Dapivirine Vaginal Ring (PrEP Ring)

Leveraging our experience developing the monthly dapivirine vaginal ring (DVR or “PrEP ring”), which received a positive opinion from the European Medicines Agency in 2020 for women’s HIV risk reduction, the Population Council is developing a 3-month multipurpose vaginal ring to help women protect against both HIV and unintended pregnancy. This ring is designed to slowly release the antiretroviral (ARV) drug, dapivirine and the contraceptive hormone, levonorgestrel.

The study of environmental DNA (eDNA) has provided researchers with a more accessible and sensitive way to identify the presence of specific species compared to traditional monitoring methods. eDNA enables species detection by analyzing environmental samples such as water or soil, which contain genetic material shed by organisms in a given area. Research indicates that eDNA techniques have become a valuable method for monitoring threatened and invasive species, proving particularly reliable for detecting aquatic species compared to traditional techniques. The depletion of the stratospheric ozone layer has led to increased levels of harmful UV-B radiation reaching the Earth's surface, posing significant challenges for plant health and development. This study investigates the oxidative stress responses in Arabidopsis thaliana when exposed to UV-B irradiation. Arabidopsis thaliana, a well-established model organism, is particularly sensitive to UV-B, making it ideal for understanding the effects of UV-B-induced oxidative stress. UV-B radiation, a mutagenic component of sunlight, disrupts key metabolic, molecular, and developmental processes, leading to DNA damage, impaired metabolism, and morphological changes. Hurler Syndrome is a rare pediatric neurodegenerative disorder, in which patients experience a multitude of symptoms starting around age 3. These symptoms include skeletal abnormalities, cognitive impairment, stunted growth, heart disease, and many others. The life expectancy of a child diagnosed with Hurler Disease is around 10 to 20 years. Currently, there is no cure for Hurler Syndrome and most treatments are targeted at relieving symptoms. Hurler Syndrome is an inherited lysosomal storage disorder defined by an accumulation of mucopolysaccharides, dermatan sulfate (DS) and heparan sulfate (HS) in tissues. Objective. Balance impairment is one of the most debilitating consequences of traumatic brain injury (TBI). To study the neurophysiological underpinnings of balance impairment, the brain functional connectivity during perturbation tasks can provide new insights. To better characterize the association between the task-relevant functional connectivity and the degree of balance deficits in TBI, the analysis needs to be performed on the data stratified based on the balance impairment. Mammalian auditory hair cells (HCs) are not naturally regenerative and must preserve their elaborate structure to ensure lifelong hearing. Stereocilia, the actin-based projections at the HC surface that detect sound vibration, are particularly vulnerable to damage incurred from noise and aging. We show that the tip-localized protein module GPSM2–GNAI required for stereocilia development is also involved in stereocilia maintenance in mature cells. Inactivating Gpsm2 in adult mouse HCs results in low-frequency hearing deficits and stereocilia height reduction proportional to the region reported to turn over actin at the distal tip. Heightened energetic and nutrient demand during lactogenic differentiation of the mammary gland elicits upregulation of various stress responses to support cellular homeostasis. Here, we identify the stimulator of interferon genes (STING) as an immune supporter of the functional development of mouse mammary epithelial cells (MECs). An in vitro model of MEC differentiation revealed that STING is activated in a cGAS-independent manner to produce both type I interferons and proinflammatory cytokines in response to the accumulation of mitochondrial reactive oxygen species. Genetic heterogeneity in tumors can show a remarkable selectivity when two or more independent genetic events occur in the same gene. This phenomenon, called composite mutation, points toward a selective pressure, which frequently causes therapy resistance to mutation-specific drugs. Since composite mutations have been described to occur in sub-clonal populations, they are not always captured through biopsy sampling. Here, we provide a proof of concept to predict composite mutations to anticipate which patients might be at risk for sub-clonally driven therapy resistance. Hair cells of the inner ear and lateral-line system rely on specialized ribbon synapses to transmit sensory information to the central nervous system. The molecules required to assemble these synapses are not fully understood. We show that Nrxn3, a presynaptic adhesion molecule, is crucial for ribbon-synapse maturation in hair cells. In both mouse and zebrafish models, the loss of Nrxn3 results in significantly fewer intact ribbon synapses. We show in zebrafish that, initially, Nrxn3 loss does not alter pre- and postsynapse numbers but, later, synapses fail to pair, leading to postsynapse loss. Neurosteroids are endogenous steroids within the brain that act as modulators in neurotransmission. Among their targets are the gamma-aminobutyric acid (GABA) receptors, which are responsible for inhibitory signaling in the brain. The interaction between neurosteroids and GABA receptors is complex, influencing mood regulation and pain perception. Endogenous neurosteroids have been used in conditions such as postpartum depression (PPD), where fluctuation in neurosteroid levels contribute to the onset of depressive systems. Conversely, exogenous neurosteroids, such as brexanolone and zuranolone, have emerged as therapeutic agents for PPD. Pyruvate carboxylase (PC) is implicated in a variety of diseases, including type-2 diabetes, cancer, and bacterial/viral infections. However, there are currently no molecular tools capable of precisely manipulating PC activity both in vitro and in vivo. This dissertation describes the identification and characterization of 1,3-disubstituted imidazolidinetriones as a novel class of potent, selective, and permeable allosteric inhibitors of Staphylococcus aureus PC. Based on kinetic, structural, and biophysical data, this class of inhibitors is hypothesized to bind at the noncatalytic “exo binding” site on PC. The purpose of this action research project was to determine if incorporating total participation techniques into a fifth-grade literacy classroom would increase reading comprehension scores. This researcher was driven to find a teaching strategy that would boost student engagement, participation and academic achievement. It has been proven that student centered learning increases student participation. Evidence has shown increasing student participation and engagement leads to an increase in academic achievement. A quantitative study was used to determine the effectiveness of total participation techniques in a fifth-grade literacy classroom. Comprehension assessment data was collected and analyzed. Bullying is a phenomenon that exists across cultures, societies, and even species. It can be found everywhere, including the workplace. The present study investigates workplace bullying amongst an approximated census sample of Louisiana academic library workers. Using the Negative Acts Questionnaire—Revised (NAQ-R), the study explores the relationships between experiencing workplace bullying and librarian status, tenure status, years in the profession, age, gender, and race. Although no statistically significant relationships were found between variables, the findings add to a growing body of literature that explores the interrelations of demographic and environmental contexts and bullying experiences.

Fundus photography is a radiation-free, non-invasive imaging modality widely employed to assess both ocular and systemic diseases in population-level applications. Traditional methods for extracting cardiovascular biomarkers rely on quantifying vascular features in these images to identify indicators of disease. However, these approaches often depend on manual clinical expertise, which can be costly and time-consuming, making them impractical for large-scale datasets. Deep learning offers a compelling alternative to traditional techniques by enabling the automated processing of extensive datasets with high accuracy. This has been offered for thelast ten years and preliminary data has shown that it can provide symptomaticrelief to patients. Scientists are beginning to investigate atmospheric methane removal approaches, which would accelerate the conversion of methane to a less radiatively potent form or physically remove methane from the atmosphere and store it elsewhere. Developing and, if appropriate, deploying atmospheric methane removal approaches will require an understanding of relevant legal considerations and governing structures that could impact whether, when, where, and how specific projects take place. This paper examines the treatment of atmospheric methane removal approaches under international and U.S. domestic law.
