The goal of this research project was to develop an infant air-jet dry powder inhaler (DPI) platform for high-efficiency nose-to-lung (N2L) aerosol administration to infants through optimization of individual system components followed by expansion of the system to higher doses and different formulations using a new approach. Primary challenges associated with aerosol delivery to infants using the N2L route include the extremely narrow extrathoracic airways, short inspiratory times (~0.2 sec), low safe airway pressure (H2O), and very low volumes of air (~10 mL) available to form the aerosol. The epidemic of drug use in the United States and elsewhere in the world has resulted in tragic loss of life and substantial economic costs. Novel psychoactive substances (NPS) are one of the contributors to this problem, and the lack of information about many of these drugs compounds their risk. This dissertation proposes a strategy to use intracranial self-stimulation (ICSS) as a behavioral screening tool to assess emerging drugs of abuse for their abuse potential in order to generate a proactive threat assessment. Amyotrophic lateral sclerosis (ALS) is a late-onset fatal neurodegenerative disease that causes loss of upper and/or lower motor neurons, and currently has no treatment or cure available. Over 90% of cases occur spontaneously with unknown causes, highlighting the complexity of the disease, and only 10% of cases are linked to heritable genetic mutations. Numerous ALS-linked genes are conserved through evolution, and model organisms may therefore provide opportunities to understand disease pathology at a molecular or cellular level, proving instrumental in identifying therapeutic targets. Utilizing the methodology of participant observation combined with semi-structured interviews, this ethnographic study aims to analyze the socio-historical development of the Richmond chapter of the Virginia Caucus of Rank-and-file Educators (VCORE), a left-wing opposition group inside the Virginia Education Association (VEA). This study aims to assess VCORE’s politics, origins, growth, transformation, organizational structure, and cultural practices, focusing upon the role VCORE members played in the lead-up to and aftermath of the 2020-2022 campaign to reinstate collective bargaining rights of public education employees in Richmond.

