Polymeric Nanocarriers Functionalized With Peptides For Improved Glioblastoma Targeting And Blood–Brain Barrier Permeability, Cristian Antonio Wieczorek Villas Boas, Aaron Priester, Buck E. Rogers, Anthony J. Convertine

We report the synthesis of polymeric conjugates designed to penetrate the blood–brain barrier (BBB) and selectively bind glioblastoma (GBM) cells through reversible addition–fragmentation chain transfer (RAFT) polymerization. The resulting materials were engineered to contain peptide macromonomers for cell-specific targeting and integrated DOTA units to facilitate radiolabeling with copper-64 (64Cu), yielding radiolabeled conjugates with greater than 95% radiochemical purity. In biodistribution assessments conducted in mice, C1C2 peptide-conjugated polymers showed significantly improved accumulation in brain tissue, supported by brain perfusion analyses confirming efficient BBB penetration. Additionally, flow cytometry evaluations demonstrated specific affinity of GBM-targeted polymer formulations toward U87 glioblastoma cells. …

Cerebral edema (CE) is a major component of early brain injury from aneurysmal subarachnoid hemorrhage (SAH). Despite the utility of blood-based protein markers, biomarkers targeting cerebral edema have yet to be developed. We performed a targeted proteomic search analyzing 141 proteins in plasma samples taken from 80 adult patients within 48 hours after aneurysmal rupture to identify plasma biomarkers of cerebral edema. The protein expression profiles were analyzed for associations with a higher level of the subarachnoid hemorrhage early brain edema score measured in admission computed tomography scans. Cryptic habitats on coral reefs consist of crevices, tunnels, and holes that are estimated to comprise 30%–75% of the total reef surface area. These large habitats are vastly understudied because they are often difficult to access. Crustose coralline algae (CCA) are thought to perform important ecological functions, including calcium accretion, both inside cryptic habitats and on the exposed (top) reef. Using GoPro cameras, we surveyed 250 coral crevices from 13 reefs on West Maui, Hawai‘i. We compared relative CCA cover between crevice microhabitats and the well-studied top-reef and identified abiotic and biotic factors that correlated with CCA abundance. We found …

Objectives: In the San Francisco Bay Area, Late Holocene resource intensification models predict an increase in diet breadth and a reduction in foraging efficiency associated with an increase in population, sedentism, and territoriality among fisher-hunter-gatherer populations. Here we compare bone stable carbon (δ13C) and nitrogen (δ15N) isotope values of sites along the eastern bay shore to interpret how ecogeographical partitioning of resources by latitude and distance from the bay reflects differential access and control of resources. Validated Hots-Based Worksheets Enhance Higher-Order Thinking Skills And Impact Geometry Learning Outcomes, Aulia Al Tsani Izzati, Siti Faizah, Mardhatillah Mardhatillah
Jurnal Pendidikan: Teori, Penelitian, dan Pengembangan
This study aims to develop and evaluate worksheets that promote higher-order thinking skills (HOTS) in teaching perimeter and area to primary school students. The worksheets were highly valid, with a score of 94.63%, and were shown to be well aligned with learning objectives through extensive validity and reliability testing. They contain various question types, including multiple-choice, short-answer and essay questions, which encourage students to think critically and creatively. While many students improved their analytical skills, 39% needed to meet the minimum competency standard (KKM). This underscores the necessity for supplementary strategies to support all learners and enhance learning outcomes.

