Altered cell surface glycosylation is a hallmark of cancer; among aberrant glycan structures, hypersialylated proteins contribute to disease progression. The enzyme ST6 β-galactoside α2,6-sialyltransferase 1 (ST6GAL1) mediates α2,6-linked sialylation of N-glycosylated proteins and is upregulated in many cancers, including prostate cancer (PrCa). We propose that ST6GAL1 may be released by cancer cells in small extracellular vesicles (sEVs) in the PrCa tumor microenvironment to potentially modulate cell surface sialylation in recipient cells. We isolated sEVs from PrCa cells by density gradient separation and characterized them by nanoparticle tracking analysis using ZetaView and immunoblotting analysis. Conceptions of autism and neurodiversity vary widely, with differing views on how they are portrayed in contemporary culture. Although autism scholars have long advocated for greater involvement of autistic individuals in knowledge creation, meaningful co-production between autistic stakeholders, researchers, and the broader community remains limited. To explore differences in autism knowledge between autistic and non-autistic people, we used a qualitative method called visual elicitation. We invited the public to upload an image they felt represented autism and explain their choice. Structural Analysis Of Pseudomonas Phage Dev Virion-Associated Rna Polymerase (Gp71), James Scherer

DEV is a Pseudomonas aeruginosa bacteriophage part of a newly reclassified genus called Schitoviridae. This group of bacteriophages is also referred to as “N4-like” phages, after the prototypical Escherichia coli phage N4, originally isolated from sewage water in 19661. N4, and other phages in the group, contain a large, virion-associated RNA polymerase (vRNAP). This vRNAP is vital to the proliferation of the phage, as it performs the transcription of the early gene products and potentially acts as a motor that helps eject the viral genome into the host bacteria. Alzheimer's disease (AD) is a neurodegenerative disease characterized by the accumulation of misfolded proteins in the brain. Recently, the impact of blood components in the progression of this disease has come to attention. This study investigates the effects of infusing blood from young and old wild-type mice into transgenic mice that model AD brain amyloidosis. Impaired memory and Aβ accumulation were observed in mice infused with blood from old donors. A proteomic analysis in the brain of these mice identified alterations in components related to synaptogenesis and the endocannabinoid system. PTDSS1 (phosphatidylserine synthase 1) encodes an enzyme that facilitates production of phosphatidylserine (PS), which mediates a global immunosuppressive signal. Here, based on in vivo CRISPR screen, we identified PTDSS1 as a target to improve anti-PD-1 therapy. Depletion of Ptdss1 in tumor cells increased expression of interferon-γ (IFN-γ)-regulated genes, including B2m, Cxcl9, Cxcl10, and Stat1, even in the absence of IFN-γ stimulation in vitro. Loss of Ptdss1 in tumor cells also led to increased expression of MHC-I, enhanced cytotoxicity of CD8+ T cells, and increased frequency of an iNOS+ myeloid subset. Hemispheric Asymmetry Of Phase Partition In Mixed-Phase Clouds Based On Near Global-Scale Airborne Observations, Ching An Yang, Minghui Diao, Yang Shi, Xiaohong Liu

Mixed-phase clouds contribute to substantial uncertainties in global climate models due to their complex microphysical properties. Former model evaluations almost exclusively rely on satellite observations to assess cloud phase distributions globally. This study investigated mixed-phase cloud properties using near global-scale in situ observation data sets from 14 flight campaigns in combination with collocated output from a global climate model. The Southern Hemisphere (SH) shows significantly higher occurrence frequencies and higher mass fractions of supercooled liquid water than Northern Hemisphere (NH) based on observations at 0.2 and 100 km horizontal scales. Such hemispheric asymmetry is not captured by the model. The …

Context: We examined predation by the saltwater crocodile (Crocodylus porosus) on the flatback turtle (Natator depressus), an Australian endemic, at Cape Domett, a significant nesting site in Cambridge Gulf, northern Australia, where crocodile populations have recovered since legislative protection was implemented in the 1970s. Aims: The investigation aimed to document crocodile predation on flatback turtles across hatchling and adult life stages, assess predation strategies, and evaluate ecological impacts on this genetically distinct turtle stock. The purpose of this pilot project was a quality improvement initiative aimed to assess various interventions designed to enhance treatment adherence amongst pediatric patients in an understaffed, underserved, rural area. It had three goals: boost interdisciplinary understanding of asthma, promote self-management amongst pediatric patients with asthma, and reduce additional clinic and emergency department visits, as well as hospitalizations.

