The clinical availability of photon-counting computed tomography (PCCT) has ushered in a new era of CT imaging. Spectral imaging coupled with superior contrast resolution, and ultrahigh spatial resolution (200 μm) offered by PCCT has the potential to revolutionize value-driven imaging. The potential of multicolor PCCT has generated excitement, and renewed interest, in novel contrast agent development for comprehensive disease interrogation, prediction and monitoring of treatment outcomes. Purpose: Cyclic nucleotide-gated (CNG) channels are ligand-gated ion channels that transduce light signals into electrical signals in the retinal photoreceptors. Pathogenic variants in CNG channel genes are reported to cause inherited retinal dystrophies (IRDs). The current study used targeted panel sequencing to describe the mutational spectrum of CNG channel genes in familial cases of IRDs from eight consanguineous Pakistani families.

Methods: The current study included consanguineous Pakistani families with at least two affected members. DNA was extracted from whole blood samples by the phenol-chloroform method. Two affected members from each family were initially analyzed using targeted panel sequencing of 344 …

Background: Aortopathies, such as aortic aneurysm and dissection (AAD), are associated with enhanced aortic wall permeability and endothelial dysfunction. We previously demonstrated that nanoparticle contrast-enhanced computed tomography (nCECT), which detects enhanced aortic wall permeability, could enable non-invasive detection of early AAD before its progresses to fatal aortic rupture. This study investigated the effect of nanoparticle contrast agent (NPCA) size and dose on detection of aortopathy by nCECT.

Background: The allelic variations of the apolipoprotein E (APOE) gene play a critical role in regulating lipid metabolism and significantly impact cardiovascular disease risk (CVD). This study aimed to evaluate the impact of exercise on cardiac structure and function in mouse models expressing different APOE genotypes using photon-counting computed tomography (PCCT) and deep learning-based segmentation.

Intracellular lipid binding proteins (iLBPs) play crucial roles in lipid transport and cellular metabolism across the animal kingdom. Recently, a fat‐to‐neuron axis was described in Caenorhabditis elegans, in which lysosomal activity in the fat liberates polyunsaturated fatty acids (PUFAs) that signal to neurons and extend lifespan with durable fecundity. In this study, we investigate the structure and binding mechanisms of a lifespan‐extending lipid chaperone, lipid binding protein‐3 (LBP‐3), which shuttles dihomo‐γ‐linolenic (DGLA) acid from intestinal fat to neurons. The recent termination of the U.S. Government's neglected topical diseases (NTD) mass treatment program, following similar cuts by the UK, threatens decades of progress. Without a strategic reset, NTDs may re-emerge, exacerbating poverty across Africa and beyond. We call for a broadened donor base and reframing NTD control as a shared global interest, especially as NTDs rise in G20 nations due to climate change and urbanization. This moment demands investment in sustainable tools, like NextGen vaccines and biologics, and greater leadership from G20 countries. While USAID's exit is a setback, opportunities arise for decolonizing global health and fostering new partnerships.

Background: Patients admitted with acute ischemic stroke (AIS) may experience accompanying acute ST-Segment myocardial infarction after AIS. The cardiovascular risks, incidence, complications, and outcomes of acute STEMI in patients hospitalized with AIS remains underexplored.

Methods: We evaluated 2,804,819 patients that presented with AIS who were listed in the National Inpatient Sample from 2016 to 2021. AIS and STEMI were defined according to the ICD-10 Diagnostic Codes. Patients with Non-STEMI were excluded. The risk of specific complications and outcomes were expressed as percentages. Stent thrombosis remains a major complication following percutaneous coronary intervention, with significant morbidity and mortality implications. Despite advancements in drug-eluting stents and optimized pharmacotherapy, real-world registry data indicate that definite or probable stent thrombosis occurs in approximately 0.5% of percutaneous coronary intervention cases, with a 30-day mortality rate approaching 25% and a long-term risk exceeding 30% at 10 years. Stent thrombosis is classified based on timing into acute, subacute, late, and very late thrombosis, with subacute and very late stent thrombosis being the most prevalent. To establish standardized practices, our multidisciplinary expert position statement serves as a comprehensive guide for the appropriate indications and utilization of 3D-ICE in various structural heart procedures. Objective: Weight loss in older men with obesity and hypogonadism accelerates musculoskeletal decline, yet the underlying metabolic mechanisms remain unclear. Testosterone replacement therapy (TRT), when added to lifestyle therapy (LT), mitigates this decline, but its metabolic basis has not been defined. We examined skeletal muscle metabolomic adaptations to LT with or without TRT, focusing on glycolysis, the pentose phosphate pathway (PPP), the tricarboxylic acid (TCA) cycle, and carnitine metabolism to identify dominant pathways of metabolic adaptation.

Design: Randomized, double-blind, placebo-controlled trial (LITROS).

Methods: Eighty-three men aged 65 years or older with obesity (BMI ≥30 kg/m2), hypogonadism (testosterone < 10.4 nmol/L), and frailty (Physical Performance Test score ≤31) were randomized to 26 weeks of LT plus TRT (LT+TRT) or LT plus placebo (LT+Pbo). A metabolomic substudy was performed in 44 participants, who underwent serial biopsies of the vastus lateralis for targeted LC-MS/MS analysis of intermediates in glycolysis, PPP, TCA cycle, and carnitine metabolism.