Elementary school students are classified as playing-age children, they like to do activities while playing. This research aims to determine the influence of the Game-Based Learning (GBL) learning model on students' cognitive activities and abilities. This type of research is quasi-experimental, one-group pre-test and post-test design. The population of this study were Madrasah Ibtidaiyah students at Darussalam Seuksa School, Thepa, Southern Thailand. The research sample for grade 5 students was 28 students. This article explores our modern world and our role in shaping collective experiences. By our thoughts, choices and behaviours (our character), we influence our shared reality. Higher education has an imperative responsibility to engage students ethically and develop their character. I offer a teaching metaphor and case study to meaningfully develop students’ self-awareness of character via an undergraduate leadership course. I employed the ‘seed’ metaphor to illustrate how nurturing students’ self-awareness and agency mirrors the growth of a seed within a carefully cultivated ecosystem. Reading text on electronic device displays can be difficult in varying ambient light conditions, particularly for users with age-related vision changes. Current solutions require manual font size adjustments, which are static, cumbersome, and separate from automatic brightness controls. This disclosure describes a method for adjusting the font size on a display in response to changing conditions, without requiring explicit user interaction. For example, an ambient light sensor may measure surrounding light levels, and user-specific data, such as age and/or a vision quality rating, can be optionally provided. The aims of this study were to determine the possible cross-resistance and horizontal gene transfer associated with halquinol in Escherichia coli and Salmonella spp. from pigs. Two of each E. coli and Salmonella isolates exhibiting low halquinol MIC (8 and 16 µg/ml, respectively) and susceptible to antibiotics tested were selected for in vitro stepwisehalquinol exposure experiment. Another E. coli (n=19) and Salmonella (n=6) isolates with high halquinol MICs (16-64 and 32-64 µg/ml, respectively) containing plasmids were used for transferability testing of halquinol resistanceencoding gene(s) using filter mating method. Using a mixed-methods approach that combines quantitative literature review and qualitative case study analysis, this capstone investigates the developmental pathways of serial killers. The study investigates a variety of biological, psychological, and sociological elements—such as genetic predispositions, neurological disorders, childhood traumas, and environmental stressors—interact to shape serial killers. Reading and writing in the 21st century have evolved from traditional text-based formats to multimodal literacy, integrating linguistic, visual, auditory, and spatial modes to enhance communication and comprehension. While multimodal reading has been widely studied, multimodal writing remains underexplored, despite its growing importance in K–12 education across disciplines. Multimodal composing demands advanced self-regulation as students navigate multiple digital tools and platforms. Self-regulated learning strategies, particularly the self-regulated strategy developmental model, offer a promising approach to support students in planning, monitoring, and revising multimodal compositions. There is a growing interest in developing safer and more effective decontaminating agents for organophosphate-based pesticides and nerve agents. In this study, we present an effective method for the nonaqueous decontamination of these compounds using small-molecule-based decontaminating agents under ambient conditions. Our approach utilizes aryl and heteroaryl carboxaldehyde hydrazones and hydrazides to effectively hydrolyze nerve agent simulants into their nontoxic degradation products. The effectiveness of this method was evaluated using a range of nerve agent simulants, including dimethyl 4-nitrophenyl phosphate (DMNP), dimethyl methyl phosphonate (DMMP), and triphenyl phosphate (TPhP). This dissertation consists of three essays on Gamification, Innovation, and Cyber Security. The first essay investigates gamified stock trading. Gamification of stock trading is a novel practice by brokers to incorporate game-like features to increase clients’ engagement with trading. This study examines how the market reacts to the introduction of 142 gamification features in the mobile trading apps of 17 major U.S. brokers. I find that the gamification of trading can be viewed as a double-edged sword. It alters and worsens retail traders’ strategy by reducing their returns and increasing return volatility. This dissertation explores the nature of social identity by examining both the collective identity of groups and the social selves of individuals. I start by offering a Humean model of collective identity. On this model, the metaphysical nature of group persistence depends on our psychological dispositions and our explanatory goals. On this view, collective identity is a fiction—something that humans ascribe to a group—rather than something that exists independently. I argue that this Humean model has advantages over structural and intentional theories of group persistence. I then move onto the topic of social construction and its incompatibility with methodological individualism. …

