Contributions Of Arginine And Nitrosative Stress To Huntington’S Disease Progression, Ian D. Gaudet

Electronic Theses and Dissertations

Neuroinflammation is a well-established component of Huntington's disease (HD) pathogenesis, yet the intricate molecular mechanisms that govern this process remain a topic of ongoing research and debate. This dissertation elucidates a novel pathway linking arginine metabolism and nitrosative stress to core HD pathology. We found that the arginine transporter SLC7A2 is selectively upregulated in various HD cellular models and patient samples. Our findings reveal that this upregulation drives an overactive inflammatory response, leading to abnormally high inducible nitric oxide synthase (iNOS) induction and subsequent nitric oxide (NO) production, which causes excessive protein nitrosylation and an increase in mitochondrial fragmentation.

Globally, an estimated 2.38 million children and adolescents (0–19 years) are living with HIV, with 80% residing in sub-Saharan Africa. In Uganda, adolescents remain at high risk, with approximately 6,600 new infections among 10–19-year-olds in 2023. Despite expanded access to antiretroviral therapy (ART), adolescents Living with HIV (ALHIV) continue to experience poor outcomes, including low viral suppression and high mortality, largely driven by non-adherence. However, ART adherence represents only one dimension of their lived experience. ALHIV must also navigate intersecting adversities, including poverty, stigma, orphanhood, and exposure to sexual and gender-based violence, while meeting academic expectations, particularly in boarding school …

Sex differences, in terms of prevalence, symptoms and disease progression, are established in the aetiology of complex neurodegenerative diseases, including amyotrophic lateral sclerosis, Parkinson's disease and Alzheimer's disease, but the underlying biology driving these differences remains poorly understood. There is emerging evidence from genetic and functional analyses affirming the role of the immune system in such diseases, but a thorough assessment of sex differences in the link between the immune system and neurodegenerative diseases remains lacking. Bdelloid rotifers are renowned for their extraordinary resilience. They can survive desiccation and long-term freezing and revive once favorable conditions return. Such remarkable survival abilities suggest that the evolution of specialized molecular adaptations sustain pH homeostasis under extreme environmental stress. In the present study, we identify and functionally characterize an isoform of the voltage-gated proton channel (Hv) from the bdelloid rotifer Adineta ricciae, which we name ArHv2. Unlike its paralog ArHv1 and human Hv1 that activate only at positive membrane potentials and contribute modestly to proton extrusion under normal conditions, the ArHv2 activates even at negative potentials, enabling rapid and …

Background: Cancer and its treatments commonly result in adverse effects that impair function, well-being, and quality of life, necessitating access to skilled physical therapists (PTs). Although oncology is an essential component of physical therapist practice, entry-level education in cancer rehabilitation has historically been limited. In 2025, the Essential Competencies for Physical Therapy Entry-Level Education in Oncology were developed through a mixed-methods, modified Delphi process, establishing 19 competencies across six domains and defining minimum expectations for new graduates. To support translation of these competencies into academic curricula, a practical implementation resource was needed.

Comparative Philosophy

Interdisciplinary approaches to teaching China in Western academic contexts often reproduce Eurocentric frameworks that reduce Chinese philosophical and cultural traditions to subordinate objects of analysis. This article proposes an alternative model grounded in intercultural philosophy and the Filosofías del Sur, arguing that China should be approached not as a distant object of representation, but as an epistemic interlocutor capable of transforming philosophical education. The article develops this argument through the recovery of the República del Reino de la China, included in the 1595 edition of Repúblicas del Mundo, as a key example of early intercultural mediation from the European South. …

Comparative Philosophy

Language and cultural barriers pose major obstacles to teaching Chinese philosophy to Western philosophy students. Without a cultural and linguistic background, students often struggle to read the original Chinese philosophical texts. Translation also involves issues of interpretation that are difficult to be settled in undergraduate-level courses. To address this situation, this paper introduces a question-oriented pedagogy called “intercultural questioning”. Instead of aiming at a comprehensive understanding of the entire Chinese philosophical tradition, the question-oriented pedagogy reveals the implicit assumptions in one’s conceptual frameworks and unexamined questions in Western philosophy through the East-West dialogue. Why Minds Need Bodies: Organismic Unity And Limits Of Artificial Consciousness, Renée Snow

