Bioinformatic Analysis Of Pogz Variants In Relation To White Sutton Syndrome, Hannah Rollins

White-Sutton syndrome (WHSUS) is a rare neurodevelopmental disorder caused by mutations in the Pogo Transposable Element with ZNF Domain (POGZ) gene, which encodes pogo-transposable element with ZNF domain, a chromatin regulator essential for proper mitotic progression and DNA repair. This study uses a bioinformatic framework to evaluate the structural and functional impact of missense mutations in the conserved amino acid region (positions 500–800) of the POGZ protein. Protein modeling, variant effect prediction, conservation analysis, and molecular dynamics simulations were employed to gain an understanding of the effects of POGZ missense mutations on protein structure and movement with specific emphasis on …

ObjectiveTo develop evidence-based clinical practice guidelines for non-invasive neuromodulation devices in acute and preventive migraine treatment.MethodsA systematic review was conducted across six databases from 1946 to April 2025. Randomized controlled trials evaluating Food and Drug Administration-cleared or Conformité Européenne (CE)-marked non-invasive neuromodulation devices were included. The quality of evidence was assessed using the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) methodology, and recommendations were developed through consensus following GRADE Evidence-to-Decision frameworks. The working group comprised 15 senior members and six junior members.ResultsFrom 1536 initial records, 15 studies met the inclusion criteria and were finally used to develop evidence-based recommendations. …

Genetic entropy is a hypothesis proposing that genomes deteriorate over time due to the gradual accumulation of slightly harmful mutations that natural selection cannot effectively remove. This paper explores the scientific debate surrounding this claim by examining differing perspectives on the theory of genetic entropy. Proponents of genetic entropy, such as John Sanford, argue that most mutations are deleterious and fall within a “near-neutral” range, allowing them to accumulate since their effects are too small for natural selection to detect. Science, Sand, And Faith: Exploring Competing Explanations Of Coconino Sandstone, Emily Schneider

The Coconino Sandstone, a Permian rock layer found in the western United States, is a topic of debate among standard geologists and flood geologists. The conventional geological interpretation claims the sandstone is an eolian deposit, meaning the sediments were deposited by wind (McKee, 1979; Selley, 1968). In their view, key elements of the sandstone, like the cross-stratified beds, high degree of frosting, and the animal trackways, confirm the Coconino’s similarity to modern eolian dunes (McKee, 1979). As this rock layer is sandwiched between layers typically interpreted by flood geologists as being deposited during the flood, the idea that the Coconino …

The insect evolution Lesson Plan is a four-day course, to be taught sequentially through a week. All the materials presented are meant to be for ninth to tenth grades. Textile waste has become one of the most visible consequences of the fashion industry’s linear production and consumption model, where materials are rapidly extracted, transformed, consumed, and discarded. This paper critically examines how sustainable fashion can move beyond isolated recycling efforts toward a more integrated circular economy framework. Rather than treating textile waste as only an end-of-life problem, the study analyzes it as a systems issue shaped by overproduction, weak product design, limited recovery infrastructure, fragmented regulation, and consumer habits that normalize short garment lifespans. Advanced age is one of the most recognizable risk factors for dry eye. Dry eye disease affects millions worldwide and can result from age-related lacrimal gland dysfunction, which correlates with a decline in lacrimal gland secretory cell function and chronic inflammation. This study investigated the potential of calorie restriction to maintain lacrimal gland and ocular surface health. Adult female C57BL/6 J mice were subjected to a 40% calorie restriction for 4 months, starting at 6-7 months and continuing until 10-11 months. These mice were compared to controls fed ad libitum. T cell-mediated autoimmune type 1 diabetes (T1D) is under complex polygenic control in both humans and the NOD mouse model. However, in both species, particular major histocompatibility complex (MHC; designated HLA in humans) haplotypes provide the primary T1D risk factor. Both MHC/HLA class I and II variants interactively contribute to T1D by respectively driving autoreactive CD8 and CD4 T cell responses that cooperatively destroy insulin-producing pancreatic β cells. While NOD mice have provided important insights to the pathogenic basis of T1D, the model has so far provided only a limited means to identify possible clinically translatable disease intervention approaches. INTRODUCTION: Developing effective therapeutics for Alzheimer's disease (AD) requires a better understanding of the molecular drivers of the disease. Our previous work nominated DLGAP2 as a modifier of age-related cognitive decline and risk for AD. We tested the hypothesis that overexpression of DLGAP2 in the hippocampus would protect against cognitive and synaptic deficits in a susceptible F1 5XFAD model.
METHODS: DLGAP2 was overexpressed in the hippocampus of F1 hybrid 5XFAD and non-transgenic littermates using a viral approach. Cognitive function, electrophysiological properties, and dendritic spine morphology were assessed at 6 and 14 months of age.

