Cold atmospheric plasma (CAP) has potential as a new cancer treatment tool due to its room temperature biocompatibility and possible cancer cell selectivity with low side effects. This review examines the current research stage of CAP therapy for Prostate Cancer (PCa) treatment with a focus on CAP device standardization and CAP therapy clinical translation. The current CAP preclinical data is explored through the lens of PCa cellular response, PCa cells selectivity, CAP treatment type and CAP combination therapies. Lipid nanoparticle (LNP)-encapsulated nucleoside-modified mRNA vaccines elicit robust CD4+ T cell responses, yet the mechanisms underlying this T cell priming remain unknown. Antigens presented to CD4+ T cells on major histocompatibility complex class II (MHC II) are traditionally acquired by antigen presenting cells (APCs) from extracellular sources. Here we show that vaccine specific CD4+ T cell responses instead rely on antigen directly expressed within APCs, without extracellular transit. Murine APCs treated with mRNA-LNP vaccines activate T cells more efficiently when presenting antigen produced internally, rather than acquired externally. Background: Antibiotic overuse in Neonatal Intensive Care Units (NICUs) is a major contributor to antimicrobial resistance and adverse neonatal outcomes. This study aims to evaluate baseline antibiotic utilization (AU), identify factors influencing variability, and assess the impact of neonatal characteristics and sepsis incidence. Methods: A multicenter retrospective analysis examined AU in seven NICUs from 2019 to 2023, involving 25,532 neonatal admissions during national antibiotic stewardship program implementation. Data encompassed neonatal clinical parameters, sepsis incidence, and AU metrics, including days of therapy (DOT) per 1000 patient-days. Statistical analyses included correlation assessments and multivariate regression to identify determinants of antibiotic use. Results: …

Creating genetic sensors for noninvasive visualization of biological activities in optically opaque tissues holds immense potential for basic research and the development of genetic and cell-based therapies. Magnetic resonance imaging (MRI) stands out among deep tissue imaging methods for its ability to generate high-resolution images without ionizing radiation. However, the adoption of MRI as a mainstream biomolecular technology has been hindered by the lack of adaptable methods to link molecular events with genetically encodable contrast. Here, we introduce modular aquaporin-based protease-activatable probes for enhanced reporting (MAPPER), a platform for the systematic creation of genetic sensors for MRI. To develop MAPPER, …

Purpose: RAPGEF2 encodes a guanine nucleotide exchange factor (GEF) that activates small GTPases and has not been linked to a Mendelian disorder. RAPGEF2 is highly intolerant to loss-of-function variants. We report 5 de novo heterozygous variants in RAPGEF2 in unrelated individuals with developmental delay, attention deficit hyperactivity disorder, epilepsy, dysmorphic features, or other manifestations. We used a Drosophila model to assess the functional impact of the identified human variants.

Methods: We generated a Kozak-GAL4 null allele of the Drosophila ortholog of RAPGEF2, PDZ-GEF, and used the allele to determine the gene expression pattern as well as the loss-of-function phenotypes. We …

Sensory perception is shaped by experience, giving stimuli behavioral significance. Basal forebrain (BF) cholinergic neurons in mice, which are crucial for arousal and motivation, also regulate sensory processing. Within BF nuclei, glutamatergic (vGlut2BF) neurons receive cholinergic input and modulate behaviors, but their roles in encoding sensory significance remain unclear. Using in vivo calcium imaging, we found that vGlut2BF neurons initially poorly encoded odor identity. However, their response to conditioned odors increased following associative learning, and their population activity more distinctly encoded paired stimuli, reflecting emergent value representation. Patients with poorly differentiated thyroid cancer (PDTC) and anaplastic thyroid cancer (ATC) face a much poorer prognosis than those with differentiated thyroid cancers. Around 25% of PDTCs and 35% of ATCs carry the BRAFV600E mutation, which constitutively activates the MAPK pathway, a key driver of cell growth. Although combining BRAF and MEK inhibitors can shrink tumors, resistance often develops. The exact cause of this resistance remains unclear. We previously found that in PDTC and ATC cells, the BRAFV600E mutation is strongly linked to the expression of ETV5, a transcription factor downstream of the MAPK pathway. Introduction: Blood-brain barrier (BBB) dysfunction has been associated with vascular dementia (VaD). However, the underlying mechanisms causing BBB dysfunction remain unclear, especially regarding cerebral hypoperfusion. This study aimed to investigate the effects of asymmetric hypoperfusion on BBB permeability using a mouse model of VaD.

