Hereditary angioedema is an autosomal dominant disorder caused by defects in C1-esterase inhibitor (C1-INH), resulting in poorly controlled activation of the kallikrein-kinin system and bradykinin overproduction. C1-INH is a heavily glycosylated protein in the serine protease inhibitor (SERPIN) family, yet the role of these glycosylation sites remains unclear. To elucidate the functional impact of N-glycosylation in the SERPIN domain of C1-INH, we engineered 4 sets consisting of 26 variants at or near the N-linked sequon (NXS/T). Among these, 6 are reported in patients with hereditary angioedema and 5 are known C1-INH variants without accessible clinical histories. We systematically evaluated their …

The purpose of this transcendental phenomenological study was to investigate the lived experiences of former adult drug users and their sobriety journeys after graduating from a variety of faith-based or spiritual drug and alcohol recovery programs. The research included a systematic review and narrative synthesis of existing literature on faith-based and spiritual recovery programs to address the gap concerning the lived experiences of former users and an evaluation of the strategies and techniques vital for facilitating recovery. The purpose of this phenomenological study was to describe men's experiences with the stigmatization of childlessness in the context of marriage in Ile-Ife, Nigeria. The experience of childlessness as a universal phenomenon has been predominantly studied from the female perspective, and men are primarily neglected. The heteronormative ideal of women as 'mother/nurturer' is supported by only collecting women's fertility data in contrast to relatively suboptimal statistics in men's studies. This dissertation explores the view of ten African men set in the Yorùbá culture suffering from childlessness and its attendant stigmatization, mainly as a result of infertility. The purpose of this phenomenological study was to examine the lived experiences of successful African American undergraduate males from single-parent homes, and the maternal messages that contributed to their academic success and resilience. Research shows that African American males face an icreased risk of academic underachievement, particularly those raised by single mothers. The participants consisted of African American undergraduate males who grew up in a single-parent home headed by their mother. This study investigates the imposter phenomenon, racially based trauma, and how the two correlate among high-achieving Black women. High achieving for this writing refers to Black women who attend college or graduate school, those in professional positions that require a degree, advanced degree, or professional licensure (i.e., corporate executives, medical doctors, attorneys, mental health clinicians, etc.), and entrepreneurs. Many current studies have focused on the unique and complex experiences that high-achieving Black women face while working to achieve goals and gain success. This mixed-methods study examined the prevalence of Moral Injury (MI) among Ukrainian healthcare professionals (UHCPs) and their willingness to engage in evidence-based, psycho-spiritual interventions targeting MI that has resulted from the Russian invasion and the COVID-19 pandemic. MI is a psychological construct resulting from actions that violate an individual’s moral code and can overlap with Post Traumatic Stress Disorder (PTSD) symptoms. While MI has been extensively researched in the military, it has gained attention in healthcare settings. Utilizing the Moral Injury Symptom Scale-Healthcare Professional (MISS-HP), this study quantitatively identified a moderate prevalence of MI among doctors and nurses in Ukraine. …

BACKGROUND: Multidomain lifestyle interventions may have the potential to slow biological aging as captured by deficit accumulation frailty indices. We describe the distribution and composition of the 49-component frailty index developed by the U.S. POINTER clinical trial team of investigators and assess its cross-sectional associations with sociodemographic factors and markers chosen to be representative of behaviors targeted by the trial's multidomain interventions.
METHODS: We draw baseline data from the 2 111 volunteers enrolled in U.S. POINTER who were ages 60-79 years and at increased risk for cognitive decline. Frailty components were grouped into 9 domains. Associations that frailty index scores …

Glioblastoma is the deadliest primary brain tumor, largely due to inevitable recurrence of the disease after treatment. While most recurrences are local, patients rarely present with a new discontiguous focus of glioblastoma. Little is currently known about the genetic profile of discontiguous recurrences. In our institutional database, we identified 22 patients with targeted exome sequencing of pairs of initial and recurrent IDH-wildtype glioblastoma. Recurrences were classified as contiguous or discontiguous based on the presence or absence of T2 FLAIR signal connection to the initial site of disease on MRI. Common features of the aging heart are dysregulated metabolism, inflammation, and fibrosis. Elevated oxidative stress is another hallmark of cardiac aging that can exacerbate each of these conditions. We hypothesize that by increasing natural antioxidant levels (glutathione), we will improve cardiac function. Twenty-one-month-old mice were fed glycine and N-acetyl cysteine (GlyNAC; glutathione precursors)-supplemented or control diets for 12 weeks. Heart function was monitored longitudinally, and the exercise performance was determined at the end of the study. Background: Preformulation studies have a significant role in anticipating formulation problems and identifying logical paths in dosage form technology.
Aim: This study aimed to evaluate the preformulation criteria of the aquatic ethanol extract of Kigelia africana fruit as topical antibacterial therapy.
Methods: The preformulation parameters of the extract were evaluated by testing its organoleptic characteristics, melting point, pH, spectroscopic assay, stability, solubility, and partition coefficient.

