Classify Cancers Types Using Mirnas Profiles And Machine Learning, Wafaa Khazaal Shams
AUIQ Technical Engineering Science
Cancer classification is important for early diagnosis and effective treatment. This study proposes a hybrid framework that combines Fisher Discriminant analysis (FD) with Linear Support Vector Machine (LSVM) and Long Short-Term Memory (LSTM) networks. MicroRNAs (miRNAs) expression profiles for three types of cancer are used to evaluate the model. These are: lung- adenocarcinoma, ovary and pancreas cancers. The classification process is done using 5-fold cross valuation. Results show significant performance for LSTM compared to LSVM with and without features reduction. However, using FD analysis results in high accuracy reached 0.97and 0.96 for LSTM and LSVM respectively, using 50 miRNAs. The …

High School Teachers’ Experiences Using Data-Driven Decision Making To Implement Supplemental Accelerated Instruction, Stephen Courtney Smith, Sr.
Walden Dissertations and Doctoral Studies
The problem that was addressed in this study was that high school teachers in a Texas school district struggle to use data-driven decision making (DDDM) to meet the requirements of Texas HB 1416, which mandates specific interventions to address instructional loss. Grounded in the theory of DDDM, by Mandinach et al., the purpose of this basic qualitative study was to explore high school teachers’ experiences using DDDM to implement supplemental accelerated instruction (SAI) for students who failed to demonstrate proficiency on state exams. Data were collected using semistructured interviews with 12 high school teachers from a Texas school district. Using …

Male Caregiving Experience Alters Hippocampal Neuroplasticity And Transcription Independent Of Reproduction In A Biparental Species., Maria E Colt, Priyanka Agarwal, Erica R Glasper, Heidi Fisher
Faculty Research 2025
In mammals, measurable changes in brain and behavior accompany the transition to parenthood. In the biparental California mouse, Peromyscus californicus, fathers experience enhanced neuroplasticity, including increased hippocampal dendritic spine density, reduced anxiety, and improved memory. Here, we first investigate whether siring offspring or pup interaction drives structural neuroplasticity in fathers and find that hippocampal spine density is positively associated with caregiving experience, even in the absence of reproduction. Next, we evaluate the transcriptional response to caregiving in the hippocampus of these males and identify 158 differentially expressed genes between fathers and non-fathers, many of which are associated with neurogenesis, neuronal …

The Role Of The Poly-N Repeat In The Plasmodium Falciparum Histone Acetyltransferase Protein, Pfgcn5, Kely Johana Rubiano Cardona
Arts & Sciences Graduate Student Theses and Dissertations
P. falciparum possesses the most AT-rich genome sequenced to date, with long AAT repeats coding for asparagines (N) in intragenic regions. As a result, nearly a quarter of its proteome is composed of proteins with poly-N repeats. Since the discovery of its genome, the function of these repeats has remained as one of the biggest mysteries in the biology of this parasite. Furthermore, it has long been known that proteins with N-rich domains aggregate easily, particularly during heat stress (HS). Given that the hallmark of this disease is recurrent febrile episodes, it is not well understood how the parasite survives …

Bclaf1 Is A Novel Regulator Of Hematopoietic Stem Cells, Stephanie Crowley
Bclaf1 Is A Novel Regulator Of Hematopoietic Stem Cells, Stephanie Crowley
Arts & Sciences Graduate Student Theses and Dissertations
Hematopoietic stem cells (HSC) maintain both a self-renewal capacity to persist throughout life and an ability to differentiate into mature cell lineages. HSC self-renewal programs are often ectopically activated in leukemia cells to sustain developmental arrest and neoplastic proliferation. Recently, the transcriptional regulator Bclaf1 (Bcl2 associated transcription factor 1) was shown to support proliferation and block differentiation of AML blasts. Bclaf1 is ubiquitously expressed in hematopoietic cells but its function in normal hematopoiesis has not been elucidated. Our data demonstrate that BCLAF1 promotes HSC development and function. To study the role of BCLAF1 in normal HSCs, we used murine models …

Determining The Origin Of An Isolated Great Lakes Plant Species, Agoseris Glauca (Asteraceae), Seth Paul Hiers
Masters Theses
Small, isolated populations often have low genetic diversity and low potential for adaptation in response to environmental changes. Alternatively, isolated communities are often more likely to display local adaptations and over time speciate. The Great Lakes Region is home to over 50 vascular plant species that are disjunct from the species’ core populations. Agoseris glauca (Asteraceae) is a Great Lakes disjunct and Michigan state threatened species geographically isolated from its core range by 900 km. This study uses Genotyping by Sequencing to assess the population genetics of A. glauca for two Michigan disjunct populations and five western populations in the …

