IMPORTANCE: The prevalence, pathophysiology, and long-term outcomes of COVID-19 (post-acute sequelae of SARS-CoV-2 [PASC] or "Long COVID") in children and young adults remain unknown. Studies must address the urgent need to define PASC, its mechanisms, and potential treatment targets in children and young adults.

OBSERVATIONS: We describe the protocol for the Pediatric Observational Cohort Study of the NIH's REsearching COVID to Enhance Recovery (RECOVER) Initiative. RECOVER-Pediatrics is an observational meta-cohort study of caregiver-child pairs (birth through 17 years) and young adults (18 through 25 years), recruited from more than 100 sites across the US. This report focuses on two of …

Human fetal membranes (amniochorion) that line the intrauterine cavity consist of two distinct cell layers; single-layer amnion epithelial cells (AEC) and multilayer chorion trophoblast cells (CTC). These layers are connected through a collagen-rich extracellular matrix. Cellular remodeling helps support membrane growth and integrity during gestation and helps to maintain pregnancy. Preterm prelabor rupture of the human amniochorionic (fetal) membrane (pPROM) is antecedent to 40% of all spontaneous preterm birth. Oxidative stress (OS) induced activation of the p38 MAPK due to various maternal risk exposures and the amniochorion cells' senescence are reported pathological features of pPROM. Our transcriptomics analysis implicated dysregulated …

There is a paucity of literature on the effect of COVID-19 on hospital processes. We hypothesized that COVID-19 was associated with decreased cancer physiatry referrals in 2020. This is a retrospective cohort study of consecutive patients from April to July 2019 and 2020 admitted at an academic quaternary cancer center. The main outcomes were number of hospital admissions, rate, and characteristics of inpatient rehabilitation admissions and change in percentage of physiatry referrals as the primary endpoint. Results showed that in 2019, there were 387 referrals from 10,274 inpatient admissions (3.8%; 95% confidence interval, 2.4-4.2), compared with 337 referrals from 7051 …

The essential branched-chain amino acids (BCAAs) leucine, isoleucine, and valine play critical roles in protein synthesis and energy metabolism. Despite their widespread use as nutritional supplements, BCAAs' full effects on mammalian physiology remain uncertain due to the complexities of BCAA metabolic regulation. Here a novel mechanism linking intrinsic alterations in BCAA metabolism is identified to cellular senescence and the senescence-associated secretory phenotype (SASP), both of which contribute to organismal aging and inflammation-related diseases. Altered BCAA metabolism driving the SASP is mediated by robust activation of the BCAA transporters Solute Carrier Family 6 Members 14 and 15 as well as downregulation …

BACKGROUND: Previous research showed that 5-hydroxytryptophan (5HTP), a metabolic precursor of serotonin, reduces allergic lung inflammation by inhibiting eosinophil migration across endothelial monolayers.

OBJECTIVE: It is unknown if serotonin receptors are involved in mediating this 5HTP function or if serotonin receptor (HTR) single nucleotide polymorphisms (SNPs) associate with lung function in humans.

METHODS: Serotonin receptor subtypes were assessed by qPCR, western blot, confocal microscopy, pharmacological inhibitors and siRNA knockdown. HTR SNPs were assessed in two cohorts.

RESULTS: Pharmacological inhibition or siRNA knockdown of the serotonin receptors HTR1A or HTR1B in endothelial cells abrogated the inhibitory effects of 5HTP on eosinophil …

Human embryonic stem cells (hESCs) cultured in media containing bone morphogenic protein 4 (BMP4; B) differentiate into trophoblast-like cells. Supplementing media with inhibitors of activin/nodal signaling (A83-01) and of fibroblast growth factor 2 (PD173074) suppresses mesoderm and endoderm formation and improves specification of trophoblast-like lineages, but with variable effectiveness. We compared differentiation in four BMP4-containing media: mTeSR1-BMP4 only, mTeSR1-BAP, basal medium with BAP (basal-BAP), and a newly defined medium, E7-BAP. These media variably drive early differentiation towards trophoblast-like lineages with upregulation of early trophoblast markers CDX2 and KRT7 and downregulation of pluripotency markers (OCT4 and NANOG). As expected, based on …

