Distinct Odorant Receptor Response Patterns To Aliphatic Odorants In Freely Behaving Mice, Claire A. De March, Patrick Breheny, William B. Titlow, Hiroaki Matsunami, Timothy S. Mcclintock

Physiology Faculty Publications

In mammals, odors are encoded by a combinatorial code determined by the pattern of responses across hundreds of odorant receptors expressed monogenically and monoallelically in olfactory sensory neurons. The compositions of these receptor response patterns are largely unknown and overlap between them has yet to be explored. Activity-dependent reporter gene expression in freely behaving S100a5-tauGFP mice allowed capture of activated olfactory sensory neurons and identified 168 receptors responsive to moderate concentrations of 1 or more of 12 aliphatic (5 to 8 carbons) ketones, alcohols, and carboxylic acids. These 12 response patterns are remarkably different, with only 19% of the receptors …

Feeding- And Light-Cycle Synergistically Regulate Mouse Blood Pressure Daily Rhythm Via Bmal1-Dependent And Independent Mechanisms, Tianfei Hou, Wen Su, Aaron Chacon, An-Hsuan Lin, Zhenheng Guo, Ming C. Gong

Physiology Faculty Publications

Cardiovascular health requires the orchestration of the daily rhythm of blood pressure (BP), which responds to changes in light exposure and dietary patterns. Whether rhythmic light and feeding can modulate daily BP rhythm directly or via modulating intrinsic core clock gene Bmal1 is unknown. Using inducible global Bmal1 knockout mice (iBmal1KO), we explored the impact of rhythmic light, rhythmic feeding, or their combination on various physiological parameters. Daily rhythms of BP, heart rate, and locomotor activity were monitored via radiotelemetry, while food intake patterns were tracked using the BioDAQ system. Respiratory exchange ratio (RER) and energy expenditure (EE) were assessed …

Prosecutorial Reform And The Myth Of Individualized Enforcement, Justin Murray Jan 2025

Prosecutorial Reform And The Myth Of Individualized Enforcement, Justin Murray

Articles & Chapters

The American prosecutor’s legitimacy faces unprecedented challenges. A new wave of reformist prosecutors has risen to power promising to transform the criminal justice system from within, sparking fierce backlash from defenders of the prosecutorial status quo. Central to this conflict is a debate over the nature of prosecutorial discretion, influenced by a set of claims and assumptions that this Article terms the myth of individualized enforcement. This myth posits that prosecutors base discretionary decisions on case-specific facts and equitable circumstances rather than generalizable criteria or categorical nonenforcement practices, such as the policies some reformist prosecutors have adopted that disfavor prosecuting …

Physiology Faculty Publications

Abstract: Exercise is a potent stimulus for combatting skeletal muscle ageing. To study the effects of exercise on muscle in a preclinical setting, we developed a combined endurance–resistance training stimulus for mice called progressive weighted wheel running (PoWeR). PoWeR improves molecular, biochemical, cellular and functional characteristics of skeletal muscle and promotes aspects of partial epigenetic reprogramming when performed late in life (22–24 months of age). In this investigation, we leveraged pan-mammalian DNA methylome arrays and tandem mass-spectrometry proteomics in skeletal muscle to provide detailed information on late-life PoWeR adaptations in female mice relative to age-matched sedentary controls (n = 7–10 …

Nonresolving Neuroinflammation Regulates Axon Regeneration In Chronic Spinal Cord Injury, Andrew N. Stewart, Christopher C. Bosse-Joseph, Reena Kumari, William M. Bailey, Kennedy A. Park, Victoria K. Slone, John C. Gensel Jan 2025

Nonresolving Neuroinflammation Regulates Axon Regeneration In Chronic Spinal Cord Injury, Andrew N. Stewart, Christopher C. Bosse-Joseph, Reena Kumari, William M. Bailey, Kennedy A. Park, Victoria K. Slone, John C. Gensel