The worldwide Jewish community is notoriously nomadic because of persecution and displacement through several millennia. The diasporic experience of global Jewish communities led to traumatic events such as historical expulsion and genocide. These experiences embody all five components of occupational injustice. The recent rise in antisemitism has triggered intergenerational trauma among individuals, families, and communities. This paper, grounded in trauma-based research, aims to guide occupational therapists treating Jewish clients and/or teaching Jewish students. The paper outlines trauma-informed implications for therapist-client collaboration in the context of supporting occupational justice for diverse populations. Background: Breast cancer is the leading cause of female cancer worldwide, which can lead to many negative sequelae, with up to a third of women developing breast cancer-related lymphedema. Breast cancer-related lymphedema requires chronic maintenance and is associated with worse physical and mental health outcomes compared to women without lymphedema. Occupational therapists are well-positioned to address the complex needs of women with breast cancer-related lymphedema. This review aimed to identify existing research that reports outcomes of lymphedema management programs addressing both physical and mental health needs of women with breast cancer-related lymphedema.

Alterations in mitochondrial function are the linchpin in numerous disease states including in the development of chemotherapy-induced neuropathic pain (CIPN), a major dose-limiting toxicity of widely used chemotherapeutic cytotoxins. In CIPN, mitochondrial dysfunction is characterized by deficits in mitochondrial bioenergetics (e.g., decreased ATP production) that are thought to drive the degeneration of the peripheral nerve sensory axon terminal sensory arbors in the skin (the intraepidermal nerve fibers; IENFs) and induce abnormal spontaneous discharge in peripheral nerve sensory axons. In this study, I recounted my experience using mantra meditation during a phenomenological study for the purposes of bracketing. The efficacy and purpose of bracketing have been debated from Husserl (1931), whose aimed was to achieve objectivity, to Heidegger (1962) who advocated for immersion of the researcher, through the French school (Merleau-Ponty, 1964) of middle ground, by whom bracketing was seen as the process to unearth and suspend biases for the better understanding of participants’ experiences (Arsel, 2017; Creswell & Creswell, 2017; Creswell & Poth, 2016; Fischer & Guzel, 2023). Background: Breast cancer (BC) is one of the most common malignancies in women, characterized by the malignant proliferation of breast epithelial cells that significantly impairs patients’ quality of life. Astaxanthin (ASN), a marine-derived xanthophyll carotenoid, is known for its antioxidant and anti-inflammatory properties. Emerging evidence suggests that ASN may exert anti-neoplastic effects by inhibiting tumorigenesis and suppressing the growth and progression of BC cells. This systematic review aims to critically evaluate the therapeutic potential of ASN as a pharmacological agent in the prevention and management of breast cancer.

Introduction: The annual prevalence of elder mistreatment (EM) in cognitively intact older adults is estimated to be 11%, yet the annual prevalence in older adults with Alzheimer's disease and related dementias (AD/ADRD) is estimated to be as high as 75%. Associated with a decrease in quality of life and increase in risk of mortality, EM represents a significant public health burden. Home-based primary care (HBPC) providers are uniquely positioned to address the critical need for robust EM screening and reporting, especially among individuals with AD/ADRD. The automatic recognition of medical concepts and temporal expressions in narrative clinical text enhances the utility of electronic health records (EHRs) and supports clinical decision-making and research. This study presents research about the nature and prevalence of adverse childhood experiences (ACEs), the mental and physical impacts to individuals exposed to ACEs, and incorporating mindfulness practices into therapeutic approaches to treating them, particularly Mindfulness-Based Narrative Therapy (MBNT). It is important because ACEs have become a global crisis, with the potential for interfering with psychological and physical development, early mortality, and other harmful life outcomes. This study provides a theoretical framework suggesting that nonviolent communication (NVC) can alleviate the violence frequently experienced in the inner dialogue of women with Attention Deficit/Hyperactive Disorder (ADHD). Due to the high correlation between ADHD symptoms in women and elevated rates of anxiety, depression, engagement in self-harm, and internalized self-criticism, exploring methods to mitigate negative self-dialogue is essential. Research on NVC indicates that individuals can cultivate empathy and compassion in interpersonal communication, suggesting potential benefits for addressing challenges in intrapersonal communications that often stem from the internalized presentation of ADHD symptoms in women. Single-cell multiplex imaging (scMI) measures cell locations and phenotypes within a tissue and can be used to understand the tumor microenvironment. In scMI studies, it is often of interest to quantify spatial co-localization of immune cells and its association with clinical outcomes; however, it remains unknown which of the many available spatial indices have adequate power to detect spatial within-sample co-localization and its association with patient outcomes, such as survival.