Comparative Philosophy

This paper argues that functional organization alone is not sufficient for phenomenal consciousness or the existence of a unified subject of experience. While many contemporary accounts of artificial consciousness assume that replicating the causal or computational structure of the brain would be sufficient for subjective experience, this paper identifies a deeper constraint: consciousness depends on organismic unity. The argument proceeds through a triangulation of conceptual, empirical, and metaphysical considerations. First, Zuboff’s thought experiment shows that functional organization can be preserved while the unity of the subject dissolves, suggesting that causal pattern replication alone cannot secure subjectivity. We present a data-assimilative regional ocean biogeochemical model, ASTE-BGC, which simulates the physical and biogeochemical state of the North Atlantic Ocean from 2002 to 2017. Model physics are provided by a physical state estimate (ASTE), which assimilates O(109) in situ and satellite-based observations over the model domain and time period. Model biogeochemistry is simulated by a medium-complexity biogeochemical module (BLING), which simulates nine prognostic biogeochemical tracers and three ecological classes. Through a three-step optimization process which combines informal adjustments and formal mathematical optimization, we minimize model-data misfit between a “spun-up” simulated biogeochemical state and O(105) BGC-Argo …

Attention-Deficit/Hyperactivity Disorder (ADHD) is a neurodevelopmental disorder associated with less developed working memory (WM) processes, theorized to be a core contributing factor in the development of ADHD and a predictor of ADHD symptom severity (Alderson et al., 2013; Rapport et al., 2008). Objective measures inclusive of working memory tasks, when used in conjunction with self-report measures, have been previously shown to increase the accuracy of diagnosing ADHD (Marshal et al., 2021. Blue crabs, Callinectes sapidus, support valuable commercial fisheries throughout the U.S. Atlantic and Gulf coasts, and understanding variation in reproductive output has important management and conservation implications. This study examined two components of blue crab reproduction: (1) reproductive investment across populations spanning multiple latitudes (New York, New Jersey, North Carolina, Mississippi, and Texas), using a space-for-time substitution to assess the effects of varying thermal regimes on fecundity and egg quality, and (2) spawning phenology in North Carolina from …

The construction of new data centers for artificial intelligence has become a major engine of the U.S. economy. More than 700 data centers are now under construction across 40 states, with many more to come.

These are typically multi-billion-dollar projects. This boom is facing growing public opposition, which has led to an extremely volatile regulatory land-scape, especially at the state and local levels. That landscape is the subject of this article. This volatility has raised the cost of capital for data centers and led to the cancellation of many, and to heightened due diligence by lenders and others.

However, research consistently indicates that many adult patients in acute care experience inadequately managed pain, which may contribute to adverse patient outcomes. Studies have shown that nonpharmacological pain management can lead to increased patient satisfaction, shorter hospital stays, and better patient outcomes. Background: Veterans with epilepsy experience high rates of psychiatric comorbidity, most commonly major depressive disorder (MDD). Barriers to accessing mental health care—including geographic distance, transportation limitations related to seizure-related driving restrictions, and fragmented care between neurology and psychiatry, contribute to the underdiagnosis and undertreatment of MDD in this population. Neurobehavioral Therapy (NBT), a Cognitive Behavioral Therapy (CBT)-informed psychotherapy developed for individuals with seizures, has demonstrated benefits for psychological distress and quality of life. Wildland firefighters perform physically demanding work under challenging environmental conditions that place substantial demands on nutritional health and occupational performance. Despite the importance of nutrition in supporting physical performance, recovery, and long-term health, limited research has examined nutrition knowledge and dietary behaviors among California wildland firefighters or evaluated the feasibility of remotely delivered nutrition education interventions within this occupational population. Therefore, the purpose of this pilot study was to assess baseline nutrition knowledge, nutrition self-efficacy, and dietary behaviors while evaluating the feasibility of a 12-week WhatsApp-delivered nutrition education intervention among California wildland firefighters.

INTRODUCTION: Dysfunctional microRNAs and GABAergic interneurons are features of Alzheimer's disease (AD). The role of neuronal microRNA155 (miR155), elevated in both AD and Down syndrome (DS), remains unknown.

METHODS: We utilized in silico analyses of published databases, MIR155-deleted and -overexpressing human induced pluripotent stem cell (hiPSC)-derived cells, cortical organoids, and amyloid beta precursor protein (APP)/PS1-miR155 knockout mouse.

