Investigating The Dna-Binding Role Of Tbp1 In Toxoplasma Gondii: Expression And Characterization Of A Recombinant Protein, Madison T. Jeckell

Toxoplasma gondii is an obligate intracellular parasite with a complex life cycle and an unusual transcriptional machinery compared to other eukaryotes. Unlike most organisms, Toxoplasma lacks multiple well-conserved promoter motifs and has multiple TATA-binding proteins (TBPs), including TBP1, whose function in DNA binding and transcription regulation is poorly understood. This study aimed to generate and express a recombinant TBP1 protein from Toxoplasma to enable future DNA-binding assays and motif identification. Using RT-PCR, the TgTBP1 gene was amplified and cloned into a pGEX vector to produce a GST-tagged protein. The recombinant plasmid was transformed into E. coli BL21 cells, and protein …

This study explores how life science students reason about exponential functions in the context of an introductory physics course. While exponential functions are essential in both physics and biology—appearing in scenarios like population growth and temperature cooling—students often encounter challenges in interpreting them. Using a resources framework, we examined the cognitive resources these students bring to exponential reasoning, focusing on what ideas and strategies they already possess. Our methodology involved conducting think-aloud interviews with nine students from PHYS 402. This thesis explores the presence of witches and magic in the literature of the Roman Empire. The study ranges from the 1st century BCE to the 6th century CE, and relates the prevalence of female witches in the Roman literature to particularly turbulent social contexts, such as the fall of the Republic and the rise of Christianity. The piece considers the rhetorical nature of accusations of witchcraft, and how the concepts of magic and witchcraft themselves were defined and shaped by those in power to uphold imperial hegemony and structures of patriarchy. This project explored the challenges, needs, and potential solutions related to food security and food sovereignty in rural Alaskan communities. These communities have long relied on traditional subsistence practices like fishing, hunting, and foraging, but those practices have been increasingly affected by climate change, regulatory restrictions, and commercial overfishing. Key issues identified included shorter hunting seasons, changes in animal migration, unsafe ice conditions, habitat loss, complex land ownership, and permitting barriers. Large-scale commercial fisheries also put additional pressure on subsistence resources, reducing access to traditional food sources.

Linguistic research on how transgender speakers use speech traits commonly associated with gender, such as f0 and /s/, is still limited in scope. Past research has focused primarily on singular gender groups within the transgender community, particularly transmasculine speakers, leaving gaps in how speech differs among transgender speakers. Additionally, there has been little research on how transgender identity influences perception of gender in speech. Here, I use data from reading passages by fifteen university-age transgender speakers in New England to analyze variation in /s/ and f0 among transmasculine, transfeminine, nonbinary, and multigender speakers. "Traumatic brain injury (TBI) is a leading cause of death and disability worldwide. Neurodegenerative diseases that develop post-TBI can be, in part, attributed to DNA and cell-damaging reactive oxygen species (ROS) and lipid peroxidation products (LPOx). This thesis focused on overcoming the limits of current TBI material treatment approaches by employing a RAFT (reversible-addition fragmentation chain transfer) polymer approach that incorporated novel therapeutic, diagnostic and peptide-targeting monomers. An improved nanoparticle synthesis approach was also developed. The current adsorption processes investigated for biogas upgrading encounter severe challenges with regards to both adsorbents and processes, which necessitate the design and development of novel approaches to effectively separate carbon dioxide from methane and, hence, produce pipeline-grade renewable natural gas (RNG) as a fuel. These key challenges include (i) moderate CO2/CH4 selectivity, (ii) relatively long cycle times, (iii) high regeneration energy requirements, (iv) high-pressure drop, and (v) short adsorbent lifetime. This research develops and refines the Split Inversion Pulse and Recovery (SIP R) methodology for spin-lattice relaxation (T₁) measurements in NMR spectroscopy. SIP R introduces a two-scan difference technique using a split inversion pulse sequence, where the 180° pulse is split into two 90° pulses phase-shifted with respect to each other. This approach simplifies data fitting, requiring only two parameters to extract T₁ values, compared to the traditional inversion-recovery method, which needs three. The SIP-R-DS and SIP-R-S adaptations extend this framework by incorporating selective NMR resonance excitations, enhancing its applicability to cross-polarization experiments and enabling unobstructed NOE measurements. Effective ocular drug delivery has always been a challenge for clinical and research. Although many nanotechnology-based formulations have been developed and showed great potential in ocular drug delivery, this is far from enough to meet clinical requirements. These studies aimed at the design and synthesis of novel nanoparticles using multi-inlet vortex mixer (MIVM) to improve ocular drug delivery efficiency. The first project involves packaging two different anti-glaucoma drugs, brimonidine (BM) and betaxolol (BX), into solid drug nanoparticles (SDNs) by MIVM, which can achieve both functions, reducing aqueous humor production and promoting aqueous humor efflux. Exposure to organophosphate-based nerve agents and pesticides poses significant health and security threats to civilians, soldiers, and first responders. Despite extensive efforts to develop chemical detoxification agents for use in topical applications on exposed skin surfaces and for intravenous injections, there remains an unmet need for effective, non-hazardous decontaminating agents. The current state-of-the-art decontaminating agent, Dekon-139 (2,3-butanedione oxime, potassium salt), exhibits adverse effects when applied to the skin.

