Although work on the prolamins of pearl millet (Pennisetum glaucum) has revealed partial amino acid sequences for several alcohol-soluble storage proteins (Marcellino et al. 2002) the genes encoding them have not yet been isolated. We constructed a cDNA library from developing P. glaucum seed tissue and screened it using maize zein gene probes to isolate several α-prolamin-like gene sequences. The proteins encoded by these genes generally fall into two size classes: 20.6kD and 27.1kD, which we call the 21kD and 27kD pennisetins. Both proteins are similar in composition and sequence to α-prolamins from maize, sorghum and Coix. Protein bodies that …

Agriculture is a central part of Tanzania’s economy. Both within Dar es Salaam, its primate city, and in the city’s peri-urban zone, agriculture is an important part of the livelihood strategy for people of all social classes. However, because the peri-urban zone is one of transition from urban to rural, it tends to undergo more pronounced changes in land use over time than do the city and rural area it borders. This paper examines recent changes in agriculture, land use and livelihoods in the peri-urban zone of Dar es Salaam, Tanzania. As climate changes due to rising concentrations of greenhouse gases in the atmosphere, agriculture will be one of the key human activities affected. Projections show that while overall global food production in the coming decades may keep pace with the food requirements of a growing world population, climate change might worsen existing regional disparities because it will reduce crop yields mostly in lands located at lower latitudes where many developing countries are situated. Emerging infectious diseases, most of which are considered zoonotic in origin, continue to exact a significant toll on society. The origins of major human infectious diseases are reviewed and the factors underlying disease emergence explored. Anthropogenic changes, largely in land use and agriculture, are implicated in the apparent increased frequency of emergence and reemergence of zoonoses in recent decades. Special emphasis is placed on the pathogen with likely the greatest zoonotic potential, influenzavirus A.

Humans with diabetes mellitus are two to five times more likely to develop cardiovascular disease and do so at a much younger age than non-diabetics. This study utilized the Syrian F1B hamster, a model of human atherosclerosis, to: (1) determine whether one intraperitoneal injection of streptozotocin (STZ) could induce a diabetic-like hyperglycemic state, (2) compare the development of atherosclerotic lesions between non-treated (N), hyperglycemic (G), hyperlipidemic (L) and combined hyperlipidemic/hyperglycemic (L+G) hamsters, and (3) determine whether lesion development was accelerated due to plasma levels of glucose, triglycerides, total cholesterol, non-HDL-C, HDL-C, or lipid hydroperoxides, or by the TC/HDL-C or TG/HDL-C …

Transient Changes In Blood-Brain Barrier Integrity, Thermotolerance, And Heat Shock Protein Expression Following Brief, Hyperthermia In An In Vitro Model, Courtney Klein

Hyperthermia is being studied as a means of disrupting blood-brain barrier (BBB) integrity. An in vitro BBB model was exposed to 10 s of 45, 48, or 51°C, or 5 s of 54°C, and TEER was used to assess integrity. Loss of BBB integrity increased as temperature increased (r = 0.88, P<0.0001). Thermotolerance was examined by applying a second hyperthermia to models 24 h afterward. Models demonstrated thermotolerance, indicated by less loss (P<0.001) of integrity compared to controls. The degree of thermotolerance increased as pre-conditioning temperature increased (r = 0.42, P<0.0001). Hsp27 and Hsp70 were analyzed by Western blot to examine their contribution to this thermotolerance. Hsp27 was not changed (P>0.10) by the pre-conditioning heat treatments. Hsp70 expression increased (P<0.05) in response to pre-conditioning for 5 s at 54°C. This thermotolerance cannot be explained by changes in Hsp27 (r = 0.17, P>0.10) and Hsp70 (r = -0.17, P>0.10).

