It is well known that microplastics are ubiquitous within the marine environment and that organisms are interacting with them. Advancements in technology have made possible the development of highly personalized and efficacious medicines. Various forms of immunotherapy have demonstrated clinical utility in the treatment of blood-based cancers (e.g., leukemia, lymphoma, etc.). Cells in the tumor microenvironment (TME) of solid tumors can suppress the immune response of T cells, therefore hindering the efficacy of T-cell immunotherapies. Consequently, a growing number of studies are investigating methods of modulating the TME for improving immunotherapeutic outcomes in this context. Exposure to per- and polyfluoroalkyl substances (PFAS) has been associated with toxicities in wildlife and negative health effects in humans. Decades of fire training activity at Joint Base, Cape Cod, Massachusetts incorporated the use of aqueous film forming foam (AFFF) which resulted in long-term PFAS contamination of sediments, groundwater, and hydrologically connected surface waters. To explore the bioaccumulative potential of PFAS residing in complex environmental mixtures, a mesocosm was established to evaluate bioconcentration of PFAS from AFFF-contaminated groundwater by flow-through design. This thesis reports on the implementation of a Genetic Algorithm for finding heuristic solutions to the Capacitated Vehicle Routing Problem (CVRP) on High-Performance Field Programmable Gate Arrays (FPGAs). The heuristic approach to the class of NP-hard problems related to the Travelling-Salesman-Problem (TSP) has a long tradition in computer science, with this thesis being directly based on an implementation of a specialized Genetic Algorithm for a Graphic Processing Unit (GPU). With the rise of offshore wind energy in the United States, there has been increasing concern about how offshore wind turbine installation and operation is affecting benthic and pelagic marine life (Bailey et al. 2010). Increased, localized ocean noise resulting from project-related activities, such as vessel traffic, surveying, and turbine installation, has the potential to disturb or injure nearby marine life. Food sovereignty emphasizes the rights of people to have control over their food system. Relocalizing food production and limiting dependence on global industrial supply chains is necessary to increase food sovereignty in regions dominated by conventional food systems. In the US, the New England region aims to relocalize production by producing 50 percent of food consumed in the region by the year 2060. Rhode Island can contribute to this goal by expanding production and strengthening local food systems, thus serving as a model for New England and other urbanizing regions globally. When seeking information on how to clean away mold from clothing and textiles, searching the internet can lead to an overwhelming amount of confusing and vague information. A lack of reputable references and consistent measurements across methodologies makes attempting to clean textiles, valuable or not, potentially damaging to the textile itself. Background: Renal ischemia-reperfusion injury (IRI) causes acute kidney injury, as well as liver injury resulting in altered hepatic metabolism. Decreased hepatic cytochrome P450 levels can impact drug clearance and increase the risk for adverse drug reactions. Currently, there is no treatment available to improve liver cyp450 metabolism during renal IRI. Treprostinil (Remodulin®) is a FDA approved prostacyclin analog that our lab has shown to have hepatoprotective effects during renal IRI. This study investigates the efficacy of treprostinil on improving hepatic cyp450 metabolism during renal IRI in vivo.

Understanding neural networks has been challenging since their inception. Neural networks are non-linear and learn representations without the user defining them, making the internal workings obscure. Methods for understanding networks have been researched for many years. This study uses centered kernel alignment (CKA) to measure the similarity between the representations of data between layers in a network. The method is invariant to invertible orthogonal transformation, invariant to isotropic scaling, and exhibits behavior demonstrating its superiority over other methods.

During the expeditions, thousands of specimens were collected of several taxa, including mammals. As far as we know, no complete summary of the expeditions and the mammals collected has been created. Knowledge of the expeditions and evaluation of the mammals collected compared to current literature could have conservation implications. Many pet owners feel strongly about their animals. Some feel so strongly that they desire their pets to accompany them to the grave. This Article addresses the validity of pet euthanasia provisions in decedents’ wills.
