Salix section Subviminales, including only three species, is endemic to East Asia. In this study, we revised three species, S. gracilistyla, S. graciliglans, and S. blinii in Korea based on their plastomes and extensive morphological analyses to provide a comprehensive understanding of their taxonomic relationships. Based on our findings, we identified the morphological characteristics of S. blinii endemic to Korea, including its small size and distinct leaf shapes, which are well distinguished from both S. gracilistyla and S. graciliglans. Most of the morphological characteristics of S. gracilistyla and S. graciliglans overlapped, particularly leaf shape, floral structure, and growth habits. Next, …

Soft power emanates from a country's cultural, political beliefs, and foreign policies. Glycosylation controls immune evasion, tumor progression, and metastasis. However, how tumor cell sialylation regulates immune evasion remains poorly characterized. ST6GalNAc-I, a sialyltransferase that conjugates sialic acid to the glycans in glycoproteins, was overexpressed in an aggressive-type KPA (KrasG12D/+ Trp53R172H/+ Ad-Cre) lung adenocarcinoma (LUAD) model and patient samples. Proteomic and biochemical analysis indicated that ST6GalNAc-I mediated NECTIN2 sialylation in LUAD cells. ST6GalNAc-I-deficient tumor cells cocultured with T cells were more susceptible to T cell-mediated tumor cell killing, indicating a key role for NECTIN2 in T cell dysfunction. Mice injected with St6galnac-I-knockdown syngeneic cells showed reduced lung tumor incidence and Nectin2/Tigit-associated immunosuppression. …

This dissertation explores the role-space of educational interpreters at IC Cossato, an Italian public school known for its Bilingualism Project promoting LIS-Italian bilingualism and biliteracy (Rivisto, 2016; Teruggi, 2003). Role-space theory (Lee, 2023; Llewellyn-Jones & Lee, 2013, 2014) conceptualizes interpreters’ roles as dynamic and co-constructed with interlocutors along three axes: Presentation of Self, Interaction Management, and Participant Alignment. Through ethnographic interviews (Spradley, 2016), observations and field notes (Emerson et al. 2011; Glesne, 2016), and policy artifacts (Hornberger & Johnson, 2011), I investigate how three interpreters in IC Cossato’s elementary school perceive and enact their roles, often diverging from their …

Increasing exploitation of water resources has led to declining water quality in aquatic environments. Freshwater ecosystems face various environmental pressures, such as the impacts of climate change and intensifying land-use practices. This study aimed to assess the ecological preferences of phytoplankton communities and determine the ecological status (ES) of several reservoirs and lakes in Ceyhan River Basin across different seasons. A canonical correspondence analysis was conducted to understand the associations between phytoplankton assemblages and stressors in freshwater ecosystems, revealing a strong correlation of 97%. Sickle cell disease (SCD) is a genetic disorder characterized by sickle red blood cells (RBCs). Sickle RBCs cause cerebral vasculopathies including vaso-occlusive events, leading to ischemia-reperfusion injury and hypoxic tissue environment. To date, the physiological blood flow velocities in cerebral vessels of preclinical SCD models has not been evaluated under hypoxic-reoxygenation. In our study, we used transcranial ultrasound techniques to measure abnormal blood flow velocities in the internal carotid (ICA) and middle cerebral arteries (MCA) of transgenic sickle cell mice (SS) challenged with hypoxia-reoxygenation. Over the course of this project our group members Rebecca, Max, PJ, Amanda, and Risa worked closely with Jim Knight and Brian Auman from Turtle Creek Park to help research, plan, and execute a tree transplanting event that would be efficiently implemented and environmentally sound. This work involved extensive preparatory work in the park including soil testing, bare leaf tree identification, tree and shrub species characterization, and recommendation. Meiotic chromosome segregation errors in human oocytes are the leading cause of miscarriages and aneuploid pregnancies, and these errors increase dramatically as women age. Using Drosophila oocytes as a model system, this dissertation identifies the NAD⁺-dependent deacetylase Sirt1 as a key regulator of chromosome segregation and meiotic arm cohesion. Loss of Sirt1 activity during meiotic prophase leads to premature loss of arm cohesion and increased segregation errors in Drosophila oocytes. In addition, elevated acetylation of its substrate, histone H4K16, indicates that Sirt1 activity declines in oocytes during aging. Strikingly, dietary administration of the Sirt1 activator SRT1720 preserves Sirt1 deacetylase activity …