Endosalpingiosis (ES) and endometriosis (EM) refer to the growth of tubal and endometrial epithelium respectively, outside of their site of origin. We hypothesize that uterine secretome factors drive ectopic growth. To test this, we developed a mouse model of ES and EM using tdTomato (tdT) transgenic fluorescent mice as donors. To block implantation factors, progesterone knockout (PKO) tdT mice were created. Fluorescent lesions were present after oviduct implantation with and without WT endometrium. Implantation was increased (p<0.05) when tdt oviductal tissue was implanted with endometrium compared to oviductal tissue alone. Implantation was reduced (p<0.0005) in animals implanted with minced tdT oviductal tissue with PKO tdT endometrium compared to WT endometrium. Finally, oviductal tissues was incubated with and without a known implantation factor, leukemia inhibitory factor (LIF) prior to and during implantation. LIF promoted lesion implantation. In conclusion, endometrial derived implantation factors, such as LIF, are necessary to initiate ectopic tissue growth. We have developed an animal model of ectopic growth of gynecologic tissues in a WT mouse which will potentially allow for development of new prevention and treatment modalities.

I Can-B Money Savvy! Teaching Adults With Intellectual And Developmental Disabilities To Make Purchasing Decisions Online, Kaley B. Adams

Electronic Theses and Dissertations 2020 - Present

Online shopping is a common feature of modern adult life and is made easy with targeted advertisements, a large buy now button, and pre-entered credit card information. However, these features do not support making purchasing decisions based on what is best for the individual. Decision making about buying items requires an understanding of basic mathematics, one’s current finances and circumstances, and information about the product, then choosing whether to purchase the item. Adults with intellectual and developmental disabilities (IDD) need explicit instruction on how to make informed decisions regarding online purchases. The current study sought to evaluate the effectiveness of …

Identifying Epigenetic Modifications To Combat Resistance To The Chemotherapeutic Agent Doxorubicin, Anna Lesley Kingham Jan 2024

Identifying Epigenetic Modifications To Combat Resistance To The Chemotherapeutic Agent Doxorubicin, Anna Lesley Kingham

Electronic Theses and Dissertations 2020 - Present

There has been substantial progress in cancer research that has markedly enhanced patient outcomes. However, chemotherapy resistance persists and often leads to multidrug resistance, rendering cancer cells unresponsive to multiple chemotherapy drugs, presenting a significant challenge in the effective treatment of the disease. Dysregulation in gene expression patterns caused by abnormalities in epigenetic mechanisms have been identified as contributing factors to the development and progression of cancer. Epigenetic research offers potential to discover drugs that target specific epigenetic modifications to regulate gene expression patterns in the context of chemotherapy resistance. I hypothesize that histone modifications on histone H3 and histone …

Image-Based Sexual Abuse In The Workplace: Examining The Effects And Responses Of Professional Women, Veronica Tyler Stewart Jan 2024

Image-Based Sexual Abuse In The Workplace: Examining The Effects And Responses Of Professional Women, Veronica Tyler Stewart

Electronic Theses and Dissertations 2020 - Present

Although the sensationalized term “Revenge Porn” is used by media outlets to spark conversation about consent and digital privacy, the nonconsensual distribution of intimate media, or “image-based sexual abuse” (IBSA), is the preferred term by scholars for its more accurate depiction of the variety of modes, methods, and damages. I argue that targeted women experience many of the same damages to their socioemotional, interpersonal, and professional well-being that targets of traditional, offline, sexual violence experience, and that the nature and affordances of digital technology often allow these harms to transcend the once isolated contexts in which offline cases of sexual …