Physiology Faculty Publications

Chronic spinal cord injury (SCI) lesions retain increased densities of microglia and macrophages. In acute SCI, macrophages induce growth cone collapse and facilitate axon retraction away from lesion boundaries. Little is known about the role of sustained inflammation in chronic SCI or whether chronic inflammation affects regeneration. We used the colony-stimulating factor-1 receptor inhibitor, PLX-5622, to deplete microglia and macrophages months after complete crush SCI in female mice. Transcriptional analyses revealed a significant inflammatory depletion within chronic SCI lesions after PLX-5622 treatment. Both transcriptional analyses and immunohistochemistry revealed that Iba1+ cells repopulate to predepleted densities after treatment removal. Neuronal-enriched …

Physiology Faculty Publications

Significance: Cerebral blood flow (CBF) imaging is crucial for diagnosing cerebrovascular diseases. However, existing large neuroimaging techniques with high cost, low sampling rate, and poor mobility make them unsuitable for continuous and longitudinal CBF monitoring at the bedside. Aim: We aimed to develop a low-cost, portable, programmable scanning diffuse speckle contrast imaging (PS-DSCI) technology for fast, high-density, and depth-sensitive imaging of CBF in rodents. Approach: The PS-DSCI employed a programmable digital micromirror device (DMD) for remote line-shaped laser (785 nm) scanning on tissue surface and synchronized a 2D camera for capturing boundary diffuse laser speckle contrasts. New algorithms were developed …

Differential Effects Of Myosin Activators On Myocardial Contractile Function In Nonfailing And Failing Human Hearts, Joohee Choi, Patrick T. Wood, Joshua B. Holmes, Katherine L. Dominic, Cristobal G. Dos Remedios, Kenneth S. Campbell, Julian E. Stelzer

Physiology Faculty Publications

The second-generation myosin activator danicamtiv (DN) has shown improved function compared with the first-generation myosin activator omecamtiv mecarbil (OM) in nonfailing myocardium by enhancing cardiac force generation but attenuating slowed relaxation. However, whether the functional improvement with DN compared with OM persists in remodeled failing myocardium remains unknown. Therefore, this study aimed to investigate the differential contractile responses to myosin activators in nonfailing and failing myocardium. Mechanical measurements were performed in detergent-skinned myocardium isolated from donor and failing human hearts. Steady-state force, stretch activation responses and loaded shortening velocity were analyzed at submaximal [Ca2+] in the absence or presence of …

Background: Most tumors initially respond to treatment, yet refractory clones subsequently develop owing to resistance mechanisms associated with cancer cell plasticity and heterogeneity. Methods: We used a chemical biology approach to identify protein targets in cancer cells exhibiting diverse driver mutations and representing models of tumor lineage plasticity and therapy resistance. An unbiased screen of a drug library was performed against cancer cells followed by synthesis of chemical analogs of the most effective drug. The cancer subtype target range of the leading drug was determined by PRISM analysis of over 900 cancer cell lines at the Broad Institute, MA. RNA-sequencing …

Sex-Based Differences In The Long-Term Fate Of Hippocampal Neurons Born After A Traumatic Brain Injury, Hannah C. Downing, Ashley B. Glover, Jessica E. Gebhardt, Katherine L. Thompson, Kathryn E. Saatman Jan 2025

Sex-Based Differences In The Long-Term Fate Of Hippocampal Neurons Born After A Traumatic Brain Injury, Hannah C. Downing, Ashley B. Glover, Jessica E. Gebhardt, Katherine L. Thompson, Kathryn E. Saatman

Physiology Faculty Publications

Introduction: Moderate-to-severe traumatic brain injury (TBI) results in an early loss of immature hippocampal granule cells and the activation of typically quiescent neural stem cells (NSCs) in the dentate gyrus. Activation of NSCs leads to a robust increase in proliferation and generation of neural progenitor cells (NPCs), supporting restoration of the immature neuron population of over a period of 1–2 weeks. However, it is unclear if neurons born early after injury develop normally, survive long-term and functionally integrate into the hippocampal network. Although adult hippocampal neurogenesis is regulated in a sex-dependent manner, the majority of pre-clinical TBI studies lack the …