Genetic variants associated with autoimmune diseases are highly enriched within putative cis-regulatory regions of CD4+ T cells, suggesting that they could alter disease risk through changes in gene regulation. However, very few genetic variants have been shown to affect T cell gene expression or function. Here we tested >18,000 autoimmune disease-associated variants for allele-specific effects on expression using massively parallel reporter assays in primary human CD4+ T cells. We find 545 variants that modulate expression in an allele-specific manner (emVars). Primary T cell emVars greatly enrich for likely causal variants, are mediated by common upstream pathways and …

The mouse is a premier model system for investigating gene function and modeling human disease. For almost 40 years, Mouse Genome Informatics has worked to capture and integrate the data generated from mouse studies. A critical component of this integration is the development and use of the Mammalian Phenotype (MP) Ontology to capture the morphological and physiological effects of alterations to gene function in the mouse. As the wealth of phenotype data captured using the MP has expanded, its utility in the diagnosis of human disease has increased. This dissertation integrates isotopic, biodistance, and skeletal stress data from cemeteries, Gać and Pień to study health and survival in the Polish–Lithuanian Commonwealth (PLC). The PLC (16th–18th centuries) in Poland was marked by religious migration, social inequality, and environmental change. Chapter 2 used a multiproxy approach to identify migrant status: most at Pień were local, about half at Gać were migrants, often biologically related to locals. Chapter 3 used stable isotope analyses to find minimal sex-based dietary differences, except for Pień females’ higher millet intake, and showed that market access and feudal systems influenced subsistence strategies. The GGGGCC nucleotide repeat expansion (NRE) mutation in the C9ORF72 (C9) gene is the most common cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Neuronal activity plays an essential role in shaping biological processes within both healthy and neurodegenerative disease scenarios. Here, we show that at baseline conditions, C9-NRE-induced pluripotent stem cell-cortical neurons display aberrations in several pathways, including synaptic signaling and transcriptional machinery, potentially priming diseased neurons for an altered response to neuronal stimulation. Since the 2010’s, a technological trend has been flourishing: the development and enhancement of video games across different platforms. The early 1970s began the evolution of video game systems, from Atari and Magnavox Odyssey (1972) to Game Boy (1989) to Nintendo Switch 2 (2025) today. Weaning is widely recognized as one of the most stressful practices within the beef industry, as calves are separated from their dams and must manage changes in environment, social structure, and nutrition. This process provides indicators of stress, including increased vocalizations, reduced feed intake, pacing, and a stop in weight gain. Traditional management strategies include fenceline separation, nose flaps, and abrupt separation. Each year, the number of people diagnosed with attention deficit hyperactivity disorder (ADHD) and Autism Spectrum Disorder (ASD) rises. This rising number is due to the abundance of research done on each disorder. Because of this increase in information, many adults and children are getting a comorbid diagnosis, meaning that they are being diagnosed with both disorders at the same time. Maternal nutrition, immune function, and microbiome health work together to shape fetal development in ways that influence health across the lifespan and even into future generations. This paper synthesizes evidence from molecular biology, nutrition science, immunology, and public health to show that essential nutrients-such as omega-3 fatty acids, folate, iron, zinc, and vitamin D-directly affect fetal immune programming, neurodevelopment, placental function, and metabolic regulation. Research on the maternal microbiome further demonstrates that microbial diversity and metabolite production modify nutrient absorption, inflammatory balance, and fetal gene expression, underscoring the microbiome’s role as both a mediator and amplifier of nutritional effects.
