The COVID-19 pandemic underscored the urgent need for integrated healthcare resource planning to manage patient surges and reduce mortality. This study presents a novel optimization-based framework that jointly considers healthcare capacity planning and non-pharmaceutical interventions (NPIs), including mask mandates, social distancing, and quarantining. The framework extends traditional compartmental models by distinguishing patient categories—such as hospitalized non-ventilator patients, ventilated patients, and ICU admissions—within a mixed-integer programming (MIP) structure that remains computationally tractable. Applied to the COVID-19 outbreak in New York City, the model optimizes the allocation of hospital beds and ventilators while minimizing deaths. Latin America experienced some of the world’s highest COVID-19 mortality rates, yet data on in-hospital outcomes and their social and geographic determinants remain limited. This dissertation investigated multilevel baseline factors associated with in-hospital mortality among adults with COVID-19 at Panama’s largest public tertiary hospital (Hospital Santo Tomás), integrating survival, mediation, and spatial-temporal modeling within a causal-inference framework.

A retrospective cohort of 1,526 RT-PCR–confirmed COVID-19 hospitalizations (May 2020–November 2021) was analyzed using multivariable Cox proportional-hazards, Fine–Gray competing-risk, mediation, and Bayesian spatial models. Background and Objective: 

While vaccination remains central to controlling the COVID-19 pandemic, the emergence of SARS-CoV-2 variants with partial resistance to immune responses has highlighted the need for complementary therapeutic strategies. Among these, antiviral agents that inhibit viral entry mechanisms are of particular interest. Animal venoms, especially scorpion venoms, are a rich source of bioactive peptides with potential antiviral properties. This study aimed to evaluate peptides derived from the Moroccan scorpion Androctonus mauretanicus as inhibitors of SARS-CoV-2 spike glycoprotein, which mediates virus entry into host cells via ACE2 receptor binding.

BACKGROUND: Respiratory syncytial virus (RSV) and SARS-CoV-2 can cause acute respiratory failure in children. We compared characteristics and outcomes of children aged < 2 years with respiratory failure from infection with RSV, SARS-CoV-2, or both viruses.

METHODS: We used data from a US pediatric respiratory virus hospitalization surveillance network including children with ICU admission for acute respiratory failure (receiving high-flow oxygen or mechanical ventilation) with RSV and/or SARS-CoV-2 during November 2023-March 2024. Demographic, clinical characteristics, and hospitalization outcomes were stratified by a positive test for RSV, SARS-CoV-2, or both viruses, and compared using chi-squared or Kruskal-Wallis tests. Multivariable analyses assessed independent associations between outcomes and infection.

BACKGROUND. The SARS-CoV-2 pandemic provided a rare opportunity to study how human immune responses develop to a novel viral antigen delivered through different vaccine platforms. However, to date, no study has directly compared immune responses to all 3 FDA-approved COVID-19 vaccines at single-cell multiomic resolution.

METHODS. We longitudinally profiled SARS-CoV-2–naive adults (n = 31) vaccinated with BNT162b2, mRNA-1273, or Ad26.COV2.S, integrating plasma cytokines, antibody titers, and single-cell multiomic data (DOGMA-Seq).

RESULTS. We discovered a distinct, transient IFN program termed ISG-dim, which emerged specifically 1–2 days after the first mRNA dose in approximately 10% of myeloid cells. This state was …

Objective: To investigate the predictive value of inflammatory, liver, renal, and cardiac biomarkers in relation to the progression of multiorgan failure (MOF) in COVID-19 patients. A primary goal was to identify which of these markers served as the most discriminative predictors of disease severity.

During the Coronavirus Disease of 2019 (COVID-19) pandemic, MTB received an influx of queries regarding the teratogenicity of the virus and the vaccine. BACKGROUND: Obesity affects over 10% of the world population and has significant public health implications. With rising recognition of the long-term effects of Long COVID (LC) coupled with new agents to facilitate weight loss, it is critical to understand the influence of obesity on LC. This study assessed the association of obesity with rates of LC and degree of LC-related mental and physical health outcomes among participants up to three years after initial infection.

