Limited studies have been conducted regarding the fecal contamination and microbial dispersal through rainwater, with no known studies investigating the potential connection between antibiotic resistant microbes and water quality in rainwater. This study seeks to identify the presence of a correlation between water quality and incidence of antibiotic resistance in rainwater microbes. We hypothesize that there will be a correlation between these two variables. We additionally anticipate a high incidence of fecal contamination across rural and urban collection sites alike, with fecal contamination being more prevalent in rural areas than urban locals. Early treatment of ischemic stroke can significantly reduce disability and mortality rates. Stem cell-derived extracellular vesicles (EVs) have shown potential as therapeutics for neurological disorders. This study explored whether intranasal administration of EVs from human bone marrow mesenchymal stem cells (BM-MSCs) enhances forelimb motor function recovery in a mouse model of motor cortex stroke and investigated their mechanism of action, focusing on neuroinflammation. C57BL/6JRj mice received EV treatment of 0.1 × 109 EVs per dose per day, 48 h post-stroke and twice weekly for four weeks. Sports betting among emerging adults (ages 18-29) is linked to financial harm, academic decline, and psychological distress. Responsible gambling (RG) messages aim to reduce such harms, yet generic slogans like “Gamble Responsibly” often lack credibility, especially when sponsored by gambling operators. Guided by social influence theory, this study examined how message framing (generic vs. self-appraisal) and sponsorship (operator vs. non-operator) affects trust, behavioral intentions, and gambling desire among emerging adult sports bettors. Struggle For Economic Plurality; Economies Of Belonging: Subsistence, Indigneity, And The Contradictions And Margins Of Capitalism In The Andean Highlands, Mario Renato Cavani Quevedo

This dissertation examines the collective-oriented economy of the Viacha Highlands, a Quechua community in Cusco, Peru, to show how subsistence agriculture, reciprocity, and ritual practices constitute a coherent non-capitalist system of provisioning and belonging. Drawing on one year of ethnographic immersion, the study situates Viacha’s economy within the Community Economies framework, foregrounding its three commitments: anti- essentialism, pluriversality, and ethical–political praxis. Findings reveal that practices such as ayni-ayni (reciprocal labor exchange) and faina (communal work) resist reduction to wage labor, coexist alongside monetary and capitalist dynamics, and ground social reproduction in responsibility and care. Sustainable agriculture practices promote cover cropping, which enhances nutrient management and impacts subsequent crop productivity in field crop rotations. Precision agriculture technology, particularly Unmanned Aerial Vehicle (UAV) remote sensing, enables a detailed evaluation of crop management. The studies were conducted in a maize-soybean rotation system with winter cover cropping and fallow management. The maize experiment investigated the effect of nitrogen application rates and cover cropping, using UAV-derived Normalized Difference Red-Edge (NDRE) to monitor crop performance. Periodic fire was a primary driver of ecosystems across the Southeastern United States for millennia; however, fire suppression policies have become pervasive in the United States and led to increased fuel loads in forests as well as reduced biodiversity and reduced disease resilience. Prescribed fire presents a risk mitigation strategy and land management tool whereby practitioners can apply fire to the landscape in a controlled manner, thus reaping the benefits of fire while minimizing the risks associated with fire. The 17F(α, p)20Ne reaction has been identified as an alternate pathway for breakout from the hot-CNO cycle that can be important in some X-ray burst scenarios. The 17F(α, p)20Ne reaction rate was previously determined through a parameterized exponential S-factor with a large enhancement from thermally-excited states based on statistical models with large uncertainties. We have measured the time-inverse 20Ne(p,α)17F cross section at center-of-mass energies ranging from 5.50 to 7.10 MeV using an activation method with a proton beam on a neon gas cell. Cantu syndrome (CS) is a rare disease caused by gain-of-function (GOF) mutations of Kir6.1 or SUR2 subunits of ATP-sensitive potassium (KATP) channels. CS patients with SUR2 and Kir6.1 variants display a similar constellation of symptoms, including muscle weakness and fatigue. The effects of CS mutations on skeletal muscle KATP channels, and any consequent direct effects on contractility, are currently unclear. Here, we used two knock-in mouse models of CS, respectively, carrying GOF mutations Kir6.1[V65M] or SUR2[A478V], to assess KATP channel properties and contractility in isolated fast-twitch extensor digitorum longus (EDL) and slow-twitch soleus (SOL) muscles. Approximately 2% of the population are people with intellectual disability (PwID). PwID are prone to multimorbidity, polypharmacy and premature mortality. Constipation is a prevalent and life-threatening issue for PwID fostered by iatrogenic harm, ignorance and stigma in family, and health and care settings. Despite its clinical significance, educational tools targeting caregivers’ knowledge of constipation prevention and management in PwID are scarce. Serious games (SGs) offer a novel and engaging platform to educate carers and reduce health stigma. This thesis considers how perceptual, emotional, and behavioral patterns—often inherited, unnoticed, or obscured—can be revealed through the optical phenomena of glass. It connects visual experience with themes of grief, memory, and identity, using glass to explore the boundary between what is seen and what is felt. Through material investigations in refraction, reflection, and spatial arrangement, the studio research examines how glass can both obscure and reveal, echoing the instability of perception itself. These investigations, paired with a focus on viewer interaction, suggest that patterns hiding in plain sight become visible through intentional acts of perception and reflection.

