Nikethamide, N,N-diethyl-3-pyridinecarboxamide (Coramine®) is a central nervous system stimulant at one time used therapeutically in human medicine but no longer marketed in the US. Nikethamide is classified a Drug Class 1, Penalty Class A substance by the Association of Racing Commissioners International (ARCI) and as a Horseracing Integrity and Safety Authority (HISA) SO or banned Stimulant that lacks FDA approval. Identifications of Nikethamide or its metabolite N-Ethylnicotinamide in racing samples can result in significant penalties in horse racing. Nikethamide is rapidly metabolized to N-Ethylnicotinamide, which remains detectable in plasma and urine leading to its use as a regulatory “marker” of …

Host Defense Inside & Outside Of The Immune System: Modeling Antimicrobial Immunity By Gbps, Zhongyu Yuan Jan 2025

Host Defense Inside & Outside Of The Immune System: Modeling Antimicrobial Immunity By Gbps, Zhongyu Yuan

Yale Graduate School of Arts and Sciences Dissertations

Traditional views of host defense often focus on antibody-mediated neutralization, cytotoxic T-cell plus NK-cell-mediated killing, and phagocytic activities by cell populations originating in the bone marrow as part of the classical immune system. Additional effector activities, however, also operate in non-immune cells which compose the stroma or parenchyma of host tissues. These latter cell populations often mobilize antimicrobial activities in response to cytokines such as type II IFN (IFN-gamma; IFN-) against a wide variety of intracellular pathogens.

Among the most prominent IFN--stimulated genes are a family of immune dynamin-like GTPases termed the Guanylate Binding Proteins (GBPs). GBPs are widely expressed …

Department of Otolaryngology (ENT) Faculty Publications

Introduction

Tympanoplasty and mastoidectomy are common procedures performed by otolaryngologists that can result in complications for which patients may seek compensation. Medical malpractice case analyses may offer insight into how clinicians can avoid risk and improve patient satisfaction. We aimed to comprehensively characterize litigation after mastoidectomies and tympanoplasties in the United States.

Methods

The Westlaw Campus Research legal database was searched for all available court decisions associated with claims of medical malpractice after tympanoplasty and/or mastoidectomy in the United States between 1975 and 2022. Information on the plaintiffs' relationships to the patients, patient characteristics, states where the procedures took place, …

A Comparison Of Allergen Sensitization Profiles In Patients With Chronic Rhinosinusitis With And Without Nasal Polyposis, Lauren Trzcinski, Suhas Bharadwaj, Randall A. Bloch, Joseph K. Han, Kent K. Lam

Department of Otolaryngology (ENT) Faculty Publications

Chronic rhinosinusitis (CRS) and allergic rhinitis (AR) are common comorbid sinonasal conditions. CRS is classically divided into two distinct phenotypes: CRS with nasal polyposis (CRSwNP) and CRS without nasal polyposis (CRSsNP). The purpose of this retrospective observational study is to determine whether aeroallergen sensitization profiles in patients with comorbid CRS and AR can distinguish between CRSwNP and CRSsNP. A total of 241 patients diagnosed with comorbid CRS and AR who underwent skin prick testing or in vitro allergy testing in a single tertiary rhinology practice were included for evaluation. The rates of allergen-specific sensitizations in CRSwNP patients were compared with …

Sensor Fusion For Enhancing Motion Capture : Integrating Optical And Inertial Motion Capture Systems, Hailey Hicks

Theses

This study aimed to create and evaluate a custom sensor fusion algorithm based on Gauss-Newton optimization that combines Optical Motion Capture (OMC) and Inertial Motion Capture (IMC) measurements to combat the limitations of solely using each system. The goal was to demonstrate how inertial measurement unit (IMU) data can be seamlessly integrated into motion capture data to provide a more efficient and reliable gap filling process for future research. The algorithm takes the first and last frame of OMC data, as quaternions, and fills the rest with IMU gyroscope orientation data to simulate gaps of up to ten minutes. The …

Factors Affecting Freshwater Fish Communities In Two North Alabama Counties, Joshua Fitts Jan 2025

Factors Affecting Freshwater Fish Communities In Two North Alabama Counties, Joshua Fitts

Theses

This study examined how physical habitat characteristics influence freshwater fish diversity in streams of Madison and Limestone Counties, Alabama. Ten 150-meter stream reaches were sampled using backpack electrofishing and seining, with habitat variables measured at fixed transects. Principal component analysis identified four main habitat gradients representing flow and substrate, channel size, gravel dominance, and canopy–bedrock conditions. Although overall species richness and Shannon diversity were not significantly related to these gradients, species-specific correlations revealed clear ecological preferences. Regression models showed that root wads, herbaceous vegetation, and riprap modestly increased richness, emphasizing the role of structural diversity. These findings highlight the importance …

Potentiation Of Gelonin Cytotoxicity By Pulsed Electric Fields, Olga N. Pakhomova, Eleni Zivla, Giedre Silkuniene, Mantas Silkunas, Andrei G. Pakhomov