Odor perception is the impetus for important animal behaviors, most pertinently for feeding, but also for mating and communication. There are two predominate modes of odor processing: odors pass through the front of nose (ortho) while inhaling and sniffing, or through the rear (retro) during exhalation and while eating and drinking. Despite the importance of olfaction for an animal’s well-being and specifically that ortho and retro naturally occur, it is unknown whether the modality (ortho versus retro) is transmitted to cortical brain regions, which could significantly instruct how odors are processed. The ventral tegmental area (VTA) is well known for regulating reward consumption, learning, memory, and addiction behaviors through mediating dopamine (DA) release in downstream regions. Other than DA neurons, the VTA is known to be heterogeneous and contains other types of neurons, including glutamate neurons. In contrast to the well-studied and established functions of DA neurons, the role of VTA glutamate neurons is understudied, presumably due to their relatively small quantity and a lack of effective means to study them. In this work, we integrated a commercially-available fully-spectroscopic pixelated cadmium telluride (CdTe) detector system as a two-dimensional (2D) array detector into our existing benchtop cone-beam x-ray fluorescence computed tomography (XFCT) system. After integrating this detector, known as High-Energy X-ray Imaging Technology (HEXITEC), we performed quantitative imaging of gold nanoparticle (GNP) distribution in a small animal-sized phantom using our benchtop XFCT system. Owing to the upgraded detector component within our benchtop XFCT system, we were able to conduct this phantom imaging in an unprecedented manner by volumetric XFCT scans followed by XFCT image reconstruction in 3D. Median survival of patients with glioblastoma (GBM) treated with standard of care which consists of maximal safe resection of the contrast-enhancing portion of the tumor followed by radiation therapy with concomitant adjuvant temozolomide (TMZ) remains 15 months. The tumor microenvironment (TME) is known to contain immune suppressive myeloid cells with minimal effector T cell infiltration. Stimulator of interferon genes (STING) is an important activator of immune response and results in production of Type 1 interferon and antigen presentation by myeloid cells. Transcriptional and proteomic profiling of individual cells have revolutionized interpretation of biological phenomena by providing cellular landscapes of healthy and diseased tissues1,2. These approaches, however, do not describe dynamic scenarios in which cells continuously change their biochemical properties and downstream 'behavioural' outputs3-5. Here we used 4D live imaging to record tens to hundreds of morpho-kinetic parameters describing the dynamics of individual leukocytes at sites of active inflammation. By analysing more than 100,000 reconstructions of cell shapes and tracks over time, we obtained behavioural descriptors of individual cells and used these high-dimensional datasets to build behavioural landscapes. Background: Proton pump inhibitors (PPIs) have been shown to regulate the gut microbiome and affect the response to immune checkpoint inhibitors (ICIs). Contradictory results on survival have been observed in patients concomitantly treated with ICIs and PPIs. We performed a systematic review and meta-analysis to determine the association between PPI use and survival outcomes in ICI-treated cancer patients.
Methods: EMBASE, MEDLINE/PubMed, Cochrane Library databases, and major oncology conference proceedings were searched. Base excision repair (BER) has evolved to maintain the genomic integrity of DNA following endogenous and exogenous agent induced DNA base damage. In contrast, aberrant BER induces genomic instability, promotes malignant transformation and can even trigger cancer development. Previously, we have shown that deoxyribo-5'-phosphate (dRP) lyase deficient DNA polymerase beta (POLB) causes replication associated genomic instability and sensitivity to both endogenous and exogenous DNA damaging agents. Specifically, it has been established that this loss of dRP lyase function promotes inflammation associated gastric cancer. The ecosystem services approach seeks to recognize the benefits that people obtain from ecosystems, in order to highlight society’s dependence on natural systems. Ecosystem services fall into four categories: provisioning, regulating, cultural, and supporting services. Most of the ecosystem services research literature is focused on services that arise from biotic components of ecosystems. Ecosystems are dynamic complexes of biotic communities and their physical, non-living environment, interacting as a functional unit. The emphasis on biotic ecosystem services leaves understudied and undervalued the role of the abiotic, physical environment, especially in providing geologic ecosystem services. Glioblastoma (GBM) is a high-grade, aggressive brain tumor with dismal median survival time of 15 months. Chromosome 6q (Ch6q) is a hotspot of genomic alterations, which is commonly deleted or hyper-methylated in GBM. Two neighboring genes in this region, QKI and PRKN have been appointed as tumor suppressors in GBM. While a genetically modified mouse model (GEMM) of GBM has been successfully generated with Qk deletion in the central nervous system (CNS), in vivo genetic evidence supporting the tumor suppressor function of Prkn has not been established. The phylum Bryozoa is an incredibly diverse group of marine invertebrates with a widespread global distribution that is well suited for evolutionary studies but whose phylogenetic relationships are still poorly understood. Although recent studies on bryozoan taxonomies and phylogenies have increased, there is still a lack of assessment of species found at shallow water (<1 m) to intertidal depths. In this study, I aimed to expand the taxonomic sampling and assessment of the phylogenetic diversity of cheilostome bryozoans along the California coastline by utilizing mitochondrial DNA as well as inferring potential correlations between species presence and dispersal range both within and between rocky outer coast and sheltered harbor habitats. Illumina high-throughput sequencing was used to produce mitogenomes for cheilostome bryozoan samples collected off rocks from two rocky intertidal sites and off settlement panels from two harbor sites. Phylogenetic analyses generated evolutionary hypotheses of species relationships alongside geographic mapping of their distribution. This study identified 15 distinct species that represent 10 different families to form the first comprehensive phylogeny for multiple bryozoan families in California across a total range of approximately 973 km of coastline. Three genetically distinct species were found at multiple sites that are separated by a combination of rocky shores and sandy beaches, which indicates that the dispersal range of these species are not limited by geographic barriers along the coast of California. These results provide a future opportunity for further integration of this data with the phylogenies generated in this study to examine more robust evolutionary hypotheses for the phylogenetic and geographic relationships of Californian bryozoan species.