Skeletal Muscle Stem Cell Mitochondria Are Transferred To Muscle Fibers In Response To A Hypertrophic Stimulus, Jensen Goh, Julia G. Williams, Sarah E. Ogle, Jai K. Joshi, Logan N. Scott, Benjamin Burke, Alex R. Keeble, Nicholas T. Thomas, Christopher S. Fry, Ahmed Ismaeel, John J. Mccarthy

Physiology Faculty Publications

The fusion of skeletal muscle stem cell (MuSC) to myofibers during hypertrophy has exclusively focused on the transfer of the MuSC nucleus, leaving the fate of other MuSC organelles, such as mitochondria, largely unexplored. The objective of this study was to determine whether MuSCs transfer their mitochondria upon myofiber fusion in response to a hypertrophic stimulus. To achieve this goal, we specifically labeled MuSC mitochondria with Dendra2 fluorescence by crossing the MuSC-specific CreER (Pax7CreER/CreER) mouse with the Rosa26-Dendra2 mouse to generate the Pax7-Dendra2 mouse. To induce the fusion of MuSC to myofibers, Pax7-Dendra2 mice underwent synergist ablation surgery …

Seahorsedad: A Study Of Metaphors In Transmasculine Birther’S Instagram Posts, Alexa Anne Runnion Jan 2025

Seahorsedad: A Study Of Metaphors In Transmasculine Birther’S Instagram Posts, Alexa Anne Runnion

Graduate Student Theses, Dissertations, & Professional Papers

Dominant notions of pregnancy frame gestation as a solely female process. However, the experiences of transmasculine birthers challenge this assumption, demonstrating that pregnancy is not inherently tied to womanhood but is instead shaped by social and cultural narratives and representations. This paper explores the unique pregnancy and parental exercises of transmasculine birthers by examining public social media posts under the hashtag #seahorsedad on Instagram. Utilizing metaphorical criticism, I aim to uncover how transmasculine birthers, known within the community as “seahorse dads,” both uphold and resist traditional notions of gender, pregnancy, and parenting through a combination of textual and visual metaphor. …

Increasing Nature Connection And Decreasing Climate Change Distress In An Indigenous Population: The Development Of A Traditional Ecological Knowledge Intervention Framework, Olathe Antonio Jan 2025

Increasing Nature Connection And Decreasing Climate Change Distress In An Indigenous Population: The Development Of A Traditional Ecological Knowledge Intervention Framework, Olathe Antonio

Graduate Student Theses, Dissertations, & Professional Papers

Research suggests that nature connection is positively correlated with pro-environmental behaviors, concern for the climate, general wellbeing, and subjective levels of climate change distress. Few research studies have examined how to increase nature connectedness without increasing psychological distress. Such research is especially important for those who are disproportionately affected by climate change such as Indigenous Peoples. To address this discrepancy, we developed a culturally-grounded psychological intervention framework based in traditional ecological knowledge (TEK) for Indigenous populations and offered an adapted example for a Navajo population. The intervention was created in collaboration with TEK experts to conceptualize key psychologically-relevant TEK principles, …

Indigenous Placemaking, Colonial Placetaking, And Indigenous Data Sovereignty: Implications Of Environmental Data Collection On Tribal Lands, Paige Nizhonii Kya'iyo Johnson Jan 2025

Indigenous Placemaking, Colonial Placetaking, And Indigenous Data Sovereignty: Implications Of Environmental Data Collection On Tribal Lands, Paige Nizhonii Kya'iyo Johnson

Graduate Student Theses, Dissertations, & Professional Papers

This project investigates how Indigenous Data Sovereignty protocols apply to environmental data collected on Tribal lands by federal and state entities. The 2011 and 2014 flooding of the Missouri River and a need for further soil moisture data led the United States Army Corps of Engineers Omaha District to implement a Mesonet weather station monitoring network across the Upper Missouri River Basin (UMRB). Historically, the Army Corps of Engineers has not included Indigenous perspectives into their decision-making, and this has caused Tribes to be distrustful of partnerships with them. The UMRB monitoring network will span across the Upper Missouri River …

