Volumetric Fluorescence Microscopy For High-Throughput And High-Sensitivity Imaging: From Single Molecules To Tissues, Le-Mei Wang

Fluorescence microscopy is an indispensable tool in the biological sciences, enabling researchers to investigate intricate subcellular structures, particularly for volumetric studies. However, conventional optical microscopy for volumetric imaging remains fundamentally constrained by imaging speed and throughput. To bypass traditional serial z-scanning, we introduce an axially scan-free method using a phase layer cake to modulate the system's point spread function. This approach projects volumetric information onto a 2D plane in a single shot, offering high flexibility in tuning axial depth alongside simultaneous multicolor imaging with high spatial resolution and sensitivity. How we view air, especially clean air, has meaningful normative and practical implications. Today, it is commonly recognized as an exhaustible natural resource. But air was not always thought of as something to be conserved, developed, and used. This Article is the first to explore and interrogate that view.
The shift to seeing air as a resource started in the first part of the twentieth century. Previously, air was characterized by its limitless supply of oxygen: a pure public good. Over time, the quality of that good changed, and a precious resource emerged. That resource—especially its chemical and physical characteristics—inspired …

This article reveals that limited-life business entities—finite ventures—play a significant and underappreciated role in modern commerce. Private equity and venture capital funds, SPACs, and insurance syndicates are all organized with a limited lifespan.

One critical dispute surrounding the rights of LGBTQ people and societal recognition of their existence is the legal debate regarding very different types of gender therapy. On the one hand, several states have banned the provision of gender-affirming medical care to transgender youth, positing that it is a dangerous form of mutilation. A separate set of states has banned the practice of so-called gay or gender conversion therapy that seeks to transform youth away from their queer identities, concluding that it is a form of abuse. For centuries, the law of transit has specified when states must permit foreign persons and goods to traverse their territory on journeys beginning and ending beyond their borders. Transit is a right — not a privilege — that tempers territorial sovereignty. Though largely forgotten today, the right to transit springs from states’ responsibilities to the international community when their territory hosts infrastructure of global utility.

Government regulations of medicine sometimes treat patients differently based on their race or sex. Does heightened scrutiny apply to such regulations—as is usual when law treats someone differently based on a protected characteristic? Or is there a medical exception to equal protection?
Recently, in United States v. Skrmetti, the Supreme Court upheld a law that restricted access to medical treatments for transgender minors. Though its holding did not rely on medical exceptionalism, its discussion reinvigorated interest in a "medical exception" to equal protection: that medical decisions are exempt from the searching review applied when governmental decisions consider protected characteristics.

Convergence education promotes learning experiences that integrate science, technology, engineering, and mathematics (STEM) to address complex real-world problems. However, secondary teachers often report limited access to professional development (PD) and curricular resources that support transdisciplinary instruction. This exploratory case study examines how four secondary teachers (three chemistry; one engineering) made sense of a transdisciplinary PD model, Convergence Research in STEM and Tradition (CReST), that leverages cultural heritage artifacts (Renaissance frescoes) as boundary objects to connect chemistry, engineering, world history, and technology. Background
The dominance of Western-inspired teaching methods and instructional materials has contributed to cultural erosion in African schools. In response, increasing attention has been directed toward culturally responsive approaches to decolonising science education. One such approach is the Culturo-Techno-Contextual Approach (CTCA), which enables African students to conceptualize science as culturally embedded knowledge, integrated into their everyday experiences and practically relevant to their lived realities.
Purpose

Few elementary teachers are confident in teaching science or in their science knowledge. This study sought to address this concern by investigating the impact of a science methods course on confidence and science knowledge for pre-service elementary teachers (PSETs). In this study, 23 participants enrolled in a teacher education program at a Midwest US university participated in a quasi-experimental pretest-posttest design. The participants engaged in course content, including metacognitive awareness activities, for a full semester and qualitatively described their confidence within the pre-post assessment survey. This study examines how project-based learning (PBL) supports pre-service STEM teachers (PSTs) in understanding and applying disciplinary literacy in classroom instruction. Conducted in a semester-long PBL course at a mid-sized public university, the research explores PSTs’ shifting perceptions of literacy in STEM and the challenges and successes they experience. Using qualitative methods grounded in student voice and case study frameworks, data were collected through focus groups, reflections, and lesson plan analyses. Findings show PSTs initially held narrow views of literacy but developed more nuanced understandings through PBL. Key successes included increased confidence and use of inquiry-based strategies; challenges involved time …

This thesis examines the historical, religious, and cultural consequences of portraying Jesus as white within Christian art. Although Western art has overwhelmingly depicted Christ with white skin, archaeological, and contextual evidence supports a dark-skinned Jesus rooted in the ethnogeography of first century Palestine. The persistence of a White Jesus has shaped religious imagination and perception for centuries, reinforcing racial hierarchy and legitimizing systems of white supremacy. Through an interdisciplinary analysis combining biblical studies, Christian iconography, ethnography of ancient Semitic, Greco-Roman, and Egyptian peoples, medieval symbolism, modern mass-produced imagery, and Black thought, this study demonstrates how Jesus’ original complexion was gradually …