PURPOSE: Targeted therapies for metastatic prostate cancer are limited, highlighting the need for novel drug targets and mechanisms of action (MoA). Human kallikrein 2 (KLK2) is a prostate-specific antigen expressed across the prostate cancer disease continuum. However, it was not recognized as a therapeutic target for prostate cancer in the past due to limited evidence of its cell surface expression. In this study, we systematically characterized KLK2 expression in prostate cancer, confirmed its cell surface expression, and demonstrated the preclinical efficacy of three KLK2-targeting therapeutics with distinct MoA.

Mutations that initiate acute myeloid leukemia (AML) can cause clonal expansion without transformation (clonal hematopoiesis). Cooperating mutations, usually in signaling genes, are needed to cause overt disease, but these may require a specific fitness state to be tolerated. Here, we show that nearly all AMLs arising in a mouse model expressing 2 common AML-initiating mutations (Dnmt3aR878H and Npm1cA) acquired a single copy amplification of chromosome 7 (chr7), followed by activating mutations in signaling genes. We show that overexpression of a single gene on chr7 (Gab2, which coordinates signaling pathways) was tolerated in the presence of the Npm1cA mutation, could accelerate …

Systematic Review Of Elementary Cybersecurity Education: Curriculum, Pedagogy, And Barriers, Na Liu, Siyu Long, Florence Martin

Journal of Cybersecurity Education, Research and Practice

Abstract -As children increasingly engage with digital platforms, the need for effective cybersecurity education has become urgent. This systematic review synthesizes 81 studies published between 2017 and 2024 to examine global curricula research focus and topics, pedagogical approaches and assessment methods, and key challenges in elementary cybersecurity education. The findings reveal six major thematic categories: student awareness, parental mediation, teacher engagement, curriculum design, community and policy support, and pedagogical innovation. Among instructional strategies, game-based learning and narrative storytelling emerge as the most frequently explored. Despite this growth, major gaps remain in curriculum consistency, teacher preparation, assessment rigor, and stakeholder coordination. …

The sterile insect technique (SIT) reduces population numbers by releasing sterile males that produce non-viable progeny. Specifically, CRISPR/Cas9-based precision-guided SIT (pgSIT) generates sterile males through genetic crosses of two transgenic lines: a Cas9 strain and a guide RNA (gRNA) strain targeting male sterility and female viability or infertility. However, pgSIT requires separate maintenance of the two lines and sorting to obtain sterile males, creating possible challenges for scaling. To overcome this, we propose using Cas12a nuclease, which is inoperative at lower temperatures but active at higher temperatures. Purpose: Because surgery is the only potential cure for pancreatic cancer, high-risk premalignant pancreatic lesions often evade detection by palpation or white-light visualization, increasing the risk of recurrence. We asked whether near-infrared fluorescence imaging of tumor-associated inflammation could identify high-risk premalignant lesions, leveraging the tumor microenvironment as a sentinel of local disease and, thus, enhance surgery outcomes.

Triple-negative breast cancer (TNBC) that fails to respond to neoadjuvant chemotherapy (NACT) can be lethal. Developing effective strategies to eradicate chemoresistant disease requires experimental models that recapitulate the heterogeneity characteristic of TNBC. To that end, we established a biobank of 92 orthotopic patient-derived xenograft (PDX) models of TNBC from the tumors of 75 patients enrolled in the ARTEMIS clinical trial (NCT02276443), including 12 longitudinal sets generated from serial patient biopsies collected throughout NACT treatment and from metastatic disease. Understanding how sharks respond to catch and release (C&R) recreational fisheries is an important requirement for effective management. Post-release survival and recovery in recreational fisheries can be species- and setting-dependent. Nevertheless, no study on sharks has been conducted in European recreational fisheries, despite their popularity as target. This study presents the first example of biologging to assess C&R effects on blue sharks (Prionace glauca), porbeagle sharks (Lamna nasus) and tope sharks (Galeorhinus galeus) in the UK. Leptomeningeal metastases are the most important source of morbidity and mortality for medulloblastoma patients. Radiation of the entire brain is highly effective in the treatment and/or prevention of medulloblastoma leptomeningeal metastases. Infants treated on clinical trials with focal tumor radiation recur metastatically, whereas infants treated with only chemotherapy relapse locally. In murine medulloblastoma model systems, provision of a single dose of radiation to the tumor drives leptomeningeal dissemination. An inflammatory response after radiation-induced tumor cell death recruits a variety of immune cells. Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer with few effective targeted therapies. Taxanes and other microtubule-targeting agents (MTAs) are frontline chemotherapies for TNBC; however, the molecular pathways that cause TNBC taxane sensitivity are largely unknown, preventing selection of taxane-responsive patients and development of more selective therapeutic strategies. In this study, we identified tumor-selective vulnerabilities in TNBC harboring inactivation of the tumor suppressor PTPN12 by integrating proteogenomic characterization and synthetic lethality screening. We discovered that PTPN12 inactivation drives mitotic defects through aberrant hyperactivation of the ubiquitin ligase complex APCFZR1, a critical regulator of the cell cycle. Consistent …

Calcium ions (Ca2+) are essential second messengers intimately implicated in a variety of biological processes, ranging from short-term events such as muscle contraction to long-term effects like gene expression. Dysregulated Ca2+ signaling can disrupt cellular function and contribute to the development of various human diseases, including developmental, neurological, immunoinflammatory, metabolic, and cardiovascular disorders. To study the mechanisms and biological consequences of Ca2+ signaling, optogenetic approaches have proven invaluable, as they offer exceptional spatiotemporal resolution compared to traditional methods.