Hormonal Decay After Death: Evaluating Dhea-S As A Potential Molecular Marker For Postmortem Estimation, Marisa Lydia Novak Jan 2025

Hormonal Decay After Death: Evaluating Dhea-S As A Potential Molecular Marker For Postmortem Estimation, Marisa Lydia Novak

Graduate Student Theses, Dissertations, & Professional Papers

Post-mortem interval (PMI) estimation is a central component of forensic anthropological investigation, but traditional methods for estimating time since death lose accuracy after ~72 hours. Recent research indicates that some steroid hormones may undergo predictable degradation patterns after death, offering potential use as biochemical clocks for PMI estimation beyond this threshold. Dehydroepiandrosterone (DHEA), and its sulfate form (DHEA-S) are of particular interest due to their metabolic stability, systemic influence, and association with trauma. This study investigates whether DHEA-S hormonal degradation patterns correlate with time elapsed since death and body temperature, evaluating their potential as biochemical PMI markers. 160 mL of …

The P3 Event-Related Potential (Erp) In Visual And Auditory Tasks: Implications For Executive Functioning In Adhd Preadolescent Children, Alex Visal Seng Jan 2025

The P3 Event-Related Potential (Erp) In Visual And Auditory Tasks: Implications For Executive Functioning In Adhd Preadolescent Children, Alex Visal Seng

Open Access Master's Theses

Attention-Deficit/Hyperactivity Disorder (ADHD) is a widespread psychological disorder in children, poorly impacting executive functioning domains of inhibitory control and working memory. Event-related potentials (ERPs) provide neuroelectric insight into these cognitive processes. However, most ERP research focuses on one task type, limiting direct comparisons of cognitive demands across different task paradigms. This study examined P3 amplitude and latency in ADHD children using a visual NoGo task (response inhibition) and an auditory oddball task (working memory), and associations between the P3 ERP and behavioral measures of executive functions.

Cross-sectional data of symptomatic ADHD children (N=13; F=5; aged 9.15 ± 1.86 years) were …

The Bodhi App: A Post-Trauma Self-Regulation App For People With Intellectual And Developmental Disabilities, Liam Cannon

Open Access Master's Theses

People in the United States living with intellectual and developmental disabilities (I/DD) often experience trauma at a disproportionate rate. Due to societal and personal barriers, individuals with I/DD have problems with gaining access to therapy, thus often having to live with the negative effects of their untreated trauma. The effects of untreated trauma can cause severe impairment which affects a person’s quality of life and emotional, social, and physical development.

Post-trauma self-regulation is a way of doing activities to regulate one’s emotions to help cope with the negative effects of trauma. It has been shown as an effective method of …

From Lawns To Life: Landscaping For Biodiversity On Mason’S Island, Margit Burgess Jan 2025

From Lawns To Life: Landscaping For Biodiversity On Mason’S Island, Margit Burgess

Open Access Master's Theses

The collapse of biodiversity is an existential threat to life on Earth. Residential landscapes, dominated by open lawns maintained in turfgrass, do not support biodiversity. In the United States, turfgrass has replaced native landscapes in over 40 million acres, with 500 square miles added each year (Tallamy, 2019). Of the 78% of the U.S. land that is privately owned, approximately 135 million acres - nearly twice the size of all national parks combined - are residential (Milesi et al., 2005). With a shift in private landscape management, this land could present a largely untapped opportunity for biodiversity restoration and social …

Investigating The Effectiveness Of Best Management Practices In Reducing Fecal Bacteria In Narrow River, Rhode Island, Katherine Thompson Jan 2025

Investigating The Effectiveness Of Best Management Practices In Reducing Fecal Bacteria In Narrow River, Rhode Island, Katherine Thompson

