A Bovine Cell Line Resistant To Japanese Encephalitis Virus Entry But Permissive To Post-Entry Replication, Sang-Im Yun, Young-Min Lee

Japanese encephalitis virus (JEV) is a mosquito-borne zoonotic orthoflavivirus that poses a significant global health threat. It causes severe neuroinflammatory disease in humans and reproductive failure in swine. Because of the broad host range and cell tropism of JEV, identifying animal cell lines resistant to infection has been a persistent challenge. In this study, we demonstrate that Madin–Darby bovine kidney (MDBK) cells are resistant to JEV infection yet remain fully permissive to viral replication when transfected with viral genomic RNA. IFN signaling deficient WNV-infected mice develop greater inflammation in the brain, which correlates with loss of gut barrier integrity. The cGAS/STING-dependent innate immune pathway is central in the cellular response to cytosolic DNA derived from viral infections, genotoxic stress, or mitochondrial defects. While efficient activation of the pathway is crucial for defending against pathogens and cancer, maintaining its dormancy without stimuli is equally important to avoid autoimmunity. However, the precise control of the cGAS/STING pathway remains poorly understood. Here, we report that the ion channel TRPV2 regulates both the dormancy and activation of STING. Ollier's disease (enchondromatosis) is a dysplasia of cartilage characterized by multiple enchondromas. Current evidence supports a measurable prevalence of concurrent gliomas in patients with Ollier's disease. In patients with Ollier's disease, isocitrate dehydrogenase 1 (IDH1) and isocitrate dehydrogenase 2 (IDH2) mutations were shown to be a predisposing factor to the development of astrocytomas in conjunction with subsequent mutations in adenosine triphosphate (ATP)-dependent helicase ATRX (ATRX) and tumor protein p53 (TP53). Transdermal microneedle systems offer a minimally invasive strategy for systemic drug delivery in veterinary medicine. In this study, biodegradable microneedle patches composed of polyvinyl alcohol, collagen, and chitosan were evaluated in pigs for the delivery of two model compounds, fluorescein isothiocyanate-dextran (FITC-dextran, 4 kDa) and flunixin meglumine (FLU). Patches were applied to the ear and neck to assess the influence of anatomical site on systemic absorption. FITC-dextran delivered via a single 50 mg patch on the neck achieved approximately 1.2-1.4-fold higher plasma concentrations than oral administration and ear-applied patches, demonstrating enhanced uptake from vascularized regions. Mortality mechanisms play an important role in how oceanic microorganisms contribute to global biogeochemical cycles. Salps are widespread pelagic tunicates known to remove phytoplankton from coastal and high-latitude waters, but their interaction with microorganisms in the vast tropical and subtropical gyres is not well quantified. Using quantitative measurements of six major marine microorganisms in the guts of six distinct but co-occurring salp species from the North Pacific Subtropical Gyre, we examined the impact and dynamics of salp feeding on marine microorganisms in a vast open ocean region. The ETS family transcription factor ETV2, VEGFA, and its receptor FLK1 are essential for hematopoietic, vascular, and cardiac development. Here, we combine dual Etv2 and Flk1 lineage tracing with molecular profiling to define how mesoderm progenitors are allocated to hematopoietic, endothelial, cardiomyocyte, and smooth muscle lineages. Multiple myeloma (MM) is a plasma cell malignancy characterized by bone pain and end-organ failure. A major challenge in treating MM is therapeutic resistance. CD38-targeted immunotherapies, such as daratumumab, have significantly improved outcomes; however, variable responses, resistance, and relapse remain vital challenges. We hypothesized that loss of CD38 drives a more aggressive phenotype and resistance to therapy. To test this, we developed a CD38 knockout (KO) clone of a human MM cell line and evaluated it in immunodeficient mice. Mice with CD38 KO tumors exhibited an increased tumor burden and reduced survival compared with those with CD38 wild-type (WT) tumors. …