Infants’ interactions with toys play a crucial role in their early cognitive and motor development, yet much of the research on this topic has been conducted in controlled laboratory settings, which may not accurately reflect the diversity of toys and interactions infants experience in their natural environments. This study uniquely examined these interactions in a home setting by utilizing an online video conferencing platform. This study investigated how toy properties influence manual infant-object interactions during dyadic tabletop free play, where mothers selected the toys for the session. Insecticide use has increased as population and agricultural demand for crops grows. Commercial insecticides contain active ingredients that are often harmful to humans and the environment. A viable alternative to this concern is developing and using insecticides with low toxicity and minimum risk to the environment. In this study, we sought to empirically assess the effectiveness of an insecticide based entirely on plant essential oil active ingredients, which contain complex secondary metabolites. Plant-Microbe Interactions Across Scales: Diversity, Disease, And Stress In Herbaceous Plant Communities, Philippa Tanford

Plants interact consequentially with microbes spanning mutualism to pathogenicity, with broader effects on large-scale plant community structure. These plant communties, in turn, shape below and aboveground microbial communities through a variety of mechanisms. However, plant diversity-microbial diversity relationships remain broadly inconsistent across environments and among different plant and microbial assemblages. In my dissertation, I work to bridge some of these gaps in plant-microbial ecology in three experimental chapters assessing relationships between plant diversity and microbial diversity, including the influence of belowground-aboveground microbiome interactions, the influence of drought stress, and incorporating different spatial and experimental scales. The language of subjective, individual rights has become the primary way of speaking about the rights and wrongs of human life together. Since the end of World War II, one subset of this language—that of human rights—has achieved a global influence. The various Christian traditions of the west have, in large part, adopted this language of human rights to articulate and supplement their assorted social teachings. Yet, the adoption of human rights language has been contested by a dedicated cadre of Roman Catholic and Protestant theologians concerned about the potentially anti-Christian implications inherent in the grammar of rights. The development of motor control over sensory organs is a critical milestone, enabling active exploration and shaping of the sensory environment. Whether the onset of sensory organ motor control directly influences the development of corresponding sensory cortices remains unknown. Here, we confirm and exploit the late onset of whisking behavior in mice to address this question in the somatosensory system. Using ex vivo electrophysiology, we describe a transient increase in the intrinsic excitability of excitatory neurons in layer IV of the barrel cortex, which processes whisker input, immediately following the onset of active whisking on postnatal days 13 and 14. …

Corticotropin-releasing hormone (CRH) is a peptide associated with stress and anxiety that acts as a potent modulator throughout the nervous system. The thalamic reticular nucleus (TRN) displays high expression of the CRH receptor 1 (CRHR1), but whether CRH modulates key TRN functions, such as sleep spindle rhythmogenesis, remained unexplored. Combining polysomnographic and photometric recordings in mice, we show that CRH release in TRN during non-rapid-eye movement sleep (NREMS) oscillates with a ~50-s periodicity, anti-correlating with sleep spindle dynamics. Optogenetic manipulations of CRH release in TRN modulated NREMS fragmentation through microarousals with corresponding changes in sigma and delta power. In ex-vivo …

Viral infection induces robust reprogramming of metabolic pathways in host cells. However, whether host metabolic enzymes detect viral components remains unknown. Our group and others previously identified O-GlcNAc transferase (OGT), an important glucose metabolic enzyme, as a crucial mediator of the antiviral immune responses. Here, by studying a mouse model with a catalytically impaired OGT, we discover a catalytic activity-independent function of OGT in restraining influenza A virus (IAV) infection in addition to its catalytic activity-dependent effect on MAVS-mediated antiviral immunity. Background/Objectives: Effective prophylaxis for Mycobacterium tuberculosis (Mtb) requires greater understanding of immune correlates of protection. With renewed interest in BCG as an Mtb vaccine, particularly via the intravenous (IV) route, our objective was to characterize both innate and adaptive immune correlates of vaccine-induced pulmonary immunity as potential biomarkers for protective efficacy in a murine model of Mtb infection. Methods: Mice were given BCG via different routes and some boosted with recombinant virus constructs encoding Mtb Ag85B.