Open Access Master's Theses

Fecal bacterial contamination shapes how communities manage and use estuarine waters, with consequences for ecosystem health and public access. In Rhode Island's Narrow River, elevated concentrations of fecal coliforms and enterococci have restricted shellfishing for decades despite ongoing management efforts. From 2004 to 2018, local organizations implemented Best Management Practices (BMPs) to address water quality challenges, yet their effectiveness remains incompletely understood. This study analyzes three decades of monitoring data using linear regression, generalized additive models, and Before After Control Impact methods to evaluate BMP effectiveness and identify key drivers of contamination. Linear regression revealed spatially variable trends, with bacterial …

The Coral Carousel: A Device And Method For In-Situ Propagation Of Deep-Sea Corals, Gregory Bales Jan 2025

The Coral Carousel: A Device And Method For In-Situ Propagation Of Deep-Sea Corals, Gregory Bales

Open Access Master's Theses

The research described herein covers the development of a tool station for performing in-situ propagation of corals using a work class ROV. This includes a system for manipulating coral fragments and affixing them to a cement base plug. To validate this tool station, testing was performed both in the lab as well as at depth in the Gulf of Mexico. As restoration of shallow-water corals has grown in popularity, many techniques have been developed for propagation. However, it is difficult or inappropriate to directly apply these techniques to deep-sea corals. While some forms of diving are capable of approaching the …

Open Access Master's Theses

Artemis missions are set to send the next man and the first woman back to the moon and enable long-term missions on the lunar surface. However, exposure to lunar gravity has significant physiological effects characterized by loss of muscle mass, weakness and decreased bone density. Nutraceutical countermeasures such as Resveratrol (RSV), a naturally occurring polyphenol, have been identified as promising strategies to preserve musculoskeletal health; however, its pleiotropic effect hinders our ability to identify the primary cellular pathways through which it delivers its benefits. Furthermore, its efficacy in simulated lunar gravity is unknown. Based on the benefits observed with RSV …

Unconditional-To-Conditional Transfer And Optimization For Web-Based Skybox Gan, Crystal Kwong Jan 2025

Unconditional-To-Conditional Transfer And Optimization For Web-Based Skybox Gan, Crystal Kwong

Master's Projects

Generative adversarial networks (GANs) are known for their ability to generate high quality
images mimicking real life or even particular art styles. Yet for all their capability, casually
training a GAN on an average machine can be infeasible as GANs require an enormous amount
of time and data to train. Even with a trained GAN, model inference demands heavy
computations, making GANs difficult to deploy on applications. To address these limitations,
techniques such as transfer learning and quantization have been leveraged to speed up training of
GANs and lighten computational cost of GAN inference. This project aims to use such …

Effects Of Data Augmentation On Sponge Identification Using Computer Vision Models, George Ku Jan 2025

Effects Of Data Augmentation On Sponge Identification Using Computer Vision Models, George Ku

Master's Projects

Coral reefs can be primarily found in tropical and sub-tropical regions of our oceans, providing a thriving habitat for millions of species. Marine sponges, which can be frequently found in coral reefs, play a critical role that contributes to the maintenance of these ecosystems, including the recycling of nutrients through water filtration. However, rising ocean temperatures and acidification due to climate change have resulted in the bleaching and death of coral reefs worldwide. In order to preserve these reefs and the sponges that depend on them, scientists have been performing studies on their biodiversity. This includes collecting numerous images of …

Phishing Detection Using Continual Learning And Large Language Models, Gopi Prajeev Battula Jan 2025

Phishing Detection Using Continual Learning And Large Language Models, Gopi Prajeev Battula

Master's Projects

Adaptive phishing detection remains crucial as the nature of cyber-attacks changes over time, which renders static models obsolete. This project extends phishing detection through the implementation of continual learning approaches, namely Elastic Weight Consolidation (EWC) and Learning Without Forgetting (LWF) with RoBERTa, a Large Language Model (LLM) and compares the results of these approaches against GPT-4o-mini, another LLM. Our approach begins with fine-tuning RoBERTa on multiple phishing datasets to establish an effective baseline. EWC is then implemented to preserve vital model parameters based on their importance measured by the Fisher Information Matrix, while LWF uses knowledge distillation to retain prior …