Bioelectrics Publications

Gelonin is a ribosome-inactivating protein with extreme intracellular toxicity but poor permeation into cells. Targeted disruption of cell membranes to facilitate gelonin entry is explored for cancer and tissue ablation. We demonstrate a hundreds- to thousands-fold enhancement of gelonin cytotoxicity by pulsed electric fields in the T24, U-87, and CT26 cell lines. The effective gelonin concentration to kill 50% of cells (EC₅₀) after electroporation ranged from <1 nM to about 100 nM. For intact cells, the EC₅₀ was unattainable even at the highest gelonin concentration of 1000 nM, which reduced cell survival by only 5–15%. For isoeffective electroporation treatments using 300 ns, 9 µs, and 100 µs pulses, longer pulses were more efficient at lowering gelonin EC₅₀. Increasing the electric field strength of 8, 100 µs pulses from 0.65 to 1.25 kV/cm reduced gelonin EC₅₀ from 128 nM to 0.72 nM. Conversely, the presence of 100 nM gelonin enabled a more than 20-fold reduction in the number of pulses required for equivalent cell killing. Pulsed electric field-mediated delivery of gelonin shows promise for hyperplasia ablation at concentrations sufficiently low to minimize or avoid systemic toxicity.

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has caused a global health crisis, marked by high transmissibility and virulence. Despite widespread vaccination efforts, effective treatments for COVID-19, particularly for severe cases leading to Acute Respiratory Distress Syndrome (ARDS), remain limited. This study investigates the efficacy of KVX-053, a protein tyrosine phosphatase type IVA (PTP4A3) small molecule inhibitor, in modulating SARS-CoV-2-induced inflammation and lung injury using in vitro cell models and in vivo K18-hACE2 transgenic mice. KVX-053 reduced in vitro pro-inflammatory cytokine expression, including TNFα, CXCL10, and CXCL11, without impacting viral replication or cell viability. K18-hACE2 mice treated with KVX-053 demonstrated …

Jaguar (Panthera Onca) Population Status And Conservation Needs In Panama Revealed Using Spatial Density And Occupancy Models., Rebeca E. Becdach

Cal Poly Humboldt theses and projects

Jaguars (Panthera onca) are key predators and the largest felid in the Americas. The species is listed as near threatened globally due to habitat loss, fragmentation, and human persecution. Their population decline can negatively affect the structure and functioning of the ecosystems they inhabit. Conservation efforts for jaguars and their habitat require understanding jaguar populations and how they are impacted by human presence across their range. This information is lacking in Panama, a critical portion of the Jaguar range. Panama forms a narrow land bridge connecting Central and South America, integral to wildlife movement, including jaguars, across both …

Altrenogest is a progestogen substance used in equine medicine for the synchronization of ovulation during the spring breeding season and the control of estrus in mares. It accomplishes its actions by down-regulation of endometrial progesterone receptors and repression of estrogen receptor transcriptional activation. However, various groups have also measured androgen receptor binding activity for altrenogest in vitro; these findings combined with androgenic and anabolic properties roughly 1/20 in potency of testosterone and its structural similarity to the anabolic steroid trenbolone have created concerns among racing authorities of altrenogest’s potential abilities to alter the outcome of equine athletic performance, even though …

State Constitutional Development And The Criminal Procedure Revolution, Quinn Yeargain

Georgia Law Review

Questions about which rights, liberties, and guarantees are included in the U.S. Constitution’s protections, and how they are protected, have long produced animated debate among scholars, judges, and advocates. Yet, while today’s debates may seem familiar, they take place at a perilous time in our country’s trajectory. We are six decades removed from the Warren Court’s criminal procedure revolution, and just a few years into the Roberts Court’s full-throated embrace of originalist jurisprudence. Whether many of the protections we have come to know and recognize from the Warren Court—the exclusionary rule, Miranda warnings, and Gideon’s mandate—are subjected to re-evaluation …

Loss Of Lgl1 Increases Expression Of C-Jun And Dcx In The Svz And Rms Of The Mouse Brain, Madison Leigh Gutierrez

Cal Poly Humboldt theses and projects

Glioblastoma (GBM), the most aggressive and treatment-resistant subtype of glioma, accounts for the majority of brain tumor–related mortality. A defining feature of GBM is its ability to invade healthy brain tissue through extensive cell migration, which severely limits the effectiveness of surgical and pharmacological treatments. Increasing evidence suggests that GBM cells activate gene programs known to drive normal neural stem and progenitor cell (NSPC) migration; however, the precise molecular mechanisms shared between NSPCs and GBM cells remain poorly understood. Lethal (2) giant larvae 1 (Lgl1), a conserved cortical cytoskeletal protein, has emerged as a key regulator of polarity and migration. …

Artificial Intelligence (AI) is crucial to numerous functions required for driving automation systems, including the computer vision techniques used to detect the roadway environment and make real-time decisions. However, the images used as inputs to the AI system may be maliciously perturbed, or manipulated, causing the AI system to make an incorrect classification. In this study, we examined humans’ perception of the AI’s computer vision capability of classifying various road sign images, including the original images, images with two different types of malicious attacks, and images that are scrambled randomly at the pixel level. Our results showed that participants rated …
