Background: Lead (Pb) persists in urban soils, where its partitioning among geochemical fractions governs mobility, bioavailability, and human health risk. These fractions are strongly controlled by soil physicochemical properties, necessitating site-specific remediation strategies. Methods: This study developed a site-specific Pb immobilization framework linking amendment selection to soil geochemistry and sustainability considerations. Pb-contaminated residential soils from three U.S. cities, San Antonio (alkaline), Baltimore (acidic), and Detroit (near-neutral), were treated with gypsum, biochar + lime, and alum, respectively. Changes in Pb speciation were tracked using sequential extraction over 7, 30, and 90 days. Results: All amendments significantly reduced exchangeable Pb (F1) and …

Background: Practice patterns of surgical management of rectal adenocarcinoma vary among US regions. Nationwide, there has been an overall trend toward the performance of anterior resection (AR) over abdominoperineal resection (APR) in the surgical management of invasive rectal adenocarcinoma when feasible. It is suspected that this trend is adopted at a slower rate in underserved regions of the United States. Methods: Data from the NCDB for 2004–2019 was queried for patients with stage I-III rectal adenocarcinoma who underwent anterior resection (AR) or abdominoperineal resection (APR). Comparisons of AR versus APR were made among year, region, approach, and stage. Results: Of …

Background
The Zanzibar archipelago has been a pre-elimination region for malaria thanks to rigorous control interventions, but recent surges in malaria cases have been observed. The contribution of non-falciparum species to the current malaria situation is unknown.
Method
This study investigates the epidemiology of falciparum and non-falciparum malaria species in Zanzibar. Leveraging plasma extracted from dried blood spots (DBS) collected during reactive case detection (RCD) activities across Unguja island from May 2022 to January 2023, we measured immunoglobulin G (IgG) responses to Plasmodium MSP1-19kD antigens using a multiplex bead-based immunoassay. Additionally, active infections were detected using species-specific real-time PCR.

On Robust Estimation Of Multivariate Location And Scale With Applications, Soumya Chakraborty
On Robust Estimation Of Multivariate Location And Scale With Applications, Soumya Chakraborty
Doctoral Theses
The principal objective of this thesis is, in a nutshell, to provide robust estimators of multivariate location and scale which have reasonable to high model efficiency but avoid high computational complexity so as to be practically useful in real problems. We utilize the minimum density power divergence (DPD) and the related philosophy to invoke robustness. There are some computational issues while minimizing the DPD in different multivariate set-ups. We will work on this problem rigorously and come up with three types of estimation procedures which are explicitly or implicitly related to the minimum DPD methodology, keeping the computational issue in …

INTRODUCTION: Persistent postsurgical pain (PPSP) affects up to 15% of patients after major surgery, impairing physical function, quality of life and increasing risk for long-term opioid use. Current PPSP prediction models rely on static or retrospective data and fail to incorporate dynamic perioperative factors. The Personalised Prediction of Persistent Postsurgical Pain (P5) study aims to develop individualised, multimodal prediction models by integrating preoperative behavioural, psychophysical and neurocognitive assessments and high-frequency symptom monitoring.
METHODS AND ANALYSIS: P5 is a prospective, single-centre cohort study enrolling 2500 adults aged 18-75 undergoing major surgery at a tertiary academic hospital. Participants complete baseline surveys, cognitive …

Over the last decade or more, our collaborative research has provided noteworthy information on the helminth parasites of reptiles of Arkansas. Because several species still need to be surveyed from the state, some for the first time, there has been a general void of information on their parasites. We therefore examined 10 reptiles, including seven species/subspecies of turtles, as well as three species/subspecies of snakes for helminth parasites. Found were 24 taxa of parasites, including two monogeneans, 14 digeneans, one tapeworm, and seven nematodes. We document several new host and distributional records for these parasites from some reptiles of the …

The state of Maryland has experienced significant but commonly overlooked impacts on water quality and quantity. Future climate change is predicted to further reduce water availability, with nontraditional water sources such as treated recycled wastewater (TRW) as one potential way to mitigate impacts. We conducted semi-structured interviews with 17 Maryland stakeholders—including farmers, extension agents, state agency personnel, and academics—to explore their perceptions and experiences with 1) past, existing and future weather variability; 2) freshwater and reusable water use and management in agriculture; and 3) related factors impacting agricultural water management. Qualitative coding of interview transcripts produced 27 categories containing 291 …

Owing to their immunoprotective properties, natural killer (NK) cells are critical for the innate immune response to pathogens, as well as a new wave of cancer immunotherapy that harnesses natural cytotoxicity. We sought to study the genetic and epigenetic drivers behind human-specific NK cell receptors, so that we can better understand the underlying cellular function. Here, we present a transcriptomic, proteomic (CITE-seq), and chromatin (single nuclei ATAC-seq) profiling of human peripheral NK cell subsets, which was then compared with genomic databases. Through integrative multi-omics, we demonstrate that CD56bright versus CD56dim NK cell subsets have differential distal regulatory element …

