Alternatively Spliced Map4 Isoforms Have Key Roles In Maintaining Microtubule Organization And Skeletal Muscle Function, Lathan Lucas, Larissa Nitschke, Brandon Nguyen, James A Loehr, George G Rodney, Thomas A Cooper
Faculty, Staff and Students Publications
Skeletal muscle cells (myofibers) are elongated non-mitotic, multinucleated syncytia that have adapted a microtubule lattice. Microtubule-associated proteins (MAPs) play roles in regulating microtubule architecture. The most abundant MAP in skeletal muscle is MAP4. MAP4 consists of a ubiquitous MAP4 isoform (uMAP4), expressed in most tissues, and a striated-muscle-specific alternatively spliced isoform (mMAP4) that includes a 3,180-nucleotide exon (exon 8). To determine the role of mMAP4 in skeletal muscle, we generated mice that lack mMAP4 and express only uMAP4 due to genomic deletion of exon 8. We demonstrate that loss of mMAP4 leads to disorganized microtubule architecture and intrinsic loss of …

Curcumin Reduces Malondialdehyde And Improves Antioxidants In Humans With Diseased Conditions: A Comprehensive Meta-Analysis Of Randomized Controlled Trials, Mohammad Alizadeh, Sorayya Kheirouri
BioMedicine
Objective: This systematic review and meta-analysis was conducted to collate the effects of curcumin on MDA and antioxidant markers in individuals with diseased conditions. In this study the research question was “does curcumin supplementation improves oxidative stress and antioxidant defense enzymes in human subjects compared to a group without curcumin supplementation?
Methods: This research included randomized controlled trials published in English in any year, in which intervention with curcumin was compared to either placebo, or standard of care or no intervention. Pubmed, Embase, Cochrane Central, Scopus and Google Scholar were searched. Meta-analysis was performed using RevMan (version 5.3), with standardized …

BioMedicine
Colorectal cancer (CRC) is distinguished by epigenetic elements like DNA methylation, histone modification, histone acetylation and RNA remodeling which is related with genomic instability and tumor initiation. Correspondingly, as a main epigenetic regulation, DNA methylation has an impressive ability in order to be used in CRC targeted therapy. Meaningly, DNA methylation is identified as one of most important epigenetic regulators in gene expression and is considered as a notable potential driver in tumorigenesis and carcinogenesis through gene-silencing of tumor suppressors genes. Abnormal methylation situation, even in the level of promoter regions, does not essentially change the gene expression levels, particularly …

The Rna Degradation Pathway Is Involved In Pparα-Modulated Anti-Oral Tumorigenesis, Nai-Wen Chang, Yi-Ping Huang
BioMedicine
Background: The activation of peroxisome proliferator-activated receptor alpha (PPARα) has been shown to reprogram tumor metabolism and exhibits great potential for treating anti-oral tumorigenesis.
Methods: In this study, we used a pathway-based strategy to explore possible functional pathways involved in the anticancer activity of PPARα in oral cancer cells through next-generation sequencing (NGS) and bioinformatic approaches.
Results: We found that 3919 genes were upregulated and 1060 genes were downregulated through PPARα activation. These genes were mainly involved in the proteasomal, mRNA surveillance, spliceosomal, RNA transport, and RNA degradation pathways, as indicated by GO and KEGG enrichment analysis. Importantly, a total …

Oral Acute And Sub-Acute Toxic Effects Of HydroalcoholicTerminalia ChebulaRetz AndAchillea WilhelmsiiExtracts In Balb/C Mice, Mahnaz Jafari, Kourosh Manochehri Naeini, Zahra Lorigooini, Rasool Namjoo
BioMedicine
Background: This study examined the acute and sub-acute toxic effects ofTerminalia chebulaandAchillea wilhelmsiiextracts on the murine model.
Methods: In both phases, mice were assigned to intervention and control groups. At the end of study, the liver, kidney, and heart tissues were collected for histopathological studies.
Results: In the acute phase of the study, the safe dose was ≤5000 mg/kg for both extracts. In sub-acute phase, LD50 (95% CI) ofAchillea wilhelmsiiextract was determined ≥5000 mg/kg and that ofTerminalia chebulaextract 2754.436 (2438-3114) mg/kg. The highest dose ofT. chebulaextract induced few histopathological changes.
Conclusion: It will be useful to gain information on the …

The Potential Therapeutic Effect For Melatonin And Mesenchymal Stem Cells On Hepatocellular Carcinoma, Yasser Mohamed, Mohamed A. Basyony, Nabila I. El-Desouki, Walied S. Abdo, Mohammed A. El-Magd
BioMedicine
Background/aim: Herein, we investigated the potential therapeutic effect of Melatonin (Mel) and/or mesenchymal stem cells (MSCs) on rat model of HCC.
Materials and Methods: Female mature rats were divided into 5 groups (n= 10/group): normal (Nor), HCC group intraperitoneally injected with 200 mg/kg DEN, and 3 treated groups; HCC + Mel (Mel) group given Mel intraperitoneally 20 mg/kg, twice a week, HCC + MSCs (MSCs) group intravenously injected by 1 × 106 cells, and HCC + MSCs (Mel +MSCs) group.
Results: Rats in HCC group showed most deteriorated effect in form of increased mortality and relative liver weight, elevated …

Development Of Broad Spectrum Mycosporine Loaded Sunscreen Formulation From Ulva Fasciata Delile, Saurabh Bhatia, Satish Sardana, Ajay Sharma, Celia Bertha Vargas De La Cruz, Bhupal Chaugule, Laleh Khodaie
BioMedicine
Ba.ckground: Sunscreen formulations primarily offer protection against UV induced damages however nowadays it also maintains skin natural physiological conditions. Current global market is flooded with numerous sunscreen products which offer protection to skin against several UV induced damages. However most of these sunscreen formulations offers narrow spectrum protection against UV and also suffer from stability as well as toxicity related issues.
Methods: Present work aims to isolate mycosporine amino acid (Mgy) from green alga namelyUlva fasciata (U. fasciata)and study its sunscreen potential against widely used domestic marketed formulation. Stability evaluations were also performed for almost 90 days.
Results: Results demonstrated …

BioMedicine
Background: Protective effects ofGynura bicoloraqueous extract (GAE) at three concentrations upon nerve growth factor (NGF) differentiated-PC12 cells against H2O2induced injury were examined.
Methods: NGF differentiated-PC12 cells were treated with GAE at 0.25%, 0.5% or 1%. 100 μM H2O2was used to treat cells with GAE pre-treatments. After incubating at 37 °C for 12 hr, experimental analyses were processed.
Results: H2O2exposure decreased cell viability, increased plasma membrane damage, suppressed Bcl-2 mRNA expression and enhanced Bax mRNA expression. GAE pre-treatments reversed these changes. H2O2exposure reduced mitochondrial membrane …
