Gecko-inspired adhesives offer strong, reversible, and directionally tunable adhesion, yet fabrication methods often depend on cleanroom lithography or proprietary molds, limiting scalability and accessibility. This study presents a low-cost, modular fabrication strategy combining high-resolution digital light processing 3D printing with 1000 lines/mm optical diffraction gratings to create hierarchical elastomeric adhesives. The resulting structures feature macroscale micropillars and embedded sub-micron surface topography, enabling effective contact splitting without advanced microfabrication. Mechanical testing reveals a nonlinear increase in shear performance with contact area, with maximum shear forces exceeding 80 N at 103.2 cm2. SETD2 is the primary methyltransferase responsible for generating H3K36me3, an epigenetic mark that is essential for transcriptional regulation and chromatin integrity. SETD2 mutations are frequently observed in various cancers and tend to cluster within its catalytic SET domain. Despite the clinical relevance of SETD2 missense mutations in cancer, their biochemical and structural consequences remain insufficiently characterized. Here, we present the enzymatic and structural characterization of the SETD2 L1609P mutant enzyme identified in leukemia. Mutation of KRAS in endothelial cells (KRAS-EC) leads to intracerebral hemorrhage (ICH) in brain arteriovenous malformations (bAVM), resulting in severe disabilities or even death. However, it is unclear what causes this hemorrhagic conversion of bAVM. Here, using a locally established, clinically-relevant sporadic bAVM mouse model, created by overexpressing mutant KRAS (KRASG12V) in the brain EC, we demonstrate that KRAS-EC act as trigger for microglia (MG) activation and infiltration of macrophages (Mϕ). Using three-dimensional immunostaining approach with cleared human and mouse bAVM tissues, we demonstrate an abundance of MG/Mϕ around the bAVM nidus. The presence of MG/Mϕ are correlated to the …

The aspects of translation acceptability are not related only to the accuracy and clarity of the translation. They also cover the naturalness of translation. This study aims to reveal the degree of translation naturalness of the Quranic imperative verses. This evaluative study compares the translations of the verses by the Ministry of Religious Affairs of the Republic of Indonesia (MORA) and the one made by Muhammad Thalib (MT). The study used embedded case study research. The findings of the study show that in general, the translations of the verses in the two versions have an acceptable degree of naturalness. However, …