Pandemic Perspectives: The Influence Of Disaster Experience And Technology Use Over The Life Course On Covid-19 Perception And Response Among Older Adults, Jordanne Kessel

Electronic Theses and Dissertations 2020 - Present

No group was more physically vulnerable to the COVID-19 pandemic than older adults. However, differing life histories and structural realities make for widely varying pandemic experiences. Using a life course approach, this study situates the COVID-19 pandemic and use of communication tools into context of older adults’ life experience with disasters and technology. Merging the scholarly fields of disaster sociology and aging studies, the purpose of this research is to find how life course experience and technology use impacted older adults’ perception of, and response to, COVID-19. Accordingly, I ask how does previous disaster experience and technology usage influence older …

Perceived Network Responsiveness As A Moderator Of The Effects Of Social Media Use On Wellbeing, Angalee Wilson

Electronic Theses and Dissertations 2020 - Present

Social media’s role in daily life is on the rise, but as social media use increases, so do questions about its potential benefits or harms. This set of studies examined the relationship between social media use, wellbeing, and perceived network responsiveness, a measure of an individual’s perception about the degree to which their online social networks are caring, validating, and understanding. Two pre-registered correlational studies which employed surveys of undergraduate students (Study 1a: N = 218, Study 1b: N = 179) found that perceived network responsiveness was positively correlated with life satisfaction and negatively correlated with loneliness. A pre-registered experimental …

Political Reforms And Women’S Rights In The Kingdom Of Saudi Arabia, Sama Khalil Abuhasabo

The Arabian Peninsula was under the influence of the Ottoman Empire from 1517 until its collapse in 1918. During this time, three attempts were made to establish a Saudi state, the last of which began in 1902 and ended with the unification of the third Saudi State in 1932. During this period, three Saudi States were formed. The first Saudi State was established in 1745 when the al-Diriyah Agreement was introduced. Real Earnings Management Effect On Customer Service And Service Quality, Brett Powers

Electronic Theses and Dissertations 2020 - Present

I investigate the link between real earnings management and customer satisfaction. Following the passing of the Sarbanes-Oxley Act, which requires public companies to ensure the accuracy of their financial reporting, the use of non-accrual based methods to achieve financial earnings targets has become commonplace. Nonaccrual-based methods consist of decisions that curtail firm activities, which increase net income; however, these methods come at the cost of future operations. These opportunistic behaviors in the service, retail, and hospitality industries impact customers and a firm's long-term value. I hypothesize that the outcome will be lower customer satisfaction and service quality. I use firms’ …

Regional Social Capital And Backer Sentiment For Successful Crowdfunding Campaigns, Joseph Grimes

Electronic Theses and Dissertations 2020 - Present

Local social capital, defined as the level of community interaction and social participation of a region, has been theorized to positively affect economic outcomes and discourage opportunistic behaviors in various settings. I examine whether local social capital is related to positive outcomes for entrepreneurs and their financial backers in the settings of reward crowdfunding and small business lending. In my first study, I look at how local social capital influences the creators of successful reward crowdfunding campaigns. These creators, in turn, may influence the sentiment of their investors, or backers, towards their projects through missed delivery deadlines and poor communication. …

Reusing And Recording: The Past And Future Survival Of The Jupiter Inlet Light Station Radio Beacon Building, Hailie Crowder

Electronic Theses and Dissertations 2020 - Present

For much of the 20th century, mariners in the United States were able to utilize the radio beacon system to aid in navigation; however, in spite of its importance in U.S. nautical history, there has been very little historical or archaeological research published about the system. The Jupiter Inlet Light Station Radio Beacon Building, located at what is today known as the Jupiter Inlet Lighthouse Outstanding Natural Area (JILONA), was part of this coastal network of radio beacons. This thesis involves the methodologies of historical research and terrestrial laser scanning and serves several purposes: to provide JILONA with information about …