Methods: Asymmetric bilateral common carotid artery stenosis (ACAS) was induced using ligature rings. BBB integrity was assessed on 3 days post-surgery using Evans blue, IgG, and albumin extravasation. A modified tissue processing protocol optimized endogenous marker detection. Regional and sex-based differences in BBB permeability were analyzed.

The 1979 Cortland Conference marked a decisive shift in the adventures of Telos by opening up a debate on narcissism that would pave the way for later contests that, in turn, symbolize in the eyes of many, the errant swerve of Marxism and post-Marxist critical theory. In this Opinions in the Profession article, we explore remediation treatment strategies using exercise for recovery from post-traumatic brain injury and aim to justify the use of such treatment in the occupational therapy profession to increase skills for functional gain. Common treatment strategies used by occupational therapists after brain injury are compensatory strategies, indicating the client learns a new way to perform the task. Though effective, it is also necessary to consider how remediation and neuroplasticity are essential. The Open Journal of Occupational Therapy

Background: Decreased sound tolerance (DST) is among the most common and distressing forms of sensory over-responsivity experienced by children with autism spectrum disorders. DST includes hyperacusis and misophonia, among others, and can limit participation in daily activities. Recently, a new screening tool, the Pediatric Hyperacusis and Misophonia Questionnaire (PMHQ-4) has been developed to allow health professionals to better identify and differentiate these conditions using a single assessment. Additional clinical tools are needed to further characterize DST symptoms to facilitate treatment planning after diagnosis.

Branched-chain amino acid transaminase-1 (BCAT1) initiates the catabolism of branched-chain amino acids (BCAAs), which are essential for neurologic function. However, the role of BCAT1 in neurodevelopment is largely unknown. Here, we identify compound heterozygous BCAT1 variants in a patient with a severe progressive neurodevelopmental syndrome. To investigate the functional consequences, we established patient variant (BCAT1: c.792T>A p.Phe264Leu; c.1042G>A p.Glu348Lys) and BCAT1 knockout hiPSC models. Both disease models show profound defects in cortical neuron differentiation and neurite outgrowth. Furthermore, metabolic analysis revealed evidence of mitochondrial dysfunction associated with increased levels of tricarboxylic acid (TCA) cycle intermediates, glutamate, and glutamine. …

[Objective ] This study aims to elucidate the transformation process of black ginseng saponins and their immunomodulatory mechanisms.[Method ] An ultrasound-assisted enzymatic method was employed for the transformation of black ginseng saponins,and the mass concentration of black ginseng saponins was determined.Furthermore,the immunomodulatory effects of black ginseng saponins were investigated through animal experiments.[Result ] Under the conditions of an enzymatic hydrolysis temperature of 40 ℃,an enzyme mass fraction of 8%,enzymatic hydrolysis time of 60 min,and ultrasonic treatment time of 25 min,the mass concentration of black ginseng saponins increased from 1 615.40 μg/mL to 3 351.38 μg/mL,which represented an increase of 107.47%.The …

Pasture-based ruminant systems link nitrogen (N) nutrition with ecosystem N cycling. Grazing ruminants convert fibrous forages into milk and meat but excrete 65 to 80% of ingested N, creating excreta hotspots that drive ammonia volatilization, nitrate leaching, and nitrous oxide (N2O) emissions. This review synthesizes ecological and ruminant nutrition evidence on N flows, emphasizing microbial processes, biological N2 fixation, plant diversity, and urine patch biogeochemistry, and evaluates strategies to improve N use efficiency (NUE).