By investigating the historical, geographical, and socio-cultural influences that shape the music in these regions, this study highlights the unique musical identities of each group while uncovering their commonalities and cultural exchanges. Children’s books are some of the earliest forms of entertainment and education a person will interact with in their lifetime. Simultaneously weaving a tale and teaching a lesson, children’s books, therefore, have the great potential to communicate important lessons and mindsets at young ages. Utilizing narrative theory (Fisher, 1984) and ecocriticism (Glotfelty, 1996), while also considering environmental ideologies and narrative components (Bloomfield, 2024), this study explores the functions of environmental children’s books and how they might communicate environmental messaging and encourage ecological thinking. Alzheimer’s disease (AD) is an irreversible and devastating neurodegenerative disease characterized by progressive synaptic, dendritic, glial, and neuronal loss, collectively leading to impairments in learning, memory, and deterioration of cognitive and behavioral functions. AD is the most common form of dementia accounting for 60-80% of all cases. AD affects approximately 7 million Americans and is the 6th leading cause of death in the US. Furthermore, by 2050, the US is projected to spend over $1.1 trillion on AD-related treatments. Fungal exposure can cause acute and chronic illnesses or exacerbate existing illnesses. Some fungi are opportunistic pathogens and can lead to serious systemic fungal infections. In 2020, more than 150 million cases of severe fungal infections were reported globally resulting in an estimated 1.7 million deaths. Coccidioidomycosis, or Valley fever, is classified as a serious fungal infection caused by the fungi Coccidioides immitis and C. posadasii. These particular fungi are prevalent in the soil of the Southwestern United States, including Las Vegas, Nevada. In 2024, there were a total of 269 cases of Valley fever reported in Nevada, and …

Walnut (Juglans regia L.) is one of the most economically important species. It is of great importance to the food industry due to its nutritional value, and high content of many beneficial components such as macro and microelements. In the current study, the aim was to determine markers of single nucleotide polymorphism (SNP) associated with the gene(s) controlling the accumulation of Selenium (Se) and Magnesium (Mg) in walnuts. Se and Mg concentrations were measured in 154 genotypes of Juglans regia in 2021 and 2022. A total of 16,473 SNP markers were used for association mapping to analyze the marker-trait associations. …

The widespread utilisation of deep sequencing has dramatically enhanced and revolutionised our understanding of virus diversity, particularly in complex environments such as soil and biological samples. This research explored mycovirus sequence analyses in the ectomycorrhizal fungus Amanita vidua, utilising deep sequencing alongside total double-stranded RNA (dsRNA) extractions. Our investigation unveiled a unique member of the proposed family, Narliviridae, designated as Amanita vidua narlivirus 1 (AvNV1), in an A. vidua specimen. AvNV1, classified under Amabiliviricetes, possesses a distinct genome structure and sequence attributes. This study aims to investigate any possible interaction between two highly conserved regulators of cellular SNO levels: S-nitrosoglutathione reductase (GSNOR1) and thioredoxin (TRX). These two enzymes control global S-nitrosylation levels and thus impact the cellular redox state during immune responses. A mutant line (Δtrx1trx2sfa1) of Saccharomyces cerevisiae MaV203 strain was generated to explore this possibility rather than using a classical strain. We identified and characterized the physical interaction between GSNOR1 and Arabidopsis cytosolic h isoforms in a yeast two-hybrid screen. Out of the seven TRX isoforms, AtGSNOR1 displayed a high interaction with AtTRXh3, AtTRXh4, and AtTRXh5 (AtTRXh5>AtTRXh4 & AtTRXh3). …

