Successful delivery of plasmid DNA into the microbial cells is fundamental in recombinant DNA technology. Natural bacterial transformation is limited to only certain species due in part to the repulsive forces between negatively charged DNA and bacterial membranes. Most common method of DNA delivery into bacteria is artificial transformation through heat shock and electroporation. These methods require sophisticated instruments and tedious steps in preparation of competent cells. Transformation by conjugation is also not applicable to all plasmids. Nanoparticles have been used successfully in therapeutics for drug delivery into animal cells. They are starting to gain popularity in plant sciences as …

My thesis project research consisted of single-cell RNA-sequencing and bioinformatics using R programming and Neuroscience Multi-omic Data Archive (NeMO) Analytics to examine the impact of aging, brain region, and other variables on the genotypes and phenotypes of oligodendrocytes (OLGs) and oligodendrocyte precursor cells (OPCs). The overarching goal was to increase understanding of glial cell fate and function. After determining bioinformatical findings, follow-up wet-bench experiments were performed to validate results. RNA in situ hybridization and immunohistochemistry were performed to confirm differential gene expression across cell types and brain regions. Hydrolyzed feather meal (HFM) is a feedstuff high in rumen undegraded protein (RUP) that can be used as an effective source of metabolizable protein for dairy cattle. Because the production process may vary, the rumen degradability and intestinal digestibility of HFM may also vary. Additionally, some processes may incorporate additional blood into the final product to result in feather meal with poultry blood. To determine the rumen degradability and intestinal digestibility of these products, several laboratory assays can be used; the common assays are the mobile bag (MOB), modified three-step (MTS), and Ross (ROS) assays. There is very little research that explores the public attitude toward marine biodiversity loss as an environmental issue. This study aims to provide insight into the disparities in marine conservation education in order to better provide meaningful education opportunities for coastal communities. The data for this research was collected using a series of semi-structured interviews that were conducted as focus groups of participants from Horry County, South Carolina. Residents from North Myrtle Beach, Myrtle Beach, Loris, and Bucksport were recruited to three focus groups based on their geographical residency. Consultation/liaison psychiatrists care for people with co-existing medical and psychologic difficulties. The coronavirus pandemic is stressful for patients and their psychiatric caregivers. Patients have lost support systems and harbor fears about exposing family to the virus. COVID-19 sometimes exacerbates previous psychiatric conditions, while diminishing intimacy with physicians. Everyone is oversaturated with COVID-19-related news focusing on rising concerns about the illness and about jobs, school, and housing insecurities.

Early-onset heart failure (HF) continues to be a major cause of morbidity and mortality in people living with human immunodeficiency virus type one (HIV-1) infection (PLWH), yet the molecular causes for this remain poorly understood. Herein NOD.Cg- PrkdcscidIl2rgtm1Wjl/SzJ humanized mice (Hu-mice), plasma from PLWH, and autopsied cardiac tissues from deceased HIV seropositive individuals were used to assess if there is a link between the glycolysis byproduct methylglyoxal (MG) and HF in the setting of HIV-1 infection. The piece highlighted that AMR should be addressed in the G7 Summit agenda and that our world leaders must recognize that research and investment into feasible solutions are urgently needed. Heparin-induced thrombocytopenia (HIT) is associated with severe and potentially lethal thrombotic complications. NETosis was recently shown to be an important driver of thrombosis in HIT. We investigated the role of reactive oxygen species (ROS) and nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 2 (NOX2) and their contributions to thrombus development in HIT. We showed that neutrophil activation by HIT immune complexes induced ROS-dependent NETosis. Analysis of thrombi formed in a microfluidics system showed ROS production in both platelets and neutrophils, and abundant neutrophil extracellular traps (NETs) and ROS distributed throughout the clot. Neutrophil-targeted ROS inhibition was sufficient to block HIT-induced NETosis …

Objectives: This study aimed to explore mechanism of colistin resistance amongst Klebsiella pneumoniae isolates through plasmid mediated mcr-1 gene in Pakistan. Carbapenem and Colistin resistant K. pneumoniae isolates (n = 34) stored at - 80 °C as part of the Aga Khan University Clinical Laboratory strain bank were randomly selected and subjected to mcr-1 gene PCR. To investigate mechanisms of resistance, other than plasmid mediated mcr-1 gene, whole genome sequencing was performed on 8 clinical isolates, including 6 with colistin resistance (MIC > 4 μg/ml) and 2 with intermediate resistance to colistin (MIC > 2 μg/ml).

Objective: Patients with rheumatoid arthritis (RA) often suffer from bone complications due to persistent joint inflammation, especially incident fracture. Nowadays, Chinese herbal medicines (CHMs) have provided safe and effective therapy for treating skeletal conditions, but it is unclear whether CHMs can prevent fracture onset among RA individuals. This study aimed to determine the association between the use of CHMs and the risk of fracture among them.

Somatic mutations are rare in pediatric acute myeloid leukemia (pAML), indicating that alternate strategies are needed to identify targetable dependencies. We performed the first enhancer mapping of pAML in 22 patient samples. Generally, pAML samples were distinct from adult AML samples, and MLL (KMT2A)-rearranged samples were also distinct from non-KMT2A-rearranged samples. Focusing specifically on superenhancers (SEs), we identified SEs associated with many known leukemia regulators. The retinoic acid receptor alpha (RARA) gene was differentially regulated in our cohort, and a RARA-associated SE was detected in 64% of the study cohort across all cytogenetic and molecular subtypes tested. The type and size of equipment used affects the harvested yield and nutritive value of the forage crop as well as production costs. Through interactions with other parts of the farm, these effects can impact market value of the forage, animal intake and performance, delays in other farm operations, other production costs, and ultimately farm profit. Forage conservation permits a better supply of quality feed when forage production is low. While haymaking and ensiling have been practiced for generations, research is still needed to 1) understand the processes affecting quality during harvesting and storage and 2) develop practical means to minimize losses and enhance quality.
Several trends in harvesting forages for silage are notable. Kernel processing of maize, once confined to Europe, has become popular in North America.