Alzheimer’s Disease (AD) is the most common neurodegenerative disorder and the leading cause of dementia characterised by the accumulation of beta amyloid (Aβ) plaques and neurofibrillary tangles (NFT). Several monoclonal antibodies against amyloid in early AD have shown the utility of reducing brain amyloid in large phase 3 studies, resulting in modest clinical benefit. To further slow, or even halt disease progression, targeting additional pathobiological pathways is likely to be necessary. ASTN1 encodes astrotactin 1, a neuronal-glial ligand in the developing brain that promotes neuronal migration along radial glia in brain structures with laminar organization, such as the cerebral cortex, hippocampus, and cerebellum. In mouse models, disruption of Astn1 results in neuronal migration deficits, a mild reduction in cerebellar volume, and balance and coordination deficits. In humans, bi-allelic ASTN1 variants have been identified in nine individuals with neurodevelopmental disorders (NDDs) with or without brain malformations. ASTN1 additionally interacts with astrotactin 2 (ASTN2) to implement neuronal migration; ASTN2 deletions associate with NDDs with reduced penetrance. Microplastic contamination is a pervasive global issue, yet its impact on ecologically significant cephalopod populations remains poorly understood. This study quantified microplastic prevalence and distribution in longfin squid (Doryteuthis pealeii), a key trophic link in the Gulf of Mexico. Following hydrogen peroxide digestion of digestive tract and mantle tissues, microplastics were isolated via vacuum filtration and characterized using microscopy. Fourier-transform infrared (FTIR) spectroscopy confirmed polymer identities. Microplastic contamination was exceptionally high, with 96.4% of individuals containing particles (mean = 36.6 ± 20.0 particles per individual, range = 0–94, n = 110 squid). Maintenance of bone mass is coordinated with adipose tissue function through the secretion of hormones and endocrine factors that act on the opposing tissue. Sclerostin, a small glycoprotein produced by osteocytes embedded within the bone matrix, potently suppresses bone formation by antagonizing Wnt/β-catenin signaling while stimulating adipose tissue accumulation via the same mechanism of action. Since sclerostin-deficient mice develop pockets of multilocular adipocytes in subcutaneous adipose, we investigate the influence of sclerostin on thermogenic and β3-adrenergic stimuli-induced white adipose tissue beiging. Sickle cell disease (SCD) is a genetic disorder that affects millions of individuals worldwide. Chronic anemia, hemolysis and vasculopathy are the most common symptoms that arise from hemoglobin (Hb) polymerization, which is the primary event of SCD. The above process can lead to erythrocyte sickling. Therapeutic options for SCD remain limited, and novel therapies are still being evaluated for their effectiveness in particular populations. Sphingosine-1-phosphate (S1P), a significant bioactive sphingolipid, acts as a potent signal mediator, modulating several cellular functions. The global poultry industry continues to face significant challenges due to Salmonella infections, which pose severe public health concerns and economic losses. Recently, the reemergence of antimicrobial resistance has led to the restriction of antibiotic use in poultry, especially as growth promoters, thus accelerating the search for sustainable alternatives. Among these, probiotics have gained attention as potential candidates for improving poultry health and mitigating Salmonella colonization in the gut. Friedreich's ataxia (FA) is a leading form of hereditary ataxia caused by autosomal recessive mutations in frataxin (FXN). GAA triplet repeat expansions lead to lower levels of FXN expression, abnormal influx of iron into mitochondria, and damage to the nervous system. Patients typically present before the second decade with loss of muscular function, speech impediments, and cardiomyopathy. At later stages, vision loss typically manifests. The poor clinical outcomes of pediatric high-grade glioma (pHGG) highlight the urgent need for new therapies. Oligodendrocyte lineage transcription factor 2 (OLIG2) is a pro-mitotic transcription factor highly expressed in glioma stem cells and may represent a novel therapeutic target. To evaluate the therapeutic efficacy of an OLIG2 inhibitor CT-179 in pHGG, we determined the OLIG2 mRNA expression in 10 patient-derived orthotopic xenograft (PDOX) models. In vitro activities of CT-179 were analyzed in monolayer and neurosphere cells (0–10 µM) with and without radiation (XRT) (0–8 Gy), brain penetration was evaluated in tumor-bearing PDOX mice, and in vivo efficacy was determined …

Oligodendrocyte lineage transcription factor 2 (OLIG2) is a pro-mitotic transcription factor highly expressed in glioma stem cells and may represent a novel therapeutic target. To evaluate the therapeutic efficacy of an OLIG2 inhibitor CT-179 in pHGG, we determined the OLIG2 mRNA expression in 10 patient-derived orthotopic xenograft (PDOX) models. Friedreich's ataxia (FA) is a leading form of hereditary ataxia caused by autosomal recessive mutations in frataxin (FXN). GAA triplet repeat expansions lead to lower levels of FXN expression, abnormal influx of iron into mitochondria, and damage to the nervous system. Patients typically present before the second decade with loss of muscular function, speech impediments, and cardiomyopathy. At later stages, vision loss typically manifests. Work is under way to develop gene therapies that address the cardiac and CNS manifestations, but their routes of administration do not lead to efficient transduction of the retina. INTRODUCTION: On April 24, 2025, the International Law and Social Science Interest Group debuted at the ASIL Annual Meeting with a panel that posed a deceptively simple question: how can social science tools advance the work of international lawyers? Over the last decade or more, our collaborative research has provided noteworthy information on the helminth parasites of reptiles of Arkansas. Because several species still need to be surveyed from the state, some for the first time, there has been a general void of information on their parasites. We therefore examined 10 reptiles, including seven species/subspecies of turtles, as well as three species/subspecies of snakes for helminth parasites. Found were 24 taxa of parasites, including two monogeneans, 14 digeneans, one tapeworm, and seven nematodes. Autonomous vehicles (AVs) are increasingly deployed in complex and safety-critical environments, raising fundamental questions about how ethical principles should be operationalized within real-time control systems. While existing research has extensively addressed ethical decision-making at the planning and behavioral layers, ethical intent is commonly translated into predefined trajectories and delegated to low-level controllers under the assumption of safe execution. This separation introduces a critical gap between ethical reasoning and physical feasibility, particularly in scenarios involving nonlinear vehicle dynamics, actuation limits, constrained perception horizons, and time-critical interactions with vulnerable road users. To understand the relative importance of cis and trans effects on regulation, we crossed multi-parent recombinant-inbred lines (RILs) to a common tester and measured allele-specific gene expression in the offspring. Testing the difference of allelic imbalance between two RIL × Tester crosses is a test of cis or trans, depending on the RIL alleles compared. The study design also enables to separation of two sources of trans variation, genetic and environmental, detected via interactions with cis effects. We demonstrate the effectiveness of this approach in a long-read RNA-seq experiment in female abdominal tissue at two time points in Drosophila melanogaster. …

