Assessment Of Dose-Dependent Cytotoxicity Of Caffeine On Human Colorectal Adenocarcinoma And Foreskin Fibroblast Cell Lines, Sanjay Srinivasan
Theses/Capstones/Creative Projects
Caffeine is a stimulant well-known for its alertness enhancement and energy functions. Beyond its neurological effects, caffeine also influences cellular qualities, including viability and membrane integrity. This study aimed to investigate the cytotoxic effects of caffeine on two distinct human cell lines: Human Colorectal Adenocarcinoma (HT-29) and Foreskin Fibroblasts (HFF). Cells were exposed to varying concentrations of caffeine to evaluate whether its effects were dose dependent. Cell viability was assessed using the Trypan Blue exclusion and Alamar Blue assays. Results showed that HFF cell death was more evident than the HT-29 Cells, displaying a potential therapeutic for cancer treatment.

Mcf-7 Dense Breast Cancer Cells Modulate Stress Through Senescence, Lara Donadio, Moses Alfonso, Josiah Philip, Tyler Zatarain
Molloy Multidisciplinary Undergraduate Research Conference
Breast cancer remains a leading cause of cancer-related mortality among women worldwide, with dense breast tissue being recognized as a significant risk factor for its development. This dense breast tissue, characterized by a higher concentration of mammary glandular tissue, represents additional sites for tumor development. The following study explores how the MCF-7 immortal cell line, representing dense breast cancer, modulates stress responses through cellular senescence. Cellular senescence is a defense mechanism that halts the cell cycle and acts as a tumor suppression mechanism, preventing out-of-control cell proliferation triggered by cytological stress. To simulate these stress conditions, we applied two agents: …

Hypochlorous acid (HOCl) is a potent nonradical oxidant involved in various physiological processes, particularly within the human immune system. In this study, we introduce a novel, rapid, and highly efficient fluorometric method for the detection of HOCl. The method utilizes a near-infrared (NIR)-based fluorescent probe, NIR-QBH, which is characterized by its high sensitivity and chemical stability. NIR-QBH, containing olefinic C=C bonds, exhibits strong NIR emission at 660 nm (λex = 618 nm). The detection mechanism relies on the oxidation of the C=C bond in the NIR-QBH structure by HOCl, resulting in the formation of non-fluorescent products. With a detection limit …

Cytotoxic immune cells mediate precise attacks against diseased cells to maintain organismal health. Their operational unit of killing and host defense is lytic granules (LGs), which are specialized lysosomal-related organelles. Precision in cytotoxicity is achieved by converging the many LGs to the microtubule-organizing center (MTOC) and polarizing these to the diseased cell for secretion. We identify unappreciated intimate relationships between the Golgi, MTOC, and LGs after cytotoxic cell activation, as well as the trans-Golgin protein GCC2 on the LG surface. GCC2 serves to tether LGs to the Golgi following convergence, and both GCC2 and the Golgi are required for the …

Chimeric antigen receptor T cells (CART) targeting CD19 through CD28.ζ signaling induce rapid lysis of leukemic blasts, contrasting with persistent tumor control exhibited by 4-1BB.ζ-CART. We reasoned that molecular dynamics at the CART immune synapse (CARIS) could explain differences in their tumor rejection kinetics. We observed that CD28.ζ-CART engaged in brief highly lethal CARIS and mastered serial killing, whereas 4-1BB.ζ-CART formed lengthy CARIS and relied on robust expansion and cooperative killing. We analyzed CARIS membrane lipid rafts (mLRs) and found that, upon tumor engagement, CD28.ζ-CAR molecules rapidly but transiently translocated into mLRs, mobilizing the microtubular organizing center and lytic granules …

7-Ketocholesterol: Occurrence, Biological Activities, And Plausible Remediation, Shubhrima Ghosh
Research Outputs: 2025-Present
Oxysterols, or cholesterol oxidation products, are potentially harmful derivatives of cholesterol arising from heat treatment in food systems and enzymatic as well as non-enzymatic processes in the body. Of these, 7-ketocholesterol (7KC) is a major oxysterol implicated in age-related diseases such as atherosclerosis, macular degeneration, neurodegenerative disorders, cancers, and some inflammatory diseases. The cytotoxic pathways of 7KC often involve typical apoptotic markers such as the production of reactive oxygen species (ROS), mitochondrial and peroxisomal dysfunction, followed by the release of cytochrome c, activation of caspase-9 followed by caspase-3, and other secondary messenger activations leading to cell death. The remedial approach …

CD1d-restricted invariant Natural Killer (iNK) T cells are a suitable candidate for allogeneic Chimeric Antigen Receptor (CAR) T cell therapy as they do not cause graft-versus-host disease (GvHD) due to the monomorphic nature of CD1d proteins. However, the phenotypic and functional heterogeneity of iNK T cells from adult donors (AD) may lead to the inconstant CAR-iNK T cell products. Cord blood-derived (CB) iNK T cells, in contrast, exhibit inter-donor homogeneity in phenotype including uniform CD4 expression and are enriched in memory iNK T cell populations. Thus, we evaluated the preclinical therapeutic potential of iNK T cells derived from cord blood …

CD4+ T cells can either enhance or inhibit tumour immunity. Although regulatory T cells have long been known to impede antitumour responses1-5, other CD4+ T cells have recently been implicated in inhibiting this response6,7. Yet, the nature and function of the latter remain unclear. Here, using vaccines containing MHC class I (MHC-I) neoantigens (neoAgs) and different doses of tumour-derived MHC-II neoAgs, we discovered that whereas the inclusion of vaccines with low doses of MHC-II-restricted peptides (LDVax) promoted tumour rejection, vaccines containing high doses of the same MHC-II neoAgs (HDVax) inhibited rejection. Characterization of the inhibitory cells induced by HDVax identified …

Chimeric antigen receptor T cells (CART) targeting lymphocyte antigens can induce T cell fratricide and require additional engineering to mitigate self-damage. We demonstrate that the expression of a chimeric antigen receptor (CAR) targeting CD5, a prominent pan-T cell antigen, induces rapid internalization and complete loss of the CD5 protein on T cells, protecting them from self-targeting. Notably, exposure of healthy and malignant T cells to CD5.CART cells induces similar internalization of CD5 on target cells, transiently shielding them from cytotoxicity. However, this protection is short-lived, as sustained activity of CD5.CART cells in patients with T cell malignancies results in full …

OBJECTIVES: Glioblastomas (GBM) are the most common primary invasive neoplasms of the brain. Distinguishing between lesion recurrence and different types of treatment related changes in patients with GBM remains challenging using conventional MRI imaging techniques. Therefore, accurate and precise differentiation between true progression or pseudoresponse is crucial in deciding on the appropriate course of treatment. This retrospective study investigated the potential of apparent diffusion coefficient (ADC) map values derived from diffusion-weighted imaging (DWI) as a noninvasive method to increase diagnostic accuracy in treatment response.
METHODS: A cohort of 21 glioblastoma patients (mean age: 59.2 ± 11.8, 12 Male, 9 Female) …

Triple-negative breast cancer (TNBC) is an aggressive subtype characterized by the absence of estrogen receptor, progesterone receptor, and human epidermal growth factor receptor type 2 expression. It is known for its high malignancy, invasiveness, and propensity for metastasis, resulting in a poor prognosis due to the absence of beneficial therapeutic targets. Natural products derived from mushrooms have gained significant attention in neoplastic therapy due to their potential medicinal properties. The therapeutic potential of Ganoderma lucidum in breast cancer has been highlighted by our group, suggesting its use as an adjuvant treatment. The present study aims to assess the potential antineoplastic …
