Genomic analysis of parasites can deepen our understanding of their transmission, population structure, and important biological characteristics. Onchocerciasis (river blindness), caused by the parasitic nematode Onchocerca volvulus, involves adult worms residing in subcutaneous nodules that produce larval-stage microfilariae (mf), which are routinely detected in the skin for diagnosis. Whole-genome studies of mf are limited; most analyses have focused on the mitochondrial genome. We conducted a genome-wide analysis with 94% median nuclear genome coverage, analyzing 171, 37, and 98 mf from 16, 3, and 5 individuals from Ghana, Liberia, and the Democratic Republic of Congo, respectively. Recent studies have highlighted the importance of mitochondria in NP cells and articular chondrocyte health. Since the understanding of mechanisms governing mitochondrial dynamics in these tissues is lacking, we investigated the role of OPA1, a mitochondrial fusion protein, in their homeostasis. OPA1 knockdown in NP cells altered mitochondrial size and cristae shape and increased the oxygen consumption rate. In a translationally relevant ovine model of gestational hyperandrogenism we have previously reported cardiometabolic disruption in female offspring (males were not extensively studied). We hypothesized that gestational hyperandrogenism would lead to sex-specific disruption in offspring's growth, cytokine and metabolic milieu, potential mediators of cardiometabolic disease (CMD). 100 mg Testosterone propionate (T) or vehicle (C) was administered intramuscularly twice weekly between gestational days (GD) 30-90. Two cohorts of offspring were generated, including both males and females. Fetal weight and systemic metabolomics were analyzed in the fetal cohort (GD120). Introduction:
Total temporomandibular joint replacement (TMJR) is a well-established surgical solution for patients with severe TMJ disorders. It aims to relieve chronic pain, restore jaw mobility, and significantly enhance quality of life. This systematic review evaluates QoL outcomes following TMJR, analyzes complication profiles, compares custom versus stock prostheses, explores pediatric applications, and highlights technological innovations shaping the future of TMJ reconstruction.
Methods:

Background:
Prebiotic fiber has been examined as a way to foster gut bacteria less associated with obesity. Tests of prebiotic fiber in reducing obesity have occurred mainly in animals, adults, and Caucasians when the highest obesity rates are in African American and Latinx youth. Response to prebiotic fiber is determined by the pre-existing intestinal microbiota. The type of microbiota varies based on diet and physical activity (PA), so it is important to examine acceptability and response to prebiotic fiber in those most at risk for obesity.

Redefining Rural Representation In Short Story Anthologies: Using Short Stories To Highlight Rural Counterstories, Brittany Gargano-Smith, Jenna Spiering

This article explores the importance of incorporating diverse rural short stories into English language arts (ELA) curricula to foster empathy, understanding, and connection among students. It addresses underrepresentation and lack of variety in rural stories within publishing and traditional curricula, which affects rural students’ sense of identity and belonging. Through exploration of rural short story anthologies, the authors highlight the significance of counterstories that depict the intersectional dimensions of rural life, including geography, race, ethnicity, sexual orientation, socioeconomic background, and abilities. By amplifying contemporary rural voices and complicating stereotypes, educators can create more inclusive learning environments that validate diverse experiences.

Single-cell multiomic techniques have sparked immense interest in developing a comprehensive multi-modal map of diverse neuronal cell types and their brain-wide projections. However, investigating the complex wiring diagram, spatial organization, transcriptional, and epigenetic landscapes of brain-wide projection neurons is hampered by the lack of efficient and easily adoptable tools. Here we introduce Projection-TAGs, a retrograde AAV platform that allows multiplex tagging of projection neurons using RNA barcodes. By using Projection-TAGs, we performed multiplex projection tracing of the cortex and high-throughput single-cell profiling of the transcriptional and epigenetic landscapes of the cortical projection neurons in female mice. IMPORTANCE: Associations between childhood obesity and brain microstructural differences have been observed. It remains unknown whether these associations are driven by sex-specific excessive weight gain. Restriction spectrum imaging characterizes brain tissue microstructural health via water diffusion, where the restricted normalized isotropic (RNI) compartment assesses neuronal and glial cellularity, which may reflect neuroinflammation, synaptic pruning, or both.

OBJECTIVE: Our objective was to determine if oxytocin discontinuation in the active phase of labor impacts the rate of cesarean delivery compared to continuation of oxytocin.
DATA SOURCES: This study was a systematic review and meta-analysis of randomized controlled trials. A research librarian performed a database search using a combination of standardized terms and keywords related to oxytocin discontinuation and stages of labor from database inception until February 2024. In recent years, large-scale disaster events have dominated headlines. As the world faces an increasing number of such disasters, it is crucial to understand their impacts beyond immediate monetary losses. Disasters not only directly impact individuals and businesses but may also reshape the spatial patterns of the regional economy. Through a case study of Hurricane Harvey in Texas and Louisiana, this study examines how disaster events influence regional economic specialization patterns and contribute to uneven impacts across economic supersectors and geographies. BACKGROUND: Tracheostomy is a common procedure in intensive care medicine, involving the insertion of an artificial airway through the front of the neck into the trachea. The size of tracheostomy tube in relation to the trachea is important, influencing clinical outcomes and patient experience. Ultrasound is readily available at the bedside and airway applications have been reported, including airway measurement. However, the presence of air within airway lumina presents challenges to ultrasound and causes artefact. Guidance is lacking on tracheal sonography and measurement.