Asthma, a chronic condition characterized by airway inflammation, affects over four million children in the US, impacting their quality of life. BACKGROUND: Human Papillomavirus (HPV), the most common sexually transmitted infection, is linked to cervical, anal, vulvar, and oropharyngeal cancers. Despite its impact, only 16% of women aged 27-45 have received an HPV vaccine dose, partly due to recent vaccine recommendation changes. This project aimed to improve HPV knowledge and vaccination rates among women in this age group at a private obstetrics and gynecology practice in Southwestern Indiana.

METHODS: Targeting unvaccinated women aged 27-45, this quality improvement initiative (January–April 2024) provided verbal education on HPV and assessed patient knowledge via a Likert scale survey. Staff received training on vaccine …

“A Field Beaten Bare Of Grass”: Samuel Youd’S The Death Of Grass (1956) And The Scouring Of The Shire, Kristine Larsen

Journal of Tolkien Research

One of the novels Tolkien admits to liking (according to his published letters) was Samuel Youd’s thriller The Death of Grass (1956), published under the pen name John Christopher. Among the novel’s aspects that might have particularly resonated with Tolkien included environmental concerns, analogs with Tolkien’s World War II experience (especially rationing), and an emphasis on fairy-story Consolation and Recovery, among others. But in addition, parallels with “The Scouring of the Shire” and other late episodes in The Lord of the Rings clearly deserve further analysis. This paper therefore engages with Youd’s novel through that specific lens.

Purpose: Skeletal stability after orthognathic surgery remains challenging in Class III malocclusion. This study examined how preoperative mandibular plane angle (FMA) and its surgical changes affect postoperative outcomes.

Patients and Methods: Sixty adult Taiwanese patients with skeletal Class III malocclusion treated by bimaxillary surgery without genioplasty were retrospectively analyzed. Cone-beam computed tomography (CBCT) at preoperative (T0), immediate postoperative (T1), and orthodontic debonding, ≥ 1-year postoperative (T2) stages was used to measure sagittal (Pog-CP), vertical (Me-FH), and transverse (Go–Go) changes. Pearson correlation and multiple regression analyses were performed.

Previously, we identified the only dinitrogen reduction mechanism known to date as an ancient feature conserved from nitrogenase ancestors, which we directly tested by resurrecting and integrating synthetic ancestral nitrogenases into the genome of Azotobacter vinelandii (Garcia et al., 2023), a genetically tractable, nitrogen-fixing model bacterium. Here, we extend this paleomolecular approach to investigate the structural evolution of nitrogenase over billions of years of evolution by combining phylogenetics, ancestral sequence reconstruction, protein crystallography, and deep-learning based predictions. The ferric uptake regulator (Fur) is a global transcription factor that reversibly binds an iron-sulfur [2Fe-2S] cluster via the cysteine residues (site 3) in response to elevation of intracellular free Fe content in Escherichia coli. Here, we report that when E. coli Fur is expressed in E. coli cells grown in M9 medium supplemented with Fe or zinc (Zn), purified Fur binds a [2Fe-2S] cluster or Zn(II), respectively. While apo-form Fur is a monomer and has no DNA-binding activity, both the [2Fe-2S] cluster-bound Fur and the Zn(II)-bound Fur are homodimers and have a similar binding activity for the DNA sequence …

Background and Objectives: Pakistan is among the countries with the highest prevalence of consanguineous unions (CU), with reported rates ranging between 55% and 65%. This high level of consanguinity contributes significantly to the burden of recessive genetic disorders. The Chitrali population, an isolated community residing in the Hindu-Kush mountains of northwestern Pakistan, exhibits distinct genetic and cultural characteristics. This study aimed to assess the prevalence and patterns of CU within this population.

