Freeze-Dried, Not Frozen: Lyophilized Mesenchymal Stromal Cell-Derived Extracellular Vesicles And Therapeutic Function In Neuroinflammatory Models, Janet R Ashley, Aidan M Collier, Scott D Olson, Charles S Cox

Faculty, Staff and Student Publications

Background: Mesenchymal stromal cell (MSC)-derived extracellular vesicles (EVs) offer a promising acellular therapy for immune modulation, but clinical translation is hindered by variability and storage constraints. Lyophilization (freeze-drying) could enable shelf-stable EV therapeutics, although its effects on biological activity remain incompletely defined.

Study design: We compared the immunomodulatory effects of frozen and lyophilized EVs from bone marrow (BM), adipose tissue (AD), and umbilical cord (UC) MSCs using lipopolysaccharide-stimulated rodent splenocytes and microglial models and human peripheral blood mononuclear cells (PBMCs). Variables included batch scale (small vs large), cytokine priming, and MSC source. Tumor necrosis factor alpha (TNF-α) secretion was measured …

PRDM1, encoding a transcription factor (TF), regulates plasma cell and CD8+ T-cell terminal differentiation and Th2 lineage specification, while its role in human NK-cell differentiation and homeostasis is largely unknown. Here, we employed a multi-omics approach to dissect the transcriptional control of PRDM1 on human NK-cells. PRDM1 is important in NK-cell terminal differentiation based on gene expression profiling and its targeting of key regulators in the process. PRDM1-deleted NK-cells displayed a less mature phenotype simulating the CD56bright NK-cell population accompanied by upregulation of stem-like gene signatures. PRDM1-bound genes were enriched in T/NK-cell receptor signaling, activation, and NK-cell effector …

South Asian Nutrition Guide For Heart Health: A Practical Guide To Sodium, Saturated Fat, Packaged Foods, Recipes And Meal Planning, Ashwini Wagle Jan 2026

South Asian Nutrition Guide For Heart Health: A Practical Guide To Sodium, Saturated Fat, Packaged Foods, Recipes And Meal Planning, Ashwini Wagle

Open Educational Resources

South Asians, including people with ancestry from India, Pakistan, Bangladesh, Sri Lanka, Nepal, Bhutan, and the Maldives, have a higher risk of cardiovascular disease and diabetes than many other populations. They often develop heart disease at younger ages and sometimes at lower body weights. (Jose, 2024; Patel et al., 2021; Volgman et al., 2018).

This increased risk is not caused by a single factor. Evidence points to a combination of inherited, metabolic, lifestyle, and social influences. Common contributors include insulin resistance, abdominal or visceral fat, type 2 diabetes, high triglycerides, low HDL cholesterol, elevated lipoprotein(a), family history, stress, migration-related changes …

Neural Basis Of Private And Social Decision-Making: New Insights From Transcranial Ultrasound Stimulation And Multivariate Analysis, Mathilde Lojkiewiez Jan 2026

Neural Basis Of Private And Social Decision-Making: New Insights From Transcranial Ultrasound Stimulation And Multivariate Analysis, Mathilde Lojkiewiez

School of Psychology Theses

Value-based decision-making is a complex process that requires the integration of many pieces of information, or decision variables, to choose among several options. These decision variables include the expected value of an option, its associated risk, or the anticipation of the emotions that one could experience when seeing the outcomes of all possible options. In some cases, values attributed to options are learnt from experience, and additional strategies can be employed to refine them. One of them is to use information from the social environment in which decisions are made. Observational learning allows one to learn from observing others making …

The Effects Of Altered Partial Pressures Of Gas On Cognitive And Neural Function, Daniel Graham Jan 2026

The Effects Of Altered Partial Pressures Of Gas On Cognitive And Neural Function, Daniel Graham

School of Psychology Theses

Oxygen is central to neural function, yet the precise mechanisms and effects by which varying oxygen levels, whether through Hypoxia, Hyperoxia, or Hyperbaric Oxygen Therapy (HBOT), shape cognition and brain activity remain incomplete. This thesis adopts a novel, multi-modal framework that integrates normobaric gas manipulations, cognitive testing, Transcranial Magnetic Stimulation (TMS), Electroencephalography (EEG), Functional Magnetic Resonance Imaging (fMRI), and HBOT to examine how gas and pressure variability influences cognitive, motor, and neural processes. Chapter 3 presents the findings of a preliminary experiment investigating normobaric oxygen manipulations on cognition. Standardised cognitive assessments revealed domain-general impairments (i.e. memory and executive function) under …

Virtually Painless: An Investigation Into The Therapeutic Potential Of Virtual Natural Environments For Chronic Low Back Pain, Alexander Smith

School of Psychology Theses

Chronic pain conditions represent one of the greatest causes of disability in the UK, affecting up to 28 million people nationwide. Despite its prevalence, chronic low back pain (cLBP) in particular remains poorly served by existing treatments. Growing evidence suggests that engagement with natural environments provides analgesic and restorative benefits, yet access to nature is profoundly limited for people living with chronic pain. This thesis investigated whether a codesigned, immersive virtual reality (VR) nature experience could deliver meaningful pain relief to individuals with cLBP, drawing on Attention Restoration Theory (ART) and Stress Reduction Theory (SRT) as key theoretical frameworks. The …