Pet owners generally have the legal power to humanely euthanize their pets. In addition, the primary focus of the law of wills is to effectuate the wishes of the decedent. These two facts seem to counsel in favor of carrying out a testamentary instruction to humanely euthanize a companion animal. The generally-accepted law and economics theory of trademarks fails to explain why a brand owner would ever walk away from a trademark that generates financially lucrative returns. In 2020, that is exactly what happened again and again as brand owners pledged to abandon racially explicit marks in the weeks following George Floyd’s murder. As citizens became more attuned to the experiences of those depicted in racial marks, the owners of Aunt Jemima, Uncle Ben’s, the Cleveland Indians, the Redskins, the Dixie Chicks, Lady Antebellum and others announced these brands’ days were numbered. Animus and discrimination are the two legal lenses through which in-equality is typically assessed and understood. Insufficient attention, however, is paid to the role of status in animating inequality, even in landmark cases thought to be equality-promoting. More than an animating force between intractable po-litical conflicts, status also informs the development of equality law in the United States. When courts, advocates, and policymakers affirm, ignore, miss, or con-cede to status hierarchies instead of dismantling them, those groups that perceive a decrease in their status relative to others will only use “equality-promoting” doctrine to rebalance status hierarchy in their favor. When individuals have questions about Federal benefits, services, and legal rules, they are increasingly seeking help from government chatbots, virtual assistants, and other automated tools. Current forms of automated legal guidance platforms include the U.S. Citizenship and Immigration Services’s “Emma,” the U.S. Department of Education’s “Aidan,” and the Internal Revenue Service’s “Interactive Tax Assistant.” Most scholars who have studied artificial intelligence and Federal government agencies have not focused on the government’s use of technology to offer guidance to the public. Assisted Reproductive Technology (ART) and determining parentage have a common feature: each is governed by state law or the lack of such laws. This lack of statutory regulations presents significant legal challenges to gay men who wish to start a family. Because same-sex male couples seeking to become fathers through ART and surrogacy are the most likely demographic to be impacted when determining parentage, laws that influence the direction of surrogacy will undeniably facilitate whether both males will be deemed a father. Age-related physiological changes are most notable and best-studied late in life, while the nature of aging in early- or middle-aged individuals has not been explored as thoroughly. In C. elegans, many studies of movement vs. age generally focus on three distinct phases: sustained, youthful movement; onset of rapidly progressing impairment; and gross immobility. We investigated whether this first period of early-life adult movement is a sustained "healthy" level of high function followed by a discrete "movement catastrophe"-or whether there are early-life changes in movement that precede future physiological declines. The global response to Coronavirus Disease 2019 (COVID-19) is now facing new challenges such as vaccine inequity and the emergence of SARS-CoV-2 variants of concern (VOCs). Preclinical models of disease, in particular animal models, are essential to investigate VOC pathogenesis, vaccine correlates of protection and postexposure therapies. Here, we provide an update from the World Health Organization (WHO) COVID-19 modeling expert group (WHO-COM) assembled by WHO, regarding advances in preclinical models. Infections with herpesviruses, including human roseoloviruses, have been proposed to cause autoimmune disease, but defining a causal relationship and mechanism has been difficult due to the ubiquitous nature of infection and development of autoimmunity long after acute infection. Murine roseolovirus (MRV) is highly related to human roseoloviruses. Herein we show that neonatal MRV infection induced autoimmune gastritis (AIG) in adult mice in the absence of ongoing infection. MRV-induced AIG was dependent on replication during the neonatal period and was CD4+ T cell and IL-17 dependent. Cancer patients frequently develop chemotherapy-induced peripheral neuropathy (CIPN), a painful and long-lasting disorder with profound somatosensory deficits. There are no effective therapies to prevent or treat this disorder. Pathologically, CIPN is characterized by a "dying-back" axonopathy that begins at intra-epidermal nerve terminals of sensory neurons and progresses in a retrograde fashion. Calcium dysregulation constitutes a critical event in CIPN, but it is not known how chemotherapies such as paclitaxel alter intra-axonal calcium and cause degeneration. Here, we demonstrate that paclitaxel triggers Sarm1-dependent cADPR production in distal axons, promoting intra-axonal calcium flux from both intracellular and extracellular calcium stores. During dendritic cell (DC) development, Myc expression in progenitors is replaced by Mycl in mature DCs, but when and how this transition occurs is unknown. We evaluated DC development using reporters for MYC, MYCL, and cell cycle proteins Geminin and CDT1 in wild-type and various mutant mice. For classical type 1 dendritic cells (cDC1s) and plasmacytoid DCs (pDCs), the transition occurred upon their initial specification from common dendritic cell progenitors (CDPs) or common lymphoid progenitors (CLPs), respectively. N6-methyladenosine (m6A) is an abundant mRNA modification and affects many biological processes. However, how m6A levels are regulated during physiological or pathological processes such as virus infections, and the in vivo function of m6A in the intestinal immune defense against virus infections are largely unknown. Here, we uncover a novel antiviral function of m6A modification during rotavirus (RV) infection in small bowel intestinal epithelial cells (IECs). The pioneer factor hypothesis (PFH) states that pioneer factors (PFs) are a subclass of transcription factors (TFs) that bind to and open inaccessible sites and then recruit non-pioneer factors (non-PFs) that activate batteries of silent genes. The PFH predicts that ectopic gene activation requires the sequential activity of qualitatively different TFs. We tested the PFH by expressing the endodermal PF FOXA1 and non-PF HNF4A in K562 lymphoblast cells. While co-expression of FOXA1 and HNF4A activated a burst of endoderm-specific gene expression, we found no evidence for a functional distinction between these two TFs. In the past decade, several studies have estimated the human per-generation germline mutation rate using large pedigrees. More recently, estimates for various nonhuman species have been published. However, methodological differences among studies in detecting germline mutations and estimating mutation rates make direct comparisons difficult. Here, we describe the many different steps involved in estimating pedigree-based mutation rates, including sampling, sequencing, mapping, variant calling, filtering, and appropriately accounting for false-positive and false-negative rates.
