Sex has generated incredible life cycle diversity across eukaryotes. Differences in the timing of meiosis and fertilization have generated tremendous variation in the duration of haploid and diploid phases. To understand what drives eukaryotic sexual diversity, there remains a need to determine the rates of sexual and asexual reproduction in natural populations. Moreover, life cycle diversity is predicted to be correlated with the reproductive system. Temporal Lobe Epilepsy (TLE) is the most prevalent form of focal epilepsy and the most treatment-resistant type. This condition is characterized by recurrent, unprovoked seizures typically originating in the hippocampus. Epigenetic modifications such as DNAme changes have been implicated in brain functions such as synaptic plasticity, learning and, memory, and cognition. The oxidation of DNA 5-methylcytosine (5-mC) and its oxidized form, 5-hydroxymethylcytosine (5-hmC) catalyzed by Ten-Eleven Translocation (TET) family of dioxygenases and linked to epilepsy's hyperexcitable state. Therapeutic options for the treatment of Surfactant Protein-B (SP-B) deficiency, an autosomal recessive disorder that leads to fatal respiratory distress, are extremely limited with the exception of double lung transplantation. One method for treating this monogenic lung disease is with adeno-associated virus (AAV) gene therapy treatment that delivers the hSFTPB transgene into the lungs. The transient nature of AAV transgene expression led to a need for sustained gene expression, which could possibly be achieved through redosing. Self-derivation through integration is the process of integrating novel facts and producing new knowledge never directly taught. Knowledge integration has been studied with the presentation of two novel facts. However, in educational settings, individuals are required to integrate new information with prior knowledge learned days, months, or years earlier. Prior knowledge robustly predicts learning outcomes, but less is known about self-derivation through the integration of new information with prior knowledge. Thus, in Study 1, we examined adults' (n = 25) memory integration of new facts with prior knowledge. The participants had 52% accuracy in self-derivation. In Study 2 (n = …

LGBTIQ+ research acknowledges shared experiences of groups marginalized due to gender identities, sexualities, and sex characteristics. This universalist coalition approach has resulted in much affirmational research and progressive policy development. However, it risks homogenizing the unique experiences and needs of specific groups; a risk lessened by a particularist subgroup approach. In this theoretical paper, we reflect on the challenges of a coalition or subgroup approach by considering interdependencies and boundaries between sex, gender, and sexuality-based identities. Parent mental health challenges in the postpartum and early parenthood have profound implications for parent, child, and family well-being. Little research has focused on postpartum mental health challenges and barriers to help-seeking among queer birthing people, including members of this community who may be particularly vulnerable to mental health difficulties, such as queer cis women partnered with men, trans/nonbinary parents, and queer parents who are young, low-income, and/or of color. Wild populations of New England cottontails (NECs, Sylvilagus transitionalis) are declining and occupy a small proportion of their historic range. To conserve this species, wild-caught cottontails participate in a reintroduction breeding program. To increase the program’s productivity, this study described breeding behavior in pairings with (n = 3 pairings) and without (n = 9 pairings) reproductive success. Females were paired with two males consecutively and behaviors were recorded using 24 h continuous video footage. Activity peaked between 19:00 and 06:00. Copulation was only observed in successful pairings, and the gestation period was 31 days. Successful pairings had …

Adaptive immunity, comprised in part by the antibody repertoire, is the cornerstone of our ability to remain healthy amongst the plethora of pathogenic challenges we encounter daily. Despite the importance of understanding antibody repertoire development, much remains unknown about the composition and evolution of circulating antibodies in response to viral challenges. To better understand antibody repertoire development, we profiled the serum antibody repertoire using a combination of next-generation sequencing and serum antibody proteomics in the predominantly naïve response against SARS-CoV-2 spike and the recalled response to influenza hemagglutinin. Responsiveness to chimeric antigen receptor (CAR) T cell therapy correlates with CAR T cell expansion and persistence in vivo. Multiple working strategies improve CAR T cell persistence by increasing stem-like properties or by sustaining CAR T cell activity with combination therapies. Herein, we describe the intrinsic ability of CAR T cells to differentiate into memory T cells, the effect of cytokine armoring, and neoadjuvant CD4 depletion therapy on CAR and tumor-specific endogenous memory T cells. This study aims to assess the competitive advantage of foreign trade in agri-food products of five Association of South East Asia Nations (ASEAN) countries, namely Indonesia, Malaysia, Thailand, Singapore, and the Philippines between 2003–2022 by dividing the period into the global crisis in 2009, the trade war between China and the United States (US) in 2018, and the COVID-19 pandemic in 2020. The study built an econometric model using the Revealed Comparative Advantage (RCA) and Revealed Symmetric Comparative Advantage (RSCA) indicators. The green sea urchin (Strongylocentrotus droebachiensis) fishery was once prosperous in the Gulf of Maine, peaking in the 1980s and early 1990s. However, since the early 1990s, the fishery has dramatically declined despite having harvesting restrictions. To sustain the industry, production through aquaculture is necessary, but hatchery production methods are not yet optimized, with settlement success and post-settlement survival as low as 1-5%, and the spatial scale for field grow-out may be limited in some regions due to climate change. Background: Previous research has shown that the Serum Amyloid A (SAA) protein family is intricately involved in inflammatory signaling and various disease pathologies. We have previously demonstrated that SAA is associated with increased colitis disease severity and the promotion of tumorigenesis. However, the specific role of SAA proteins in breast cancer pathology remains unclear. Therefore, we investigated the role of systemic SAA1 and SAA2 (SAA1/2) in a triple-negative breast cancer mouse model.

Microplastics, which are defined as plastic particles less than 5mm in size, have additionally become increasingly recognized as a harmful pollutant in aquatic systems. Constructed treatment wetland systems (CTWSs) are highly effective at removal and mitigation of pollutants, including anthropogenic nutrient inputs. The dimeric nuclear factor kappa B (NF- κB) transcription factors (TFs) regulate gene expression by binding to a variety of κB DNA elements with conserved G:C-rich flanking sequences enclosing a degenerate central region. Toward defining mechanistic principles of affinity regulated by degeneracy, we observed an unusual dependence of the affinity of RelA on the identity of the central base pair, which appears to be noncontacted in the complex crystal structures. We demonstrate a single-component hydrophilic photocrosslinkable copolymer system that incorporates all critical functionalities into one chain. This design allows for the creation of uniform functional organic coatings on a variety of substrates. The copolymers were composed of a poly(ethylene oxide)-containing monomer, a monomer that can release a primary amine upon UV light, and a monomer with reactive epoxide or cyclic dithiocarbonate with a primary amine. These copolymers are easily incorporated into the solution-casting process using polar solvents. Furthermore, the resulting coating can be readily stabilized through UV light-induced crosslinking, providing an advantage for controlling the surface properties of various substrates. …

In this study, we aim to quantify coating uniformity and correlate fluorescence intensity to drug loading for drug-coated angioplasty balloons (DCB) coated with 5, 10, 15, or 20 layers of poly(lactic-co-glycolic acid) nanoparticles (NPs) entrapped with quercetin. Uniformity was quantified from histograms and horizontal line profiles of microscopic fluorescent images acquired with sample specific parameters, and cracks in the coating were measured and counted. The fluorescence of images acquired with global parameters was correlated with quercetin loading measured via gravimetric/HPLC analysis. More layers on DCBs may be associated with less uniform coatings, as indicated by differences in histogram standard deviations. …
