RESEARCH QUESTION: What is the role and mechanism of action of transcription factor AP-2 gamma (TFAP2C) in porcine early embryo development?
DESIGN: TFAP2C siRNA were injected into porcine oocytes, which subsequently underwent IVF. Different stages of embryos were collected for RNA sequencing, quantitative polymerase chain reaction, immunofluorescence staining to explore the affects in gene expression and epigenetic modification. Porcine fetal fibroblasts were transfected with siRNA, and cells were collected for chromatin immunoprecipitation and dual luciferase reporter assays.
RESULTS: The deficiency of TFAP2C led to disorders in early embryonic development; 1208 genes were downregulated and 792 genes were upregulated in TFAP2C …

Heightened energetic and nutrient demand during lactogenic differentiation of the mammary gland elicits upregulation of various stress responses to support cellular homeostasis. Here, we identify the stimulator of interferon genes (STING) as an immune supporter of the functional development of mouse mammary epithelial cells (MECs). An in vitro model of MEC differentiation revealed that STING is activated in a cGAS-independent manner to produce both type I interferons and proinflammatory cytokines in response to the accumulation of mitochondrial reactive oxygen species. Hair cells of the inner ear and lateral-line system rely on specialized ribbon synapses to transmit sensory information to the central nervous system. The molecules required to assemble these synapses are not fully understood. We show that Nrxn3, a presynaptic adhesion molecule, is crucial for ribbon-synapse maturation in hair cells. In both mouse and zebrafish models, the loss of Nrxn3 results in significantly fewer intact ribbon synapses. We show in zebrafish that, initially, Nrxn3 loss does not alter pre- and postsynapse numbers but, later, synapses fail to pair, leading to postsynapse loss. Cis-regulatory elements (CREs) control gene expression, orchestrating tissue identity, developmental timing and stimulus responses, which collectively define the thousands of unique cell types in the body1–3 . While there is great potential for strategically incorporating CREs in therapeutic or biotechnology applications that require tissue specificity, there is no guarantee that an optimal CRE for these intended purposes has arisen naturally. Here we present a platform to engineer and validate synthetic CREs capable of driving gene expression with programmed cell-type specificity. Neurosteroids are endogenous steroids within the brain that act as modulators in neurotransmission. Among their targets are the gamma-aminobutyric acid (GABA) receptors, which are responsible for inhibitory signaling in the brain. The interaction between neurosteroids and GABA receptors is complex, influencing mood regulation and pain perception. Endogenous neurosteroids have been used in conditions such as postpartum depression (PPD), where fluctuation in neurosteroid levels contribute to the onset of depressive systems. Conversely, exogenous neurosteroids, such as brexanolone and zuranolone, have emerged as therapeutic agents for PPD. These compounds act as positive allosteric modulators (PAMs) on GABA receptors, enhancing inhibitory neurotransmission and …

It investigates both the historical and present-day use of psychoactive substances, specifically psilocybin, in connecting with the divine, attaining transcendental enlightenment, and shaping identity within specific civilizations. Although psilocybin is illegal in Indonesia, these mushrooms can be found in the Gili Islands, raising questions about how they fit into local spiritual traditions and the legal loopholes that allow their use. A spinal cord injury (SCI) compresses the spinal cord, killing neurons and glia at the injury site and resulting in prolonged inflammation and scarring that prevents regeneration. Astrocytes, the main glia in the spinal cord, become reactive following SCI and contribute to adverse outcomes. The anti-inflammatory cytokine transforming growth factor beta 3 (TGFβ3) has been shown to mitigate astrocyte reactivity; however, the effects of prolonged TGFβ3 exposure on reactive astrocyte phenotype have not yet been explored. Wind shear-induced Tornado behavior under full thermodynamic formulation using the ideal gas equation is investigated. The computational fluid dynamics model that includes the energy equation is used to obtain solutions of a natural-scale tornado under pressure and temperature boundary conditions that account for their variations vs. altitude in the atmosphere. The present results show significant differences from previous isothermal simulations and those using the Boussinesq model for buoyancy under small temperature differences. Results show that the downdraft observed in tornados originating from supercells, often accompanied by rain and hail can be captured using the present model that simulates the genesis, …

