The objective of this dissertation was to develop a comprehensive, data-driven spatial framework for characterizing the complex cultural landscape of the National Coal Heritage Area (NCHA) in West Virginia. By transitioning away from traditional, heuristic spatial mapping, this research integrates advanced spatial statistics, machine learning, and GIS-based methodologies to objectively quantify the physical, visual, and cultural dimensions of the post-mining environment. Designing robots traditionally relies on centralized, top-down methods that constrain adaptability and resiliency, producing systems optimized for specific tasks. However, such systems often fail in complex environments, such as extraterrestrial, disaster, or natural settings, where designers cannot foresee every scenario a robot may encounter. Achieving autonomy in these contexts requires robots that are resilient and capable of adapting to the unknown. Multicellular organisms exhibit this adaptability through self-organization, where complex structures and behaviors emerge from local interactions among their cells. Such organisms can be viewed simultaneously as a single entity and a swarm of cooperating cells, i.e., a Swarm-of-One.

Apicomplexan parasites, responsible for toxoplasmosis, cryptosporidiosis and malaria, invade host cells through a unique gliding motility mechanism powered by actomyosin motors and a dynamic organelle called the conoid. Here, using cryo-electron microscopy, we determined structures of four essential complexes of the Toxoplasma gondii conoid: the preconoidal P2 ring, tubulin-based conoid fibers, and the subpellicular and intraconoidal microtubules. Our analysis identified 40 distinct conoid proteins, several of which are essential for parasite lytic growth, as revealed through genetic disruption studies. This study focuses on ribosome-inactivating proteins (RIPs), a class of potent toxins that includes ricin, α-sarcin, and shiga toxin, which irreversibly damage large subunit rRNA. Although the catalytic activity of ricin toxin A chain (RTA) is well characterized, the precise mechanisms linking ribosome damage to altered cellular morphology remain poorly understood. In this study, we engineered a tetracycline-regulatable expression system to express RTA in Saccharomyces cerevisiae. We characterized the effects of RTA expression on yeast growth, viability, and reproductive potential. The infraorder Mygalomorphae represents one of the earliest-diverging lineages of spiders, but its fossil record remains sparse relative to the modern diversity of the group. This is a reflection of both ecological and preservational biases, such as the predominantly fossorial habits of many mygalomorphs and the strong bias of the spider fossil record toward amber deposits. Compression fossils from rock deposits are therefore especially important for documenting the evolutionary history of the group. Managing at-risk species or populations requires understanding the demographic and genetic drivers of long-term population trends. The eastern massasauga rattlesnake (Sistrurus catenatus) is a federally threatened species facing multiple interacting threats across its range. Habitat loss from wetland drainage and agricultural land conversion has caused range contractions and geographic isolation of populations. Because remaining populations are often small and widely separated, most are now genetically isolated, threatening their long-term viability. White-tailed deer (Odocoileus virginianus) are one of the most culturally and economically important big game species in the United States. At the turn of the 20th century white-tailed deer were almost extirpated due to market hunting and unsustainable forestry practices that destroyed habitat. Acid mine drainage (AMD) is a legacy contaminant, impairing Appalachian watersheds despite decades of regulatory work and investment in remediation. This study analyzes and evaluates the performance of seven AMD treatment systems (5 passive and 2 active) as well as thirteen untreated sources in the headwaters of Deckers Creek. Monthly influent and effluent samples measuring pH, major metals, and acidity were collected from January 2024 through 2026 to assess variability in treatment performance, and engineering site visits were conducted for each treatment system. Historical mining, intensive timber harvesting, agriculture, and atmospheric acid deposition have left lasting impacts on soil health throughout the Central Appalachian region, reducing soil productivity, carbon storage, and ecosystem resilience. This thesis evaluated restoration strategies across two long-term research systems in West Virginia to determine how land-use history and soil amendments influence soil health, carbon dynamics, and ecosystem recovery. Snowshoe hares (Lepus americanus) occupy a large range across the United States and Canada. While snowshoe hare populations are generally considered stable in the interior of their range, populations along range edges are experiencing declines, possibly driven by climate-related factors such as seasonal coat color mismatch. West Virginia represents one of the southernmost extents of the species’ range, yet little is known about the population genetic status or population sizes of snowshoe hares in the state. Combinatorial optimization problems (COPs) require searching over a finite solution space subject to constraints, with the goal of satisfying an objective function. They arise in operations research, scheduling, resource allocation, circuit design, and many other fields. Many problems in combinatorial optimization (including satisfiability and network design) are NP-hard. Invasive species pose significant ecological, economic, and management challenges globally. Purple loosestrife (Lythrum salicaria), Japanese knotweed (Fallopia japonica), tree of heaven (Ailanthus altissima), and mile-a-minute (Persicaria perfoliata) are widespread invasive plants across the eastern United States and present persistent challenges, particularly in remote and inaccessible landscapes. Effective management of these species requires an integrated approach that combines early detection, targeted intervention, public engagement, and spatial modeling. This secondary data analysis aimed to determine the nature of the relationship between obstructive sleep apnea (OSA) risk, biological age acceleration, and nonspecific chronic low back pain (CLBP). In total, 199 adults aged 18-82 years who filled both STOPBANG and pain questionnaires subsetted for secondary analysis. Based on the STOPBANG questionnaire, 104 had a low OSA risk and 95 had an intermediate or high OSA risk. Dunedin Pace of Aging Computed from the Epigenome (DunedinPACE), Horvath's, Hannum's, PhenoAge, and GrimAge clocks were used to determine biological age and pace of biological aging. BACKGROUND AND OBJECTIVES: Navigation is a complex, multisensory process in which subtle changes may emerge before the onset of symptomatic Alzheimer's disease (AD). This study investigated whether navigation abilities were associated with amyloid and tau pathology in cognitively normal older adults.