The Slater Fire of 2020 burned in Karuk aboriginal territory overseen by the Klamath National Forest. It burned over 200 homes to the ground and ravage over 100,000 acres of forest. This thesis argues that state-enforced fire suppression policies and methods are tools of settler-colonial erasure and the continuation of genocidal violence towards Karuk people. It analyzes the conflict between interests of the colonial state on one side and Indigenous resistance and survival on the other. Marine protected areas (MPAs) were created with the purpose of helping conserve and restore diminished populations of marine organisms. Measuring the effectiveness of MPAs requires long-term monitoring, investigating the abundance and size distributions of the species that utilize the conservation areas, and comparing the results to neighboring reference sites that are not currently protected. Bisphenol A (BPA), a manufactured compound found in consumer products, is known to adversely affect early brain development by disrupting normal estrogen signaling. Recently, BPA was reported to suppress expression of a gene encoding the neuron-specific chloride ion transporter, KCC2. Human and animal studies show suppressing KCC2 can cause neuronal and behavioral hyperactivity. Therefore, some adverse effects of BPA may be due to KCC2 suppression and consequent neuronal hyperactivity. This study aimed to determine whether BPA exposure during brain development alters KCC2 expression. Using hermeneutical methodology, this paper examines some of the legal fictions that form the foundation of Federal Indian Law. The text of the U.S. Supreme Court’s 1823 Johnson v. M’Intosh opinion is evaluated through the lens of the Convention on the Prevention and Punishment of the Crime of Genocide to determine the extent to which the Supreme Court incorporated genocidal principles into United States common law. The genealogy of M’Intosh is examined to identify influences that are not fully apparent on the face of the case. Roosts, locations at which birds and other animals come together to rest and preen while remaining vigilant for predators, are a common facet of the nonbreeding biology of many shorebirds. Roosts fall on a continuum from traditional to ephemeral; traditional roosts are used consistently by the same individuals, whereas ephemeral roosts are used inconsistently and may be used only once or a few times. Studies show that roost habitat preference is the result of an individual shorebird’s attempt to optimize the costs and benefits associated with time constraints, energetic demands, and danger risks as they relate to overall fitness.

The King Range colony is the northernmost successful and expanding breeding site for northern elephant seals (Mirounga angustirostris). To evaluate the composition and growth of this colony, I conducted systematic surveys recording counts, age classes, births, and resighting seals with individually identifiable flipper tags. The timing of life-history events (breeding, molting, and resting) of all elephant seal sex and age groups at the King Range was consistent with observations at other colonies. Colony counts during breeding and molting have increased steadily over the 4-year study period (2018-2021), and pup production has increased by 87% since the first year …

Advancements in bioacoustics field studies have further elucidated spatial, temporal, and behavioral aspects of otherwise-cryptic species, as well as offering insights into species communication. The discovery of high-frequency vocalizations in North American flying squirrels in particular has allowed researchers to use ultrasonic acoustic recorders to detect these cryptic species in the wild. Investigations into vocalizations of northern flying squirrels (Glaucomys sabrinus) and southern flying squirrels (G. volans) have highlighted call type variation between species and the potential use of recorders as a tool to examine vocal activity patterns. This thesis explored the feasibility of electrifying a food market in an urban city in Nigeria with a hybrid biogas-powered mini-grid. Under the Energizing Economies Initiative of the Rural Electrification Agency of Nigeria, nine markets in the country currently receive constant access to electricity via hybrid mini-grid systems. As a majority of these systems are diesel-solar-battery systems, this thesis explored the use of biogas generators as a substitute for diesel generators in hybrid mini-grids for food markets. The Aleutian cackling goose (Branta hutchinsii leucopareia) experienced a population low of 790 geese in 1974 and has recovered to the current estimate of 186,100 geese in 2021. Private livestock producers view geese as pasture competitors and use hazing and hunting to scare geese from private lands to adjacent public lands. I investigated if geese provide an ecosystem service via dropping fertilizing effects to improve pastures in northern California.

Juvenile Chinook Salmon (Oncorhynchus tshawytscha) migrating through the Sacramento – San Joaquin Delta (the Delta) in central California have higher mortality rates than salmonids migrating through other west coast estuaries. Some hypotheses for high mortality rates include entrainment into water export facilities, physical alterations of the river system, and predation. Of these factors, predation is considered the least understood but may have the largest impact. Predation from large populations of non-native piscivorous fishes in the Delta is believed to effect abundances of Central Valley Chinook salmon, however sufficient data supporting this hypothesis is scarce.