The synthesis of high-quality graphene from sustainable carbonaceous feedstocks offers an avenue to reduce the environmental and economic costs associated with conventional graphite-based production. This research investigates the co-utilization of biochar and graphite as precursors for graphene fabrication in an ultrasonic reactor. Biochar was produced from biomass via downdraft gasification at 850 °C, yielding a high fixed-carbon, partially graphitized material with favorable surface area and porosity. Graphite, selected for its crystalline structure, was investigated with biochar to assess the influence of precursor composition on exfoliation efficiency, layer thickness distribution, and defect density. BACKGROUND: Hyperkalemia, generally defined as serum potassium levels greater than 5.0 mEq/L, poses significant clinical risks, including cardiac toxicity and muscle weakness. Its prevalence and severity increase in patients with chronic kidney disease (CKD), diabetes mellitus, and heart failure (HF), particularly when compounded by medications like angiotensin converting inhibitors, angiotensin receptor blockers, and potassium sparing diuretics. Hyperkalemia arises from disruptions in potassium regulation involving intake, excretion, and intracellular-extracellular distribution. In CKD and acute kidney injury, these regulatory mechanisms are impaired, leading to heightened risk. Metabolic-associated steatotic liver disease is a metabolic disorder with limited pharmacological treatment options and only one approved medication to date. MASLD can be asymptomatic in its earlier stage and hence, prevention of this disease progression is the most effective approach. Recently, green-synthesized nanoparticles, particularly silver (AgNPs) and gold nanoparticles (AuNPs), have gained interest due to their eco-friendly production, biocompatibility, and bioactive properties. However, natural browning enzymes can limit its use in baking. This study aims to determine droplet deposition resulting from dynamic change in payloads and flight speeds of Unmanned Aerial Vehicle (UAV) during operation. The study was conducted at Universiti Putra Malaysia, Serdang, Malaysia. The Advansia A1 hexacopter was first set to fly at combinations of 3 payloads (3.3, 6.6, and 9.9 kg) with 3 flight speeds (3, 5 and 7 m/s) to collect individual rotor RPM, which will later be used to set the rotors’ speed during indoor testing experiment. This paper examines the use of horticulture and botany in Ebony Patterson's 2023 exhibition at the New York Botanical Garden, …things come to thrive…in the shedding…in the molting… as it relates to the history of NYBG, the Bronx, and the populations that historically and currently reside in the borough. Using plants, blown glass, and glittering styrofoam forms, Patterson productively intervenes in the relationship between an historic, prestigious institution and a changed borough that, since the 1990s, has been walled off from the Garden.
