Dissecting Tumor Evolution And Immune Composition In Prc2 Loss (H3k27me3 Loss) Vs Retained Mpnst, Kuangying Yang
Arts & Sciences Graduate Student Theses and Dissertations
Malignant peripheral nerve sheath tumors (MPNSTs) are aggressive sarcomas that arise sporadically or in the setting of neurofibromatosis type 1 (NF1). Although benign plexiform neurofibromas (PNs) are common in NF1, only a subset undergo malignant transformation, and the molecular events that define high-risk progression remain incompletely understood. A major axis of biological heterogeneity in MPNST is loss of Polycomb Repressive Complex 2 (PRC2), commonly detected through loss of H3K27me3 and frequently mediated by alterations in SUZ12 or EED. This dissertation integrates three manuscript-based studies to define genomic resources, tumor-intrinsic vulnerabilities, and immune microenvironmental remodeling in NF1-associated peripheral nerve sheath tumors, …
Data-Driven Staging Of Neurodegenerative Pathology Related To Alzheimer’S Disease, Thomas Earnest
Data-Driven Staging Of Neurodegenerative Pathology Related To Alzheimer’S Disease, Thomas Earnest
McKelvey School of Engineering Graduate Student Theses & Dissertations
Alzheimer’s Disease (AD), the most common cause of dementia, is a dual proteinopathy exhibiting deposits of extracellular amyloid-beta (Aβ) and intracellular tau in brain tissue. Models which describe the spatial and temporal trajectories of these pathologies have been invaluable for understanding AD and have the potential to support diagnosis, disease monitoring, and treatment development. Disease staging systems, which provide protocols for assessing individual disease severity based on their extent of brain pathology, have been crucial for explaining and diagnosing AD. However, these systems have been primarily informed by post-mortem histological assessments and are not directly applicable to disease assessment in …

The Expert Myth: Impostor Phenomenon And Research Identity In Counselor Education, Kristi B. Cannon, Kelly Coker, Trent Hepting, Lidian Ramirez-Fiol, Robyn T. Simmons
Journal of Counselor Preparation and Supervision
Imposter Phenomenon (IP) affects counselor educators across career stages, yet it is often conceptualized as an individual deficiency rather than a systemically reinforced experience. This article frames IP in counselor education through the lens of the Expert Myth—a systemic pattern within academic culture that reinforces the expectation that faculty must consistently present as fully knowledgeable, productive, and self-assured to be regarded as credible. We examine how institutional cultures, publish-or-perish pressures, and inadequate mentorship structures create and reinforce imposter feelings, particularly among marginalized faculty and those navigating researcher identity development. Drawing on existing literature, we propose that meaningful intervention requires systemic …

Rapid Detection And Application Of Rock Mass Joint Features Based On Improved Yolo Model, Li Huixing, Zhang Jing, Zhang Jiannan, Fang Hao, Chen Tao, Wang Changshuai
Journal of China & Foreign Highway
In view of the problems that existing monitoring methods for rock mass joints mainly rely on manual identification, with low detection efficiency and strong subjectivity, a detection and segmentation method for rock mass joints was proposed in this paper. The model improved the YOLOv8 algorithm by introducing the multi-scale feature module (MSBlock) and channel prior convolutional attention (CPCA) mechanism to enhance the image recognition accuracy of the existing YOLOv8 network model. By introducing the MSBlock algorithm, features at different scales and levels were fused, which could both capture the fine local details in the image and recognize the image features …

Plasma-Generated Ozone For Odor Removal, Jordan Marot
Plasma-Generated Ozone For Odor Removal, Jordan Marot
Seton Hall University Dissertations and Theses (ETDs)
Ozone (O3) is gaining popularity for its use in industrial and environmental applications, including water treatment, air purification, disinfection, and odor remediation. As a powerful oxidizing agent, ozone is highly effective in destroying harmful bacteria and degrading hazardous gas pollutants such as hydrogen sulfide (H2S). Dielectric barrier discharge (DBD) reactors are widely used for commercial ozone production under atmospheric-pressure conditions. However, ozone yield and selectivity are strongly influenced by operating parameters, particularly gas flow rate, which affects residence time, energy transfer, and competing byproduct chemistry.
This study explores the influence of the air flow rate on …

