Background/Objectives: Hepatocellular carcinoma (HCC) remains a significant global health issue due to its high mortality rate and resistance to standard treatments. Sorafenib, the first-line systemic therapy for unresectable HCC, shows limited effectiveness due to resistance and severe side effects. Recent studies suggest that combining sorafenib with immunotherapy, particularly natural killer (NK) cells, may improve treatment outcomes. Methods: This study examined the effectiveness of sorafenib combined with NK cells pretreated with interleukin-12 (IL-12) and interleukin-18 (IL-18) in a rat HCC model. Tumor progression and treatment outcomes were assessed using MRI and histological analysis. Results: The results show that …

BACKGROUND: Sex differences in brain development are thought to lead to sex variation in social behavior. Sex differences are fundamentally driven by both gonadal hormones and sex chromosomes, yet little is known about the independent effects of each on social behavior. Further, mouse models of the genetic liability for the neurodevelopmental disorder MYT1L Syndrome have shown sex-specific deficits in social motivation. In this study, we aimed to determine if gonadal hormones or sex chromosomes primarily mediate the sex differences seen in mouse social behavior, both at baseline and in the context of Myt1l haploinsufficiency.
METHODS: Four-core genotypes (FCG) mice, which …

Lung macrophages play a pivotal role in pulmonary fibrosis, with monocyte-derived alveolar macrophages driving disease progression. However, the mechanisms regulating their pro-fibrotic behavior and survival remain unclear, and effective therapeutic strategies are lacking. Here we show that triggering receptors expressed on myeloid cells 2 are predominantly expressed on monocyte-derived alveolar macrophages in fibrotic mouse lungs and are significantly elevated in lung macrophages from patients with idiopathic pulmonary fibrosis. Deletion or knockdown of this receptor disrupts intracellular survival signaling, promotes macrophage apoptosis, and attenuates their pro-fibrotic phenotype. We further demonstrate that a lipid mediator and a high-avidity ligand of this receptor, …

Fatty acid synthase (FAS) is predominantly expressed in the liver and adipose tissue. It plays vital roles in de novo synthesis of saturated fatty acids and regulates insulin sensitivity. We previously demonstrated that serum circulating FAS (cFAS) is a clinical biomarker for advanced atherosclerosis, and that it is conjugated to low-density lipoproteins (LDL). However, it remains unknown whether cFAS can directly impact atheroprogression. To investigate this, we evaluate whether cFAS impacts macrophage foam cell formation - an important cellular process leading to atheroprogression. Macrophages exposed to human serum containing high levels of cFAS show increased foam cell formation as compared …

We measured the BASAL breathing frequency following an overnight fast in adult,non-pregnant/non-lactating, inactive mammals ranging in body mass from 15 to5520 kg. The data included results from 338 individual animals from 34 speciesthat were divided into terrestrial, semi-aquatic (Otariidae and Phocidae) and aquaticmammals. Following attempts to limit the collection of breathing frequency usinga basal definition and to correct the analysis phylogenetically, our results suggestthat there are differences in the allometric mass-exponent between terrestrial andaquatic/semi-aquatic mammals. An allometric regression model, whereby both bodymass and breathing frequency were transformed using log10 , suggested that theallometric mass exponent for terrestrial mammals (−0.303) was …

Theses and Dissertations
Hepatocellular carcinoma (HCC) is the second lethal cancer worldwide, with over 60 million new patients added annually. Hepatitis B infection and toxins, e.g., aflatoxin, contribute to the incidence of HCC. Egypt and China show the highest prevalence of viral hepatitis and HCC in the world.
Long non-coding RNAs (lncRNAs) are non-protein-coding transcripts larger than 200 nucleotides. These transcripts work at the transcriptional, post-transcriptional, and epigenetic levels to regulate gene expression by interacting with various biomolecules, such as RNA, DNA, and proteins. They serve essential functions in chromatin remodeling and cell survival, differentiation, and proliferation. The role and biological interactions of …

The research aims to assess consumer preferences and attitudes toward CRISPR applications in the food sector and estimate consumer willingness to pay for a food product developed using CRISPR technology to enhance food safety and confer health benefits. Additionally, the research assesses the impact of information provision on perceptions, attitudes and willingness to pay. Specifically, we consider the potential effect of both the nature of information provided (i.e., whether it emphasizes the differences between GM and CRISPR technology or not) and the delivery format (i.e., text versus video) used to present content-equivalent information. In this context, we also assess consumer …

Identification Of Novel Argonaute Proteins Using A Metagenomic Mining Approach, Lobna Abdallah Ghonaim
Theses and Dissertations
Gene editing is one of the most promising tools in science. It enables precise modifications of an organism's genetic material. Metagenomics is considered a powerful tool that unlocks the broad genetic potential found in uncultured microbial communities. Exploring the genetic diversity of uncultured microbial communities helps identify novel functional proteins with unique properties and make the best use of these diverse microbial ecosystems.
We developed and employed a metagenomic-based approach to mine more than 1000 metagenomes for prokaryotic argonaute proteins (pAgos), a potential gene editing machinery encoded in bacterial and archaeal genomes. Our workflow involved strict quality control, sequence assembly, …

