Extracellular vesicles (EVs) are generated by all cells. Systemic administration of allogenic EVs derived from epithelial and mesenchymal cells have been shown to be safe, despite carrying an array of functional molecules, including thousands of proteins. To address whether epithelial cells derived EVs can be modified to acquire the capacity to induce immune response, we engineered 293T EVs to harbor the immunomodulatory molecules CD80, OX40L and PD-L1. We demonstrated abundant levels of these proteins on the engineered cells and EVs. Complete Genome Sequence Of A Penicillin-Resistant Fusobacterium Necrophorum Subsp Funduliforme Isolate From A Tonsillitis Patient, Bibek G C, Anders Jensen, Chenggang Wu

We report the complete genome sequence of a penicillin-resistant Fusobacterium necrophorum subsp. funduliforme isolate, AJ79, from a tonsillitis patient. The AJ79 genome consists of a chromosome (2,440,359 bp) and plasmid (9,887 bp), providing insights into the genetic basis of penicillin resistance in F. necrophorum and its implications for treating tonsillitis.

Lakeshore vegetation supports many valuable aspects of lake ecosystems: foraging areas for birds, rearing grounds for fish, nutrient cycling, carbon storage, and aesthetic, cultural, and spiritual values. Yet, the vegetation of many lakeshores has been lost or heavily impacted by humans. What can be done to restore lakeshore vegetation to ensure that lakes remain healthy for future generations? In this guide, we summarize best practices for lakeshore revegetation (i.e., the process of actively reintroducing vegetation, usually native species, to a restoration site through seeding and planting) in the Great Basin.

Protein aggregates are emerging therapeutic targets in rare monogenic causes of cardiomyopathy and amyloid heart disease, but their role in more prevalent heart-failure syndromes remains mechanistically unexamined. We observed mislocalization of desmin and sarcomeric proteins to aggregates in human myocardium with ischemic cardiomyopathy and in mouse hearts with post-myocardial infarction ventricular remodeling, mimicking findings of autosomal-dominant cardiomyopathy induced by the R120G mutation in the cognate chaperone protein CRYAB. In both syndromes, we demonstrate increased partitioning of CRYAB phosphorylated on serine 59 to NP40-insoluble aggregate-rich biochemical fraction. While CRYAB undergoes phase separation to form condensates, the phosphomimetic mutation of serine 59 …

Background: N-terminal pro-B-type natriuretic peptide (NT-proBNP) and high-sensitivity cardiac troponin T (hs-cTnT) are associated with cardiovascular outcomes and are recommended for measurement in type 2 diabetes (T2D). However, the effects of an intensive lifestyle intervention (ILI) targeting weight loss on cardiac biomarkers and the prognostic association of changes in these biomarkers with risk of adverse cardiovascular outcomes in T2D are not well-established.

Myotonic dystrophy type 1 (DM1) is an autosomal dominant disease caused by a CTG repeat expansion in the dystrophia myotonica protein kinase (DMPK) gene. The expanded CUG repeat RNA (CUGexp RNA) transcribed from the mutant allele sequesters the muscleblind-like (MBNL) family of RNA-binding proteins, causing their loss of function and disrupting regulated pre-mRNA processing. We used a DM1 heart mouse model that inducibly expresses CUGexp RNA to test the contribution of MBNL loss to DM1 cardiac abnormalities and explored MBNL restoration as a potential therapy. BACKGROUND: The receptor tyrosine kinase TIE2 and its ligands, angiopoietins (ANGPTs), promote angiogenesis. In addition to expression on vascular endothelial cells, TIE2 is expressed on M2-like pro-tumorigenic macrophages. Thus, the TIE2 inhibitor rebastinib was developed as a potential therapy to address multiple cancers. The objective of this study was to determine the effects of rebastinib alone and combined with chemotherapy in a syngeneic murine model of ovarian cancer.