Gastrointestinal Fistulas-What Gastroenterologists Need To Know In 2025, Monjur Ahmed
Division of Gastroenterology and Hepatology Faculty Papers
Gastrointestinal fistulas are increasingly being encountered in our clinical practice because of the increased burden of Crohn's disease, bariatric surgeries, interventional endoscopic procedures, nonsurgical trauma, and war and disaster zones worldwide. Presentation depends on the location and specific type of the fistula. Symptomatic ones can have a tremendous impact on social life and can cause dehydration, electrolyte imbalance, malnutrition, increased morbidity, and mortality. Different imaging studies and endoscopic procedures are done to establish the diagnosis. Treatment modalities to close the fistula depend on the underlying disease and the type of fistula. They include conservative treatment, medical therapy, endoscopic interventions, and …

Implementation Of A Forensic Ancestry Inference Indel Panel, Jacqueline Molina
Implementation Of A Forensic Ancestry Inference Indel Panel, Jacqueline Molina
Student Theses
Ancestry-informative markers (AIMs) are genetic variants that exhibit significant frequency differences between populations, making them valuable tools for inferring biogeographical ancestry in forensic investigations. Insertion/deletion polymorphisms (INDELs) offer particular advantages for degraded forensic samples due to their short amplicon sizes and binary allelic nature. As forensic laboratories adopt newer instrumentation, validation of established assays becomes critical to ensure analytical continuity and reliability. This study evaluated the comparative performance of the SeqStudio™ Genetic Analyzer in detecting a 46 AIMs INDEL multiplex panel, previously established with the ABI 3130 Genetic Analyzer. Four commercial blood samples were analyzed using both platforms to assess …

Cardiometabolic Outcomes In The Obese African American Population With Rheumatoid Arthritis, Cecilia Jillo Walan
Walden Dissertations and Doctoral Studies
Rheumatoid arthritis (RA) disproportionately impacts African American adults, particularly those with obesity, due to overlapping cardiometabolic risks and health care disparities. The intersection of RA, obesity, and chronic conditions such as cardiovascular disease (CVD) and hypertension underscores the need for targeted public health interventions. This study investigated the association between RA and cardiometabolic outcomes among African American adults with obesity, aiming to identify significant predictors within this high-risk population. This quantitative study employed a cross-sectional design to analyze data from the 1999–2018 National Health and Nutrition Examination Survey undergirded by the social ecological change framework with N=23644 African American adults …

The ribosome is the central hub for protein synthesis and is heavily targeted by antibiotics. Ribosomal mutations, antibiotic treatment, and nutrient starvation can alter translational efficiency and lead to stressed cells. Ribosome deficiency plays a critical role in stress responses and disease progression; yet, how it affects bacteria-host interactions remains poorly understood. In this study, we show that a ribosome-deficient strain exhibits a surprising morphological change from rod shape to filamentous in Salmonella cells growing inside host macrophages. Such filamentation depends on an acidic condition within macrophages and in a defined medium mimicking macrophage conditions. Further genetic analyses revealed that …

Background: Aneurysmal subarachnoid hemorrhage (aSAH) is notoriously known for its high mortality and morbidity. Approximately one-third of the patients who survive aneurysm rupture are reported to develop delayed cerebral ischemia (DCI), which contributes to a poor clinical outcome. Currently, there are no biomarkers for identifying which aSAH patients are at risk of developing DCI. We aimed to determine the feasibility of cerebrospinal fluid (CSF) exosomal microRNAs (miRNAs) for predicting DCI post-aSAH.
Methods: aSAH patients were prospectively enrolled, and CSF samples were collected at two time points (< 24 h and 72 h post-aSAH) from individuals undergoing external ventricular drainage. Exosomal miRNAs were isolated from the CSF for analysis. In the initial group of patients (discovery cohort), an exploratory analysis was conducted using a CSF panel containing 84 miRNAs, assessed by quantitative real-time PCR (RT-qPCR). Based on this analysis, 27 miRNAs were selected for further evaluation in a second group of patients (validation cohort). Among these, 10 miRNAs had previously been reported in SAH-related CSF studies, supporting their relevance for continued investigation.
Results: In this study, RT-qPCR analysis of 84 miRNAs in CSF samples from …

RET solvent-front G810C/R/S mutations confer resistance to the currently approved RET protein tyrosine kinase inhibitors (TKIs) selpercatinib and pralsetinib. Moreover, RET fusion-positive lung adenocarcinoma frequently metastasizes to the brain. To address these challenges, it is imperative to develop a RET TKI that is effective against solvent-front mutations and exhibits intracranial activity. We synthesized alkynyl nicotinamide-based RET TKIs and tested their efficacy in cell cultures in inhibiting selpercatinib/pralsetinib-resistant RET solvent-front mutants G810C/R/S found in cancer patients, and in BaF3/KIF5B-RET(G810C) cell-derived subcutaneous and intracranial tumors in vivo. We also evaluated alkynyl nicotinamide RET TKIs in KIF5B-RET-induced lung tumors in immune competent …