Brain myeloid cells accumulate neutral lipids in multiple human neurodegenerative disorders and relevant mouse models. These lipids are often assumed to be contained in lipid droplets (LDs). While studies have been performed in cell culture and Drosophila models to characterize glial LDs, the roles of microglial LD biogenesis in mammalian tauopathy are unclear. To address this issue, we induced the deletion of diacylglycerol acyltransferases (DGATs) 1 and 2, enzymes critical for LD formation, from microglia in the PS19 mouse model of tauopathy. This Children in Cities research project examines an approach to storying research that foregrounds collaborative meaning-making between educators and young children in their city. Through exploring how educators and young children might “think otherwise” about teaching, learning, and participation with history, Place noticing and entanglements between humans, more-than-human, and non-humans, three stories and a metaphor surface in the data. Drawing on three stories, metaphor is employed as an analytic and integrative device to deepen interpretation of lived experiences. An examination of the nature of the human person provides a crucial foundation for understanding human life and identity. Conceptions of personhood have far-reaching implications beyond philosophy, influencing politics, ethics, religion, and epistemology. The notion that the organization of large-scale social structures are rooted in domestic ones has a longstanding history in both Chinese and Greek classical political philosophy. Aristotle and Confucius independently reasoned that a good family, one with a strong father, mapped onto a good state, one with a strong king - though they differed on the details. While there is a strong literature juxtaposing these two titans, less attention has been paid to how their ideas compare with other philosophical traditions. The Coherence Of Navayāna Pragmatism: Ambedkar And The Pragmatist Reconstruction Of Buddhist Philosophy, Danish Hamid

This paper examines the philosophical coherence of B.R. Ambedkar’s reconstruction of his (Navayāna) Buddhism in relation to John Dewey’s pragmatism. Building on Scott Stroud’s reading of Ambedkar as a Deweyan Pragmatist (2017, 2023), it argues that Dewey’s evolutionary naturalism, his conception of growth, and pragmatic redefinition of concepts provide Ambedkar with the methodological tools for a naturalistic and socially reconstructive reinterpretation of Buddhist doctrines such as anicca (impermanence), dhamma (Buddha’s teaching), nirvāṇa (enlightenment), karuna (compassion), etc. Ambedkar transforms impermanence from a metaphysics of flux into an ethics of progressive growth, and recasts dhamma as a social morality grounded in fraternity …

Right-wing commentators (on tv, radio, newspapers, documentaries, or the internet) are often considered a source of misinformation and radicalization in American politics. Understanding the role of such commentators in the political sphere has taken on new significance since the election of Trump, who regularly takes talking points as well as political advice from prominent figures in right-wing media. In his 1949 work Hamlet's Ghost, Richard Flatter wrote of the ghost of Hamlet's father that the play ultimately belongs to him, to the ghost.   Modernity, like the play, belongs to its ghosts, to its dead fathers haunting their wayward sons, the metaphysical specters it imperfectly endeavors to exorcise. Shakespeare's plays are situated upon the threshold of two worlds: a declining traditional order torn apart by political rivalry and ascendant early-modern capitalism. The history plays dramatize infighting among warring factions of privileged aristocrats as well as revolutionary forces propelled by the rising commons and the politics of commodity. Legitimate authority is rarely secure among the kings perched upon Shakespearean thrones. In Shakespeare's Henriad and King John, crowns are contested from the moment they are placed upon royal heads, inspiring Kantorwicz's political theology: the corporeal body of short-lived kings is distinct from the sovereign's sublime body that reigns without cease. …

This essay examines two parallel, historically contemporaneous, depictions of the Freudian master-slave dialectic. John Frankenheimer's first film, The Young Savages (1961), reconstructs Hank Bell's (Burt Lancaster) repressed transformation from Italian racial other into a white ethnic. In doing so, the film approaches the possibility that race itself may be a kind of social construction. To get at this notion, the film explores the meaning of race in an overtly psychoanalytic language. In fact, The Young Savages echoes the argument of Jacques Lacan's nearly contemporaneous essay, "The subversion of the subject and the dialectic of desire in the Freudian unconscious." Lacan's …

The neglect of Goethe, his work in general, and Faust in particular, in English-speaking countries, is notorious. While Shakespeare's plays have been prominent and widely performed in German-speaking countries, the same is not true for Goethe in the English-speaking world. Focusing non-exclusively on Faust, and cutting a link to Thomas Mann's Doctor Faustus, I will endeavor to delineate problematic aspects of American ideology, and what it would take, within U.S. sociology, to be cognizant of the latter.   Georg Lukacs wrote about Goethe and Faust, and so did Theodor W. The impaired skin regeneration, scarring, and delayed healing make the management of burn injuries a challenging task. We designed a photopolymerized hydrogel of gelatin-grafted with glycidyl methacrylate (GMA) for burn management applications. Hydrogel was incorporated with Morin, a plant flavonoid that was originally isolated from the Moraceae family, with known anti-antioxidant and antifibrotic activities. The physicochemical characterization of the resultant hydrogel, including its gelation time and swelling properties, was conducted. The characterization results indicated that the hydrogel development was successful, exhibiting well-established porosity, as evidenced by the SEM images.
