This research explores Isothermal Titration Calorimetry as a method for measuring beta-galactosidase activity directly and continuously in milk, sweet whey, sweet whey permeate, acid whey, and acid whey permeate. Beta-galactosidase in various concentrations was injected into each of the liquid dairy products spiked with lactose to verify if the heat rate from the enzymatic reaction could be observed. In addition, a consistent concentration of beta-galactosidase was injected into various concentrations of lactose in the products, to observe the heat rates from the enzymatic reaction. After groundbreaking work by multiple archaeologists in the latter half of the 20th century, caves in the Maya world are currently acknowledged as fundamentally ritual rather than domestic spaces. However, a more nuanced read of the anthropological literature and conversations with Indigenous collaborators in the past and present pushes us to move still farther and see caves not as passive contexts to contain ceremonies directed elsewhere but animate beings with unique identities and personalities in their own right. Mixed-conifer forests play an important role in the Southwestern USA’s regional carbon cycle by sequestering and storing large amounts of atmospheric carbon onto the landscape. Despite this, little is known about how large, stand-replacing fire alters ecosystem carbon sink-source dynamics. In this study, we used eddy covariance to quantify how stand-replacing fire altered forest sink strength, and the sensitivity of carbon uptake to soil and atmospheric drought in a mixed-conifer forest located in The Valles Caldera National Preserve, NM. We found that the post-burn ecosystem returned to an annual net sink of carbon in the year immediately following the fire, …

Recovery environments are a crucial role in any individual’s journey to achieve sobriety. A safe environment will allow people who have a history of substance use to work their program effectively and decrease their relapse potential. The purpose of this paper is to identify multiple recovery-based programs for new professionals and providers entering the field of substance abuse treatment that allow their clients to have the best opportunity to succeed with their personal goals. Disease lesion mimic mutants (DLMMs) are characterized by the spontaneous development of necrotic spots with various phenotypes designated as necrotic (nec) mutants in barley. The nec mutants were traditionally considered to have aberrant regulation of programmed cell death (PCD) pathways, which have roles in plant immunity and development. Most barley nec3 mutants express cream to orange necrotic lesions contrasting them from typical spontaneous DLMMs that develop dark pigmented lesions indicative of serotonin/phenolics deposition. Barley nec3 mutants grown under sterile conditions did not exhibit necrotic phenotypes until inoculated with adapted pathogens, suggesting that they are not typical DLMMs. The F2 progeny …

Elephants have shown remarkable olfactory capabilities. Their sense of smell impacts their foraging choices, behavior, and ultimately, survival. Being able to detect a target odor can allow elephants to locate specific resources, identify threats, and find receptive conspecifics. Previous studies have shown that elephants can consistently detect target odors, but have not identified the limits of this detection. Thus, to investigate the extent of elephants’ odor detection capabilities, we tested Asian elephants in a two-step odor discrimination task. Habitat fragmentation is a common result of urbanization and species living in these fragments are at risk of extirpation. We conducted a 12-y (2008–2019) capture-mark-recapture study on snakes living in a 593-m section of Rock Castle Creek flowing through an urban area in central Virginia, USA. Our study site was occupied primarily by Queensnakes (Regina septemvittata). We used data from 168 individual Queensnakes to examine several aspects of their population ecology including survival rates (0.52), reproductive effort (21.3% juvenile to adult ratio), growth rates (68% and 30.6% increase for 1 to 2 y-old and 2 to 3+ y-old snakes, respectively), …

This thesis discusses the history of catharanthine and related compounds, and their potential anti-addictive properties. Current research is exploring possible mechanisms of these properties. Past studies have found catharanthine has effects on neurons that project to the mesocorticolimbic system, an area implicated in addiction. We have seen that catharanthine decreases evoked dopamine (DA) release but increases basal DA release. This is the first study to investigate catharanthine’s effect on DA transmission in vivo. Green roofs can provide food resources to several insect groups. For pollinators found in cities, as are wild bees and hoverflies, the existence of a wide variety of green infrastructures is crucial to ensure their development and survival. In order to investigate if wild bees and hoverflies use green roofs and how local and landscape factors influence their abundance and diversity, sampling of these insects was done in 2017 using cornet traps on extensive green roofs of two types: 1) urban green roofs (30% of green spaces in a 200m radius). Infectious disease is a primary source of mortality for most mammal species, but scientists have little understanding of factors driving variation in infection and immunity between individuals, populations, and species in the wild. Life history theory provides an evolutionary framework for delineating distribution of available energy to competing physiological demands, including growth, reproduction, and maintenance. Early life reproduction should be favored over late life survival, but, in long-lived species, reproductive success is strongly tied to survival to old age. A myriad of genetic and other abnormal changes underlies the aggressiveness and dissemination properties observed in pancreatic cancer (PC). Aberrant protein glycosylation is a commonly observed feature in PC. The modification of protein O-glycosylation is mediated by glycosyltransferases, which attach and sequentially elongate monosaccharides on Serine/Threonine (Ser/Thr) motifs. Aberrant glycosylation is recognized as an emerging hallmark of cancer where a disruption in normal glycosylation results in irregular O-glycans.