Associations Between Big Five Personality Traits, Facets, And Sexual Fantasies, Emily Cannoot, Amy C. Moors, Wiliam J. Chopik

The present study investigated associations between Big Five personality traits, their facets, and the frequency and variety of sexual fantasies in a sample of 5,225 adults (M = 58.30 years old; 56.5% men). People high in conscientiousness and agreeableness report fewer sexual fantasies across exploratory, intimate, impersonal, and sadomasochistic domains; these effects were primarily driven by respectfulness and responsibility facets. Negative emotionality, particularly the depression facet, was associated with more frequent sexual fantasizing. Extraversion and open-mindedness showed minimal relationships with sexual fantasy frequency. Dysregulated ribosome biogenesis and p53 mutations are known to play oncogenic roles in various cancers, including pancreatic cancer. In this study, we demonstrated the therapeutic potential of BMH-21, a pharmacologic inhibitor of RNA polymerase I, against pancreatic cancer by uncovering a novel molecular mechanism involving RPA194-mediated ubiquitination of mutant p53 without affecting the ubiquitination of wild-type p53. Our key findings are that (i) BMH-21 selectively induces apoptosis and cell growth inhibition of pancreatic cancer cells with no effect on normal human pancreatic ductal epithelial cells; (ii) BMH-21 degrades RPA194; (iii) BMH-21 inhibits recruitment of both RPA194 and RPA135 on rDNA …

The clinical success of chimeric antigen receptor (CAR) T-cell therapies has revolutionized oncology, yet the high costs and logistical complexities of ex vivo manufacturing remain significant barriers to global patient access. In vivo cell therapy, which involves the direct injection of lentiviral vectors (LVVs) to engineer cells within the patient’s body, offers a promising, cost-effective alternative. However, transitioning from ex vivo to in vivo applications necessitates a fundamental shift in LVV biomanufacturing to ensure safety and efficacy. Radiation therapy is integral to the treatment regimens of over 50% of cancer patients. However, it is not biologically targeted to just the tumor, leading to adverse effects in peritumoral normal tissue. Selective uptake of gold nanoparticles (AuNPs) by tumors realizes tumor-specific radiosensitization via increased secondary electron release from high atomic number gold atoms. Here, we review AuNP-mediated radiosensitization and outline strategies to optimize tumor-targeted AuNP delivery. Modifying the physicochemical characteristics of AuNPs, including size, shape, charge, and surface chemistry, can increase their ability to evade the reticuloendothelial system (RES) and penetrate the dense tumor stromal architecture, thereby promoting tumor …

Drug repurposing is the process of reusing existing pharmaceuticals for novel clinical purposes, which offers advantages such as streamlined clinical trial access and reduced drug development costs. Clarithromycin (CAM), a member of the macrolide antibiotics family, is a promising candidate for repurposing in cancer therapy due to its known preclinical and clinical immunomodulatory and anticancer properties. In the current study, we investigated whether CAM could be repurposed as a preventive treatment for KRAS-mutant lung cancer, a subtype of lung adenocarcinoma that is strongly associated with heavy smoking.
