The major female ovarian hormone, 17β-estradiol (E2), can alter neuronal excitability within milliseconds to regulate a variety of physiological processes. Estrogen receptor-α (ERα), classically known as a nuclear receptor, exists as a membrane-bound receptor to mediate this rapid action of E2, but the ionic mechanisms remain unclear. Here, we show that a membrane channel protein, chloride intracellular channel protein-1 (Clic1), can physically interact with ERα with a preference to the membrane-bound ERα. Clic1-mediated currents can be enhanced by E2 and reduced by its depletion. In addition, Clic1 currents are required to mediate the E2-induced rapid excitations in multiple brain ERα …

Science, Technology, Engineering, and Mathematics (STEM) disciplines include various programming needs across institutional levels. In high school settings, STEM programs are designed to encourage critical thinking and interest in STEM careers. At postsecondary institutions, STEM disciplines allow students to earn credentials to enter different career sectors. However, some STEM educators are transforming how they perceive their discipline by incorporating innovative practices into their work. Instructional materials (i.e., textbooks, lab manuals, and other ancillary materials) allow educators to diversify their teaching style. Implementing OERs further emboldens educators to incorporate different resources in their classes. However, as innovation spurs positive feedback, challenges …

Online credit recovery (OCR) allows US students at risk of dropping out of school to retake courses to meet graduation requirements and eventually graduate from high school. The problem addressed in this study is that despite participating in online English II credit recovery (ELA II OCR), students in a rural school district in North Florida still had a high failure rate on the Florida Standards Assessment English Language Arts (FSA-ELA) exam. This article is part of a clinical practice guideline update on the risk assessment, diagnostic imaging, and microbiological evaluation of complicated intraabdominal infections in adults, children, and pregnant people, developed by the Infectious Diseases Society of America. In this article, the panel provides recommendations for diagnostic imaging of suspected acute cholecystitis and acute cholangitis. The panel's recommendations are based on evidence derived from systematic literature reviews and adhere to a standardized methodology for rating the certainty of evidence and strength of recommendation according to the Grading of Recommendations Assessment, Development, and Evaluation (GRADE) approach.

Wavefront shaping allows focusing light through or inside strongly scattering media, but the background intensity also increases which reduces the target's contrast. By combining transmission or deposition matrices for different regions, we construct joint operators to achieve spatially resolved control of light in diffusive systems. The eigenmode of a contrast operator can maximize the power contrast between a target and its surrounding. A difference operator enhances the power delivery to a target while avoiding the background increase. This work opens the door to coherent control of nonlocal effects in wave transport for practical applications.

The MRI-derived brain network serves as a pivotal instrument in elucidating both the structural and functional aspects of the brain, encompassing the ramifications of diseases and developmental processes. However, prevailing methodologies, often focusing on synchronous BOLD signals from functional MRI (fMRI), may not capture directional influences among brain regions and rarely tackle temporal functional dynamics. In this study, we first construct the brain-effective network via the dynamic causal model. Subsequently, we introduce an interpretable graph learning framework termed Spatio-Temporal Embedding ODE (STE-ODE). Parental incarceration has a significant impact on children’s and adolescents’ lives, including emotional and mental well-being, which can carry to their adulthood. Although parental incarceration has been researched, the lived experiences and mental health needs of Latino children and adolescents of incarcerated parents have not been explored. The purpose of this qualitative phenomenological study was to understand the recollections of young Latino adults about their lived experiences and mental health needs as children and adolescents with incarcerated parents. Acute lymphoblastic leukemia expressing the gamma delta T-cell receptor (γδ T-ALL) is a poorly understood disease. We studied 200 children with γδ T-ALL from 13 clinical study groups to understand the clinical and genetic features of this disease. We found age and genetic drivers were significantly associated with outcome. γδ T-ALL diagnosed in children under 3 years of age was extremely high-risk and enriched for genetic alterations that result in both LMO2 activation and STAG2 inactivation. Mechanistically, using patient samples and isogenic cell lines, we show that inactivation of STAG2 profoundly perturbs chromatin organization by altering enhancer-promoter looping, resulting in …

Multiple factors in the design of a chimeric antigen receptor (CAR) influence CAR T-cell activity, with costimulatory signals being a key component. Yet, the impact of costimulatory domains on the downstream signaling and subsequent functionality of CAR-engineered natural killer (NK) cells remains largely unexplored. Here, we evaluated the impact of various costimulatory domains on CAR-NK cell activity, using a CD70-targeting CAR. We found that CD28, a costimulatory molecule not inherently present in mature NK cells, significantly enhanced the antitumor efficacy and long-term cytotoxicity of CAR-NK cells both in vitro and in multiple xenograft models of hematologic and solid tumors. Biodiversity is declining across all natural environments, however, freshwater systems are thought to be among the most threatened. Quantifying and understanding trends in freshwater biodiversity has become more urgent as rapid biodiversity loss continues. To provide a robust ecological assessment, more than one bioindicator should be used, along with multiple biodiversity metrics. Macrophytes and diatoms are important components of aquatic systems, especially in shallow lakes. Chitosan was synthesized from Apis Mellifera dead bees by traditional and cryogenic methods, and the degree of deacetylation, viscosity, and molecular mass of the synthesized sample was studied. Chitosan was synthesized by deacetylation of chitin under cryogenic conditions. NaOH concentration 40%, temperature 90 ± 3 °C, time 120 min. The degree of deacetylation, and molecular mass was determined using an Ubbelode viscometer. DDA was determined by conductometric titration under standard conditions. The degree of deacetylation of Apis Mellifera chitosan was 87%. synthesized by cryogenic method DDA of chitosan was 83% synthesized by traditional method. A hallmark of addiction is the ability of drugs of abuse to trigger relapse after periods of prolonged abstinence. Here, we describe an epigenetic mechanism whereby chronic cocaine exposure causes lasting chromatin and downstream transcriptional modifications in the nucleus accumbens (NAc), a critical brain region controlling motivation. We link prolonged withdrawal from cocaine to the depletion of the histone variant H2A.Z, coupled with increased genome accessibility and latent priming of gene transcription, in D1 dopamine receptor-expressing medium spiny neurons (D1 MSNs) that relate to aberrant gene expression upon drug relapse. The major female ovarian hormone, 17β-estradiol (E2), can alter neuronal excitability within milliseconds to regulate a variety of physiological processes. Estrogen receptor-α (ERα), classically known as a nuclear receptor, exists as a membrane-bound receptor to mediate this rapid action of E2, but the ionic mechanisms remain unclear. Here, we show that a membrane channel protein, chloride intracellular channel protein-1 (Clic1), can physically interact with ERα with a preference to the membrane-bound ERα. Clic1-mediated currents can be enhanced by E2 and reduced by its depletion. In addition, Clic1 currents are required to mediate the E2-induced rapid excitations in multiple brain ERα …

Foveal and peripheral vision are two distinct modes of visual processing essential for navigating the world. However, it remains unclear if they engage different neural mechanisms and circuits within the visual attentional system. Here, we trained macaques to perform a free-gaze visual search task using natural face and object stimuli and recorded a large number of 14588 visually responsive units from a broadly distributed network of brain regions involved in visual attentional processing. Foveal and peripheral units had substantially different proportions across brain regions and exhibited systematic differences in encoding visual information and visual attention.