Multispecies swards have been recognized as more sustainable than traditional ryegrass systems, in addition to providing a range of phytochemicals to the ruminant diet. In this regard, polyphenols may be linked to higher levels of bioactive fatty acids in milk and enhanced nutraceutical properties in dairy products. This study compares the fat composition and oxidative stability of milk produced by cows fed on multispecies swards (MULTI) with milk from two conventional swards [a perennial ryegrass monoculture (PRG) and a mixture of perennial ryegrass and white clover (PRG + WC)] during the Irish summer. Milk from cows fed on MULTI had …

Development Of A Conditional Plasmid For Gene Deletion In Non-Model Fusobacterium Nucleatum Strains, Peng Zhou, Bibek G C, Chenggang Wu
Faculty, Staff and Student Publications
Fusobacterium nucleatum is an opportunistic pathogen with four subspecies: nucleatum (FNN), vincentii (FNV), polymorphum (FNP), and animalis (FNA), each with distinct disease potentials. Research on fusobacterial pathogenesis has mainly focused on the model strain ATCC 23726 from FNN. However, this narrow focus may overlook significant behaviors of other FNN strains and those from other subspecies, given the genetic and phenotypic diversity within F. nucleatum. While ATCC 23726 is highly transformable, most other Fusobacterium strains exhibit low transformation efficiency, complicating traditional gene deletion methods that rely on non-replicating plasmids. To address this, we developed a conditional plasmid system in which …

Recent studies have highlighted the crucial role of microglia (MG) and their interactions with the gut microbiome in post-stroke neuroinflammation. The activation of immunoregulatory pathways, including the aryl hydrocarbon receptor (AHR) pathway, is influenced by a dynamic balance of ligands derived from both the host and microbiota. This study aimed to investigate the association between stroke-induced dysbiosis and the resultant imbalance in AHR ligand sources (loss of microbiota-derived [indole-based] and increase of host-derived [kynurenine-based]) after stroke. Microbiota-derived AHR ligands decreased in human plasma and remained low for days following an ischemic stroke highlighting the translational significance. Transient-middle-cerebral-artery-occlusion was performed in …

Commensality In The Eateries Of Roman Pompeii, Amaya Yolanda Eckersley
Dissertations and Theses
Despite a nearly 300-year excavation history at Pompeii and many identified eating and drinking establishments, social behavior in these public or semi-public spaces has eluded significant research. Elite Roman literature is the traditional guideline for understanding Roman mealtimes and what they meant for social behavior. This literature, however, fails to accurately represent its sub-elite inhabitants. These sub-elites of Rome largely had their meals prepared outside of their homes. Pompeii is no exception and provides a massive boon for researchers willing to consider archaeological evidence. The buildings and evidence buried at Pompeii have ultimately shown a wide array of tabernae related …

BACKGROUND: Chronic alcohol consumption leads to lipid accumulation, oxidative stress, cellular damage, and inflammation in the liver, collectively referred to as alcohol-associated liver disease (ALD). FAF2/UBXD8/ETEA (Fas-associated factor 2) is a ubiquitin ligase adaptor protein that plays a crucial role in the ubiquitin-mediated degradation of misfolded proteins in the endoplasmic reticulum. A recent genome-wide association study indicated an association between FAF2 and ALD; however, the exact contribution of FAF2 to ALD pathogenesis remains unclear. METHODS: FAF2 was knocked down using AAV-delivered shRNA in C57/BL6 mice. Mice were subjected to a chronic-plus-single binge ethanol feeding (NIAAA) model. Nine hours after gavage, …

Shock Effects Including Maskelynite Formation In Three Shergottite Meteorites From Mars, George Ayeh Anim
Dissertations and Theses
Martian meteorites result from large impact events on the surface of Mars that cause intense shock metamorphism. This study examines the shock metamorphic features of three shergottite meteorites, Dho 019, NWA 12241, and NWA 15628, to better understand shock impact events. The research uses petrographic analysis, scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and electron backscatter diffraction (EBSD) to analyze the meteorites. A key focus of this research is the formation of maskelynite, a glass with plagioclase composition, whose formation mechanism has been the substance of debate among researchers. This study proposes that maskelynite forms through a transient intermediary …

The United States' Recycling Model Is In Desperate Need Of A Makeover, Lindsay A. Williamson
The United States' Recycling Model Is In Desperate Need Of A Makeover, Lindsay A. Williamson
Widener Commonwealth Law Review
Most Americans do not hesitate to throw something away, but a lack of recycling is causing landfills to overflow with products that could be reused and repurposed. In 2017, it was estimated that the average American creates 4.51 pounds of garbage per day, nearly double the amount of the 2.68 pounds Americans produced on average in 1960. Recycling in the United States has become problematic as recycling companies turn away recyclables that have been contaminated with food and other nonrecyclables. The effect is that not nearly enough recyclable products are being repurposed, with many of them ending up in landfills. …

Yes, Your Broken Tailight Does Matter: Why Enforcement Of Minor Traffic Violations Is Integral For A Safe Society, Richard E. Berner
Widener Commonwealth Law Review
