacterial activity. In this PROSPERO-registered systematic review, we evaluated the in vivo clinical evidence for the effectiveness of DACC-coated dressings in chronic, hard to heal wound-related outcomes. The Grading of Recommendations, Assessment, Development, and Evaluation (GRADE) Framework was adopted as the template in constructing this systematic review. The PICO format (Population [or patients], Intervention, Comparison [control], Outcome/s) was used to identify key clinical questions in determining patient outcomes under two domains (infection control and …

‘More Love Letters, Please!’: Portuguese Love And The Literary Legacy Of Sincere Feeling, Manuela MourãO Oct 2024

‘More Love Letters, Please!’: Portuguese Love And The Literary Legacy Of Sincere Feeling, Manuela MourãO

English Faculty Publications

This essay traces the formation of the myth of Portuguese Love, and demonstrates both its transcultural reach and its importance for literary history. Sparked by the tragic love of Pedro and Inês de Castro, the notion of Portuguese Love was crystallized in seventeenth-century France by Lettres portugaises, influenced European letters for centuries, and became an enduring myth in Portuguese culture. Anchored by Roland Barthes’s critique of the contemporary disparagement of sentimental love, the essay argues that Portuguese Love’s continued affirmation, from nineteenth-century literary iterations to current manifestations, demonstrates the power of a discourse of genuine feeling to counter ideological …