RESULTS: MIR155HG (miR155 host gene) colocalizes with APP in a neuron-specific, topologically associated domain (TAD) in chromosome 21. In human neural stem cells (NSCs), neurons, and cortical organoids, MIR155 deletion enhanced NSC proliferation and GABAergic interneuron generation. Over the past decade, the Model Organism Development and Evaluation for Late-Onset Alzheimer's Disease (MODEL-AD) consortium has transformed preclinical Alzheimer's disease (AD) research by addressing critical limitations in traditional mouse models that failed to translate to human disease. By leveraging human genetic discoveries, MODEL-AD has developed > 70 genetically informed mouse models, standardized phenotyping pipelines, and an open-access data infrastructure aligned with late-onset AD biology. These models incorporate human risk variants, environmental factors, and aging to better capture disease complexity, including emerging recognition of mixed pathologies such as vascular contributions, Lewy body disease, and TDP-43 proteinopathy. Microenvironment remodelling impacts tumour growth and metastasis, but whether remodelling promotes pre-malignant clonal fitness remains unknown. Here, using single-cell RNA-sequencing of the bone-marrow microenvironment in a mouse model of DNMT3A-mutant clonal haematopoiesis (CH), we identify mesenchymal stromal cells (MSCs) in a molecular state of cellular senescence. Elevated bone-marrow MSC senescence is also observed in humans with CH driven by several common somatic mutations. MSC senescence is induced by mutant haematopoietic cells in a contact-independent manner through production of soluble factors including TNF-α and IL-6. Vascular-related factors are now considered major contributors to most forms of dementia, including Alzheimer's disease. However, the degree to which vascular deficits contribute to risk, onset, and progression of cognitive impairment and dementia has only recently been appreciated. Our understanding of the mechanisms by which vascular deficits drive cognitive decline in dementia is still limited. Further, the testing of therapeutic approaches to prevent vascular deficits to treat dementia are few. This review shares the ongoing work of the global Worldwide Innovative Network (WIN) Consortium for Precision Medicine to synthesize emerging cancer treatment data and to define the requirements for a common global cancer database that can truly support precision oncology. We performed a narrative review of emerging cancer treatment data, molecular profiling technologies, and existing clinicogenomic databases, focusing on how tumors are characterized, how subgroups are defined, and how demographic, lifestyle, and environmental factors are captured. INTRODUCTION: Post mortem proteomic analysis of Alzheimer's disease (AD) brain tissue has identified novel target genes and proteins for potential therapeutic development. Moesin (MSN) and CD44 were identified as candidate targets. Using human induced pluripotent stem cells (hiPSCs)-derived neurons, we assayed how reducing gene expression of MSN and CD44 affected amyloid beta secretion, tau phosphorylation, and transcriptional state.

METHODS: Knockdown of MSN and CD44 in hiPSC-derived neurons was performed using short hairpin RNA (shRNA). Amyloid precursor protein processing and intracellular tau phosphorylation was measured using chemiluminescent ELISA assays. Global gene expression was analyzed by bulk RNA sequencing (RNA-seq).

The purpose of this phenomenological study was to explore educators’ lived experiences implementing schoolwide Positive Behavioral Interventions and Supports (PBIS) discipline systems and to understand how those experiences influenced school culture in private PK–12 schools. Historically, PBIS focused on specific outcomes, such as reductions in discipline referrals, suspensions, and behavioral incidents. Though these indicators are critical, they do not consider in what way educators experience disciplinary systems, nor how their lived experiences affect routine interactions with students. Educators play a key role in implementing PBIS; their experiences foster consistency, trust, and sustainability. The purpose of this qualitative phenomenological study was to research secondary STEM educators’ perceptions of PD needs. This study focused on the lived experiences of educators actively teaching in STEM subject disciplines in grades nine through 12. The essential question for this research study was: How do Tennessee secondary STEM teachers perceive the relevance, applicability, and impact of PD opportunities? The research was rooted in Knowles’ (1984) theory of adult learning, Andragogy, and Learning Forward’s (2022) Standards for Professional Learning.

Continuum robots are a class of continually deformable robots inspired by snakes and tentacles that are commonly used in medical, search and rescue, and exploration applications. The robots are highly nonlinear and difficult to accurately model and control. Precise control of the pose of a continuum robot is typically reliant upon a system model. The function of a material is defined not only by its chemical composition, but also by the structural organization of its constituents across multiple length scales, from the architecture of individual building blocks to their collective arrangement. Developing strategies to access, engineer, and reconfigure that structure on demand represents a central opportunity in the design of adaptive materials. Here, we present a thermoresponsive polymer-colloid platform that enables high-precision tuning of colloidal structures using only thermal stimuli.
