Rightsizing Regulations To Foster Innovation And A Healthy Business Environment: Insights From Elite Executives, Brian Charles Mulligan, Nelson Granados

This study examines insights from business executives on how TRAILS—that is, taxes, regulations, assessments, interventions, litigation, and subsidies—influence entrepreneurial behavior and firm performance. Using an applied constructivist grounded theory design, we conducted 17 in-depth interviews to identify how top business executives view the effects of regulations on business performance. We adopt the premise that the right level and set of regulations lead to a healthier business environment. We find that right-sized TRAILS catalyze entrepreneurial dynamism, whereas wrong-sized TRAILS impose hidden opportunity costs, stifle innovation, and erode institutional trust. Selection is a fundamental evolutionary force that shapes patterns of genetic variation across species. However, simulations incorporating realistic selection along heterogeneous genomes in complex demographic histories are challenging, limiting our ability to benchmark statistical methods aimed at detecting selection and to explore theoretical predictions. stdpopsim is a community-maintained simulation library that already provides an extensive catalog of species-specific population genetic models. Here, we present a major extension to the stdpopsim framework that enables simulation of various modes of selection, including background selection, selective sweeps, and arbitrary distributions of fitness effects (DFE) acting on annotated subsets of the genome (for instance, …

According to the World Health Organization (WHO), mosquitoes are the deadliest animals on Earth, responsible for more human deaths annually than any other species. Mosquito-borne illnesses continue to pose severe risks to global health. In 2015 alone, there were an estimated 214 million malaria cases worldwide. Similarly, a 2016 report from the Centers for Disease Control and Prevention (CDC) revealed that Puerto Rico’s Department of Health received over 62,500 suspected cases of Zika, with 29,345 confirmed positive cases. In 2019, Southeast Asia experienced its worst dengue outbreak in recorded history. Of the approximately 4,500 mosquito species distributed across 34 genera, …

Dopamine (DA) midbrain neurons are involved in a wide array of key brain functions including movement control and reward-based learning. They are also critical for major brain disorders such as Parkinson's disease or schizophrenia. DA neurons projecting to distinct striatal territories are diverse with regard to their molecular makeup and cellular physiology, which are likely to contribute to the observed differences in temporal DA dynamics. Among these regions, the dorsolateral striatum (DLS) displays the fastest DA dynamics, which might control the moment-to-moment vigor and variability of voluntary movements. While efforts are being made to curb the high rate of coastal land loss in Louisiana, and even to restore ecosystem services and functions, it is generally accepted that restoration of these areas and functions is not a one-to-one value. It may take decadal time scales to reach efficient functional processing of nutrients and carbon storage rates, which may never reach true capacity comparable to similar naturally-organized systems. Chromatin-based mechanisms contribute to the exquisite regulation of gene expression during animal development. But how those mechanisms evolved remains elusive. Here we investigate chromatin regulatory features in the closest relatives of animals, choanoflagellates. In a model choanoflagellate Salpingoeca rosetta, we compare chromatin accessibility and histone modifications to gene expression. Accessible genomic regions in S. rosetta primarily correspond to gene promoters, and we find no evidence of distal gene regulatory elements that resemble enhancers deployed to regulate developmental genes in animals. Remarkably, the histone modification H3K27me3 decorates genes with cell type-specific expression, revealing a functional similarity in S. rosetta and animals. …

During development, brain regions follow encoded growth trajectories. Compared to classical brain growth charts, high-definition growth charts could quantify regional volumetric growth and constituent cell types, improving our understanding of typical and pathological brain development. Here, we create high-resolution 3D atlases of the early postnatal mouse brain, using Allen CCFv3 anatomical labels, at postnatal days (P) 4, 6, 8, 10, 12, and 14, and determine the volumetric growth of different brain regions. We utilize 11 different cell type-specific transgenic animals to validate and refine anatomical labels. LED lighting has transformed city infrastructure with its efficiency and longevity, but its dispersed and highly diverging emission poses challenges like light pollution and artificial light at night (ALAN), which disrupts circadian rhythms in humans and animals. Volume or Bragg phase transmission holographic optical elements (HOEs) offer a low-cost solution to reducing LED light pollution in cities by efficiently shaping and redirecting light using thin, lightweight films and coatings. Scholars differ as to the degree to which Jewish law is formalistic. This chapter aims to make sense of the debate surrounding formalism in Jewish law by distinguishing between three types of legal formalism: classic formalism, modern formalism, and detail-following formalism. Viewed through the lens of modern formalism, Jewish law is likely less formalistic than commonly assumed as it is not primarily concerned with constraining decision makers. Nor is Jewish legal discourse typified by its unified conceptual schemas that proceed from abstract concepts to bottom-line decisions in the way that classical formalism proceeds. Mucopolysaccharidosis IVA (MPS IVA) is a lysosomal storage disease with an autosomal recessive trait caused by the deficiency of N-acetylgalactosamine-6-sulfate sulfatase (GALNS) enzyme, which leads to the accumulation of chondroitin-6-sulfate and keratan sulfate, primarily in cartilage and its extracellular matrix, resulting in a direct impact on cartilage and bone development, as well as subsequent systemic skeletal dysplasia. ERT and HSCT are current treatment options, but they have a limited effect on bone lesions. BACKGROUND: The attenuation of cellular phenotypic switchingdriving PAH vascular remodeling remains an unmet therapeutic need. As eNAMPT (nicotinamide phosphoribosyltransferase)/TLR4 signaling significantly contributes to PH pathobiology, an eNAMPT-neutralizing ALT-100 mAb was utilized to rescue monocrotaline (MCT) and hypoxia/Sugen (Hy/Su) preclinical PH rat models and to evaluate eNAMPT/TLR4 involvement in endothelial cell (EC), smooth muscle cell (SMC) and monocyte/macrophage phenotypic switching.