DNA replication and RNA transcription processes compete for the same DNA template and, thus, frequently collide. These transcription-replication collisions are thought to lead to genomic instability, which places a selective pressure on organisms to avoid them. Here, we review the predisposing causes, molecular mechanisms, and downstream consequences of transcription-replication collisions (TRCs) with a strong emphasis on prokaryotic model systems, before contrasting prokaryotic findings with cases in eukaryotic systems. Current research points to genomic structure as the primary determinant of steady-state TRC levels and RNA polymerase regulation as the primary inducer of excess TRCs. Monocytes and macrophages are sentinels of the immune system that distinguish themselves from other cells by expressing the beta isoform of the folate receptor (FRβ). Because FRβ does not bind folate until the monocyte/macrophage is exposed to immunosuppressive cytokines, the question naturally arose whether FRβ might also perform an immune-related function. To examine this matter, we compared the properties of wild type (WT) and FRβ knockout mice. We observe that FRβ knockout (KO) mice display autoimmune symptoms that can include alopecia, enlarged spleens, and dermatitis, despite having normal cellular folate levels. Following in the tradition of ethno-psychological research proposed by Diaz-Guerrero (1986, 1989, 1993, 1994, 1995), this manuscript delineates the process toward obtaining statistically and conceptually valid and reliable personality measures, that at the same time are also culturally relevant and sensitive to the Mexican culture. The methods and construct development for the creation of multidimensional measures of achievement orientation, empathy, locus of control, assertiveness, masculinity, femininity, and self-concept are presented. This thesis provides an analysis of Australian and Chinese news media perspectives on this relationship. This dissertation examines the effects of linguicism on nonnative speakers of English who are currently studying in higher education in the United States or other English-dominant countries. Linguicism, also called linguistic discrimination, others and discriminates against people based on their oral or written language use. Rooted in master narratives of Standard English, monolingualism, and the native speaker, linguicism, unlike other forms of discrimination, such as gender or racial discrimination, is subtle and unmarked, thus permeating the consciousness of even the most well-intentioned of people.

Speculation about the developmental shifts in the magnitude of peer influence has outstripped empirical research. Friends are assumed to be particularly influential at the onset of adolescence, as adult oversight declines and the salience of peers increases. Few studies, however, have explored age-group differences in the magnitude of friend influence over academic achievement and physical activity. The present longitudinal study compares friendship dyads in late childhood (grades 4-5) versus early adolescence (grades 6-7), describing the strength of their mutual influence across peer- and self-reports of academic achievement and physical activity. Algorithmic processing and classification of racism-related content have become increasingly prevalent across Meta's platforms. While these algorithms aim to create safer online environments, their effectiveness and impact on fairness remain understudied. This research examines how Meta's algorithmic processes for classifying racist content affect fairness, bias, and social justice across Facebook, Instagram, and WhatsApp.

Using qualitative research methodology, the study conducted focus groups with diverse platform users to understand their experiences with content moderation. The research also analyzed theoretical frameworks related to algorithmic bias, fairness metrics, and social justice in digital spaces.

By the time of the 2022 midterm elections, housing affordability had become a prominent policy issue in Florida, as post-COVID economic conditions, including rising rents and inflation, heightened political attention and debate at the local level. Two counties—Palm Beach County and Orange County—became focal points of this debate, each introducing a ballot referendum offering divergent solutions to local housing affordability challenges. Palm Beach County pursued a market-driven approach, passing a $200 million housing bond to incentivize private-sector development, while Orange County attempted a regulatory intervention through a rent stabilization ordinance, directly challenging Florida’s dominant pro-growth housing framework.

The Rise And Resilience Of Women Of Color Executives In Nonprofit Organizations: A Phenomenological Study Of Systemic Barriers, Support Systems And Leadership Identity, Nicole Edwards

This phenomenological study explored the lived experiences of women of color executive leaders in nonprofit organizations, focusing on perceptions of systemic barriers, support systems, and leadership identity. Using a mixed-methods, phenomenological design, data were collected from 31 women of color (N = 31) through a Likert-type survey and from six women (n = 6) selected for semi-structured interviews.

Quantitative analysis revealed that perceived systemic barriers significantly predicted leadership identity (p = .013), while support systems did not. Further analysis uncovered a significant positive correlation between positional status and perceived support (r = .39, p = .050), indicating …

The current study evaluated the psychometric properties of the newly developed Multi-Ethnic Acculturation Scale (MAS), a multidimensional instrument designed to assess behavioral and cognitive aspects of cultural adaptation among Hispanic/Latino individuals in the United States. A sample of 208 adults from diverse Hispanic backgrounds completed the MAS alongside two validated acculturation measures: the Short Acculturation Scale for Hispanics and the Vancouver Index of Acculturation. Exploratory factor analyses revealed a partially distinct but moderately correlated factor structure for both American and Latino cultural orientation subscales, supporting a bidimensional conceptualization of acculturation. This dissertation examines the equity effects of Title I funding on student achievement in Broward County Public Schools, one of the largest school districts in the United States. Title I, established under the Elementary and Secondary Education Act (ESEA) of 1965, aims to address educational disparities by providing financial resources to schools that serve economically disadvantaged students. Despite over five decades of federal intervention, significant achievement gaps persist, particularly among marginalized populations.
