Policy declarations from the G7 and other high-level meetings call for increased incentives for antimicrobial research and development (R&D). Governments fund push incentives like CARB-X and GARDP, and G7 countries are now designing pull incentives—financial rewards given to manufacturers post-market authorization that are intended to encourage the creation and introduction of novel antimicrobials. Germany has declared previously at the G7 that it has developed a pull incentive that will increase revenues from sales of important new antimicrobials, principally by exempting them from some aspects of health technology benefit assessments and reference pricing, which should result in higher prices. This policy …

Engineering and Technology Management Faculty Publications and Presentations
Translational science aims to bridge the gap between scientific discoveries and practical application yet faces challenges in achieving high success rates. This study examines the effective engagement of translational scientists, who play a key role in the translation process. It uses Fuzzy Cognitive Maps (FCMs) to analyze the scientists' mental models and identify common concepts related to success factors, such as ability, resources, opportunity, and entrepreneurial competencies. It then compares the mental models of the scientists and finds that mental models exhibit similarities, yet variations exist such as density and connectedness. Scenario analysis with FCM is then used to identify …

Ambulatory and Primary Care Articles
Herpes simplex virus (HSV) and varicella-zoster virus (VZV) are common viruses that are present in the general population. However, it is uncommon for both viruses to coincide at the same time and location. These viruses infect the nervous system to establish latency and have been associated with neurological disorders. We discuss a case of co-occurring VZV reactivation and recurrent HSV infection with subsequent VZV encephalitis following an insult to the neurologic system.

Graduate Student Dissertations, Theses, Capstones, and Portfolios
Malaria is a significant public health problem which mainly affects malaria endemic regions, such as parts of Africa, South America, and Western Asia. However, all hot and humid environments are susceptible to spread leading to a global scale. Malaria affects an estimated 200 million people each year, with about 600,000 cases resulting in death. Malaria is passed through the bites of infected female anopheles mosquitos, with one of the six Plasmodium genus parasites that are malarial causing. Once the parasite enters the blood stream it infects erythrocytes which can then spread throughout the entire body. Disease is often characterized by …

Antifungal echinocandins inhibit the biosynthesis of β−1,3-glucan, a major and essential polysaccharide component of the fungal cell wall. However, the efficacy of echinocandins against the pathogen Aspergillus fumigatus is limited. Here, we use solid-state nuclear magnetic resonance (ssNMR) and other techniques to show that echinocandins induce dynamic changes in the assembly of mobile and rigid polymers within the A. fumigatus cell wall. The reduction of β−1,3-glucan induced by echinocandins is accompanied by a concurrent increase in levels of chitin, chitosan, and highly polymorphic α−1,3-glucans, whose physical association with chitin maintains cell wall integrity and modulates water permeability. The rearrangement of …

Prion diseases result from the misfolding of the physiological prion protein (PrPC) to a pathogenic conformation (PrPSc). Compelling evidence indicates that prevention and/or reduction of PrPSc replication are promising therapeutic strategies against prion diseases. However, the existence of different PrPSc conformations (or strains) associated with disease represents a major problem when identifying anti-prion compounds. Efforts to identify strain-specific anti-prion molecules are limited by the lack of biologically relevant high-throughput screening platforms to interrogate compound libraries. Here, we describe adaptations to the protein misfolding cyclic amplification (PMCA) technology (able to faithfully replicate PrPSc strains) that increase its throughput to facilitate the …

Adaptive Immune Profiling Of Sin Nombre Hantavirus And Severe Acute Respiratory Syndrome Coronavirus 2 Infection, Tonilynn M. Baranowski
Biomedical Sciences ETDs
Hantaviruses are causative agents of potentially fatal respiratory illness and/or organ failure in humans worldwide. In the United States, the predominant hantavirus species, Sin Nombre hantavirus (SNV), causes a severe disease pathology, called hantavirus cardiopulmonary syndrome (HCPS). Infection with SNV and subsequent clinical manifestations of HCPS lends itself to case fatality rates approaching 40%, even under supportive care. This is partially due to the lack of FDA-approved vaccines and antivirals, in addition to a deficit in our knowledge regarding the human immune response to SNV. Further investigation of the adaptive immune response may reveal key targets to mediate disease outcome. …

Pins aims to portray the societal landscape observed and experienced by me as an international student, highlighting sociological concerns like hatred and hate crimes instead of focusing on surface-level metrics such as the risk and fatality rates of COVID-19. Through this work, I want to remind the audience of the multifaceted nature of society and the emotions evoked by COVID-19, prompting reflection on the role of art during times of crisis. While transforming my original medium of metal objects into an installation art format, I explored concepts such as spacing, installation, cognition, narrative, and multiple senses. UNLABELLED: Acute encephalitis syndrome (AES) causes significant morbidity and mortality worldwide. In Nepal, Japanese encephalitis virus (JEV) accounts for ~5-20% of AES cases, but ~75% of AES cases are of unknown etiology. We identified a gemykibivirus in CSF collected in 2020 from an 8-year-old male patient with AES using metagenomic next-generation sequencing. Gemykibiviruses are single stranded, circular DNA viruses in the family
IMPORTANCE: Viral encephalitis is a devastating disease, but unfortunately, worldwide, the causative virus in many cases is unknown. Therefore, it is important to identify viruses that could be responsible for cases of human encephalitis. Here, using metagenomic sequencing …
