This playbook examines the implementation of New York State’s Humane Alternatives to Long-Term Solitary Confinement (HALT) Act and presents evidence demonstrating that the law has not been fully or consistently enforced by the New York State Department of Corrections and Community Supervision (NYSDOCCS). Drawing on findings from the New York State Inspector General, the Correctional Association of New York, the Justice Center, court rulings, and community testimony, the analysis documents persistent violations, including excessive and illegal use of solitary confinement, inadequate documentation, failures to protect vulnerable populations, and insufficient accountability for staff misconduct. Microbial electrolysis cells (MECs) present a pathway for energy recovery in wastewater treatment by converting organic substrates into methane or hydrogen. Traditional MEC designs typically rely on physical electrode contacts and expensive materials, which restrict their scalability. This study introduces the first induction-based MEC that uses mutual inductance between an external copper wrap and an internal coil to generate a fluctuating electrical potential, removing the need for expensive electrodes. The induction-stimulated reactor showed faster total organic carbon removal and a higher CH4/CO2 ratio compared to a conventional anaerobic digester, indicating improved carbon-to-methane conversion, although total gas yield remained similar. Adolescence represents a critical window of frontal cortex (FC) maturation and corticolimbic circuit refinement that supports stress and emotion regulation. Disruption of these processes by early-life insults such as prenatal alcohol exposure (PAE) or adolescent stress increases vulnerability to anxiety and mood disorders, in part through circadian–immune dysregulation. The circadian transcription factor Bmal1 and the proinflammatory cytokine tumor necrosis factor-alpha (TNFα) have emerged as key regulators of neuroimmune signaling and synaptic communication, yet their interactions across developmental risk contexts remain poorly understood. The genesis of this project stems from a lived experience performing with a professional choir, whose sung physical shaping of the English language—its crystalline clarity, profound expressivity, and the pre-expressive elements inherent in the delivery—was deeply and viscerally compelling. This overwhelming reaction was in response to their bodies; specifically, the sound their bodies produced that evoked a sense of corporeality, or tangible bodily presence. There was beauty to be found in the way their mouths physically formed the language, both within and removed from the semantic context.

Pulmonary hypertension (PH) is characterized by increased pulmonary arterial constriction, pulmonary arterial remodeling, and right ventricular dysfunction. While metabolic and mitochondrial derangements are increasingly recognized as central pathogenic features, the underlying mechanisms and effectors are not yet fully understood. Acid-sensing ion channel 1a (ASIC1a) is a proton-gated cation channel involved in the development of chronic hypoxia (CH)-induced PH. ASIC1a represents a potential candidate involved in metabolic-mitochondrial dysfunction, as metabolic byproducts can act as ligands for ASIC1a. Additionally, ASIC1a has recently been identified as a mitochondrial ion channel involved in neuronal death following oxidative stress. Therefore, we hypothesize that ASIC1a functions …

Many remarkable discoveries, especially in the last decade, have shown scientists just how important RNA is. In this study, the secondary structure of RNA, G-Quadruplexes, is the topic of interest and how they are affected by salts and S1 protein. To obtain the S1 protein it was transformed from E. Coli using a bacterial plasmid. The S1 protein was then purified from the bacteria through the use of many purification methods. KRASG12D mutation is a prevalent gain-of-function mutation that drives pancreatic cancer tumorigenesis, but the underlying mechanisms that promote KRAS-induced cell proliferation and tumor formation remain elusive. To uncover the molecular pathways that facilitate KRASG12D-driven malignant transformation, we measured the transcriptomic alterations at various time points after induction of KRASG12D expression in human pancreatic normal epithelial cells. KEGG pathway enrichment of the differentially expressed genes (DEGs) showed that the major DEGs were located in pathways that regulate nicotinate/nicotinamide metabolism, TNF signaling, and microRNAs associated with cancer. Among these molecular alterations, the NAD-dependent deacetylase gene SIRT3 was significantly down-regulated by KRASG12D. Conversely, …