Corvids are opportunistic omnivores and facultative scavengers, but there are no direct records of Cyanocorax species scavenging on carrion. We documented opportunistic feeding behavior of the White-throated Magpie-Jay (Cyanocorax formosus) on a road-killed Common Pauraque (Nyctidromus albicollis) in northwestern Costa Rica. On 25 August 2023, we observed three White-throated Magpie-Jays feeding on the carcass of a Common Pauraque, likely killed by collision with a vehicle, along a gravel road near the Taboga Forestry Reserve. This provides de first documented evidence of the White-throated Magpie-Jay scavenging on roadkill, and of carrion consumption by a Central American corvid. Given the White-throated Magpie-Jay's …

Undergraduate research experiences (UREs) are high-impact practices that enhance retention, motivation, and self-efficacy in science, particularly for students with limited prior exposure to scientific environments. However, many students-especially those from underrepresented or marginalized backgrounds-enter these experiences with low research self-efficacy, stemming from limited familiarity with academic language, scientific literature, and research communication. The Research Induction Curriculum (RIC) was designed to address this challenge by providing early, structured training in scientific literacy through scaffolded journal club discussions. Cannabis vaping is a relatively new route of administration that is increasingly common among youth and young adults. Cannabis vaping has been associated with cases of acute lung injury, especially among youth, and most often involves the heating and aerosolization of high-potency products having cannabinoid concentrations of ~ 50 % - 98 % w/w. Proponents argue that vaping is safer than smoking due to the characteristically lower temperatures and fewer plant materials. In eukaryotes, the telomere serves as a safeguard against DNA damage and loss during replication and other events. To do so, telomeric DNA folds into a variety of protective secondary and tertiary structures whose formation and conformational dynamics have not been well characterized. We have identified a slow conformational rearrangement in telomeric single stranded DNA that results in a G-Quadruplex (G4) that cannot be unfolded by a complementary strand. The current dissertation presents an overarching pipeline for the development and production of custom-made scaffolds for periodontal tissue regeneration using finite element analysis (FEA), machine learning (ML), and additive manufacturing (AM).CAD anatomically realistic scaffolds were generated from CT-based maxilla and mandible models. The FEA simulation results under masticatory (100 N), parafunctional (500–550 N), and traumatic (800–850 N) loads showed region-specific distribution of stresses and strains, which guided structural reinforcement strategies. Over 1000 3D-printed PCL scaffolds were printed using various pore diameters (200–300 µm) and filament diameters. Targeted cancer therapies are known to cause cardiovascular (CV) complications in human patients. Agents such as osimertinib, a tyrosine kinase inhibitor (TKI), as well as immune checkpoint inhibitors (ICIs) have been demonstrated to induce severe cardiotoxicities, including heart failure and myocarditis. Nevertheless, the mechanisms of TKIs and ICIs-induced cardiotoxicity remain poorly understood. Herein, we developed the first in vivo preclinical model of osimertinib-induced cardiotoxicity using a transverse aortic constriction (TAC) mouse model. Osimertinib-treated mice displayed severe cardiac dysfunction, elevated markers of heart failure and fibrosis, and failed to undergo compensatory hypertrophic remodeling, resulting in cardiomyocyte death. The shells of Pecten maximus (king scallop) are composed primarily of polycrystalline calcitic calcium carbonate, with a crucial aragonitic prismatic myostracum layer that facilitates soft tissue attachment and contributes to mechanical strength. Despite its importance, the impact of environmental stressors, such as metal contamination, on the myostracum remains underexplored. Hence, this study's main goal was to shed light on the microstructure and crystallography of king scallop shells, particularly the myostracum region, using a combination of scanning electron microscopy (SEM), electron backscatter diffraction (EBSD) analysis, and Raman spectroscopy.
