Predicting the likelihood parents will become physically abusive towards their children remains a high priority. Early parenting stress may increase the extent to which parents negatively appraise their children's behaviours, which in turn may influence their abuse risk. This study explored whether parental negative attributions mediated the association between early parenting stress and later abuse risk. A sample of 186 first-time mothers and 146 of their male partners reported on their parenting stress when infants were 6 months old and their negative attributions regarding child behaviours at 18 months. Suicide risk assessments require individuals to differentiate among types of suicidal thoughts (e.g., passive vs. active); yet most research relies on aggregate metrics that obscure this differentiation. Indeed, more severe forms of suicidal thinking are thought to involve the simultaneous experience of multiple forms of thoughts at high intensities. The present study draws on concepts from the affect differentiation literature to (a) examine different ways to operationalize differentiation in suicidal thinking and (b) compare differentiation in suicidal thinking with differentiation in affect. Advances in statistical techniques for analyzing biological data have driven the need for larger sample sizes, which are ultimately made possible through team science efforts. Findings from these efforts have led to a significant shift in approaches to biological data collection over the past decade, from traditional approaches relying on single measures or assays collected by individual investigators in small lab-based studies to high-dimensional methodologies embedded within large cohorts or multi-site studies. Research has shown that kinship lineage and inheritance norms determine kin membership for children by specifying whether they belong to their maternal or paternal family, as well as whether they can inherit properties of their biological father or mother. The notion of kinship membership has shaped parenting and child welfare practices in Ghana, including child maintenance payments and parental neglectful behaviours. Sexual offending by young people causes significant harm to victims, families and communities. Treatment engagement and completion are therefore important for reducing future harm; however, young people who drop out of, or refuse to engage in, treatment present a unique challenge for clinicians and justice systems. Drawing on Bronfenbrenner’s (1979) ecological systems theory, this study explored clinicians’ perspectives on the barriers to treatment engagement and completion for young people who have sexually offended in the Australian context. This study aimed to explore whether health effects of dietary nitrate depend on its source, by investigating associations between plant and animal-sourced dietary nitrate groups with markers of inflammation and CVD risk factors. Among 100 non-smoking adults (mean age 49 ± 13 years, 31% male), dietary nitrate intake was assessed using food frequency questionnaire (n=100) and 3-day food diary (n=89), combined with nitrate food composition databases. Nitrate intake was classified into plant, naturally occurring animal and additive-permitted meat-sourced groups. Mitochondrial diseases are a complex group of conditions exhibiting significant phenotypic and genetic heterogeneity. Genomic testing is increasingly used as the first step in the diagnostic pathway for mitochondrial diseases. We used next-generation sequencing followed by bioinformatic data analysis to identify potentially damaging variants in the POLRMT gene (NM_005035.4) in six new affected individuals. Structural protein analysis predicted the detrimental impact of variants on POLRMT protein structure. Patients show extended phenotypic abnormalities often presenting early in life with features including global developmental delay, cognitive impairment, short stature and muscular hypotonia. Orthopedic osteosarcomas and metastatic lesions can be difficult to treat with systemically delivered chemotherapy agents alone. Following removal of the primary tumor, implantation of cement to fill the lesion is often done to stabilize weight-bearing bones. One of the commonly used treatments to address osteosarcoma is systemic delivery of doxorubicin. Therefore, research on the incorporation and release of efficacious doxorubicin for local delivery from cement is important. Poly(methyl methacrylate) (PMMA)-based bone cements are the gold standard in orthopedics but have inherent disadvantages. Copy number alterations (CNAs) are hallmarks of cancer, yet investigation of their oncogenic role has been hindered by technical limitations and missing model systems. Here we generated a genome-wide DNA methylation and CNA atlas of 106 genetic mouse models across 31 pediatric tumor types, including 18 new models for pediatric glioma. We demonstrated their epigenetic resemblance to human disease counterparts and identified entity-specific patterns of immune infiltration. We discovered that mouse tumors harbor highly recurrent CNA signatures that occur distinctly based on the tumor subgroup and driving oncogene and showed that these CNAs share syntenic regions with the matching human …

BACKGROUND: Cell migration and invasion are well-coordinated in development and disease but remain poorly understood. We previously showed that the neural crest (NC) cell migratory wavefront shares a 45-gene panel with other cell invasion phenomena. To rapidly and systematically identify critical genes, we performed a high-throughput siRNA screen and statistical and deep learning analyses to determine changes in NC- versus non-NC-derived human cell line behaviors.

BACKGROUND: The Natural History Study of the Safety, Effectiveness, and Practice of Treatment for People with Haemophilia (ATHN 7) monitors use of contemporary haemophilia therapies, including emicizumab, a bispecific antibody substituting for activated factor (F)VIII in people with haemophilia A (HA).

AIM: To report participant characteristics and real-world safety and effectiveness of emicizumab in ATHN 7.

OBJECTIVE: Oxytocin remains the mainstay for induction and for the management of labor arrest during trial of labor after cesarean (TOLAC). Therefore, the clinical question should be not whether to use oxytocin, but how to optimize its administration. This meta-analysis aimed to assess which oxytocin protocol, in terms of initial dose, amount of increase, timing to next increase, and maximum dose, may minimize the risk of uterine rupture (UR) during TOLAC.

This study foregrounds the voices of transgender (trans) and non-binary students in a large-scale mixed-methods investigation of gendered grouping practices and curriculum provision in secondary school physical education (PE) in England. Theoretically, the research brought together an interdisciplinary framework for educational inclusion and the trans inclusion staged model to pursue conceptualisations and expressions of inclusion that disrupt cisnormative policies and structures. A survey of 14 co-educational secondary schools generated data from 36 students who identified as trans (n = 18) or non-binary (n = 18). Short-chain fatty acids (SCFAs) produced by gut microbial fermentation influence host metabolism and neuroinflammatory processes implicated in Alzheimer’s disease (AD). However, the relationship between fecal SCFAs, microbial taxa, and cerebral amyloid-β (Aβ) burden in cognitively unimpaired individuals remains unclear. Fecal SCFAs were quantified using GC-MS, and microbial species were profiled by shotgun metagenomics in 87 participants. Associations between SCFAs, demographics, APOE ε4 status, and Aβ burden were tested using nonparametric statistics and multivariable regression. Microbial–SCFA links were evaluated using Spearman correlations and multivariate ordinations, with mediation analysis exploring potential indirect pathways.