METHODS: MCT-PH or Hy/Su-PH rats received IgG or ALT-100 mAb (subQ, beginning week 4) with measurements of PH severity and lung tissue scRNAseq at day 42.

Nasal colonization by Staphylococcus aureus or Streptococcus pneumoniae is associated with an increased risk of infection by these pathobionts, whereas nasal colonization by Dolosigranulum species is associated with health. Human nasal epithelial organoids (HNOs) differentiated at air-liquid interface (ALI) physiologically recapitulate human nasal respiratory epithelium with a robust mucociliary blanket. Due to their natural stem-like properties, HNO lines are a long-term experimental resource that offers genetic diversity based on the different donors. Background: The rising prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD), particularly among pediatric populations, requires identification of modifiable risk factors to control disease progression. Per- and polyfluoroalkyl substances (PFAS) have emerged as potential contributors to liver damage; however, their role in MASLD remains underexplored. This study aimed to develop a translational framework integrating human and in vitro data to elucidate the effect of PFAS on MASLD development.

Fas-activated serine/threonine kinase (FASTK) proteins comprise one of the largest families of mitochondrial post-transcriptional regulators. Members are classified based on their conserved C-terminus, which shows homology with the PD-(D/E)XK superfamily of endoribonucleases. However, it is still uncertain which of these FASTK members are catalytic. The six human FASTK homologs rely on their RNA-binding activity to regulate distinct stages of mitochondrial gene expression, including early processing of nascent RNA, 3'-end messenger RNA (mRNA) maturation, ribosomal RNA (rRNA) modification, mRNA stability, and translation. Microglia are the resident immune cell of the brain, and alterations in microglia signaling have been implicated in many neurodegenerative disorders. While microglia responses to central cues and other brain cell types are well documented, studies are increasingly investigating the impact of peripherally derived signals on microglia function. A diverse array of peripheral cues, including dietary components, hormones, and bacteria metabolites and components from the microbiome cross the blood brain barrier and directly influence microglia state through ligand-receptor interactions. The Thai Journal of Veterinary Medicine

This study investigated the biofilm (BF) formation of a bovine-derived Escherichia coli (E. coli) strain (designated DL1907) and analyzed the factors influencing its formation. The BF-forming capacity, optimal formation time and temperature, as well as the effects of different culture media, pH levels, NaCl concentrations, and glucose concentrations, were assessed using the crystal violet method. The results demonstrated that the strain had a strong BF-forming capacity, achieving the most intact BF structure after 36 h of culture at 37°C. Trypticase Soy Broth (TSB) medium promoted the strongest BF formation compared to Luria-Bertani (LB) and Müller-Hinton Broth (MH) media. …

The Thai Journal of Veterinary Medicine

Cryptosporidium spp. is the most frequent agent in Algeria that causes neonatal diarrhea in calves, with high symptomatic diarrhea in pre-weaned dairy calves. The study aims to measure the prevalence, distribution, and genotype of Cryptosporidium-infected calves in one of Algeria's high milk production areas (North Central and Northeast). From 35 dairy cattle farms were selected 172 (77 with diarrheic and 95 without diarrheic symptoms) calves aged less than 60 days. Twenty-three farms (65.71%) were detected with cryptosporidiosis using a modified Ziehl-Neelsen stain, whereby 68 calves (39.53%) were obtained positive. PANDION: The Osprey Journal of Research and Ideas

This scientific illustration depicts the spiny orb weaver spider (Gasteracantha cancriformis), a distinctive arachnid known for its characteristic hardened, spiny abdomen. Spiny orb weaver spiders enjoy warm, tropical, and subtropical environments and are primarily found in the Southeastern U.S., the Caribbean, and South America. They live in wooded areas, citrus groves, and shrubby gardens. I’ve encountered spiny orb weaver spiders in my food forest where they like to set up their orb-shaped webs and feast on flies, mosquitoes, and moths.

Darkling beetles (Tenebrio molitor) are holometabolic insects. Darkling beetle is the common name of the large family of beetles Tenebrionidae which has more than 20,000 species. The T. molitor species gets its name from the yellowish appearance of the larvae. During its lifespan, a darkling beetle experiences a complete metamorphosis in four distinct stages: Egg, larva, pupa, and adult. The eggs hatch within 4 to 19 days after the female oviposits. It takes about 3 – 4 months for an egg to develop into an adult, or the beetle stage.

Horseshoe crabs (Limulus polyphemus) are fascinating animals known as “living fossils.” Older than dinosaurs, horseshoe crabs are arthropods, closely related to spiders and scorpions. Many shorebirds, including red knot, depend on the eggs of horseshoe crabs during migration. Horseshoe crabs are captured and bled alive for their blue blood. Rich in limulus amebocyte lysate (LAL), their blood is used for the Limulus Amebocyte Lysate (LAL) test, which is used worldwide to detect bacterial toxins in pharmaceuticals and medical devices.