Ecological disturbances are discrete events that alter or transform the physical, chemical, or biological characteristics of ecosystems. Disturbance can cause animal populations to decline and, according to the risk-disturbance hypothesis and population collapse framework, these declines can be predicted by declines in animal body condition. However, no research has empirically examined the general relationship between body condition and abundance, nor their relationship in response to disturbance. We present a novel method of measuring thrust of aquatic animals using in situ video data of swimming motions. To demonstrate its utility, the method was applied to several large elasmobranch species, which are typically highly challenging to measure. Using motion tracking software, we analyzed video footage of wild and captive sharks to track their instantaneous position and speed. In order to estimate the force output, we used the tail/body motion based on the swimming modes of the fish to calculate the water displaced by this motion during locomotion. The Department of Primary Industries and Regional Development (DPIRD) monitors and reports on the vegetation condition of pastoral rangelands in Western Australia. Two levels of reporting are provided: every 5 years a full report (this report) details the state, trend and risk of decrease of vegetation condition in the pastoral rangelands using information derived from remotely sensed and on-ground data; in the intervening years, short reports are provided based on remotely sensed data.
This full report is based on remotely sensed vegetation cover data, rainfall data, livestock data and station-level rangeland condition assessment (RCA) data available in November 2024. Data …

Maternally derived antibodies (MDAs) play a vital role in protecting neonates from infectious diseases, but their presence at the time of vaccination can interfere with vaccine-induced immune responses, thereby reducing vaccine effectiveness. MDA interference has been documented in pigs vaccinated with whole inactivated virus (WIV) vaccines against swine influenza A virus (IAV). In this thesis, we evaluated the efficacy of a lipid nanoparticle encapsulated DNA (LNP-DNA) vaccine against swine IAV in the presence and absence of MDAs, comparing it to a WIV vaccine. The retinal determination genetic network controls the development of the visual system in all seeing animals through the molecular regulation of cells to adopt an eye tissue fate. The compound eye of the fruit fly, Drosophila melanogaster, is an excellent model system to study the complex mechanisms within the network that regulate specification of cellular identity during embryogenesis. In Drosophila, the two Pax6 paralogues, eyeless and twin of eyeless, sit at the very pinnacle of the network and their expression early in development activates critical downstream components of the retinal determination pathway. With limited land and water resources available it has become critical to maximize output for agricultural products while maintaining sustainable farming practices. Ricefish co-culture can be used to maximize limited land and water resources for production of rice and fish concurrently and reduce the need of chemical inputs. BACKGROUND: Thoracic (TAA) and abdominal (AAA) aortic aneurysm are life-threatening diseases characterized by dilation, inflammation, and structural weakness; development of pharmacological therapies is desperately needed. CNP (C-type natriuretic peptide) plays a key role in vascular homeostasis, mediating vasodilator, anti-inflammatory, and antiatherogenic actions. Since such processes drive AA, we determined the role of endogenous CNP in offsetting pathogenesis. METHODS: Tissue from patients with AA was analyzed to determine the consequences on CNP signaling. Senescent cells emerge with aging and injury. The contribution of senescent cells to DNA methylation age (DNAmAGE) in vivo is uncertain. Furthermore, stem cell therapy can mediate “rejuvenation”, but how tissue regeneration controlled by resident stem cells affects whole tissue DNAmAGE is unclear. We assessed DNAmAGE with or without senolytics (BI01) in aged male mice (24–25 months) 35 days following muscle healing (BaCl2-induced regeneration versus non-injured). Young injured mice (5–6 months) without senolytics were comparators. DNAmAGE was decelerated by up to 68% after injury in aged muscle. This thesis explores how bold and unconventional architectural designs are first rejected and later embraced by the public. Through a comparative study of the Sydney Opera House, the Louvre Pyramid, and the Walt Disney Concert Hall, it examines how public opinion shifts over time. Using media reports, interviews, and public comments, the study tracks emotional responses (positive, neutral, or negative) assigned a score to measure change over the years. The research applies Everett Rogers’ Diffusion of Innovation Theory to explain this shift in five stages: knowledge, persuasion, decision, implementation, and confirmation. BACKGROUND: Hormone fluctuations and inconsistencies across the menstrual cycle paired with a shortage of literature on female physiology in exercise have made understanding on female exercise performance troublesome. Performance-based research has fallen behind and the increase in women participation and current evidence does not warrant general guidance on modulating specific exercise across the menstrual cycle. Maintenance of mitochondrial transport and clearance at the pre- and post-synaptic terminal is an essential function of neuromuscular junctions to meet their high energy demands. Damaged or dysfunctional mitochondria can put stress on these synaptic junctions, ultimately leading to the degradation of the synapse and subsequent cell death. Dysfunctional mitochondria are also often present in neuromuscular diseases such as ALS. One possible mechanism by which neurons may eliminate dysfunctional mitochondria is through the muscle-synthesized BDNF (msBDNF) /TrkB-mediated Bcl-2 nineteen-kD interacting protein (BNIP3) mitophagy pathway, although the relationship is not well-defined. Brain derived neurotrophic factor (BDNF) is a neurotrophin essential to neuron health and maintenance in both the central nervous system and periphery. It acts as a positive feedback system for the body’s motor neurons and the muscles they innervate, strengthening the synapse between them. Skeletal muscle-synthesized BDNF transmission occurs across the neuromuscular junction, the site where motor neuron axons synapse with muscle fibers. BDNF transmission to tropomyosin receptor kinase B (TrkB) at the motor neuron axon terminal causes clathrin mediated endocytosis of the complex and an intracellular signaling cascade that recruits dynein motor proteins for retrograde transport of the BDNF/TrkB …