This article analyzes the FDA's proposed policy plan issued in May 2025 regarding the approval of Covid-19 boosters for different groups of persons. It offers two constructive recommendations that will help prevent constitutionality challenge on impermissible intrusion of the freedom of health.

The first is to avoid using the confusing term "benefit" but instead focus on the statutory parameters of "safety, purity, and potency." The affected group can make a strong case that their free choice of vaccination should not be deprived, as long as the vaccine qualifies for the statutory thresholds of being safe, pure, and potent. ObjectivesThe chronic kidney disease (CKD) patients were at high risk for severe clinical complications during the COVID-19 pandemic. Our objectives were to evaluate comorbidity prevalence; predict mortality risks for CKD patients during the pandemic; assess how various health factors interact to influence mortality; and provide insights for targeted prevention strategies.MethodWe analysed data from 186,396 CKD patients in Mexico during the entire pandemic (Jan 2020- May 2023). Healthcare disparities exist in the U.S. between different races. This study investigated the frequency of COVID-19 infections and hypertension cases among five different racial groups (White, Black, Asian, Native American, and Native Hawaiian). The study also examined the correlation between COVID-19 infections and hypertension with the hypothesis that because of disease predisposition and socioeconomic barriers, Black populations would have the highest rates of COVID-19 infections and hypertension. This study employed an age-structured Susceptible-Infected-Recovered-Vaccinated-Deceased (SIRVD) model to design optimal COVID‑19 intervention strategies by integrating epidemiological dynamics with economic costs. Using empirical data from South Korea, we estimated transmission parameters and analyzed quarantine-based non-pharmaceutical interventions (NPIs) alongside targeted vaccination scenarios. Our results indicated that a stronger form of mild NPI (upper range of Level I), when combined with targeted vaccination that prioritized seniors, achieved substantial reductions in infection peaks and overall economic burden. These findings underscore the effectiveness of age-specific strategies in epidemic management and offer critical insights for policymakers seeking balanced, resource-efficient approaches.

While humoral responses to vaccination and infection have largely focused on IgG, there is an evolving literature on ways in which IgA may drive neutrophil activity in lung injury secondary to viral infections, including COVID-19. We evaluated 238 immune-naïve, critically ill patients in the first year of the COVID-19 pandemic. Employing a laboratory-developed, multiplexed antibody testing, clinical history, medical record, and machine learning, we pursued relationships between systemic IgA and disease severity. Introduction

Vaccine hesitancy is a global issue, and the important role of healthcare workers in addressing vaccine hesitancy is well documented. However, there is limited theory-guided research on the uptake of COVID-19 vaccinations among healthcare workers in low- and middle-income countries (LMICs). This study examined COVID-19 vaccine uptake among healthcare workers in Sierra Leone and assessed the relevance of the 5C model—which posits that vaccine uptake is shaped by confidence, complacency, constraints, calculation and collective responsibility—in explaining their vaccination attitudes, intentions and behaviours.

Importance: Treatment for cognitive dysfunction due to postacute sequelae of long COVID (ie, symptoms of fatigue, malaise, weakness, confusion that persist beyond 12 weeks after an initial COVID infection) remains a significant unmet need.

Objective: To test evidence-based rehabilitation strategies for improving cognitive symptoms in persons with long COVID.

Design, setting, and participants: This was a 5-arm, multicenter, randomized clinical trial of 3 remotely delivered interventions conducted between August 17, 2023, and June 10, 2024. The study took place at 22 trial sites and included the screening of individuals with cognitive long COVID.

In this retrospective study, we compared the cause of death among hospitalized patients who died with a diagnosis of COVID-19 across two periods of the pandemic two years apart: an early period (admissions June to October 2020), when the population was immunologically naïve, and a later period (admissions April to October 2022), when an estimated 96% of the population had immunity from prior infection, vaccination, or both. We investigated whether plasma levels of lung injury biomarkers are associated with progression to respiratory failure among adults hospitalized with COVID-19 pneumonia without respiratory failure at admission.