Sterile acute kidney injury (AKI) is common in the clinic and frequently associated with unexplained hypoxemia that does not improve with dialysis. AKI induces remote lung inflammation with neutrophil recruitment in mice and humans, but which cellular cues establish neutrophilic inflammation and how it contributes to hypoxemia is not known. Here we report that AKI induced rapid intravascular neutrophil retention in lung alveolar capillaries without extravasation into tissue or alveoli, causing hypoxemia by reducing lung capillary blood flow in the absence of substantial lung interstitial or alveolar edema. Kidney disease, the ninth leading cause of death in the United States, suffers from poor diagnostic efficiency (10%). Traditional biopsies use molecular reagents to enhance diagnostic power but are limited by overlapping spatial and chromatic signals, product quality variability, and additional processing. To address these challenges without disrupting routine diagnostics, we implement label-free imaging modalities-stimulated Raman scattering (SRS), second harmonic generation (SHG), and two-photon fluorescence (TPF)-within a single setup. We identify morphological, lipidomic, and metabolic biomarkers in control and diabetic kidney samples at subcellular resolution. Meningeal lymphatics serve as an outlet for cerebrospinal fluid, and their dysfunction is associated with various neurodegenerative conditions. Previous studies have demonstrated that dysfunctional meningeal lymphatics evoke behavioral changes, but the neural mechanisms underlying these changes have remained elusive. Here, we show that prolonged impairment of meningeal lymphatics alters the balance of cortical excitatory and inhibitory synaptic inputs, accompanied by deficits in memory tasks. These synaptic and behavioral alterations induced by lymphatic dysfunction are mediated by microglia, leading to increased expression of the interleukin 6 gene (Il6). Mechanisms that mediate allograft tolerance differ between organs. We have previously shown that Foxp3+ T cell-enriched bronchus-associated lymphoid tissue (BALT) is induced in tolerant murine lung allografts and that these Foxp3+ cells suppress alloimmune responses locally and systemically. Here, we demonstrated that Foxp3+ cells that reside in tolerant lung allografts differed phenotypically and transcriptionally from those in the periphery and were clonally expanded. Using a mouse lung retransplant model, we showed that recipient Foxp3+ cells were continuously recruited to the BALT within tolerant allografts. Chronic back pain (CBP) affects 1 in 10 people worldwide, reducing mobility, productivity, and quality of life. Despite its prevalence, non-addictive treatments remain limited due to an incomplete understanding of chronic pain mechanisms. This dissertation examines peripheral neuroimmune interactions in CBP using a urokinase-type plasminogen activator (uPA)-induced mouse model (uPA-CBP). uPA, a pro-inflammatory serine protease, was injected into the L2/L3 lumbar spinous ligament, inducing prolonged mechanical and cold hypersensitivity. Flow cytometry of dorsal root ganglia (DRG) showed increased CD45+CD11b+ myeloid cells in males and CD45+CD3+CD4+ lymphocytes in females. We conducted a community-science project in Northern California - the Seed Pile Project - with three broad goals: 1) to test which locally-native, seed-dispersed plants would thrive without intervention in human-occupied spaces, 2) to engage the public with the often-ignored biodiversity along our roadsides, sidewalks, and other highly-disturbed spaces, and 3) to create community around rewilding our urban areas. We conducted the project initially as a pilot project in 2020-2021, then as a community-science project in 2021-2022 and 2022-2023. San Joaquin kit foxes (Vulpes macrotis mutica) are endemic to arid habitats in the San Joaquin Valley region of central California. This species is listed as Endangered due to profound habitat loss throughout its range caused by agricultural, industrial, and urban development. Interestingly, kit foxes currently occur in three cities with the population in the city of Bakersfield being large (several hundred foxes) and self-sustaining. Demographic and ecological attribute values for this population are among the highest observed in San Joaquin kit fox populations. The global increase in the number of managed honey bees (Apis mellifera) has raised concerns about their impact on native pollinator species, including bumble bees (Bombus spp.). This study examines the extent of floral resource overlap between honey bees and bumble bees in urban areas within the Greater Toronto Area (GTA), Ontario, Canada, to better understand how competition for floral resources may influence bumble bee foraging behavior and niche partitioning. The identity of the fur covering the sculpture named Object in the Museum of Modern Art has remained a mystery since its creation in 1936 by Meret Oppenheim. DNA testing can possibly provide an answer, but to preserve the integrity of the sculpture, only a very small piece of the fur can be sampled. Prior to testing Object, testing using museum samples was performed to determine whether DNA of a sufficient quantity and quality can be extracted and sequenced.