The Witch Hunt exhibition conjures a powerful dialogue between history, identity, and empowerment, bringing together sixteen international artists to channel the spirit of the witch. This show traces a vast genealogy of women—from the 15th century witch hunts to Californian artistic legacies of the 1970s, to the exhibitions opening in 2021, and into present day. The participating artists emerge as symbols of resistance through their creations which transform domesticity, sexuality, and labor into sites of inquiry all while grappling with the intersectional concerns of womanhood. Although we are making progress in overcoming infectious diseases and cancer, one of the major medical challenges of the mid-21st century will be the increasing prevalence of stroke. Occlusions in large vessels are especially debilitating, yet effective treatment—needed within hours to achieve best outcomes—remains limited because of geographic accessibility. To examine the association between neighborhood-level social determinants of health and stillbirth.
METHODS:
We performed a retrospective cohort study of deliveries that occurred at a gestational age of at least 20 weeks within a five-hospital system (2012-2022). Electronic health record data mapped to the Observational Medical Outcomes Partnership Common Data Model were geocoded using addresses at the time of delivery and linked to neighborhood health indices, which included the Area Deprivation Index (ADI), Maternal Vulnerability Index (MVI), and Social Vulnerability Index (SVI) at the Census tract level. Objective: To quantify the extent to which rising rates of severe maternal morbidity (SMM) in the United States from 2016 to 2022 are attributable to increasing prevalence of maternal risk factors, including advanced maternal age, obesity, pregestational diabetes mellitus, gestational diabetes, chronic hypertension, and hypertensive disorders of pregnancy (HDP).
Methods: We conducted a retrospective cohort study using the Nationwide Readmissions Database from 2016 to 2022, representing more than 25 million weighted delivery hospitalizations. Transition metals such as iron and manganese are critical for cellular metabolism, redox balance, and immune function, and they also influence host–pathogen interactions. Vaginitis is characterized by mucosal inflammation that may alter local trace-element homeostasis. We hypothesize that inflammatory conditions such as vaginitis may be associated with altered cervicovaginal iron and manganese levels. Maternal anemia in pregnancy is associated with many adverse maternal, fetal, and neonatal outcomes. The role of social determinants of health (SDOH) in the development of maternal anemia and other adverse outcomes is continually evolving. One such newer tool, the Social Vulnerability Index (SVI), measures an individual's community-level SDOH, with higher SVI denoting higher vulnerability. Gestational exposure to phthalates is higher among non-Hispanic black (NHB) women than non-Hispanic white (NHW) women and may contribute to racial/ethnic differences in birthweight z-scores.
Objective
We aimed to investigate whether gestational phthalate and replacement biomarkers mediate the association between race/ethnicity and birthweight z-scores.
Methods
We analyzed data from 271 pregnancies in the Human Placenta and Phthalates study, a prospective cohort recruited from 2017 to 2020. The U.S. is currently facing a shortage of health professionals from underrepresented backgrounds. One potential pathway to decrease this shortage is through the Deferred Action for Childhood Arrivals (DACA) program, a policy that provides temporary relief from deportation for individuals who migrated to the U.S. before age 16. This study, using 15 life-history narrative interviews, investigates the experiences of Latinx young adults who were pursuing health professions education or working as health professionals. Aerospace systems frequently face competing requirements between compact packaging and high aerodynamic performance. Large propellers improve efficiency and thrust generation, yet launch constraints and confined storage volumes limit their maximum span. Conventional deployable solutions rely on hinges, locking mechanisms, and external actuation, increasing mechanical complexity and reducing reliability. This dissertation investigates an alternative approach in which elastic instabilities in thin composite shells are deliberately harnessed to enable rapid, mechanism- free deployment of aerodynamic structures. This dissertation examined the shifting culture of youth sports in the United States and its impact on academic motivation among adolescent student athletes. The study utilized Self-Determination Theory (SDT) as a framework. Traditionally, youth sports have helped students with personal growth and academic success. Dextran sodium sulphate (DSS) is widely used in vivo to induce epithelial injury; however, the molecular effects of DSS on intestinal epithelial cells, that investigate inflammatory and metabolic repair mechanisms remain unclear. Here, we establish an in vitro DSS-induced epithelial damage model using differentiated Caco-2 cell monolayer. Exposure to 2.5%(w/v) DSS for 24 hours resulted in significant upregulation of pore forming protein Claudin-2(2.677-fold) and oxidative marker, NOX-1(1.866-fold), accompanied by increased ROS production confirmed via DHE staining. Alabama contains some of the highest levels of subterranean biodiversity in North America, yet no statewide assessment of cave vulnerability has previously been conducted. A spatially explicit vulnerability assessment was developed for Alabama caves using species occurrence records, landscape characteristics, anthropogenic threat data, and groundwater vulnerability metrics. Distribution maps were generated for cave- obligate and bat species, and vulnerability was assessed using human population, land- use, groundwater quality, groundwater quantity, and modified DRASTIK models. Results identified numerous single-site endemic species, regional biodiversity hotspots, and cave systems facing elevated anthropogenic threats. Biomedical instrumentation requires low-power amplifiers to maximize battery life. Due to threshold voltage limitations, standard gate-driven operational transconductance amplifiers (OTAs) require higher supply voltages. To overcome this, a differential bulk-driven two-stage OTA is introduced for low-power applications. The proposed topology is implemented in 65nm, 90nm, and 130nm complementary metal-oxide-semiconductor (CMOS), with supply voltages ranging from 0.4V to 0.6V. Key metrics such as DC gain, gain-bandwidth product (GBWP), phase margin, common-mode rejection ratio (CMRR), slew rate, power, and noise are measured. The analysis confirms the robustness of the low-power solution provided by the proposed topology.