Human-environment interaction is a long-standing and productive line of inquiry that includes the study of cultural responses to climate change. However, demonstrating a causal relationship between climate change and the consequent culture change is rarely straightforward for numerous reasons. In this dissertation I develop an optimality model to predict how Paleoindians should have responded to changes in precipitation across the Late Pleistocene-Early Holocene. The water supply model (WSM) ranks water supplies much like the diet breadth model ranks food resources. Background and Objectives: Fluoroquinolones (FQs) are a broad-spectrum class of antibiotics routinely prescribed for common bacterial infections despite recent recommendations to use them only for life-threatening cases. In addition to their antimicrobial properties, FQs act in the central nervous system as GABAA receptor inhibitors, which could potentially affect functionality of the vagus nerve at the forefront of gastrointestinal (GI) tract function. This study explored teaching preschool age students how to use yoga strategies to calm their bodies, reducing the number of times a student demonstrates negative behavior. The fifteen participants were three through five year olds from one of the preschool classrooms at the Minnesota State University Moorhead (MSUM) Early Learning Center. Baseline data was collected for five days, resulting in a determination that four of the students would be considered the subjects for the study, one who received special education services. Learning how to read fluently was a goal the researcher had which led to the research question; How does the intervention of repeated reading impact oral reading fluency rates in students who are reading below a 2nd grade level? The purpose of the action research was to implement the intervention of repeating reading to see if the oral reading fluency rates in struggling students increased. Struggling readers were identified through the Dynamic Indicators Of Basic Early Literacy Skills assessment (DIBELS). This article critically examines autism and autistic stimming through the lenses of neurotypicals and autistics, evaluates confines currently imposed upon autistics by neurotypical society, and synthesizes what autistic leaders have expressed about their experiences striving for inclusion (Gernsbacher, 2017; Milton, 2012). Autistic-led studies are now at the forefront of the neurodiversity movement and are growing in fervor. The number of sequenced bacteriophage genomes is growing at an exponential rate. The majority of sequenced bacteriophage genomes are annotated by one or more of several freely available gene identification programs (Glimmer, GeneMark, RAST, Prodigal, etc.). No program has been shown to consistently outperform the others; thus, the choice of which program to use is not obvious. We present the Phage Commander application for rapid identification of bacteriophage genes using multiple gene identification programs. Carbon-rich habitats can provide powerful climate mitigation if meaningful protection is put in place. We attempted to quantify this around the Tristan da Cunha archipelago Marine Protected Area. Its shallows (<1000 m depth) are varied and productive. The 5.4 km2 of kelp stores ~60 tonnes of carbon (tC) and may export ~240 tC into surrounding depths. In deep-waters we analysed seabed data collected from three research cruises, including seabed mapping, camera imagery, seabed oceanography and benthic samples from mini-Agassiz trawl. Rich biological assemblages on seamounts significantly differed to the islands and carbon storage had complex drivers. We estimate ~2.3 million tC are stored in benthic biodiversity of waters <1000 m, which includes >0.22 million tC that can be sequestered (the proportion of the carbon captured that is expected to become buried in sediment or locked away in skeletal tissue for at least 100 years). Much of this carbon is captured by cold-water coral reefs as a mixture of inorganic (largely calcium carbonate) and organic compounds. Human infrastructure, particularly highway infrastructure, has proven to be a hazard for the resilience of large felid populations throughout the world. The Yucatan Peninsula is no exception and is presently confronted with the establishment of the “Tren Maya,” a continental railway that will reshape the road network of the peninsula. We describe 12 successful crossings of a 4-lane highway on the peninsula by a female puma (Puma concolor) fitted with a GPS radio-transmitter. Robotic odor source localization (OSL) is a technology that enables mobile robots or autonomous vehicles to find an odor source in unknown environments. It has been viewed as challenging due to the turbulent nature of airflows and the resulting odor plume characteristics. The key to correctly finding an odor source is designing an effective olfactory-based navigation algorithm, which guides the robot to detect emitted odor plumes as cues in finding the source. This dissertation proposes three kinds of olfactory-based navigation methods to improve search efficiency while maintaining a low computational cost, incorporating different machine learning and artificial intelligence methods.

Background:
Mobile health (mHealth) has the potential to bring preventive healthcare within reach of populations with limited access to preventive services, by delivering personalized support at low cost. Although numerous mHealth interventions are available, very few have been developed following an evidence-based rationale or have been tested for efficacy. The issue that this study addresses is to observe whether there exists a statistical relation between the stock market and Reddit’s wallstreetbets. Previous research mainly focused on the relation between the stock market and Twitter. To gather data for the study, comments were scrapped from the subreddit wallstreetbets for a period of four months, Jan 1, 2021, till April 30, 2021.