Event-based sensing presents a revolutionary paradigm shift for next-generation space surveillance systems, where power consumption is an ever-worsening constraint as temporal resolution demands increase. In addition to reduced power consumption, low latency, and wide dynamic range, the event-based sensor fundamentally only produces data when there is a change in illumination from which events are generated; no data is produced if the scene remains static. With their event-based datastream being inherently focused on the dynamic information of the scene, they are particularly well-suited to machine vision and autonomous sensing applications. This conceptual paper advances our understanding of sustainable careers by focusing on the knowledge creation activities of third-space practitioners in higher education. Specifically, we apply and extend the Process Model of Sustainable Careers to examine how different forms of knowledge production in three different institutional and national contexts – writing for publication, practice-based expertise, and curriculum development – contribute to career sustainability at the intersection of academic and professional/administrative roles and domains. Intermittent fasting (IF) has gained attention as a potential intervention for cardiometabolic health, though its long-term effects remain unclear. In this randomized clinical trial, we assessed the impact of 6 months of IF on body composition, cardiovascular risk factors, and related molecular pathways in middle-aged (30-65 years) men and women with overweight (BMI 24.8-35 kg/m²). In this trial, 41 participants were randomized to either an intermittent fasting (IF) intervention or to maintain their habitual diet. The primary outcome (circulating CRP concentration) was previously reported; here, we present exploratory analyses focusing on metabolomic and transcriptomic responses. IF led to an 8% …

The 2015 International Parkinson and Movement Disorder Society (MDS) Diagnostic criteria for Parkinson Disease are based on expert consensus opinion and defines core motor features, 'Absolute Exclusion Criteria' and a balance of 'Supportive Criteria' and 'Red Flags'. To assess validity of each criterion in pathologically-confirmed cases, a scoping literature review between 1988-2024 using search terms for clinicopathological PD and atypical parkinsonian disorders identified 28 articles. Supportive criteria were higher in PD, with excellent levodopa response and rest tremor most useful. Absolute exclusion criteria and red flags were present more often in atypical parkinsonian disorders. However, supranuclear gaze palsy, rapid progression …

Adenine base editors (ABEs) are powerful tools for gene therapy. However, efficient version of ABEs (e.g. ABE8e) always induce excessive bystander and off-target editing events and are large in size, hindering their potential in clinical disease treatment. Here, we develop a pre-trained Protein-Nucleic Acid Constrained Language Model to design ABE8e with high activity, reduced editing window and decreased size. By further engineering, the smallest ABE8e- PNLM-pcABE- with a 27% size reduction, exhibits high activity, precise 3-nt editing window, and reduced off-target events near background level in HEK293T cells. Injured ligaments are often repaired with tendon grafts which may over or under constrain the joint; there is a need for an artificial graft option with the mechanical properties of a native ligament. Crimped fibers within ligaments allow for recruitment which means the viscoelastic properties of stress relaxation and creep are not inversely related. A feasibility study using ACLs and large-scale crimp models to study viscoelasticity, led to an in-depth analysis of the scapholunate interosseous ligament. Background: Seizures are one of the most common reasons for emergency medical services (EMS) activation for children, and current EMS practice results in underdosing and delayed delivery of anti-seizure medication. A prehospital evidence-based guideline recommends using intranasal or intramuscular midazolam as first-line treatment for pediatric seizures. Despite attempts to implement these guidelines, one-third of children having a paramedic-witnessed seizure have ongoing seizures on emergency department (ED) arrival; this may be due to inadequate or delayed midazolam dosing. Replacing the error-prone, sequential calculations with age-based midazolam dosing may be simpler, faster, and more effective without compromising safety. The objective of this …

This study investigated prevention of suicide as perceived by educated adults in Ondo metropolis, Ondo State, Nigeria. Descriptive survey research design was adopted for this study. Population of the study consisted of all educated adults in Ondo metropolis, Ondo State. The sample comprised two hundred (200) educated adults randomly selected from the metropolis. Structured questionnaire was used as instrument to collect data for the study. Data collected was analysed using mean, ranking, t-test and analysis of variance (ANOVA).
