Understanding sex disparities in modifiable risk factors across the lifespan is essential for crafting individualized intervention strategies. We aim to investigate age-related sex disparity in cardiometabolic phenotypes in a large nationwide Chinese cohort. A total of 254,670 adults aged 40 years or older were selected from a population-based cohort in China. Substantial sex disparities in the prevalence of metabolic diseases were observed across different age strata, particularly for dyslipidemia and its components. Generalized additive models were employed to characterize phenotype features, elucidating how gender differences evolve with advancing age. The Pharmacogene Variation Consortium (PharmVar) serves as a global repository providing star (*) allele nomenclature for the polymorphic human CYP4F2 gene. CYP4F2 genetic variation impacts the metabolism of vitamin K, which is associated with warfarin dose requirements, and the metabolism of drugs, such as imatinib or fingolimod, and certain endogenous compounds including vitamin E and eicosanoids. This GeneFocus provides a comprehensive overview and summary of CYP4F2 genetic variation including the characterization of 14 novel star alleles, CYP4F2*4 through *17. The incidence of metabolic dysfunction-associated steatohepatitis (MASH) is on the rise, and with limited pharmacological therapy available, identification of new metabolic targets is urgently needed. Oxalate is a terminal metabolite produced from glyoxylate by hepatic lactate dehydrogenase (LDHA). The liver-specific alanine-glyoxylate aminotransferase (AGXT) detoxifies glyoxylate, preventing oxalate accumulation. Here we show that AGXT is suppressed and LDHA is activated in livers from patients and mice with MASH, leading to oxalate overproduction. In turn, oxalate promotes steatosis in hepatocytes by inhibiting peroxisome proliferator-activated receptor-α (PPARα) transcription and fatty acid β-oxidation and induces monocyte chemotaxis via C-C motif chemokine ligand 2. Drug resistance poses a crucial challenge in healthcare, with response rates to chemotherapy and targeted therapy remaining low. Individual patient's resistance is exacerbated by the intricate heterogeneity of tumor cells, presenting significant obstacles to effective treatment. To address this challenge, DrugFormer, a novel graph-augmented large language model designed to predict drug resistance at single-cell level is proposed. DrugFormer integrates both serialized gene tokens and gene-based knowledge graphs for the accurate predictions of drug response. Temporal ordering of cellular events offers fundamental insights into biological phenomena. Although this is traditionally achieved through continuous direct observations1,2, an alternative solution leverages irreversible genetic changes, such as naturally occurring mutations, to create indelible marks that enables retrospective temporal ordering3–5. Using a multipurpose, single-cell CRISPR platform, we developed a molecular clock approach to record the timing of cellular events and clonality in vivo, with incorporation of cell state and lineage information. Too Little Too Late: Why The Environmental Justice Problems Caused By The Army Corps's Nationwide Permits Program Run Much Deeper Than Permit 12, Travis O. Brandon

This Article argues that the United States Army Corps of Engineers’ review of Nationwide Permit 12 (NWP 12) is insufficient to remedy the deeper environmental justice defects embedded within the Nationwide Permit Program as a whole. NWP 12, which facilitates expedited authorization of oil and natural gas pipeline crossings under § 404 of the Clean Water Act, has enabled the construction of fossil fuel infrastructure with minimal site-specific environmental review and no meaningful opportunity for public comment. Predicting phenotypes from genotypes is a central challenge in systems biology in understanding how organisms respond to environmental and genetic changes. To address this, genome-scale metabolic models (GSMs) integrated with omics data and machine learning provide a comprehensive framework to connect genotype to phenotype. This dissertation utilizes these approaches in plants and bacterial systems, revealing key metabolic adaptations. In maize, a root-specific GSM reveals metabolic reprogramming under nitrogen stress, offering strategies for improving stress tolerance. Building on this, a multi-organ maize GSM of temperature stress identifies metabolic bottlenecks, providing targets to enhance crop resilience. Similarly, in rice, a grain-specific GSM …

Acculturation experience and attitudes toward seeking mental health services among Asian Indian Americans, an ethnic minority group, are predominantly underrepresented in current literature despite their increasing number in the United States. The aim of this study is to isolate and characterize Amoxicillin and Favipiravir biodegrading fungi as well as determine their characteristics and degradation pathways. The antibiotic-degrading fungus A. flavus was isolated from polluted wastewater samples using standard Potato Dextrose agar and Czapek–Dox medium. The biodegradation method was investigated in previous mediums with (Amoxicillin and Favipiravir), as the sole carbon sources. This article focuses on Pelargonium graveolens, a fragrant medicinal plant from the Geraniaceae family. The study examines the plant's phytochemical composition, antioxidant activity, and mineral absorptivity, with the plant being grown indoors. The study also examined the plant's ash content and antioxidant activity using a variety of techniques, and the results demonstrated that P. graveolens is effective at absorbing lead. The plant contains eight different minerals, including Cu, Mn, Co, Ni, Pb, Mg, Fe, and Ca. Statistical analysis was used to determine the level of antioxidants present, using DPPH, reducing power, and total antioxidant capacity methods. The extraction process used …

Sexual dimorphism and reproductive biology are fundamental aspects of fish breeding studies. The aim of this research was to analyze the sexual dimorphism and reproductive biology of Notopterus notopterus. A total of 74 N. notopterus were collected from the Kelekar River in Ogan Ilir Regency, Indonesia, consisting of 38 males (TL: 18–23.6 cm; BW: 35.1–92.1 g) and 36 females (TL: 19.6-26.3 cm; BW: 49.4–133.8 g). Seventeen morphometric characters, three meristic characters, and five reproductive biological parameters were analyzed. In a series of recent cases, the Supreme Court has invoked the newly named “major questions doctrine” to strike down agency regulations that protect public health and the environment. Several Justices have argued that while the name “major questions” may be new, these decisions are simply the latest iteration in a longstanding effort of the courts to curtail the explosive growth of the administrative state since 1970. The quest for materials with extraordinary properties has been a longstanding endeavor in material science and engineering, driving future technological advancement. However, the discovery of such materials is non-trivial. Recent advancements in computational methods, particularly the integration of machine learning (ML) techniques with density functional theory (DFT), have opened new avenues for accelerating the discovery of materials with exceptional and extreme properties. This dissertation focuses on developing a synergistic approach and workflow combining ML and DFT to identify materials with properties that are pushed beyond current limits, using lattice thermal conductivity (LTC) as a case study of the workflow.

Backlash on social media in response to online behaviors is swift and often decisive with significant consequences. One possible reason for this is that online actions are perceived as reflective of a person’s true self. Signaling theory provides support for this argument and suggests that people intentionally display information about themselves to communicate their beliefs to others. While existing work suggests that social media posts are intentional, social media interactions such as likes, comments, and reposts might also function similarly (i.e., as a signal of beliefs).