Background: Extreme violence and psychological abuse have been extensively documented and are pervasive in prostitution. Survivors of prostitution report high levels of posttraumatic stress disorder, dissociation, depression, and self-loathing. These are the same sequelae reported by torture survivors.
Objective: Severe forms of violence have been categorized as torture by experts. The authors note that torture is commonly suffered during prostitution and should be appropriately named.
Method: Using standardized measures and including a new measure of torture, we interviewed 45 women in the United States about their torture experiences in prostitution and their symptoms of PTSD, dissociation, childhood trauma, health status …

This case study investigates the use of Large Language Models (LLMs) and prompt engineering techniques for data augmentation,in the context of post-trial inter- view data (low-resource natural language data) from a randomized clinical trial on Pain Reprocessing Therapy (PRT) for chronic back pain. The limited sample size from the original study necessitates the generation of synthetic interview samples, needed to draw meaningful inferences from the data and to further extend the study. Application of the physical laws of energy and mass conservation at the whole-body level is not necessarily informative about causal mechanisms of weight gain and the development of obesity. The energy balance model (EBM) and the carbohydrate–insulin model (CIM) are two plausible theories, among several others, attempting to explain why obesity develops within an overall common physiological framework of regulation of human energy metabolism. INTRODUCTION: During the COVID-19 pandemic, a medical institution in the United States implemented a virtual clerkship rotation to replicate the skills that students would develop during an in-person clinical site rotation. The primary objective of this study was to identify the efficacy and benefits following the implementation of the Virtual Hospital Experiential Learning (VHEL) model, as demonstrated by increased perceived confidence and skill growth among medical students.

Drawing on a comprehensive review of existing literature and empirical studies, the research highlights the critical developmental milestones in social-emotional learning (SEL) during the early years and explore effective strategies for fostering these skills within various contexts, including home, preschool, and other early learning settings. Students with special needs across the country and the world continue to struggle to gain functional language and communication skills, and build a vocabulary to use throughout life. Research shows that augmentative and alternative communication or speech-generating devices can be a special, useful, and powerful tool for students' language skills. Purpose of Review: Entering pregnancy with obesity increases the risk of adverse health outcomes for parent and child. As such, research interventions are largely focused on limiting excess gestational weight gain during pregnancy, especially in those with obesity. Yet, while many lifestyle interventions are successful in reducing GWG, few affect pregnancy outcomes. Here we review work targeting the metabolic milieu instead of focusing solely on weight. Recent Findings: Work done in non-pregnant populations suggests that specifically targeting glucose, triglyceride, and leptin levels or inflammatory makers improves the metabolic milieu and overall health. Seasonal variability in environmental conditions is a strong determinant of animal migrations, but warming temperatures associated with climate change are anticipated to alter this phenomenon with unknown consequences. We used a 40-year fishery-independent survey to assess how a changing climate has altered the migration timing, duration and first-year survival of juvenile bull sharks (Carcharhinus leucas). From 1982 to 2021, estuaries in the western Gulf of Mexico (Texas) experienced a mean increase of 1.55°C in autumn water temperatures, and delays in autumn cold fronts by ca. 0.5 days per year. Quantitative adverse outcome pathways (qAOPs) describe the response–response relationships that link the magnitude and/or duration of chemical interaction with a specific molecular target to the probability and/or severity of the resulting apical-level toxicity of regulatory relevance. The present study developed the first qAOP for latent toxicities showing that early life exposure adversely affects health at adulthood. Specifically, a qAOP for embryonic activation of the aryl hydrocarbon receptor 2 (AHR2) of fishes by polycyclic aromatic hydrocarbons (PAHs) leading to decreased fecundity of females at adulthood was developed by building on existing qAOPs for (1) activation of the AHR leading to early …

Triploid oysters are commonly used as the basis for production in the aquaculture of eastern oysters along the USA East and Gulf of Mexico coasts. While they are valued for their rapid growth, incidents of triploid mortality during summer months have been well documented in eastern oysters, especially at low salinity sites. We compared global transcriptomic responses of diploid and triploid oysters bred from the same three maternal source populations at two different hatcheries and outplanted to a high (annual mean salinity = 19.4 ± 6.7) and low (annual mean salinity = 9.3 ± 5.0) salinity site.