Aim: The present work was aimed at formulating novel liquid self microemulsifying oil formulation and its solid form by spray drying method for improving the solubility of Ebastine, an antiallergic drug and BCS class II drug, evaluating its in vitro and in vivo potential. Results: The self emulsifying oil formulation was comprised of Rice Bran Oil, Tween 40, Capryol 90. Pseudoternary phase diagrams were used to evaluate the micro emulsification existence area, and the release rate of Ebastine was investigated using an in vitro dissolution test. Introduction: Minocycline is a tetracycline antibiotic drug that is receiving attention as an alternative for the treatment of depression. Several mechanisms have been proposed for this effect. The current study evaluated the involvement of the noradrenergic system in the antidepressant-like effect of minocycline. Materials and Methods: The total immobility time was evaluated as an index of depression in mice forced swimming test (FST). Minocycline (40 mg/kg) was injected for 3 days. The Environmental Justice (EJ) movement in the United States is comprised of diverse groups of people with a variety of environmental grievances and interests coming together to obtain equal distribution of pollution burdens across communities, reduce environmental hazards, and ensure fair enforcement of laws and policies meant to safeguard the environment for all. The 17 Principles of Environmental Justice developed in 1991 at the First National People of Color Environmental Leadership Summit remains a touchstone document today. Characterization Of 1,8-Cineole (Eucalyptol) From Myrtle And Its Potential Antibacterial And Antioxidant Activities*, Humera Khan

1,8-Cineole is a naturally occurring chemical molecule predominantly found in fragrant plants, particularly Myrtle. Its aroma is distinctive and has been the subject of numerous investigations due to its various biological actions. This study examines the characterization of 1,8-Cineole derived from Myrtle and investigates its antibacterial and antioxidant properties. This study seeks to compare 1,8-Cineole with antibiotics like Amoxicillin and Tetracycline, and moreover, to investigate its antioxidant capabilities against diverse bacterial strains (both Gram-positive and Gram-negative). 1,8-Cineole exhibits the most effective antibacterial properties, demonstrating an inhibition zone of 12.00 mm against Staphylococcus aureus and 10.00 mm against Pseudomonas aeruginosa. …

There is a growing trend in the use of severe caloric restrictive diets among normal weight young females that can jeopardize bone health. Using an animal model, the purpose of this study was to determine whether resistance training (RT) could maintain bone health during a 6-week severe caloric restrictive (CR) diet in growing female rats. The science of embodied cognition has established that thinking involves dynamic exchanges between the body and the brain and webs of interaction between humans and their environments. While learning draws on these connected ecosystems, persistent notions of the brain as separate from and hierarchically managing the body—an enduring legacy of Cartesian dualism—perpetuate static teaching and learning mindsets. Disregard for the body’s role in learning fuels disengagement, isolation, and inequity. This chapter weaves key findings from embodied learning research together with scenes from the author’s own academic journey to illustrate how leveraging brain–body connections can foster educational well-being. This article presents a paper developed by the AAG Organizing Committee on Locational Information and the Public Interest through a summit held in Santa Barbara, California in June 2022. The summit resulted in goals and ideas for addressing the issues that arise from the present environment for geodata, whereby public, private, and third-sector entities can tap into publicly available locational information with relatively little regulation on its access or use. Background: Respiratory syncytial virus (RSV) is a negative sense single-stranded RNA virus (ssRNA) that primarily causes respiratory infections in both children and older adults. We examined clinical characteristics and outcomes of hospitalized adult patients who tested positive for RSV.
Methods: This was a multicenter, retrospective, descriptive study that examined adult patients admitted with a positive RSV test over two years across eight hospitals in Orlando, Florida.
Results: A total of 120 patients were included, of which 88 (73%) were 60 years old and older. Overall, patients with RSV infection were caucasian (62%) with a median age of 70 years and …

The Philippines is considered one of the wealthiest countries in both Southeast Asia and the world when measured by biodiversity, mineral resources and natural resources. It is also one of the most highly mineralized countries on earth. However, despite these natural endowments, when measured by established economic and human development indices the Philippines remains poor. Quantifying emesis in Suncus murinus (S. murinus) has traditionally relied on direct observation or reviewing recorded behaviour, which are laborious, time-consuming processes that are susceptible to operator error. With rapid advancements in deep learning, automated animal behaviour quantification tools with high accuracy have emerged. In this study, we pioneere the use of both three-dimensional convolutional neural networks and self-attention mechanisms to develop the Automatic Emesis Detection (AED) tool for the quantification of emesis in S. murinus, achieving an overall accuracy of 98.92%.