Cultivated strawberry (Fragaria x ananassa ) fruit contain primarily pelargonidin and little cyanidin. The antioxidant potential of cyanidin is reportedly twice that of pelargonidin. A High Performance Liquid Chromatography method was developed to assay fruit anthocyanidin content in 87 Fragaria accessions, including wild diploids and octoploids, cultivars, and two segregating populations. A cyanidin to pelargonidin ratio (CPRatio) greater than one was found only in diploids Fragaria vesca ssp. americana (confirming a previous report), F. viridis, and F. iinumae, and in three wild octoploid accessions: all F. chiloensis. Two experiments were conducted to compare the deterioration of wet brewers grains (WBG) treated with an enzyme and bacterial inoculant (Silo-King GPX, Agri-King Inc., Fulton, IL). Another experiment investigated ruminal DM and CP degradability of samples taken from experiment 1. Wet brewers grains were divided into separate piles upon delivery. Silo-King GPX was added to WBG at 0 kg/kg (control), 0.45 kg/900 kg WBG, and 0.9 kg/900 kg WBG, mixed for 5 min, and then stored in bins. Piles were left for 28 d to simulate farm storage practices. The objective of this experiment was to determine the effect of various levels of lactoferrin supplementation in colostrum replacer fed as 1 or 2 doses on apparent efficiency of IgG absorption, serum IgG concentrations, and intestinal development in neonatal Holstein bull calves. Eighty-Holstein bull calves were assigned to a 2 x 4 factorial arrangement of treatments in a completely randomized block design. Bull calves were collected at two locations, Fairchild Teaching and Research Center at the University of New Hampshire (n = 48) and a commercial dairy (n = 32). The harsh environmental and poor economic conditions of the Bolivian Altiplano require intervention to assist many of those that live there to become economically self-sufficient. We attempted to find introduced dry season reserve forage grasses that could produce enough biomass to be useful as feed for livestock, and that could also produce enough seed to distribute to farmers. While some of the grasses produced reasonable amounts of biomass, none produced seed in quantities that would be even close to being economically viable. Leaf area index (LAI), defined here as one-half of the total leaf area per unit ground surface area (Chen, 1996), has been estimated at a global scale from spectral data processed from the Moderate Resolution Imaging Spectroradiometer (MODIS) sensor aboard two NASA EOS-AM spacecraft, Terra (launched in 1999) and Aqua (launched in 2002). The MOD15A2 LAI product is a 1 km global data product composited over an 8-day period and is derived from a three-dimensional radiative transfer model driven by an atmosphere corrected surface reflectance product (MOD09), a land cover product (MOD12) and ancillary information on surface characteristics. The prevalence of malformed amphibians and the association of high incidences of malformation with the presence of environmental contaminants have raised questions about the effects of environmental contaminants on the development of larval amphibians. The potential endocrine disrupting effects of pesticides suggest that the reproductive and developmental abnormalities observed in larval anurans may be due at least in part to impacts on the hypothalamic-pituitary-gonadal axis. The present studies investigate the effects of exposure to environmental contaminants including DDT and atrazine on the development, gonadal histology and reproductive steroidogenesis of larval anurans.

Thirty multiparous early lactation Holstein cows were used in a randomized complete block design to determine whether the addition of VERTAN (a specific blend of essential oils, major components: thymol, eugenol, vanillin and limonene) to corn silage based diet would increase alter protein metabolism in the rumen and increase milk yield and alter milk composition. Dietary treatments were 0 (control) and 0.08% VERTAN in diet DM. Diabetes mellitus accelerates atherogenesis, but the exact mechanisms involved are unknown. This study examined the effects of hyperglycemia on the inflammatory transcription factor peroxisome proliferator activated receptor gamma (PPARgamma), known to be involved in atherogenesis, and the ATP binding cassette A1 (ABCA1) protein that is a downstream product of PPARgamma. ABCA1 is a membrane bound transporter that is involved in reverse cholesterol transport via high density lipoprotein cholesterol (HDL-C). To study dynamic in vivo biochemical changes within the corpus luteum (CL) during luteolysis, a physiological model mimicking the onset of natural luteolysis was established, in which sheep received sequential systemic pulses of PGF2alpha (20ug/min/lhr) at mid-luteal phase of the estrous cycle. We previously demonstrated that after one pulse of PGF2alpha, the protein levels of extracellular matrix (ECM) regulators, tissue inhibitors of metalloproteinases (TIMPs) -1 and -2 decreased dramatically within one hour of PGF2alpha infusion, while matrix metalloproteinase (MMP)-2 activity increased 4 hrs post infusion. Abundance indices suggest that the moose (Alces alces) population in northern New Hampshire has stabilized despite favorable habitat and conservative harvest. Natural mortality of unknown cause is presumed a primary reason although little is known about moose reproduction and survival in New Hampshire. This study (2002-2005) was designed to investigate the dynamics that impact this population.

Analysis of harvest reproductive data (1988-2004) indicated that average field-dressed weight of adult cows increased although corpora lutea count declined from ∼1.4- 1.2/cow in the study area and statewide. Yearling ovulation rate (∼42%) and average weight (<211 kg) declined about 25% and 4%, respectively. Parturition of radio-marked cows ranged from 8 May-13 July (median=19 May) with 78% of births from 13-27 May. Calving rate of yearlings and adults (>2 yr) averaged 30 and 85%, respectively, …

The most effective method for increasing supplies of MP-Met to dairy cows to increase milk protein production is to feed rumen-protected Met ( RP-Met) products; however, not all RP-Met products are absorbed by the dairy cow in the same manner. Universally acceptable methods must be developed to determine the efficacy of different RP-Met products.

The first study was designed to compare the feeding of two sources of supplementary Met beginning 21 d prepartum and continuing for 140 d postpartum. The major protein component of the extrapallial (EP) fluid of the mollusc Mytilus edulis, named the EP protein, has been previously isolated and partially characterized. It has been proposed to play an important role in shell mineralization because of its intriguing property of Ca2+ -binding induced self-assembly.

This dissertation reports on the characterization of the primary structure of the EP protein by RT-PCR and cDNA sequencing methods. The EP protein is comprised of 213 amino acids post cleavage of a signal peptide of 23 amino acids, and is rich in His, Glu and Asp residues. The consensus site of N-glycosylation …