Characterization Of Zdhhc Palmitoyltransferase Expression During Retinoic Acid-Induced Differentiation Of Sh-Sy5y Neuroblastoma Cells, Mohammad Abdallah Matari
Seton Hall University Dissertations and Theses (ETDs)
Protein S-Palmitoylation is a reversible post-translational modification that regulates protein trafficking, membrane localization, and neuronal function through the activity of the zinc finger Asp-His-His-Cys (zDHHC) family of palmitoyl acyltransferases. Although SH-SY5Y neuroblastoma cells are widely used as an in vitro model of neuronal differentiation, the expression profile of the complete zDHHC family during retinoic acid (RA)-induced differentiation has not been systematically characterized. The objective of this study was to characterize the expression of all 23 human zDHHC genes during RA induced differentiation of SH-SY5Y cells. SH-SY5Y cells were differentiated using 10 µM all-trans retinoic acid for up to seven days. …
Expression Profiling And Bioinformatic Characterization Of Depalmitoylation-Associated Genes During Sh-Sy5y Neuronal Differentiation, Ayesha Lodhi
Seton Hall University Dissertations and Theses (ETDs)
Palmitoylation is a reversible post-translational modification involving covalent attachment of palmitoyl groups to cysteine residues. Till date, 23 members of this family have been identified in mammals, located in Golgi apparatus and endoplasmic reticulum and nearly 40% of synaptic proteins are estimated undergo palmitoylation. These proteins include vesicle fusion proteins (SNARE), signaling molecules, ion channels and neurotransmitter receptors such as gamma-aminobutyric acidreceptors, kainate receptors and glutamate receptors. While its role in neuronal function has been widely recognized, comparatively little is known about how depalmitoylation-associated enzymes are regulated during neuronal differentiation. The present study aimed to characterize changes in depalmitoylation-associated gene …

Long non-coding RNAs (lncRNAs) are emerging as important regulators of cellular adaptation to environmental and molecular stress, but the extent to which their responses remain reproducible and stress-type-dependent across human stress conditions remains unclear. Here, we performed an integrative transcriptomic meta-analysis of human stress-response datasets from ASTRA and GEO, focusing on normal, non-cancerous, wild-type human cell lines exposed to oxidative stress (H2O2), hypoxia, heat stress, or UV-induced DNA damage. Gene Ontology (GO) enrichment analysis of differentially expressed (DE) protein-coding mRNAs confirmed that the resulting stress-stratified comparison captured biologically coherent transcriptional programmes to oxidative stress signaling, hypoxic and metabolic adaptation, heat-induced …
Host-Pathogen Dynamics Underpinning Staphylococcus Aureus Growth During Intracellular Osteoclast Infection, Luke D. O'Connor
Host-Pathogen Dynamics Underpinning Staphylococcus Aureus Growth During Intracellular Osteoclast Infection, Luke D. O'Connor
WUSM Theses and Dissertations – All Programs
Bone infection is most commonly caused by the gram-positive bacterium Staphylococcus aureus. It has myriad ways in which it can circumvent or subvert killing by the host or antibiotics. Intracellular infection is one such niche that is protective from immune and antibiotic attack. In 2019, Krauss and colleagues discovered that infected osteoclasts (OCs) allow S. aureus to replicate intracellularly. There they found that the ability of S. aureus to grow inside the cell was tied to OC differentiation. The magnitude of overnight bacterial expansion positively correlated with OC maturity and duration cultured with the osteoclastogenic factor RANK ligand (RANKL). Bacterial …

“Something Else To Focus On”: Perinatal Havening And Stress Coping In Pregnant Black Women, Brittany L. Ferrell Young
WUSM Theses and Dissertations – All Programs
Black women in the United States experience maternal mortality at more than three times the rate of white women, a disparity driven by chronic stress rooted in structural racism rather than individual behavior or biology. Accessible, culturally responsive tools for managing stress during pregnancy remain scarce. This mixed-methods pilot feasibility study examined Perinatal Havening, a self-administered psychosensory technique, as a stress management intervention for pregnant Black women. Guided by Lazarus and Folkman's Transactional Theory of Stress and Coping, the study addressed three questions: how Black women experience acute and chronic stress during pregnancy, the acceptability and preliminary influence of Perinatal …