RESEARCH DESIGN AND METHODS: Among 285 cognitively unimpaired older adults, navigation and objective driving behaviors were assessed using the self-reported Santa Barbara Sense of Direction Scale (SBSOD) and a datalogger, respectively. INTRODUCTION: Alport syndrome features a defective glomerular basement membrane (GBM) due to variants in COL4A3, COL4A4, and COL4A5. The most severe forms, which lack the GBM's collagen α3α4α5(IV) network, progress from hematuria in early childhood to proteinuria, chronic kidney disease, and kidney failure by the age of ∼30. As a monogenic disease without specific treatments, and with podocytes being the only glomerular cells that synthesize collagen α3α4α5(IV), the ability to efficiently deliver genes to Alport podocytes could open up new possibilities for treatment.

SIGNIFICANCE: Adapting optical imaging technology to avian models can overcome many limitations imposed by functional magnetic resonance imaging (fMRI), which currently restricts the number of species used to study functional connectivity. BACKGROUND: Prior studies suggest that longer drive time (DT) to specialists and higher area deprivation index (ADI) are linked to worse outcomes in inflammatory bowel disease (IBD), but their impact on postileocolic resection (ICR) care in patients with Crohn's disease (CD) is not well defined.

OBJECTIVES: We aimed to evaluate disparities in post-ICR care based on DT and ADI.

METHODS: Spatial analysis was performed with ArcGIS Pro to geocode patient and medical facility addresses with StreetMap Premium locators. High DT was defined as >60 min from the center. Data were analyzed using basic statistics and multivariate logistic regression.

BACKGROUND: Sonobiopsy is a promising new technique that employs focused ultrasound (FUS) to noninvasively enrich brain tumor-derived biomarkers from spatially targeted brain location into the bloodstream. Building upon our recent first-in-human sonobiopsy trial demonstrating enrichment of circulating tumor DNA (ctDNA), here we investigated whether sonobiopsy can enhance the release and detection of tumor-derived microRNAs (miRNAs).

INTRODUCTION: Blood-based biomarkers can improve Alzheimer's disease (AD) characterization in Down syndrome (DS). This study applied hierarchical clustering and machine learning-based feature selection to identify biomarkers associated with disease progression.

METHODS: Cross-sectional blood-based biomarkers were analyzed from 211 DS participants (

RESULTS: The strongest predictors overall were neurofilament light chain (NfL), tau/amyloid beta (Aβ)40, Aβ42/Aβ40, alpha-2-macroglobulin (A2M), and interleukin (IL)-10. Within the CS group, NfL, tau/Aβ40, A2M, and IL-10 were strong predictors. In MCI, Aβ42/Aβ40, NfL, A2M, and IL-10 were strong predictors. In DS-AD, Aβ42/Aβ40, NfL, and tau/Aβ40 were the top predictors. Cluster membership varied based on disease stage.

INTRODUCTION: To investigate drivers of dementia risk disparities among older Black Americans using a community-based system dynamics (CBSD) approach.

METHODS: Over 18 months, 234 participants created 36 causal loop diagrams (CLDs) on dementia risk. An ecological analysis compared structural differences across groups based on scores on a structural and social determinants of health composite index (S/SDOH-CI).

RESULTS: Groups in the highest SDOH-CI quantile were more likely to identify key protective pathways linking mental health, substance use, exercise, medication, and community support to reduce dementia risk. INTRODUCTION: Chronic myeloid leukemia (CML) is a hematopoietic malignancy driven by constitutive tyrosine kinase activity. Dasatinib, a second-generation (2G) tyrosine kinase inhibitor (TKI), is a preferred treatment due to its superior, durable, and rapid response rates. While dasatinib has known gastrointestinal (GI) side effects, colonic polyposis is a rare complication, not well described in the literature.

CASE PRESENTATION: Upon routine colonoscopy, a 66-year-old woman with chronic-phase CML was incidentally discovered to have asymptomatic colonic polyposis after 11 months of dasatinib therapy. Histopathology revealed reactive inflammatory changes without evidence of dysplasia or neoplasia. The polyposis resolved completely after cessation of dasatinib. …

INTRODUCTION: Plasma biomarkers and mild behavioral impairment (MBI) are associated with dementia risk, but their relationships with major depressive disorder (MDD) are understudied. This study aimed to examine associations between plasma biomarkers of Alzheimer's disease (AD) and MBI among older adults with and without MDD.

METHODS: Older adults aged ≥65 were recruited from longitudinal studies of depression and AD (

RESULTS: Older adults with MDD were more likely to endorse MBI. After adjustment, Aβ42/Aβ40 and p-tau217/np-tau217 positivity were associated with MBI among those with MDD but not among those without.