Abstract – Heart failure (HF) is a complex, multifactorial clinical syndrome with diverse etiologies and phenotypic manifestations, affecting millions of people globally. HF is broadly classified into 3 subtypes: HF with reduced ejection fraction, HF with mildly reduced ejection fraction, and HF with preserved ejection fraction. Each subtype exhibits distinct pathophysiological mechanisms, clinical trajectories, and therapeutic responses, posing significant challenges in terms of diagnosis and management. Epicardial adipose tissue (EAT), a metabolically active visceral fat depot situated in close proximity to the myocardium, plays a dual role in physiological and pathological states, offering potential utility as a biomarker and therapeutic …

OBJECTIVES: The impact of the genetic risk for RA on resultant disease phenotype in RA is incompletely understood. Using individual genetic variants associated with RA and a polygenic score (PGS), we hypothesized that those with higher genetic risk for RA would demonstrate a more severe disease course.
METHODS: We genotyped participants from the Veterans Affairs RA registry (VARA), a prospective cohort of RA. We evaluated associations between PTPN22 R620W genotype, HLA-DRB1 shared epitope (SE; 0, 1, or 2 copies of a high-risk HLA allele), and a non-HLA PGS with RA disease activity scores and disease characteristics using linear and logistic …

Quantum Technologies, Part Two: Recognizing Risks And Threats To National Security And Defense, James Giordano
Strategic Insights
This commentary is Pt. 2 of the Quantum Technologies series and examines the emerging national security implications of quantum technologies and the risks posed by their development and potential use by adversaries. While quantum capabilities remain in relatively early stages of maturity, this piece explains that advances in areas such as quantum computing, sensing, and communications could significantly affect military operations, intelligence, and strategic competition. The analysis emphasizes the importance of realistically assessing both the capabilities and limitations of quantum technologies to avoid exaggerated expectations or complacency. Rather than assuming quantum technologies will immediately transform warfare, the greater risk lies …

To understand the relative importance of cis and trans effects on regulation, we crossed multi-parent recombinant-inbred lines (RILs) to a common tester and measured allele-specific gene expression in the offspring. Testing the difference of allelic imbalance between two RIL × Tester crosses is a test of cis or trans, depending on the RIL alleles compared. The study design also enables to separation of two sources of trans variation, genetic and environmental, detected via interactions with cis effects. We demonstrate the effectiveness of this approach in a long-read RNA-seq experiment in female abdominal tissue at two time points in Drosophila melanogaster. …

Dysregulated ribosome biogenesis and p53 mutations are known to play oncogenic roles in various cancers, including pancreatic cancer. In this study, we demonstrated the therapeutic potential of BMH-21, a pharmacologic inhibitor of RNA polymerase I, against pancreatic cancer by uncovering a novel molecular mechanism involving RPA194-mediated ubiquitination of mutant p53 without affecting the ubiquitination of wild-type p53. Our key findings are that (i) BMH-21 selectively induces apoptosis and cell growth inhibition of pancreatic cancer cells with no effect on normal human pancreatic ductal epithelial cells; (ii) BMH-21 degrades RPA194; (iii) BMH-21 inhibits recruitment of both RPA194 and RPA135 on rDNA …

Foundation models have emerged as powerful feature extractors in computational pathology. However, they typically omit mechanisms for leveraging the global spatial structure of tissues and the local contextual relationships among diagnostically relevant regions-key elements for understanding the tumor microenvironment. Multiple instance learning (MIL) remains an essential next step following the foundation model, designing a framework to aggregate patch-level features into slide-level predictions. We present EAGLE-Net, a structure-preserving, attention-guided MIL architecture designed to augment prediction and interpretability. EAGLE-Net integrates multi-scale absolute spatial encoding to capture global tissue architecture, a top-K neighborhood-aware loss to focus attention on local microenvironments, and background suppression …

Radiation therapy is integral to the treatment regimens of over 50% of cancer patients. However, it is not biologically targeted to just the tumor, leading to adverse effects in peritumoral normal tissue. Selective uptake of gold nanoparticles (AuNPs) by tumors realizes tumor-specific radiosensitization via increased secondary electron release from high atomic number gold atoms. Here, we review AuNP-mediated radiosensitization and outline strategies to optimize tumor-targeted AuNP delivery. Modifying the physicochemical characteristics of AuNPs, including size, shape, charge, and surface chemistry, can increase their ability to evade the reticuloendothelial system (RES) and penetrate the dense tumor stromal architecture, thereby promoting tumor …

Soft tissue sarcomas (STSs) are rare malignant tumors of mesenchymal origin with heterogeneous histologic subtypes and variable clinical behavior. Although STSs most commonly arise in the extremities, rapidly progressive sarcomas involving the distal lower extremity in adolescents have not been well documented. We report a case of an STS in a previously healthy adolescent female whose initial presentation was presumed to be a muscle strain. Over a three-month period, her symptoms worsened with progressive enlargement of a calf mass and development of widespread metastatic disease. This case highlights the importance of maintaining suspicion for malignancy in rapidly enlarging soft tissue …
